Claims (18)
1. Bindungstesteinrichtung mit einem Chromatograph!sehen Streifen zum Feststellen
des Vorliegens oder der Menge eines in einer Patientenprobe vorhandenen
Analyten, von dem Typus, welcher zur Anzeige eines positiven Testergebnisses für den Analyten ein visuell feststellbares Pluszeichen (+) oder
ein visuell feststellbares Minuszeichen (-) zur Anzeige eines negativen Testergebnisses für den Analyten entwickelt, welche Einrichtung umfaßt:
- ein aus einem chromatographischen Streifen bestehendes Substrat, welches eine solche Länge und eine solche schmale Breite aufweist, daß es befähigt
ist, Flüssigkeiten in einer im allgemeinen zur Länge des Streifens parallelen Fließrichtung der Flüssigkeit zu fördern, wobei dieser Streifen
eine Berührungsregion für die Probe, auf welcher eine flüssige Patientenprobe
und andere Testreagenzien mit dem Streifen in Berührung gebracht werden können, und eine Testregion umfaßt, welche auf diesem
Streifen auf einer stromabwärtigen Stelle in bezug auf die Berührungs-1^
region für die Probe angeordnet ist, wobei diese Testregion einen Patiententestbalken
umfaßt, der durch ein erstes Aufnahmereagens gebildet wird, das spezifisch für den zu testenden Analyten ist und auf der Testregion
des Streifens in einer im allgemeinen rechteckigen Konfiguration ijrcmobilisiert
ist, sowie einen Verfahrenskontrollbalken umfaßt, der durch ein zweites Aufnahmereagens gebildet wird, das spezifisch für einen Testmarker
ist, und der auf der Testregion des Streifens in einer im allgemeinen
rechteckigen Konfiguration immobilisiert ist, wobei der Patiententestbalken
und der Verfahrenskontrollbalken einander in der Testregion auf eine im allgemeinen senkrechte Art und Weise schneiden; und
_ wobei die Verbesserung betreffend die Abbildung des Testergebnisses umfaßt:
daß der Patiententestbaiken und der V.erfahrenskontrol!balken auf der
daß der Patiententestbaiken und der V.erfahrenskontrol!balken auf der
Testregion des Streifens, jeweils unter einem Winkel in bezug auf die
Fließrichtung der Flüssigkeit des Streifens, immobilisiert sind, um den
von einer zugesetzten, wandernden Flüssigkeit sowohl quer über den Pa-
5; tiententestbalken als auch quer über den Verfahrenskontrollbalken zurückgelegten
Weg auf ein Mindestmaß herabzusetzen.
2. Einrichtung nach Anspruch 1, worin die Winkelausrichtungen des Patiententestbalkens
und des Verfahrenskontrollbalkens in bezug auf die Fließrichtung der Flüssigkeit durch Komplementärwinkel definiert sind, welche
zwischen etwa X0 und etwa 60° variieren.
3. Einrichtung nach Anspruch 1, worin die Winkelausrichtung des Patiententestbalkens
und des Verfahrenskontrollbalkens in bezug auf die Fließrichtung der Flüssigkeit je etwa 45° beträgt.
4. Einrichtung nach Anspruch 1, worin das aus einem chromatographischen Streifen
bestehende Substrat Nitrozellulose enthält.
5. Einrichtung nach Anspruch 1, worin das erste Aufnahmereagens einen Teil
eines spezifischen Bindungspaares für den Analyten darstellt.
6. Einrichtung nach Anspruch 5, worin das erste Aufnahmereagens aus der ein
Antigen oder einen Antikörper umfassenden Gruppe ausgewählt ist.
7. Einrichtung nach Anspruch 1, worin das zweite Aufnahmereagens einen Teil
eines spezifischen Bindungspaares für den Testmarker darstellt.
8. Einrichtung nach Anspruch 7, worin das zweite Aufnahmereagens aus der ein
Antigen oder einen Antikörper umfassenden Gruppe ausgewählt ist.
9. Einrichtung nach Anspruch 1, welche weiterhin ein Gehäuse zum Umgeben und
Festhalten des Streifens umfaßt, wobei dieses Gehäuse eine Probeeintrittsöffnung,
welche neben der Probeneintrittsregion des Streifes angeordnet , ist, und ein Testergebnisfenster aufweist, welches neben der Testregion des
Streifens angeordnet ist.
10.Einrichtung nach Anspruch 1, welche weiterhin ein zugeordnetes Kissen umfaßt,
welches seinerseits ein poröses Kissensübstrat umfaßt, in welchem ein
Testmarker vorliegt, und welches Kissen neben dem Streifen, in der Probeneintrittsregion,
angeordnet ist.
11. Einrichtung nach Anspruch 10, worin das erste Aufnahmereagens einen für
den zu testenden Analyten spezifischen Antikörper umfaßt, wobei dieses zweite Aufnahmereagens inmobilisierten Analyten umfaßt; und worin der
Testmarker in dem zugeordneten Kissen einen für den zu testenden Analyten
spezifischen Antikörper umfaßt, welcher Antikörper an eine markierende Substanz konjugiert ist.
12. Einrichtung nach Anspruch 11, worin die markierende Substanz ein Selenkolloidmarker
ist.
13. Einrichtung nach Anspruch 10, worin das erste Aufnahmereagens einen für
den zu testenden Analyten spezifischen Antikörper umfaßt, und worin das zweite Aufnahmereagens einen für den Testmarker spezifischen Antikörper
umfaßt; und worin der Testmarker in dem zugeordneten Kissen einen für den zu testenden Analyten spezifischen Antikörper umfaßt, welcher Antikörper
an eine markierende Substanz konjugiert ist.
14. Einrichtung nach Anspruch 13, worin die markierende Substanz ein Selenkolloidmarker
ist.
15. Einrichtung nach Anspruch 1, welche weiterhin eine das Ende des Tests anzeigende
Region auf dem Streifen, an einer in bezug auf die Testregion stromabwärtigen Stelle, umfaßt, welche das Ende des Tests anzeigende
Region von einem Indikatorreagens gebildet wird, welches befähigt ist, ein visuell feststellbares Signal bei der Berührung mit irgendwelchen Testflüssigkeiten
zu bilden, um anzuzeigen, daß Flüssigkeiten zu dem Ende der Testregion geflossen sind und daß das in der Testregion angezeigte
Testergebnis ein endgültiges Testergebnis ist.
16. Einrichtung nach Anspruch 11, worin das zweite Aufnahmereagens einen
immobilisierten Antikörper-Analyt-Komplex umfaßt.
17. Einrichtung nach Anspruch 13, worin der Testmarker einen monoklonalen
Antikörper aus einer ersten Spezies umfaßt, welcher Antikörper spezifisch für den zu testenden Analyten ist, und worin das zweite Aufnahmereagens
einen Antikörper aus einer zweiten Spezies umfaßt, welcher Antikörper spezifisch für den monoklonalen Antikörper aus der ersten Spezies ist.
18. Einrichtung nach Anspruch 17, worin der Testmarker einen monoklonalen
Mäuse-Antikörper umfaßt, welcher spezifisch für den Analyten ist, und worin das zweite Aufnahmereagens einen spezifischen Anti-Maus-IgG-Antikörper
umfaßt.
