Description
Die Erfindung betrifft den in den Fatentansprüchen gekennzeichneten
Gegenstand.
Der Ausdruck niederer Alkylrest bedeutet unverzweigte oder verzweigte Reste mit 1 bis 7 Kohlenstoffatomen. Spezielle Beispiele
sind Methyl-, Äthyl-, Propyl- und Isopropylgruppe. Die
niederen Alkoxy- und niederen Alkylthioreste enthalten ebenfalls 1 bis 7 Kohlenstoffatome. Spezielle Beispiele sind die
Methoxy-, Äthoxy-, Propoxy-, Butoxy-, tert.-Butoxy-, Methylthio-,
Äthylthio-, Propylthio-, Butylthio- und Isobutylthiogruppe. Bevorzugt sind Alkyl-, Alkoxy- und Alkylthioreste mit
1 bis 4, insbesondere 1 oder 2 Kohlenstoffatomen.
Als Halogenatome kommen Fluor-, Chlor-, Brom- und Jodatome
in Frage. Chlor- und Bromatome sind in der angegebenen Reihenfolge bevorzugt. Vorzugsweise, jedoch nicht notwendigerweise,
sind sämtliche Halogenatome in einer Verbindung gleich.
Bevorzugt sind Verbindungen der allgemeinen Formel I, in der R einen niederen Alkylrest, insbesondere die Methyl- oder
Äthylgruppe, und R , R^ und R Wasserstoff atome bedeuten.
Als reaktionsfähige Funktion Y in den Verbindungen der allgemeinen
Formel III kommen beispielsweise Halogenatome, die Mesyl·- und Tosy!gruppe in Frage.
Ein anderes Verfahren, bei dem die Herstellung des Esters der
allgemeinen Formel III vermieden wird, ist die Umsetzung des Alkohols der allgemeinen Formel IV

mit Thionyl-bis-imidazol oder Carbonyl-bis-imidazol der Formel

in der A die -SO- oder -CO-Gruppe darstellt.
Die 11-Chlor-Derivate (Y= Cl) der allgemeinen Formel III und
die 11-Hydroxy-Derivate der allgemeinen Formel IV können
nach den in der US-PS 4 111 940, Beispiel 2 und 44a, beschriebenen
Verfahren hergestellt werden.
Die Verbindungen der allgemeinen Formel I bilden Salze. Zu den Salzen gehören die Salze mit Säuren, insbesondere die
pharmakologiseh verträglichen Säuren. Die Basen der allgemeinen
Formel I bilden durch Umsetzung mit 1 oder 2 Äquivalenten einer anorganischen oder organischen Säure die entsprechenden
Salze. Beispiele sind die Hydrohalogen!de, insbesondere die
Hydrochloride und Hydrobromide, Sulfate, Nitrate, Borate, Phosphate, Oxalate, Tartrate, Maleate, Citrate, Acetate, Ascorbate,
Succinate, Benzolsulfonate, Methansulfonate, Cyclone xansulfamat e und ToluoIsulfönate. Häufig dienen die Salze
auch zum Isolieren oder Reinigen der Produkte. Beispielsweise vjird ein Salz in einem entsprechenden Medium hergestellt
und ausgefällt, in welchem das Salz unlöslich ist. Nach der Abtrennung des Salzes wird es mit einer Base, wie Barium-

■j hydroxid oder Nat-riumhydroxid, wieder in die freie Base der .
allgemeinen Formel I überführt. Andere Salze können sodann aus den gereinigten freien Basen durch Umsetzung mit einem
oder mehreren Äquivalenten einer Säure des gewünschten Anions
5 hergestellt werden.
Die Verbindungen der allgemeinen Formel I und ihre Salze sind wertvolle Antibiotika zur Bekämpfung pathogener Pilze, Protozoen
und Bakterien. Sie können zur Bekämpfung insbesondere von Candida albicans, Trichomonas vaginalis und Trichophyton
mentagrophytes eingesetzt werden. Zu diesem Zweck können die Verbindungen oral, beispielsweise in Form von Tabletten,
Kapseln oder Elixieren, lokal, z.B. intravaginal, in Form einer Lotion oder einer Creme, verabfolgt werden.
Die Beispiele erläutern die Erfindung.
Beispiel 1
1-Äthyl-l, 5,6,11-tetrahydro-Il-(IH-imidazol-l-yl) -benzo Z"5>67-cycloheptaZ3.,2-bJpyrazaloZ?i,3-e7pyridin-hydrochlorid
(1 : 1)
Eine Lösung von 2,3 g (-0,008 Mol) ll-Chlor-l-äthyl-1,5,6,11-tetrahydrobenzo/5,6.7cycloheptaZi,
27pyrazoloZ^, 3-e-7pyrxdin in
50 ml wasserfreiem Toluol wird mit 1,7 g (0,025 Mol) Imidazol versetzt und 3 1/2 Stunden unter Rückfluß erhitzt. Danach
wird das Toluol unter vermindertem Druck abdestilliert, der Rückstand mit Wasser digeriert und mit Diäthyläther extrahiert.
Der Ätherextrakt wird getrocknet und 3 bis ^ Tage stehen gelassen. Hierbei scheidet sich nicht umgesetzte Ausgangsverbindung
und ein Nebenprodukt ab. Die erhaltene klare Ätherlösung wird unter Rühren mit einer Lösung von Chlorwasserstoff
in Äther versetzt. Das ausgefällte Hydrochlorid wird abfiltriert. Ausbeute I93'g vom P. 106 bis 109°C. Nach
ümkristallisation aus Äthylacetat wird die Titelverbindung
35 vom F. 108 bis 1100C erhalten.
