3. Verfahren nach Anspruch 1, dadurch gekennzeichnet, daß man als nicht-ionogenes grenzflächenaktives

len(23)-Iauryläther, Polyoxyäthylen(20)-cetyläther oder Alkylphenoxypolyäthoxy(30)-äthanol verwendet
4. Verwendung des Hepatitis B-Kern-Antigens erhallen nach Anspruch 1 zur Herstellung von diagnostischen
Präparaten und Vaccinen.
Aus Lancet (1971), S. 1225—1227 ist es bekannt, bei der Gewinnung von HBC Ag ein Detergens zu w-senden, |
z. B. das Handelsprodukt Tween 80, ein nicht-ionogenes grenzflächenaktives Mittel mit ca. 20 Oxyilithyleneinheiten.
Die Verwendung eines Mercaptan-Reduktionsmittels ist in dieser Druckschrift nicht erwähnt.
In. J. Immunology. Bd. 115 (1975), S. 834—838 und Bd. 114 (1975), S. 1792—1798 ist die Gewinnung von HB0Ag
unter Verwendung des Handelsprodukts Nonidet NP-40 (Detergens: mit 9 Oxyäthyleneinheiten) beschrieben,
wobei gemäß der erstgenannten Literaturstelle eine vorherige und gemäß der letztgenannten Uteratursteile
eine gleichzeitige Behandlung mit Mercaptoäthanol durchgeführt wird. Die Verwendung von Detergentien mit
etwa 15 bis etwa 35 Oxyäthyleneinheiten wird dort nicht erwähnt.
Aufgabe der Erfindung ist es, ein Verfahren zur Herstellung von HBcAg zur Verfügung zu stellen, das im
Vergleich zu herkömmlichen Verfahren in höheren Ausbeuten abläuft.

Als Ausgangsmaterial zur Herstellung des erfindungsgemäßen gereinigten Hepatitis B-Kern-Antigens (Hepatitis
B-Core-Antigen; HBcAg) wird Plasma von in bezug auf HBcAg positiven Spendern verwendet Das Plasma |
3y wird auf übliche Weise erhalten, beispielsweise durch Plasmapherese. Die Konzentration an HBcAg kann in |
bekannter Weise gemessen werden, beispielsweise durch umgekehrt« passive Hämagglutination oder Komplementfixierung.

Ration entfernt. Die Dane-Teilchen im erhaltenen Plasma werden durch isopyknische Bandenbildung isoliert. f
Die an Dane-Teilchen reiche Fraktion wird zur Entfernung von Dane-Kern-Antikörpern behandelt, vorzugswei- |
se durch Pelletisieren. Anschließend wird das Oberflächen-Antigen iwitfernt und das Kern-Antig«n freigesetzt.
Die Entfernung des Oberflächen-Antigens wird durchgeführt, indem die Dane-Teilchen mit einem nicht-ionogenen
grenzflächenaktiven Mittel mit etwa 15 bis etwa 35 und vorzugsweise etwa 18 bis etwa 33 Oxyiithyleneinheiten
im Molekül in Gegenwart eines Mercaptan-Reduktionsmittels, wie Mercaptoäthanol, Dithiothreit, Dithioerythrit
und Dithiooctansäure, in Kontakt gebracht werden. Beispiele für entsprechend nicht-ionogene grenzflä-
chenaktive Mittel sind oxyäthylierle Alkylphenole. Polyoxyäthylensorbitanfettsäureester. Polyoxyäthylensäuren.
Polyoxyäthylenalkohole. Polyoxyäthylenöle und Polyoxyäthylenoxypropylenfettsäuren. Nachstehend sind
spezielle Beispiele für derartige grenzflächenaktive Mittel angegebem:

Bei der isopyknischcn Bandenbildung wird das teilweise gereinigte Konzentrat mit einem flüssigen Medium in
Kontakt gebracht, das einen Dichtegradienten aufweist, der die Diclhte des speziellen zu isolierenden Antigens
einschließt. Das flüssige Medium wird anschließend der Ultrazentrgifugation unterworfen, um dnc Glcichgewichtsvcrteilung
der Serumbestandteile innerhalb des Dichtegradienten je nach dcii einzelnen Dichten zu
erreichen. Die Fraktionen des Mediums werden nacheinander vcrdiiangt. Die Fraktionen mit einem Gehalt an
dem gewünschten Antigen, d.h. die Fraktionen mit einer Dichte Ajon etwa 1,26 bis etwa I^Og/cm3 werden

abgetrennt. Die Konzentration der den Gradienten bildenden Lösüligen werden so gewählt, daß ein Dichtebereich
von etwa 1,0 bis etwa 1,41 g/cm3 umfaßt wird. Das flüssige Medium kann in Form eines linearen Gradienten
oder eines Stufengradienten eingesetzt werden. Vorzugsweise wird Wegen der höheren Fraktionierungskapazitat
ein Stufengradient verwendet. I
Als flüssige Medien bei der isopyknischen Bandenbildungsstufe können beliebige Dichtegradienten in den
entsprechenden Konzentrationen verwendet werden, beispielsweise solche auf der Basis von Saccharose, Kaliumbromid,
Cäsiumchlorid, Kal'iumtartrat oder Natriumbromid. Vorzugsweise wird Natriumbromid verwendet.
(Die Stufen A und B erläutern die Herstellung des Ausgangsmateriais)
A. Herstellung von Dane-Teilchen (HBV)
A. Herstellung von Dane-Teilchen (HBV)
Der Rotor einer Zentrifuge (Electronucleonics K) wird mit 8400 ml Phosphatpuffer gefüllt Nach einem
Rotorlauf bis 10 000 U/min zur Entgasung des Systems wird der nachstehend angegebene Stufengradient in den
Boden des stationären Rotors gepumpt:
1. 2400 ml 10% NaBr.p = 1,08
2. 1000 ml 20% NaBr, p= 1,17
3. 1500 ml 30% NaBr, ρ = 1,28
4. 3500 ml40% NaBr.p = 1,41
Anschließend werden 1750 ml Plasma mit einem Gehalt an HB5Ag in den Kopf den stationären Rotors
gepumpt, wodurch 1750 ml des 40prozentigen Natriumbromids am Rotorboden verdrängt werden. Sodann wird
der Rotor auf 30 000 U/min beschleunigt und 4 Stunden bei dieser Geschwindigkeit betrieben. Nach dem
Anhalten des Rotors werden 1750 ml einer 40prozentigen Natriumbromidlösung in den Rotorboden gepumpt,
wodurch das Plasma am Rotorkopf austritt. Hierauf werden weitere 1750 ml frisches Plasma mit einem Gehalt
an HB5Ag in den Rotorkopf gepumpt, wodurch ein entsprechendes Volumen an 40prozentiger Natriumbromidlösung
am Rotorboden verdrängt wird. Sodann wird der Rotor 18 Stunden bei 30 000 U/min betrieben. Nach
dem Anhalten des Rotors werden 1000 ml eines an Dane-Teilchen reichen Materiafs im Dichtebereich von 1,26
bis 130 g/cm3 gewonnen.
Die Dane-Teilchen (HBV) werden auf die nachstehend beschriebene Weise von der NaBr-Zonalfraktion
abgetrennt. Die Zonalfraktion (1000 ml) wird mit phosphatgepufferter Kochsaiziösung auf 3000 ml verdünnt
Dieses Material wird sodann in 12 Rotor-Kunststoffgefäße vom Typ 19 (etwa 250 ml/Gefäß) gebracht. Das
Material wird sodann unter Verwe-'dung eines Rotors vom Typ 19 (Beckman) zentrifugiert. Der Rotor wird 24
Stunden bei 17 000 U/min (45 000 g) betrieben, um die Dane-Teilchen zu pelletisieren. Anschließend wird der
Rotor angehalten und der Übersta? ί von den einzelnen Gefäßen dekantiert. Die Pellets aus sämtlichen 12
Gefäßen werden in einem gesamten Volumen von 5 bis 7 ml Tris-Kochsalz-Puffer aufgenommen und bei —700C
aufbewahrt. Bei diesem Material handelt es sich um das Dane-Teilchen-Konzentrat.
B. Reinigung der Dane-Teilchen
In einem SW 65-Rotor mit Zentrifugenröhrchen aus Cellulosenitrat der Abmessungen 1,27 χ 5,08 cm (1/2 χ 2")
werden 4 ml 20% Saccharose—1% Rinderserumalbumin (BSA) in Tris-Puffer vom pH-Wert 7,6 mit 1 ml
β konzentrierten Dane-Teilchen gemäß Abschnitt A übersshichtet. Die Teilchen werden 4 Stunden bei 35 000 U/
jj min zentrifugiert. Nach dem Zentrifugieren wird die überstehende Flüssigkeit dekantiert. Die erhaltenen Pellets
werden unter Verwendung eines mit Puffer angefeuchteten Wattetupfers vorsichtig in 0,5 ml Tris-Puffer mit 1 %
Rinderserumalbumin resuspendiert. Der Baumwolltupfer wird anschließend mit 0,5 ml Puffer gespült. Das
endgültige Volumen des Dane-Teilchen-Materials beträgt 1 ml. Die Dane-Teilchen werden bei — 70°C aufbewahrt.
C. Herstellung von HB0As (Kern-Antigen)
1 ml des Materials gemäß Abschnitt B wird zu 1 ml einer lprozentigen (Vol/Vol.)-Lösung von 2-Mercaptoäthanol
in entionisiertem Wasser und 1 ml einer lprozentigen (Vol/Vol.)-Lösung von Polyoxyäthylen(20)-sorbitanmonooleat
in entionisiertem Wasser gegeben. Das erhaltene Gemisch wird mäßig gerührt und in ein Wasserbad
von 37CC gestellt. Nach 1 Stunde wird das Gemisch mit TMN-1% BSA (eine Lösung mit einem Gehalt an
0,08 m Tris, 0,008 m MgCIz und 0,14 m NaCl und 1% BSA) verdünnt, wobei das Verdünnungsmittel in einer
vorher so berechneten Menge verwendet wird, daß pro ml 32 IAHA-Einheiten enthalten sind. Die Lösung wird
sodann auf 2 ml fassende Serumröhrchen mit Schraubenverschluß (0.5 ml/Röhrchen) verteilt und in einem mit
flüssigem Stickstoff arbeitenden Gefrierschrank aufbewahrt.
In 12 Gläsampullenmit einem Fassungsvermögen von 6 ml werden jeweils 1 ml gemäß Beispiel 1 hergestellte
gereinigte Dane-Teilchen, 1 ml einer lprozentigen (VoL/Vol.)-Lösung von 2-Mercaptoäthanol in entionisiertem
Wasser und 1 ml einer lprozentigen (Vol/Vol.)-Lösung der nachstehend angegebenen Produkte in entionisiertem
Wasser gegeben. Die Gemische werden mäßig bewegt und in ein Wasserbad von 37°C gestellt. Nach 1
Stunde werden die erhaltenen Gemische nach dem Immunhaftungshämagglutinationstest auf ihren HBcAg-Gehalt
analysiert.

Die vorstehenden Ergebnisse zeigen, daß nicht-ionogene grenzflächenaktive Mittel mit einem Gehalt an 9
oder 40 Oxyäthyleneinheiten solchen mit einem Gehalt an 20 bis 30 Oxyäthyleneinheiten deutlich unterlegen
sind, während grenzflächenaktive Mittel ohne Oxyäthyleneinheiten fast vollkommen unwirksam sind.
Eine erste 1-ml-Probe von gereinigten Dane-Teilchen aus der in Beispiel 2 verwendeten Charge wird mit 1 ml
Polyoxyäthylen(20)-sorbitanmonooleat und 1 ml 2-Mercaptoäthanol versetzt. Eine zweite 1-ml-Probe w'rd auf
ähnliche Weise behandelt, wobei aber anstelle von 2-Mercaptoäthanol 1 ml entionisiertes Wasser verwendet
wird. Die Proben werden vermischt, 1 Stunde bei 37°C inkubiei: und schließlich mit dem Immunhaftungshämagglutinationstest
auf ihren HBcAg-Gehalt untersucht. Die zweite Probe ohne 2-Mercaptoäthanol enthält im
Vergleich zur ersten Probe weniger ais die Hälfte an HBcAg.
In diesem Beispiel und im folgenden Beispiel ist von den HBcAg-Typen Ad und Ay die Rede. Es handelt sich
hierbei um Bezeichnungen für heterogene Untertypen des Hepatiiis B-Virus. Die Antigendeterminante A ist
allen Untertypen gemeinsam, während d und y für den jeweiligen Untertyp spezifische Determinanten darstel-
len; vgl. Melnick et al. Southern Medical Journal, Bd. 69 (1976), S.468—475, insbesondere S.471).
Gemäß Beispiel 1 hergestellte HB0Ag wird in folgender Weise an Alaun adsorbiert. 10 ml HBcAg (Typ Ad) mit
einem Gehalt an 16 bis 32 IA-Einheiten/ml werden mit 0,85 ml einer lOprozentigen Alaunlösung
(KA1(SO4)2 · 12 H2O) vermischt. Unter Rühren wird 0,1 η NaOH langsam zugegeben, um den pH-Wert auf 6,8
einzustellen. Sodann wird eine weitere Stunde bei Raumtemperatur gerührt. Anschließend wird die Lösung 10
Minute", bei 1500 g zentrifugiert. Der Oberstand wird dekantiert und das Pellet in Kochsalzlösung zum ursprünglichen
Volumen (10 ml) resuspendiert. Die Lösung w:-"d 5 bis 10 Minuten vor der Verwendung als Antigen
vermischt.
Man verfähr t wie in Beispiel 4 mit der Abänderung, daß 10 ml HBcAg (Typ Ay) verwendet werden.
Die gemäß den Beispielen 4 und 5 hergestellten Alaun-Antigene werden zur Herstellung von Seren mit einem
hohen HBcAb-Titer verwendet. Zur Bildung von HBcAb-Antisemit] werden Meerschweinchen verwendet, die in
2 Gruppen eingeteilt werden. Die erste Gruppe erhält in monatlichen Abständen 3 intramuskuläre Injektione.i
von jeweils 0,5 ml des Produkts von Beispiel 4. Die zweite Gruppe wird in entsprechender Weise mit dem
Produkt von Beispiel 5 behandelt. Es werden jeweils Hepatitis B-Antikörper-Seren mit einem hohen Titer
gebildet.

Das gemäß Beispiel 1 erhaltene HBcAg wird durch 2 1 /2stündige Zentrifugation in einem 20 bis 60pi ozentigen
Saccharosegradienten bei 200 000 g gereinigt. Das HBcAg wird sodann zur Bestimmung des Proteingehalts
untersucht
Das HBcAg wird zur Verwendung beim ELISA-Test mit 0,1 m Carbonatpisffer vom pH-Wert 9,7 auf 1 μg/ml
verdünnt. Als feste Phase wird beim Test eine Polystyrolplatte (96-Bohrloch (»well«) Cooke-Mikrotiter, U-Boden)
verwendet. AIf Enzymkonjugat wird mit alkalischer Phosphatase konjugiertes Ziegen-Antihumanimmunoglobulin
verwendet; vgl. Engvall et al, a. a. O. Die Gebrauchskonzentration wird durch Titration bestimmt. Als

Enzymsubstrat wird 0,01% p-Nitrophenylpho;sphat in 0,1 m Carbonatpuffer vom pH-Wert 9,8 mit einem Gehalt
an 0,001 m MgCh verwendet. Das Testverfahren ist von E. Nassau et al.. Tubercle, Bd. 57 (1976), S. 67 bis 70, J
beschrieben. Dieses Verfahren wird zum Nachweis von H BcAb in Plasma- und Serumproben verwendet. ; f
Beispiel 8 5
1 ml des Materials von Abschnitt B. von Beispiel 1 wird zu 1 ml einer lprozentigen (VoL/Vol.)-Lösung von
2-Mercaptoäthanol in entionisiertem Wasser und 1 ml einer lprozentigen (Vol7Vol.)-Lösung von Polyoxyäthylen(20)-sorbitanmonooleat in entionisiertem Wasser gegeben. Das erhaltene Gemisch wird mäßig bewegt und in
tin Wasserbad von 37°C gestellt. Nach 1 Stunde wird das Gemisch mit SPGA (Standard-Zellkulturmedium mit io einem Gehalt an Saccharose, Phosphat, Glutamin und Albumin; vgl. Bovarnick et al., J. Bacteriology, Bd. 59
(1950), S. 509—521, insbesondere S. 520) verdünnt, wobei die Meng« des Verdünnungsmitfels so berechnet ist,
daß sich 32 IAHA-Einheiten pro ml ergeben. Die Lösung wird sodann in 32 ml fassende Serumröhrchen mit
Schraubverschluß (0,5 ml/Röhrchen) verteilt und in einem mit flüssigen Stickstoff betriebenen Gefrierschrank
aufbewahrt. 15
2-Mercaptoäthanol in entionisiertem Wasser und 1 ml einer lprozentigen (Vol7Vol.)-Lösung von Polyoxyäthylen(20)-sorbitanmonooleat in entionisiertem Wasser gegeben. Das erhaltene Gemisch wird mäßig bewegt und in
tin Wasserbad von 37°C gestellt. Nach 1 Stunde wird das Gemisch mit SPGA (Standard-Zellkulturmedium mit io einem Gehalt an Saccharose, Phosphat, Glutamin und Albumin; vgl. Bovarnick et al., J. Bacteriology, Bd. 59
(1950), S. 509—521, insbesondere S. 520) verdünnt, wobei die Meng« des Verdünnungsmitfels so berechnet ist,
daß sich 32 IAHA-Einheiten pro ml ergeben. Die Lösung wird sodann in 32 ml fassende Serumröhrchen mit
Schraubverschluß (0,5 ml/Röhrchen) verteilt und in einem mit flüssigen Stickstoff betriebenen Gefrierschrank
aufbewahrt. 15
Claims (2)
1. Verfahren zur Herstellung von Hepatitis B-Kern-Antigen, dadurch gekennzeichnet, daß man
Dane-Teilchen mit einem nicht-ionogenen grenzflächenaktiven Mittel mit etwa 15 bis etwa 35· Oxyäthyien-
einheifen in Gegenwart eines Mercaptan-Reduktionsmittels behandelt. ^
2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, daß man als Reduktionsmittel Mercaptoäthanol |
verwendet.
