Die Erfindung betrifft neue 1-Azaxanthon-3-carbonsäuren
und deren Derivate, die ausgezeichnete pharmakologische Eigenschaften besitzen, und ein Verfahren zu
ihrer Herstellung. Insbesondere betrifft die Erfindung neue l-Azaxanthon-3-carbonsäuren der allgemeinen Formel

worin R·, Wasserstoff, Alkyl, Phenyl, Carboxyl, Hydroxyl,
Alkoxy oder eine gegebenenfalls durch ein Alkyl substituierte Aminogruppe, m O, 1 oder 2 und R^ Alkyl, Alkoxy,
Halogen, Nitro, Hydroxy, Carboxyl, Butadienylen (-CH=CH-CH=CH-) das mit jeweils angrenzenden C-Atomen einen
Benzolring bildet oder eine gegebenenfalls durch mindestens ein Alkyl substituierte Aminogruppe bedeuten, und deren
physiologisch verträgliche Salze, die ausgezeichnete

pharmakologische Wirkung wie z.B. antiallexgische und bxonchodilatorische Wirkung besitzen.
Die Verbindungen der Formel (i) können durch Hydrolyse
der Verbindung der Formel (II)
hergestellt werden, v;orin R1 , Wasserstoff, Alkyl, Phenyl,
Alkoxycaxbonyl, Hydroxyl, Alkoxy oder eine gegebenenfalls
durch ein Alkyl substituierte Aminogruppe bedeutet, m und Ii-, die vorstehend angeführte Bedeutung besitzen und R0 für

Cyan, Alkoxycarbonyl oder ^ein/ourcn wenigstens ein Alkyl
substituiertes Carboxamid steht.
Die in den vorstehenden Formeln angeführten Substituenten können in beliebigen Stellungen, d.h. in den
Stellungen 6, 7, 8 oder 9 des Azaxanthonringes substituiert sein.
Die in Formel (l) durch R-, oder R^ dargestellte Alkylgruppe
kann eine geradkettige, verzweigte oder cyclische Alkylgruppe mit 1-6 C-Atomen sein. Als typische Beispiele
für solche Alkylgruppen seien Methyl, Äthyl, Propyl, Isopropyl, Butyl, Isobutyl, sec.Butyl, tert.Butyl, Pentyl,
Cyclopentyl, Hexyl, Cyclohexyl usw. genannt. Unter diesen werden aus praktischen Gründen niedere Alkyle mit 1-3
C-Atomen bevorzugt.

Die durch R, und Pt, dargestellten Alkoxygruppen
können solche mit 1-4 C-Atomen im Alkylanteil sein, wie z.B. Methoxy, Athoxy, Propoxys Isopropoxy, Butoxy usw..
Die mit R, bezeichneten durch Monoalkyl substituierten Aminogruppen können solche mit 1-3 C-Atomen im Alkylanteil
wie z.B. Methylamino, Äthylamino, Propylamino oder Isopropylamino sein. Das mit R^ bezeichnete Halogen kann
Chlor, Brom, Jod oder Fluor sein.
Die durch Rp dargestellte, durch Alkyl substituierte
Aminogruppe umfaßt auch Mono- oder Dialkyl-substituierte
Gruppen, deren Alkylanteil 1-3 C-Atome aufweist, wie z.B. Methylamino, Äthylamino, Propylamino, Isopropylamino,
Dimethylamine, Diäthylamino oder Dipropylamino.
Die in der Formel (II) mit R1, bezeichnete Alkylgruppe
kann eine geradkettige, verzweigte oder cyclische Alkylgruppe mit 1-6 C-Atomen sein. Typische Beispiele für
solche Alkylgruppen sind Methyl, Äthyl, Propyl, Isopropyl, Butyl, Isobutyl, sec.Butyl, tert.Butyl, Pentyl, Cyclopentyl,
Hexyl, Cyclohexyl od.dgl. Unter diesen werden aus praktischen Gründen niedere Alkyle mit 1-3 C-Atomen bevorzugt.
Die durch R1, dargestellte Alkoxygruppe kann z.B. eine
solche mit 1-4 C-Atomen im Alkylanteil seinj wie z.B.
Methoxy, Äthoxy, Propoxy, Isopropoxy, Butoxy usw.
Für R1, in Frage kommende, durch Monoalkyl substituierte
Aminogruppe, kann eine solche mit 1-3 C-Atomen im Alkylanteil sein, z.B. Methylamino, Äthylamino, Propylamino
oder Isopropylamino. Die Alkoxycarbonylgruppen R', und R3

werden durch die Formel -COCR4 ausgedrückt, worin R. ein
geradkettiges Alkyl mit 1-6 C-Atomen wie z.B. Methyl, Äthyl, n-Propyl, η-Butyl, n-Hexyl od.dgl. bedeutet.
Zu den durch Alkyl substituierten Carboxamidgruppen, die mit dem Symbol FL· bezeichnet werden, gehören auch
Mono- oder Dialkyl substituierte Carboxamidgruppen, deren Alkylanteil 1-3 C-Atome aufweist. Als typische Beispiele
für diese Gruppen seien N-Methyl-carboxamid, N1H-Dimethylcarboxamid,
N-Äthylcarboxamid, Ν,Ν-Diäthylcarboxamid
und H-Propylcarboxamid genannt.
Die erfindungsgemäßen Verbindungen der Formel (I)
•können durch Hydrolysieren der Verbindungen der Formel (ll)
hergestellt werden. Die Hydrolyse kann unter sauren oder alkalischen Bedingungen durchgeführt werden, wobei saure
Bedingungen bevorzugt werden. Zu diesem Zweck kann eine Mineralsäure wie Salzsäure, Schwefelsäure, Perchlorsäure,
Phosphorsäure od.dgl. oder eine organische Säure wie Trifluoressigsäure,
Ameisensäure, Essigsäure od.dgl. eingesetzt werden. Im allgemeinen wird die Umsetzung bevorzugt
in Gegenwart von V/asser und in einem Gemisch aus einer organischen und einer Mineralsäure durchgeführt. Obgleich
die Temperatur, Zeitdauer und die anderen Reaktionsbedingungen nicht kritisch sind, wird die Umsetzung im allgemeinen
bei Temperaturen von 5o bis 15o°C innerhalb eines Zeitraumes von 1 Stunde bis zu zwei Tagen vorgenommen.
Die Verbindung der allgemeinen Formel (i), worin R^
für Carboxyl steht, kann in eine Verbindung, in welcher R, Wasserstoff bedeutet, übergeführt werden, indem man die
Verbindung, gegebenenfalls in Gegenwart eines Lösungsmittels, auf eine Temperatur erhitzt, die etwas über jener

Temperatur liegt, welche die Decarboxylierung einleitet.
Die Verbindungen der allgemeinen Formel (l) können in die entsprechenden organischen Aminsalze, Alkalimetallsalze
oder Ammoniumsalze durch Umsetzung der Verbindung (I) nach einem an sich bekannten Verfahren mit einem organischen
Amin wie z.B. Äthanolamin, Diethanolamin, dl-Methylphedrin, l-(3,5-Dihydroxyphenyl)-L- isopropylamjnoäthanol,
Isoproterenol, Dextromethorphan, Hetrazan (Diethylcarbamazin), Diäthylamin, Triäthylamin usw., einem
Alkalimetallhydroxid (wie z.B. Natriumhydroxid, Kaliumhydroxid
od.dgl.) oder Ammoniak z.B. durch Vermischen und Erwärmen in einem geeigneten Lösungsmittel, umgewandelt
werden.
Die entstandenen Verbindungen der Formel (I) oder deren Salze, insbesondere die organischen Aminsalze, besitzen
antiallergische Eigenschaften, sie sind daher als vorbeugende und heilende Mittel bei der Behandlung von
allergischem Asthma, allergischer Dermatitis, Heuschnupfen und anderen allergischen Erkrankungen bei Menschen und
Säugetieren wertvoll. Die Alkalimetallsalze und organischen
Aminsalze sind in Wasser löslich und die daraus erhaltenen wässerigen Lösungen sind stabilB so daß die für die Herstellung
von Dosierungsformen wie injizierbare Lösungen und wässerige Lösungen gut geeignet sind.
Wenn eine Verbindung der allgemeinen Formel (I) oder eines ihrer Salze zur vorbeugenden oder heilenden Behandlung
solcher allergischer Erkrankungen beim Menschen eingesetzt wird, kann sie peroral in den üblichen Dosierungsmengen von etwa 5oo mg/Tag in Form von Tabletten,, Kapseln^
Pulvern oder Lösungen oder auf andere Weise in Form von

injizierbaren Lösungen, Inhalationen, Salben od.dgl. verabreicht
werden.
Die Ausgangsverbindungen der Formel (II) werden nach folgendem Verfahren hergestellt: Die Verbindung der

kann durch Umsetzen einer Verbindung der allgemeinen Formel (IV)
worin m und FU die vorstehend angeführte Bedeutung besitzen und welche nach dem in der US-PS 3 896 114 beschriebenen
Verfahren erhältlich ist, mit V/asser in Gegenwart einer Base hergestellt werden. Beispiele für geeignete
Basen sind organische Amine wie z.B. primäre Amine, z.B. Athylamin, n-Propylamin, n-Butylamin, Benzylamin,
Anilin usw., sekundäre Amine wie z.B. Dimethylamin, Diäthylamin,
Dipropylanin, Dibutylamin, Piperidin, Pyrrolidin
usw., tertiäre Amine wie z.B. Triethylamin od.dgl., heterocyclische Basen wie z.B. Imidazol, 2-Methylimidazol,
Worpholin und anorganische Basen wie z.B. wässeriges

Ammoniak, Ammoniumacetat, Ammoniumcarbonat, Natriurnacetat,
Natriumcarbonat, Natriumhydrogencarbonat od.dgl. Die Menge der einzusetzenden Base ist nicht besonders
kritisch und kann im Bereich von einer katalytischen Menge bis zu einem großen Überschuß liegen.
Im allgemeinen wird die Umsetzung vorzugsweise in einem mit Wasser mischbaren Lösungsmittel vorgenommen. Als
Beispiele für solche Lösungsmittel seien Dimethylformamid,
Dimethylsulfoxid, Hexamethylphosphorsäuretriamid, organische
Säuren (z.B. Ameisensäure, Essigsäure, Propionsäure od.dgl.) und Äther (z.B. Tetrahydrofuran, Dioxan od.dgl.)
genannt. Während Temperatur, Zeitdauer und die übrigen Reaktionsbedingungen nicht besonders kritisch sind, wird
die Umsetzung im allgemeinen bei Raumtemperatur bis zu
etwa loo C innerhalb eines Zeitraums von einigen Minuten bis zu etwa 3 Stunden durchgeführt.
Die Verbindung der Formel (II) kann durch Umsetzung einer Verbindung der Formel (Hl) mit einer aktiven Methylenverbindung,
einem Acetylen-Carbonsäurederivat in einem organischen Lösungsmittel oder einem Cyanacetylhalogenid
'in Gegenwart eines substituierten Formamids vorgenommen
werden.
Beispiele für die in der Umsetzung zu verwendende aktive Methylenverbindung sind Methylacetoacetat, Äthylacetoacetat,
Methylcyanoacetat, Athylcyanoacetat, Cyanoacetamid, Malonsäurenitril, Äthyloxalacetat, Diäthylmalonat,
Dimethylmalonat, Äthylbenzoylacetat, Methyl-3-oxon-caproat od. dgl. Üblicherweise werden in der Praxis 1 Moläquivalente
der aktiven Methylenverbindung pro Moläquivalent Verbindung (III) eingesetzt.

Die vorstehende Umsetzung wird vorzugsweise in Gegenwart einer Base vorgenommen, als solche kann ein
organisches Amin verwendet werden. Das organische Amin kann ein primäres wie z.B. n-Butylamin, Benzylamin,
Anilin od.dgl., ein sekundäres Amin wie z.B. Diethylamin,
Dipropylamin, Dibutylamin, Piperidin, Pyrrolidin, ein
tertiäres Amin wie z.B. l,8-Diazabicyclo/~5,4,0_/-7-undecen,
Triethylamin od.dgl. oder eine heterocyclische Base wie z.B. Imidazol, 2-Methylimidazol, Morpholin
sein. Der Anteil der eingesetzten organischen Base reicht üblicherweise von einer kstalytischen Menge bis zu etwa
5 UoIäquivalenten pro Mol Verbindung (III).
Im allgemeinen ist es günstig, die Umsetzung in einem organischen Lösungsmittel durchzuführen. Als Beispiele für
solche Lösungsmittel seien Alkohole wie Methanol, Äthanol, Propanol, Butanol od.dgl., aromatische Kohlenwasserstoffe
wie z.B. Benzol, Toluol od.dgl. und Dimethylformamid genannt.
Obgleich die Temperatur, Zeitdauer und übrigen Reaktionsbedingungen nicht kritisch sind, ist es im aligemeinen
vorteilhaft, die Umsetzung innerhalb eines Temperaturbereichs von Raumtemperatur bis etwa zum Siedepunkt
des Lösungsmittels in einem Zeitraum von etwa 1 Stunde bis zu 24 Stunden vorzunehmen.
Die erfindungsgemäße Umsetzung kann auch nach folgendem
Verfahren ausgeführt werden: eine Verbindung der allgemeinen Formel (III) wird mit einem Acetylen-Caxbonsäurederivat
umgesetzt. Beispiele für geeignete Acetylen-Carbonsäurederivate
sind Dimethylacetylendicarboxylat, Diäthylacetylendicarboxylat, Methylpropiolat, Äthylpropiolat,
Cyanoacetylen od. dgl. Wird ein Prop!ölsäureester eingesetzt,
so kann das als Zwischenprodukt anfallende

Aminoacrylatdexivat isoliert oder weiter einer Ringschlußreaktion unterworfen werden. Das Acetylendicarbonsäurederivat
wird im allgemeinen in einem Mengenverhältnis von etwa 1 - Io Moläquivalenten pro Mol Verbindung
(III) eingesetzt.
Im allgemeinen wird die vorstehend beschriebene Umsetzung vorteilhaft in Gegenwart einer Base wie z.B.
eines organischen Amins vorgenommen. Beispiele für solche organische Amine sind tertiäre Amine wie Triethylamin,
Tripropylamin, Tributylamin, usw., heterocyclische Amine
wie Pyridin, Quinolin, Imidazol, 2-Methylimidazol,
Morpholin od.dgl. und sekundäre Amine wie z.B. Piperidin, Pyrrolidin, Diethylamin, Dipropylamin, Dibutylamin od.dgl.
Die organische Base wird im allgemeinen in einer Menge, die von katalytischen Mengen bis zu etwa Io Moläquivalenten
reicht pro Mol Ausgangsverbindung der Formel (II) eingesetzt.
Im allgemeinen wird die Umsetzung vorteilhaft in einem organischen Lösungsmittel vorgenommen. Beispiele für
geeignete Lösungsmittel sind Alkohole wie Methanol, Äthanol; Propanol, Butanol od.dgl., aromatische Kohlenwasserstoffe
wie z.B. Benzol, Toluol, Xylol od.dgl., Dimethylformamid u.dgl. Obgleich Temperatur, Zeitdauer und
die übrigen Reaktionsbedingungen nicht kritisch sind, wird die Umsetzung vorteilhaft innerhalb eines Temperaturbereichs
von etwa Raumtemperatur bis nahe dem Siedepunkt
des eingesetzten Lösungsmittels während einer Zeitdauer von etwa 1-24 Stunden durchgeführt.
Die Verbindung der allgemeinen Formel (II) kann nach folgendem Verfahren hergestellt Werdens Eine Verbindung

der allgemeinen Formel (ill) wird mit einem Cyanoacetylhalogenid
in Gegenwart eines substituierten Formamids
umgesetzt. Als Beispiele für geeignete Cyanoacetylhalogenide seien Cyanoacetylchlorid, Cyanoacetylbromid,
Cyanoacetyliodid, Cyanoacetylfluorid od.dgl. genannt.
Geeignete substituierte Formanide für diese Reaktion können durch Alkyl oder Aryl substituierte Formamide sein,
wie Π,Π-Dimethylforniamid, ΓΙ,Μ-Diäthylformamid, II,N-Dipr
opy If ormamid, II-Met hy 1-M- ät hy If ormamid, N-Methy 1-H-phenyIf
ormamid, Η,ΐί-Diphenylf ormamid od. dgl. Die Umsetzung
kann in Gegenwart eines substituierten Formamids allein,
d.h. unter dessen Verwendung als Reaktionslösungsmittel, oder gegebenenfalls in einem Lösungsmittelgemisch; aus dem
eingesetzten substituierten Formamid mit einen Fremdlosungsmittel,
das die Umsetzung nicht beeinträchtigt, ausgeführt werden. Dieses Lösungsmittel ist im allgemeinen ein
übliches organisches Lösungsmittel wie ein Kohlenwasserstoff, z.B. Benzol, Toluol, Xylol, Petroläther od.dgl.,
ein Äther wie z.B. Tetrahydrofuran, Dioxan, Athy lather,
Äthylenglycoldimethylather, usw., ein halogenierter Kohlenwasserstoff
wie z.B» Chloroform, Dichlorr.ethan, Dichloräthan,
Tetrachloräthan, usw. ,eines Esterswie Athylacetat, Methylacetat, Butylacetat, Acetonitril, Dimethylsulfoxid
od.dgl. Die Menge des bei der Herstellung der Verbindung der Formel (II) eingesetzten Cyanoacetylhalogenids liegt
üblicherweise innerhalb eines Bereichs von 1 - Io Moläquivalenten bezogen auf die Ausgangsverbindung (III). Obgleich
Temperaturbereich, Zeitdauer und die übrigen Reaktionsbedingungen nicht kritisch sind, wird die Umsetzung im
allgemeinen bei Temperaturen von etwa 2o bis etwa 12o C innerhalb eines Zeitraums von etwa 3o min bis 2 Tagen durchgeführt.
Auch die Menge des substituierten Formamide ist

Die Umsetzung wird vorteilhaft in einem organischen Lösungsmittel wie z.B. Chloroform, Dichlormethan, Acetons
Methylethylketon od. dgl. durchgeführt. Sie wird im allgemeinen innerhalb eines Temperaturbereichs von 0°C bis nahe
dem Siedepunkt des eingesetzten Lösungsmittels, in einem Zeitraum von etwa einigen Minuten bis einigen Stunden vorgenommen.
Die Verbindung der Formel (ll)s worin R* ^ Monoalkylamin
bedeutet, kann durch Umsetzen einer Verbindung der Formel (II), worin R1, Chlor bedeutets die durch Umsetzen
einer Verbindung der Formel (II), worin R'χ Hydroxy ist,
mit einem Gemisch aus Phosphoryltrichlorid und Phosphorpentachlorid
bei etwa loo - 12o°C innerhalb von wenigen Stunden bis zu einigen Tagen hergestellt worden war, mit
einem Monoalkylamin wie z.B. Methylamin, Äthylamins Propylamin
od.dgl. erhalten werden. Im allgemeinen wird die Umsetzung in einem organischen Lösungsmittel durchgeführt.,
Als Beispiele für das Lösungsmittel zum Lösen der Verbindung der Formel (II), worin R!, Chlor bedeutet, seien
Chloroform, Dichlormethan ododgl. genannt, ein für die
Lösung des Monoalkylamins geeignetes Lösungsmittel ist Methanol, Äthanol, Propanols Chloroform öd»dgl*

Obgleich Temperatur, Zeitdauer und die übrigen Reaktionsbedingungen bei der Herstellung einer Verbindung
der Formel (II) ,worin R1 ·, Chlor bedeutet, durch Umsetzen
mit einem Monoalkylamir: nicht kritisch sind, wird die
Umsetzung im allgemeinen innerhalb eines Temperaturbereichs
von etwa 0 C bis Raumtemperatur in einem Zeitraum von einigen Minuten bis etwa 3 Stunden durchgeführt.
Die Verbindung der Formel (II), worin R1, Hydroxy
bedeutet, kann z.B. durch Umsetzen einer Verbindung der Formel (ll), worin R1, Amino ist, mit einem Alkalisalz
eines Ilitrits wie z.B. Natriumnitrat, Kaliumnitrit od. dgl.
in einer wässerigen, anorganischen oder organischen Säure wie z.B. Salzsäure, Essigsäure od.dgl. hergestellt werden.
Eine Mischung aus 2 ml Morpholin, 3 ml Dimethylformamid
und Io ml Wasser wurde auf 6o°C erhitzt, dann wurden unter
Rühren innerhalb von 5 min 1,71 g feinteiliges 4-Qxo-4II-lbenzopyran-3-carbonitril
zugegeben. Das Gemisch wurde 1 Stunde lang auf 6o°C gehalten, der entstandene niederschlag
wurde abfiltriert, mit V/asser gewaschen, aus Essigsäure umkristallisiert und mit Chloroform gewaschen. Nach
diesem Verfahren wurden 1,32 g kristallines 2-Amino-4-oxo-4H-l-benzopyran-3-C3rboxaldehyd
mit einem Schmelzpunkt von 252 - 2550C (Zers.) erhalten.

Ein Gemisch aus 366 mg 2-Amino-4-oxo-4H-l-benzopyran-3~ carboxaldehyd, 5oo mg Methyl-3-oxo-n-caproat und 036 ml
Piperidin in 2o ml Methanol wurde 6 Stunden lang am Rückfluß gekocht. Das Reaktionsgemisch wurde bei vermindertem
Druck eingeengt und der Rückstand der Säulenchromatographie an Silikagel unterworfen. Darauf wurde mit Chloroform
eluiert und das Haupteluat gesammelt und aus Methanol umkristallisiert. Man erhielt 52 mg Methyl-2-n-propyl-lazaxanthon-3-carboxylat
in Form von gelben Kristallen mit einem Schmelzpunkt von Io5 - Io6 C.

Ein Gemisch aus 2,17 g 2-Amino-6-äthyl-4-oxo-4H-lbenzopyran-3-carboxaldehyd,
1,778 g Dimethylacetylendicarboxylat, 5o ml Methanol und 0,5 ml Triäthylamin wurde am Rückfluß 3 Stunden lang gekocht, nach dem Abkühlen
wurde der Mieder schlag abfiltriert. Darauf wurde der Niederschlag in Chloroform eingebracht und gerührt,
worauf das schwer lösliche Produkt abfiltriert wurde. Das Filtrat wurde zur Trockne eingeengt und in etwa 5o ml
n-Hexan-Chloroform-Aceton (lo ; 5 : 0,5) gelöst. Die
Lösung wurde über eine Kolonne mit 3o g Silika^gel geleitet
und mit dem gleichen Lösungsmittelsystem eluiert. Das Eluat wurde konzentriert und der Rückstand aus Methanol
umkiistall isiert. Man erhielt 59o mg Dimethyl-7-äthyl-lazaxanthon-2,3-dicarboxylat
in Form von farblosen Nadeln mit einem F.p. von 169 - 17O0C.

In 4o ml Dimethylformamid wurden 1,82 g 2-Amino-6-äthyl-4-oxo-4H-l-benzopyran-3-carboxaldehyd
gelöst, darauf folgte die Zugabe von 3,5 g Cyanoacetylchlorid. Das Gemisch wurde
unter ständigem Rühren bei 60 C 3 Stunden lang reagieren gelassen. Dann wurde das Lösungsmittel unter vermindertem
Druck abdestilliert und der Rückstand an Silikagel chromatographiert. Das gewünschte Produkt wurde aus dem
Chlorofoxmeluat entfernt und aus Acetonitril umkristallisiert.
Man erhielt I,o3 g 7-Äthyl-3-cyano-l-azaxanthon.

Das Reaktionsgemisch wurde weitere 45 min bei Raumtemperatur gerührt und dann mit einer kleinen Menge Essigsäure
versetzt, um den Überschuß an Diazomethan zu zersetzen. Das Lösungsmittel wurde abdestilliert und der
Rückstand der Säulenchromatographie an loo g Silikagel unterworfen, darauf wurde mit Chloroform-Aceton-Ameisensäure
(2o : 1 ; 0,l) eluierto Die erste Fraktion wurde aus Aceton umkristallisiert, wobei 615 mg Äthyl-7-äthyl-2-methoxy-l-azaxanthon-3-carboxylat
in Form von farblosen Nadeln mit einem Schmelzpunkt von 145 - 1460C erhalten
wurden.

Druck abdestilliert. Der Rückstand wurde allmählich unter Eiskühlung mit Äthanol versetzt um ihn zu lösen. Dann
wurde das Lösungsmittel abdestilliert und der Rückstand der Saulenchrornatographxe an Silikagel unterworfen. Das
gewünschte Produkt wurde mit Benzol eluiert und aus Isopropyläther umkristallisiert, wobei 658 mg Äthyl-7-äthyl-2-chlor~l-azaxanthon-3-carboxylat
in Form von farblosen Kristallen mit einem Schmelzpunkt von 16o - 1610C

Ein Gemisch aus 1,58 g Äthyl-3- (6-äthyl-3-f ormyl-4-οχο-4H-l-benzopyran-2-yl)aminoacrylat
(hergestellt nach Bezugsbeispiel 22), 15 ml Dimethylformamid und 5 ml Triäthylamin
wurde bei 13o°C'2,5 Stunden lang am Rückfluß gekocht. Das Reaktionsgemisch wurde eingeengt und der
Rückstand mit etwa 5o ml Äthanol versetzt. Die äthanolische Lösung wurde gekühlt und die ausgefällten Kristalle wurden
abfiltriert und aus Äthanol umkristallisiert, wobei 92o mg 7-Äthyl-l-azaxanthon-3-äthylcarboxylat in Form von
hellgelben Nadeln mit einem Schmelzpunkt von 14o - 142°C erhalten wurden.

und darauf mit 2,5 g Cyanoacetylen versetzt. Das Gemisch wurde unter Rühren bei 14o°C 15 Stunden lang erhitzt, nach Ablauf dieses Zeitraumes
wurde das Lösungsmittel unter vermindertem Druck abdestilliert. Der Rückstand wurde der Säulenchromatographie
an Silikagel unterworfen, mit Chloroform eluiert und aus Acetonitril umkx istall is iext, wobei 0,83 g 3-Cyano-1-azaxanthon
in Form von Kristallen mit einem Schmelzpunkt von 22o - 2260C erhalten wurden.

Eine Lösung von 0,5 g 7-Isopxopyl-2-amino-3-cyano-1-azaxanthon
in 8o ml Essigsäure wurde allmählich mit 1,0 g Natriumnitrii bei 7o C versetzt. Nach einer Stunde wurde
das Gemisch mit 3 ml V/asser versetzt und eine weitere Stunde auf 7ο C erwärmt. Dann wurde das Lösungsmittel unter
vermindertem Druck abdestilliert und der Rückstand mit Wasser versetzt. Der gelbe Niederschlag wurde abfiltriert,
mit Wasser gewaschen und aus Äthanol umkristallisiert,
wobei 7-Isopropyl-2-hydroxy-3-cyano-l-azaxanthon in Form von gelben Kristallen mit einem Schmelzpunkt von
> 3oo°C erhalten wurden.

Beispiel 1:
5 ml 55 /£ige Schwefelsäure wurden mit 933 mg 7-Äthyl-2-methyl-l-azaxanthon-3-äthylcaxboxylat
versetzt, das Gemisch wurde bei 13o°C 1 Stunde lang gerührt. Nach dem Abkühlen wurden dem Reaktionsgemisch loo ml Eiswasser zugegeben
und der Niederschlag wurde abfiltriert und aus Äthylacetat und Aceton umkristallisiert. Man erhielt
51o mg kristalline 7-Äthyl-2-methyl-l-azaxanthon-3-carbonsäure
mit einem Schmelzpunkt von 243 - 245 C.

Io ml Essigsäure und Io ml 55 /oiger Schwefelsäure wurde bei 13o C 4 Stunden lang gerührt, der
nach Zugabe von Wasser entstandene Niederschlag wurde abfiltriert und aus Essigsäure-Wasser umkristallisiert.
Man erhielt 1,28 g Z-Amino-Y-äthyl-l-azaxanthor-S-caxbonsäure,
F.p. 313 - 3140C (Zers.), in Form von farblosen
Nadeln.

Ein Gemisch aus 341 rng Dimethyl-7-äthyl-l-azaxanthon-2,3-dicarboxylat,
2 ml Essigsäure und 2 ml 55 /oiger Schwefelsäure wurde bei 13o°C 4 Stunden lang gerührt. Nach
dem Abkühlen wurde Wasser zugegeben und der erhaltene Niederschlag abfiltriert und vorsichtig in einem Rundkolben
über offener Flamme erhitzt, worauf er unter Schaumbildung schmolz. Nach Abklingen der Schaumbildung wurde
die geschmolzene Masse gekühlt und aus Aceton umkristallisiert. Man erhielt 124 mg T-Athyl-l-azaxanthon-S-carbonsäure
in Form von farblosen Nadeln mit einem Schmelzpunkt von 238 - 2390C.

Ein Gemisch aus 5oo mg 7-Äthyl~2-methoxy-l-azaxanthon-3-äthylcarboxylat,
Io ml Essigsäure und 5 ml 55 /oiger Schwefelsäure wurde bei 800C 2 Stunden lang gerührt, dann
mit 2o ml V/asser versetzt und weitere Io min bei dieser Temperatur gerührt. Nach dem Abkühlen wurde der entstandene
Niederschlag abfiltriert, mit V/asser gewaschen, getrocknet und in 3 ml Dimethylformamid gelöst. Die entstandene Lösung
wurde mit 3 g Silikagel versetzt und gut gerührt, dann wurde das Dimethylformamid abdestilliert. Der Rückstand
wurde der Säulenchromatographie an 5o g Silikagel unterworfen und mit einem Lösungsnittelsystera aus Chloroform-Aceton-Ameisensäure
(9 : 1 : 0,1) eluiert, wobei 39o mg Kristalle erhalten wurden. Diese wurden aus Dimethylformamid-Äthanol
umkristallisiert, und 358 mg 7-Athy1-2-methoxy-l-azaxanthon-3-carbonsaure
wurde in Form von farblosen Nadeln mit einem Schmelzpunkt von 226 - 2280C erhalten.

Beispiel 12:
In einem Gemisch von Io ml 5o böiger wässeriger Schwefelsäure
und Io ml Essigsäure wurden 0,875 g 7-Isopropyl-3-cyano-l-azaxanthon
bei 12o°C 2 Stunden lang gerührt. Nach beendeter Umsetzung wurde das Reaktxongsgemxsch
mit Wasser versetzt und der entstandene Niederschlag abfiltriert, mit Wasser gewaschen, getrocknet und aus
Äthanol umkristallisiert. Man erhielt 0,623 g 7-Isopropyll-azaxanthon-3-carbonsäure
mit einem Schmelzpunkt von 259 - 261°C.
