Phenoxyalkancarbonsäurederivat, Verfahren zu seiner Herstellung und Arzneimittel
Gegenstand der Erfindung sind neue Phenoxyalkancarbonsäurederivate,
nämlich Salze von l-Cinnamyl-4-diphenylmethylpiperazin (Cinnariziri)
und Halogenphenoxyalkancarbonsäuren, denen folgende Strukturformel I
gegeben werden kann:

Hierin bedeuten
R, ein Wass.er stoff atom, eine niedere
Alkylgruppe, oder eine weitere p-Chlorphenoxy-Gruppe
und
R, ein Wasserstoffatom oder eine niedere
R, ein Wasserstoffatom oder eine niedere
Alkylgruppe.
Unter einer niederen Alkylgruppe werden Alkylgruppen mit 1 - 4
C-Atomen, insbesondere die Methyl-, Äthyl-, Propyl- und Isopropy!gruppen
verstanden.

Wie aus zahlreichen Untersuchungen bekannt
ist, wirken p-Chlorphenoxyessigsäure, p-Chlorphenoxypropionsäure, p-Chlorphenoxyisobuttersäure
und Bis-(p-Chlorphenoxy)-essigsäure, sowie deren Ester und Amide allgemein lipidsenkend.
Die arzneilich bekanntesten Vertreter sind
der 2-(p-Chlorphenoxy)-2-methylpropionsäureäthylester (Clofibrat), sowie die entsprechende
Carbonsäure (Clofibrinsäure).
Da Clofibrat als Ester erst nach Hydrolyse zur Ciofibrinsäure seine lipidsenkende
Wirkung entfaltet, darf diese Säure auch als das eigentliche Wirkprinzip gelten.
Der therapeutischen Verwendung von CIofibrinsäure stehen jedoch einige Nachteile
entgegen, wie z. B. der unangenehme Geruch, sowie die der Säure eigene Reizwirkung auf
den Magen-Darm-Trakt.
Zur Behebung dieser Nachteile wurden bereits Salze mit organischen Basen (DOS
1 96O 273)i sowie Salze mit anorganischen
Basen, z. B. Aluminiumsalze (DOS 1 767 533), hergestellt.
Ferner ist aus verschiedenen pharmakologischen Tierversuchen bekannt, daß Ciofibrinsäure p. o.
nur unter Normaldiät zu einer guten Senkung von Serumcholesterin- und Serumtriglyzerid-Werten
führt, während unter zusätzlicher Cholesterin- sowie Cholsäure-Zufuhr bzw.
unter Pettdiät, keine ausgeprägte Lipoidsenkung mehr zu erzielen ist.
Dies beruht, wie neuere Untersuchungen zeigten, darin, daß Ciofibrinsäure ausschließlich
in den endogenen LeberStoffwechsel hemmend eingreift, bei exogener Zufuhr von
Fetten, sowie Cholesterin, aber keine Wirksamkeit mehr entfalten kann.

Wie nun überraschend gefunden wurde, zeigen Salze von Cinnarizin mit Phenoxyallcancarbonsäur'en
einen synergistischen Effekt. t der sich in einer die Wirksamkeit der Einzelverbindungen
übertreffenden Aktivität äußert.
So zeigt z. B. das Salz von Cinnarizin mit Ciofibrinsäure im Tierversuch nicht nur
unter Normaldiät eine höhere Aktivität, sondern auch unter Cholesterxnbelastung,
Cinnarizin selbst ist eine bekannte Verbindung mit antihistaminischer, sowie cerebral
und peripher vasodilatatorxscher Wirksamkeit. Ein dem Cinnarizin eigener lipidsenkender
Effekt wurde bisher nicht beschrieben.
Hingegen war bekannt, daß ähnliche Diarylalkylpiperazine,
wie Chlorcyclizin im Tierversuch, lipidsenkende Eigenschaften besitzen, wobei besonders erhöhte Serumcholesterinwerte
gesenkt wurden (Toxikol. and Appl. Pharmacol. 7_, 257, 1965).
In dieser Studie konnte gezeigt werden, da3 bei der Lipid- und Cholesterinsynthese nur der
Cl^-acetat-Einbau und nicht der C -mevalonat-Einbau
gesteigert wurde.
Die zur Herstellung der erfindungsgemäßen Salze
benötigten Ausgangsverbindungen besitzen eine geringere Toxizität. So beträgt z. B. der LD-Wert
(p. o. Maus) von Cinnarizin > 1 280 mg/kg, ·
der von Clofibrat ca. 1 48o mg/kg (Literaturangaben).

Neben der erwähnten verbesserten Aktivität zeigen die erfindungsgemäßen Salze noch folgende
Vorteile:
a) die Salze sind nahezu wasserunlöslich, aber sehr gut löslich in Lipoidlösungsmitteln,
wie Aceton, Alkohole, Essigester etc. ,
b) die Salze sind geruchlos, nahezu geschmacklos und führen zu keiner Reizung
der Schleimhäute,
c) die Salze besitzen noch eine gute antithrorabotische,
sowie insbesondere eine peripher vasodilatatorische Wirkung.
Ihre antilipämische Wirksamkeit in Verbindung
mit der peripher dilatatorischen Wirkung, macht die erfindungsgemäßen Salze geeignet zur Behandlung
von Stoffwechselerkrankungen mit erhöhten
Lipidwerten, insbesondere von Erkrankungen mit erhöhten Cholesterinwerten, die einen entscheidenden
Risikofaktor beim Entstehen der Arteriosklerose und peripheren Gefäßsklerose darstellen.
Die Herstellung der erfindungsgemäßen Verbindungen
erfolgt nach bekannten präparativen Methoden und wird in den folgenden Beispielen
beschrieben:
Beispiel 1; .
7i36 g (0,02 Mol) Cinnarizin, sowie 4,30 g
(0,02 Mol) Ciofibrinsäure werden in 30 ml Isopropanol unter Erwärmen auf Rückflußtemperatur
geJLöst und heiß filtriert. Nach dem Stehen über Nacht bei Raumtemperatur
wird das gebildete Kristallisat abgesaugt und mit 5 ml Isopropanol gewaschen.

Es werdei9,8 g (84,6 % d. Th,) Cinnarizinclofibrinat
als farbloses Kristallisat vom Fp 109,6° C erhalten.
Die Durchführung der Reaktion in Äthanol als Lösungsmittel ergibt 6,6 g (56,7 %
d. Th.) Cinnarizin-clofibrinat in farblosen Nadeln vom Fp 109,8° C.
Die Gehaltsbestimmung von Cinnarizin-clofibrinat
durch potentiometrische Titration ergibt folgende Vierte:
Titration mit Ο,Ιή Gehalt 100,6 % Cinnarizin
Titration mit 0,IN NaOH: Gehalt: 99,8 % Ciofibrinsäure
^»05 g (0,01 Mol) Cinnarizin Hydrochlorid
verden in Wasser gelöst und mit einer separat hergestellten Lösung von 2,37 (0,01 Mol)
Natriumsalz der Ciofibrinsäure im Wasser unter
Rühren versetzt. ·
Das gebildete Kristallisat wird abgesaugt, mit Wasser gewaschen und nach dem Trocknen
aus wenig Äthanol umkristallisiert.
Es werden 4,^6 g (78,2■% d. Th.) Cinnarizinclofibrinat
als farbloses Kristallisat vom 109,7° C erhalten.
Entsprechend der Herstellungsmethode nach Beispiel 1, wurden folgende weitere Salze
durch Eindampfen der Reaktionslösung hergestellt:
Cinnarizin-p-chlorphenoxypropionat,
gelbliches, zähflüssiges Öl.
Gehalt: (Cinnarizin 101.1 % d. Th./
p-Chlorphenoxypropionsäure 99» d. Th.)

Cinnarizin-p-chlorphenoxyacetat,
farbloses, zähflüssiges Öl.
Gehalt: (Cinnarizin 102,2 % d. Th./
p-Chlorphenoxyessigsäure 100,3 d. Th.)
Cinnarizin-bis-(p-chlorphenoxy)acetat,
gelbliches, zähflüssiges Öl.
Gehalt: (Cinnarizin 99,6 % d. Th./
Bis-(p-Chlorphenoxy/-essigsäure 100,9 % d. Th.)
Die unter den Beispielen 3 und 4 beschriebenen
zähflüssigen Verbindungen, konnten nicht durch geeignete Kristallisation in reiner Form erhalten
werden, da je nach Lösungsmittel das erhaltene Kristallisat mehr oder weniger durch
Cinnarizin verunreinigt war, bzw. die Gehaltsbestimmung der zähflüssigen Rohprodukte im Gegensatz
zu den Kristallisaten gut übereinstimmende Wert ergab.
Die erfindungsgemäßen Arzneimittel enthalten ein- oder mehrere Phenoxyalkancarbonsäure-Salze
der allgemeinen Formel I, gegebenenfalls in Kombination mit weiteren Phenoxyalkancarbonsäur
ed erivat en.
Die Anwendung erfolgt vorzugsweise oral, z. B. in Form von Kapseln, Tabletten oder
Dragees, die gegebenenfalls übliche pharmazeutische Trägerstoffe und Hilfsmittel enthalten.

Pflanzenöle sowie hydrierte Pflanzenöle, ferner Emulgatoren und Gerüstbildner
(Wachse, Sojalecithin), zum

Die Verabreichung der erfindungsgemäßen Verbindungen
erfolgt, je nach dem Fall, in
oralen oder rektalen Tagesdosen von 300 bis 1200 mg, vorzugsweise 400 bis 800 mg, in den üblichen pharmazeutischen Formen.

Für den Fall, daß die erfindungsgemäßen
Verbindungen bei leichteren Fällen der
Hyperlipämie mit weiteren Phenoxyalkancarbonsäurederivaten kombiniert werden,
ist die Verwendung von Clofibrat sowie
Ciofibrinsäure besonders bevorzugt.

Claims (4)
1. Phenoxyalkancarbonsäurederivate von 1-Cinnamyl-k-diphenylmethylpiperazin
(Cinnarizln) der allgemeinen formel I

in der
R. ein Wasserstoffatom, eine niedere Alkylgruppe, oder eine weitere
p-Chlorphenoxy-Gruppe
und
R2 ein Wasserstoffatom oder eine
niedere Alkylgruppe
bedeuten.
2. Cinnarizin-p-chlorphenoxyisobutyrat
Verfahren zur Herstellung der Phenoxyalkancarbonsäurederivate
der in Anspruch 1 angegebenen allgemeinen Formel (I)1 dadurch
gekennzeichnet, daß man in einem geeigneten Lösungsmittel eine Phenoxyalkancarbonsäure,
in der R. und R» die unter Anspruch 1 angegebene Bedeutung besitzen,
oder ein reaktionsfähiges Derivat hiervon, mit Cinnarizin oder dessen reaktionsfähigem
Derivat in äquimolaren Mengen zur Reaktion bringt.
4. Arzneimittel, dadurch gekennzeichnet,
daß sie eine oder mehrere Verbindungen nach Anspruch 1, gegebenenfalls zusammen
mit üblichen pharmazeutischen Trägerstoffen und Hilfsmitteln, enthalten.
