Description
- Titel: Enzymatisches Verfahren zur Bestimmung der Wasserstoffperoxid-Konzentration.
- Anwendungsgebiet: Medizinisches Laboratorium, insbesondere Klinische Chemie.
- Stand der Technik: Die Bestimmung der Wasserstoffperoxid-Konzentration spielt in der Klnischen Chemie eine zunehmend wichtige Rolle als Indikatorreaktion bei mehreren Analysenverfahren, z.B.: 1. Bestimmung der Glucose-Konzentration (Blutzucker) mit Hilfe der Glucoseoxidase (GOD-) Reaktion: Glucose + H20 + 2 Gluconsäure + H202 2. Bestimmung der Harnsäure-Konzentration mit Hilfe der Uricase-Reaktion: Harnsäure + 2 H20 + °2 Allantoin + H202 + CO2 3. Bestimmung der Cholesterin-Konzentration mit Hilfe der Cholesterinoxidase-Reaktion: Cholesterin + H20 + °2 H2°2 + A +a4-Cholestenon Bei diesen Analysenverfahren werden zur Zeit vorwiegend 2 verschiedene Indikator-Reaktionen angewendet: 1. Peroxidase-Reaktion bei der Glucose-Bestimmung: H O + DH 22 2H20+D DH2 = reduzierter Wasserstoffdonator, z.B. o-Dianisidin, ABTS Nachteil: 1. Extinktionskoeffizient des gebildeten Farbstoffs nicht bekannt. Daher muß in jeder Serie eine Standardlösung mitanalysiert werden.
- 2. Wegen mehrerer Interferenzen (zip. Ascorbinsäure) für Urinanalysen nicht brauchbar.
- 1) 2. Hantzsche Reaktion bei der Harnsäure (nach Kageyama)- und Cholesterin-Bestimmung : H202 + Methanol Formaldehyd + 2 H20 Formaldehyd + 2 CH3COCH2.COCH3 + NH3 ---- 3,5-diacetyl-1,4-dihydrolutidin +31120 Der gebildete Farbstoff wird bei 410 nm photometrisch gemessen.
- Nachteil: 1. Reaktionsverlauf sehr langsam. Bei Serum-analysen dauert die Reaktion etwa 60 Minuten.
- 2. Extinktionskoeffizient des gebildeten Farbstoffes ist nicht bekannt. Daher muß in jeder Serie eine Standardlösung mitanalysiert werden.
- 3. Interferenz durch hohe Konzentrationen an Ascorbinsäure.
- Beschreibung der vorliegenden Erfindung: Das beschriebene-Verfahren besteht aus 2 Reaktionen, der Katalse-und der Aldehyddehydrogenase Reaktion: 1. K202 + Alkohol 22 Aldehyd + 2 H,O 2 2. Aldehyd + NAD(P)+ Säure + NAD(P)H + H+ *Folgende Aldehyddehydrogenasen können verwendet werden: Aldehyd:NAD Oxidoreductase E.C. 1.2.1.3.
- Aldehyd:NADP Oxidoreductase E.C. 1.2.1.4.
- Aldehyd:NAD(P) Oxidoreductase E.C. 1.2.1.5.
- Benzaldehyd:NADP Oxidoreductase E.C. 1.2.1.7.
- Betainaldehyd:NAD Oxidoreductase 3.C. 1.2.1.8.
- Formaldehyd:NAD Oxidoreductase E.C. 1.2.1.1.
- Vorteile des beschriebenen Verfahrens: 1. Mitführen von Standardlösungen ist nicht erforderlich, da der Extinktionskoeffizient von NADH, bzw. NADPH hinreichend bekannt ist.
- 2. Rascher Reaktionsablauf (Vergl.Anwendungsbeispiel unten).
- 3. Die Reaktion ist für Urin- und Serumanalysen geeignet.
- 4. Keine Interferenzen bekannt sind.
- Chlesterin-Bestimmung 11202 + Methanol Formaldehyd + 2 H20 Formaldehyd + 2 CH3COCH2.COCH3 + N113 -" ---- 3,5-diacetyl-1,4-dihydrolutidin +3 1120 Der gebildete Farbstoff wird bei 410 nm photometrisch gemessen.

- Aldehyd:NAD(P) Oxidoreductase E.C. 1.2.1.5.
- Benzaldehyd:NADP Oxidoreductase E.C. 1.2.1.7.
- Betainaldehyd:NAD Oxidoreductase E.C. 1.2.1.8.
- Formaldehyd:NAD Oxidoreductase E.C. 1.2.1.1.
- Vorteile des beschriebenen Verfahrens: 1. Mitführen von Standardlösungen ist nicht erforderlich, da der Extinktionskoeffizient von NADH,-bzw. NADRH hinreichend bekannt ist.

- Anwendungsbeispiel: Im folgenden wird ein Reaktionsansatz zur Bestimmung der Harnsäure-Konzentration beschrieben.
- Reaktionslösungen: 1. Pyrophosphat-Puffer: Tetra-Natriumdiphosphat-10-hydrat (E.Merck, Darmstadt, Bestell-Nr. 6591) 0,1 mol/l, pH = 8,6 2. Katalase (Boehringer-Mannheim GmbH, Bestell-Nr. 15674) 20 mg/ml.
- 3. ricae (Boehringer-Mannheim GmbH, Bestell-Nr. 15074) 2 mg/ml.
- Täglich eine Verdünnung ansetzen: 20 Mikroliter + 200 Mikroliter bidest.Wasser.
- 4. NAD-Lösung (Boehringer-Mannheim GmbH, Bestell-Nr. 15300) 10 mg/ml.
- 5. Aethanol (E.Merck AG, Darmstadt, Bestell-Nr. 970).
- 6. Aldehydrdehydrogenase E.C. 1.2.1.3., isoliert aus Leber nach Leicht (3), spezifische Aktivität: 0,5 U/ mg Protein Vor jeder Serie wird ein Reaktionsgemisch aus Lsöung 1, 2, 4, 5 und 6 in folgenden Mischungsverhältnis angesetzt: Pyrophosphat-Puffer 500 Teile Katalase 0,5 Teile NAD-Lösung 20 Teile Äthanol 40 Teile Aldehyddehydrogenase etwa 25 U/ml Von diesem Reaktionsgemisch werden in eine Halbmikroküvette (Schichtdicke 1 cm) pipettiert: Reaktionsgemisch 500 Mikroliter -Probe 50 Mikroliter Uricase-Verdünnung 20 Mikroliter In dem Beispiel von Abbildung 1 wurde zunächst die Probe zu dem Reaktionsgemisch gegeben und abgewartet bis eine eventuell aufgetretene Reaktion abgelaufen ist. Danach erfolgte der start der eigentlichen Reaktion durch Zugabe der Uricase-Verdünnung. Registriert wurde mit einem Eppendorf Photometer 1101 und einem Philipps Schreiber (Analog-Meßplatz der Firma Eppendorf Gerätebau GmbH, Hamburg) bei einer Wellenlänge vo 334 nm. Die Uricase-Reaktion war nach etwa 3 Minuten beendet. Als Probe wurde eine wässrige Standardlösung (Harnsäure-Konzentration 5.000 umol/l) verwendet.
- Identische Resultate wurden mit der Aldehyddehydrogenase E.C. 1.2.1.5.
- (isoliert nach J.F.Clark und W.B. Jakoby, Journal of Biological Chemistry 245,6065, 1970, oder bezogen von Sigma Chemical Comp., St.Louis, MO 63178, USA) erhalten. Bei diesen Versuchen wurden an Stelle von Lösung 1 und 4 folgende Lösungen verwendet: 1. Pyrophosphat-Kaliumchlorid Puffer: Tetra-Natriumdiphosphat (E.Merck, Darmstadt, Bestell-Nr.6591) 0,1 mol/l und Kaliumchlorid (E.Merck, Darmstadt, Bestell-Nr. 4936) 0,1 mol/l, pH = 8,6.
- 4. NADP-Lösung (Boehringer-Mannheim GmbH, Bestell-Nr. 15600) 10 mg/ml.

Claims (2)
- PATENTANSPRUCHEAngemeldet wird ein spezifisches, enzymatisches Verfahren zur Bestimmung der H202 (Wasserstoffperoxid-)Konzentration in wässrigen und bilogischen Medien, d a d u r c h g e k e n nz e i c h n e t, daß durch Katalase katalysierte Reaktionen durch eine nachfolgende Aldehyd-dehydrogenase Reaktion (NAD oder NADP abhängig) angezeigt wird.
- 2. Analysenverfahren nach Anspruch 1, d a d u r c h g e k e n nz e i c h n e t, daß es als Indikatorreaktion bei allen Methoden, die H202 erzeugen, verwendet werden kann (z.B. enzymatische Bestimtnung der Cholesterir-, Harnsäure- und Glucosekonzentration).
