2. Verfahren zur Herstellung der Verbindungen gemäß Anspruch 1, dadurch gekennzeichnet, daß
man ein a-Chloracetanilid der allgemeinen Formel

haben, in einem wasserfreien Kohlenwasserstoff mit einem im Überschuß eingesetzten Amin der

die obengenannten Bedeutungen haben, in an sich bekannter Weise kondensiert und die erhaltenen
Produkte gegebenenfalls in die Säureadditionssalze überführt.
3. Arzneimittel, gekennzeichnet durch einen Gehalt an mindestens einer Verbindung nach
Anspruch 1 als Wirkstoff.

Diäthylamino, Pyrrolidino, Piperidino, Hexahydroazepino,
Piperazino, Morpholino und Thiomorpholino bedeutet, sowie deren Säureadditionssalze.
Die Verbindungen der Formel (I) und ihre ungiftigen Säureadditionssalze sind wertvoll für die Therapie. Es
wurde gefunden, daß pharmazeutische Zubereitungen, die in Verbindung mit einem physiologisch unbedenklichen
Träger oder Hilfsstoff wenigstens eine Verbindung der Formel (I) oder eines ihrer ungiftigen Säureadditionssalze
als Wirkstoff enthalten, wertvolle Eigenschaften, insbesondere spasmolytische, choleretische und
sedierende Wirkungen haben.
Wegen ihrer ausgeprägten Wirkung sind die folgenden Verbindungen bevorzugt:
N.N-Diäthylamino-S.S-dimethoxy-acetanilid
Morpholino-S.S-dimethoxy-acetanilid
und deren Säureadditionssalze.

und deren Säureadditionssalze.
Die Erfindung betrifft ferner die Herstellung der Verbindungen der Formel (I) nach einem Verfahren, das
dadurch gekennzeichnet ist, daß man «-Chloracetanilide, die in 3- und 5-Stellung in geeigneter Weise mit
Alkoxyresten (II) substituiert sind, in einem wasserfreien Kohlenwasserstoff, vorzugsweise Xylol, mit einem
aliphatischen oder N-heterocyclischen, im Überschuß verwendeten Amin (III) gemäß dem Reaktionsschema

die obengenannten Bedeutungen haben, in an sich bekannter Weise kondensiert.
Die Verwendung eines Kohlenwasserstoffs als Lösungsmittel
ermöglicht die Ausfällung des sich bildenden Hydrochlorids des Amins (III) und demzufolge seine
Entfernung.
Als ungiftige Säureadditionssalze kommen die in der
Therapie gebräuchlichen Salze mit anorganischen und organischen Säuren in Frage. Als anorganische Säuren
eignen sich Salzsäure, Bromwasserstoffsäure, Jodwasserstoffsäure, Schwefelsäure, Salpetersäure und Phosphorsäure,
und als organische Säuren kommen Weinsäure, Zitronensäure, Oxalsäure, Maleinsäure, Fumarsäure,
Ascorbinsäure und Cyclohexansulfonsäure in Frage.

Zur Herstellung dieses Produkts gibt man 14,6 g (0,2 Mol) Diethylamin schnell zu 11,5 g (0,05 Mol)
3,5-Dimethoxy-«-chloracetanilid in 50 ml Xylol, erhitzt
10 h am Rückfluß, filtriert nach Abkühlung das Diäthylaminhydrochlorid ab, wäscht mit Xylol, extrahiert
die Xylollösung zweimal mit je 100 ml 1 n-Salzsäure,
fällt die ölige Base mit 100 ml 2,5 n-Natriumhydroxid,
extrahiert die Base zweimal mii Äther, wäscht anschließend mit Wasser und trocknet. Das Hydrochlorid
wird unter RüVün mit ätherischer Salzsäure
ausgefällt. Man kristallisiert aus einem Pflanzenkohle enthaltenden siedenden Gemisch von i20ml Isopropanol
und 30 ml Äthanol um und erhält 11.1 g (Ausbeute 75,7%) weiße Kristalle vom SchmelzpunKt 182°C. Diese
Verbindung ist in Wasser löslich, im Äthanol wenig löslich und in Kohlenwasserstoffen unlöslich.
Beispiel 2

Der in Beispiel 1 beschriebene Versuch wird wiederholt, jedoch unter Verwendung von 17,4 g
(0,2 Mol) Morpholin anstelle von Diethylamin. Hierbei
werden 10,8 g weiße Kristalle (Ausbeute 68,4%) vom Schmelzpunkt 180 bis 1830C nach Umkristallisation aus
einem Pflanzenkohle enthaltenden siedenden Gemisch von 100 ml Isopropanol und 100 ml Äthanol erhalten.
Das Hydrochlorid ist in Wasser und Äthanol löslich
und in Kohlenwasserstoffen unlöslich.
Die Eigenschaften der vorstehend beschriebenen verschiedenen Produkte wurden in pharmakologischen
Versuchen ermittelt.
Pharmakologische Angaben für die Verbindungen

Pharmakologische Versuche haben ergeben, daß die Verbindung nach Beispiel 2 spasmolytisch ist und eine
stark sedierende und choleretische Wirkung hat. Die DL50 bei der M?,us bei intravenöser Verabreichung
beträgt bei der Verbindung nach Beispiel 1 99 mg/kg und bei der Verbindung nach Beispiel 2 420 mg/kg. Bei
der Maus, die Dosen von 50 mg/kg der Verbindung nach Beispiel 1 intramuskulär erhalten hat, wird eine
gesteigerte Erregung und gesteigerte Erregbarkeil, insbesondere eine meduläre Erregung bei der Straub-Reaktion
beobachtet. Mäuse, die intramuskuläre Injektionen der Verbindung gemäß Beispiel 2 in einer Dosis
von 210 mg/kg erhielten, zeigten die folgenden Symptome:
Sedierung, Diarrhöe, Abnahme der Atmungsfrequenz und Erweiterung der peripheren Gefäße. Bei
dieser Dosis wurde eine beruhigende und analgetische Wirkung festgestellt Die Verbindung nach Beispiel 1
zeigt eine spasmolytische Wirkung. Am isolierten Duodenum der Ratte bewirkt die Verbindung nach
Beispiel 1 in einer Dosis von 50 Mikrogramm/ml eine leichte Erschlaffung der Organe im Ruhezustand. Die
am isolierten Duodenum der Ratte untersuchte spasmolytische Wirkung der Verbindung gemäß Beispiel
2 ist durch Dekontraktionen von 77% bei einer Dosis von 4 Mikrogramm/ml und von 75% bei einer
Dosis von 40 Mikrogramrn/m! gekennzeichnet, ermittelt an den Organen, deren Kontraktion durch Bariumchlorid
verursacht wurde. Bei der Verbindung gemäß Beispiel 1 liegt die mittlere Dekontraktion gegenüber
Bariumchlorid, ausgedrückt als ED50 unter 50 mg/1 und
gegenüber Acetylcholin bei etwa 100 Mikroeramm/ml. Für die Verbindung gemälS Beispiel 2 liegt bei Organen,
deren Kontraktion mit Acetylcholin bewirkt wird, die spasmolytische Wirkung bei 16% (bei einer Dosis von 4
Mikrogramm/I), bei 32% (bei einer Dosis von 8 Mikrogramm/ml) und bei 95% (bei einer Dosis von 40
Mikrogramm/ml).
Am isolierten Ureter des Meerschweinchens drückt sich die Wirkung gegenüber Bariumchlorid bei der
Verbindung gemäß Beispiel 1 bei einer Dosis von 100 Mikrogramm/ml in einer Abnahme der Peristaltik um
30% an vier Organen und eine Blockierung an vier anderen Organen aus. Eine Dosis von 1 mg/ml blockiert
die Peristaltik vollständig. Bei der Verbindung gemäß Beispiel 2 hat eine Dosis von 420 Mikrogramm/ml am
isolierten Ureter des Meerschweinchens keine Wirkung auf spontane Kontraktionen. Sie verringert die Wirkung
von Barium um 20%, während eine Dosis von 840 Mikrogramm/ml sie um etwa 40% verringert.
Am Ileum des Meerschweinchens in situ wird bei
einer Dosis von 10 mg/kg i. v. der Verbindung gemäß Beispiel I festgestellt, daß der peristaltische Index um
57% abnimmt, jedoch führt das Produkt zu einer Hypotension, die von 40 bis 72% variiert. Durch die
Verbindung gemäß Beispiel 2 wird die Peristaltik des lleums bei sämtlichen drei Versuchstieren bei einer
Dosis von 42 mg/kg i. v. blockiert. Gleichzeitig wird eine
Hypotension von 30 bis 50% festgestellt.
Die Verbindung gemäß Beispiel 1 ist bei intravenöser Verabfolgung in einer Dosis von 10 mg/kg bei der
anästhesierten Ratte hypocholeretisch. Die Gallenabsonderung steigt maximal um 35% im Verlauf von
30 min.
Die Verbindung gemäß Beispiel 1 hat ferner nur eine sehr gespasmolytische Wirkung auf den Ureter des
anästhesierten Hundes. Bei intravenöser Verabreichung bewirkt sie Pratykardie beim anästhesierten Hund, bei
der anästhesierten Katze und beim wachen Kaninchen. Beim anästhesierten Hund hat sie praktisch keine
erweiterte Wirkung auf die peripheren Gefäße.
Die Verbindung gemäß Beispiel 2 hai weiterhin die folgenden Wirkungen:
Am Sphinkter Oddi des anästhesierten Hundes hat
die Verbindung bei einer Dosis von 42 mg/kg i. v. eine intestinale Spasmolytische Wirkung für 10 bis 30 min
und eine schnell eintretende starke spasmolytische Wirkung auf den Sphinkter Oddi (effet antispasmodique
oddien).
Am Uterus in situ bei der Ratte im östrus verringert
eine intravenös injizierte Dosis von 42 mg/kg die Spontanperistaltik des Uterus für 8 bis 18 min im
Durchschnitt um 39%.
Die Untersuchung der Cholerese zeigte, daß das Produkt bei der anästhesierten Ratte bei einer Dosis
von 42 mg/kg eine starke Hypercholerese während mehr als 2 h hervorruft.
Die örtliche vasodilatatorische Wirkung nach Injektion in die Arterien ist verhältnismäßig stark. Das
Produkt ist lOmal weniger wirksam als Papaverin. Die
Wirkungsdauer ist vergleichbar.
Das Produkt hat eine beruhigende Wirkung und verstärkt den mit Chloralhydrat oder Hexobarbital
hervorgerufenen Schiauf. Außerdem ist bei der Maus die Verringerung der Spontanmodiiität bei einer Dosis
von 105 mg/kg i. m. (63% nach 15 min, 70% nach 20 min) mit der Wirkung von 300 mg/kg i. m. Meprobamat
vergleichbar.
Die Verbindung hat in einer Dosis von 84 mg/kc i. m.
bei der Maus (d. h. bei 'Λ der LD50 i, v.) eine sedierende
Wirkung und eine analgetische Wirkung vom Typ des Ϊ Aspirins.
Beim Menschen erwies sich das Hydrochlcrid von (Dimethoxy-3,5-morpholin-acetanilid als starkes Spasmolytikum,
insbesondere bei der Behandlung von Gallenschmerzen. Es ist sehr gut verträglich.
