Die Erfinderung betrifft ein Verfahren zur Herstel^nt
allgemeine« neuer Isoxazolo-^ 5,4-b_/-pyridine, der folgenden"'Formel

niedere Alkoxyphenyl- oder Haiogenphenylgruppe bedeutet, R1 die gleiche Bedeutung wie R hat oder Wasserstoff
ist, und 5,6-Polymethylenisoxazolo-£~5j4-b7-pyridine
der folgenden Formel:

Alkoxyphenyl- oder Halogenphenylgruppe und η eine ganze Zahl von 3 bis 6 bedeutet sowie deren Säureadditiorissalze,
in der vorliegenden Anmeldung bedeutet der Begriff
■•niedere Alkylgruppe" eine Alkylgruppe mit 1 bis 4 Kohlenstoffatomen, beispielsweise eine Methyl-, Äthyl-,
Propyl- oder Butylgruppe sowie deren isomere Formen. Die • Bezeichnung "Halogenphenylgruppe" bedeutet eine halogensubstituierte
Phenylgruppe, wobei das Halogenatom beispielsweise Chlor, Brom, Jod oder Fluor ist. Mit dem
Begtiff "niedere Alköxyphenylgruppe" wird eine mit einer
oder mehreren Alkoxygruppen mit 1 bis 4 Kohlenstoffatomen substituierte Phenylgrmppe bezeichnet, z.B. eine
Methoxyphenyl-, Trimethoxyphenyl-, Äthoxyphenyl-, Propoxyphenyl-,
Butoxyphenyl-, oder Dibutoxyphenylgruppe sowie deren isomere Formen. Die Bezeichnung "niedere
Alkylphenylgruppe" bedeutet eine mit einem oder mehereren
Alkylgruppen mit 1 bis 4 Kohlenstoffatomen substituierte Phenylgruppe, beispielsweise eine Tolyl-, Xylyl-, Trimethylphenyl—,
Äthylphenyl-, oder Butylphenylgruppe sowie deren isomere Formen.
Die erfindungsgemäß herstellbaren Isoxazolo-/~5,4-b_7-pyridine
der Formeln I und Ia bestehen entweder in der nichtprotonierten Form (freie Base) oder in der protonierten
Form (Säureadditionssalz), je nach dem pH-Wert des umgebenden Mediums. Beim Neutralisieren mit geeigneten
Säuren, beispielsweise Salzsäure, Brom—wasserstoffsäure, Schwefelsäure, Phosphorsäure, Salpetersäure,
Essigsäure, Benzoesäure, Salizylsäure, Glykolsäure, Bernsteinsäure,
Nikotinsäure, Weinsäure, Maleinsäure, Apfelsäure, Pamoesäure, Methansulfonsäure, Cyclohexansulfamidsäure,
Pikrinsäure oder Milchsäure bilden sie beständige Protonate, d.h. Säureadditionssalze. Diese Säureadditionssalze
eignen sich zur Verbesserung der freien

Basen. Die freien Basen sind als Säureakzeptoreii geeignet
indem sie unerwünschte Azidität neutralisieren oder eine
Säure, die sich während einer chemischen Umsetzung bildet, beispielsweise während einer Dehydrohalogenierung,
in der Wasserstoff und Chlor, Brom oder Jod aus den "benachbarten Kohlenstoffatomen entfernt werden, absorbieren.
Die erfindungsgemäß herstellbaren Verbindungen der Formeln I und Ia bilden mit Fluorkieselsäure Salze, die als
Mottenschutzmittel gemäß den USA—Patentschriften 1 915
334 und 2 075 3 59 geeignet sind. Sie bilden ferner Salze mit Thiozyaftsäure, die mit Formaldehyd unter Bildung
harzartiger Stoffe kondensieren und als Beizhemmstoffe
gemäß den USA-Patentschriften 2 425 320 und 2 6o6 155
geeignet: sind» Sie bilden außerdem Salze mit Trichloressigsäure,
die als Herbizide geeignet sind, beispielsweise gegen Johnson-Gras, Setaria glauca, Setaria νΐατά-dis,
Bermuda—Gras und Quecken.
Die Isoxazolo£~5t4-b_7-pyridine der Formel I werden dadurch
hergestellt, daß man das bekannte 5—Amino-3—methylisoxazol
der Formel

in Gegenwart eines Säurekatalysators behandelt.
5-Amino~3-*-methylisoxazol (II) ist in der Technik bekannt.
Bs läßt sich durch Umsetzen von 3-Iminobutyronitril mit
Hydroxylamin sowie nach dem in Ann,, Bd. 624, 3. 22
(1959) beschriebenen Verfahren herstellen.
Die Dike tone der Formel III sind in der Technik gut be-*
kannt, und eine große Vielzahl derselben wurden nach bekannten Verfahren hergestellt, z.B. gemäß Organic Syntheses,
Bd. 20, S. 32 (1940) und Band VIII, S. 133 von
"Organic Reactions", John Wiley and Sons, Inc. New York, 1954. Zu den bekannten Diketonen, die sich bei dem erfindungsgemäöen
Verfahren verwenden lassen, gehören 1,3—
Diphenyl-1,3-propandion; 1 , 3-di-(p-Tolyl) 1 ,3-pr>opandion;
1,3-di-(m-Chlorphenyl)-1,3-propandion; 1,3-di-{p-Chlorphenyl)-1,3-propandion;
1,3-di-(p-Methoxyphenyl)—1,3-propandion;
1,3-di-(p-Bromphenyl)-1,3-propandion; 2,4-Pentandion;
3-Methyl-2,4-pentandion; 3-Äthyl-2,4-pentandion;
3-Butyl-2,4-pentandion; 3-Phenyl-2,4-pentandioni
3-Isopropyl-2,4-pentandion, oder-3-Isobutyl-2,4-pentandion.

Zu den geeigneten Katalysatoren gehören beispielsweise konzentrierte Schwefelsäure, Phosphorpentoxyd, Phosphorpen
tachlorid oder Polyphosphorsäure, wobei die
letztere am meisten bevorzugt wird.
Bei der Durchführung der Umsetzung zwischen 5-Amino-3-methylisoxazol
(II) und dem Diketon der Formel III, werden die beiden Reaktionsteilnehmer mit dem Säurekatalysator
gemischt und das Gemisch erhitzt, z.B. auf etwa 50 C bis etwa 175°C, vorzugsweise zwischen etwa 100 C und
etwa 140 C. Inerte Lösungsmittel sind nicht erforderlich, können jedoch gegebenenfalls verwendet werden. Das Mol—
verhältnis der Verbindungen der Formel II und Formel III
kann schwanken, jedoch erwiesen sich äquimolare Mengen d.h* im Verhältnis von etwa 1:1 zufriedenstellend. Die
Umsetzungszeit hängt von Faktoren, wie beispielsweise der Umsetzungstemperatur, den jeweils verwendeten Reak—
tionsteilnehmern, der relativen Menge der Reaktionsteilnehmer,
sorgfältiges Mischen u. dgl. ab. Die optimale Umsetzungszeit ist deshalb für jede Gruppe von Reaktions—
bedingungen verschieden. Gewöhnlich sind Umsetzungszeiten von etwa 10 Minuten bis etwa 1 Stunde geeignet. Nach Beendigung
der Umsetzung wird das Reaktionsgemisch mit Wasser verdünnt und mit einer Base (z.B. Ammoniumhydroxyd)
neutralisieret. Das auf diese Weise erhaltene Isoxazolo- ^"S^-b^-pyridin der Formel I wird in Form seiner freien
Base aus dem Reaktionsgemisch isoliert, wobei Übliche Verfahren angewandt werden, wie z.B. Filtration, Lösungsmittelverdampfung,
Lösungsmittelextraktion, Chromatographie oder Kristallisation oder eine Kombination
dieser Verfahren. Jede der so erhaltenen freien Basen kann beispielsweise durch Umkristallisation aus einem
geeigneten Lösungsmittel oder Lösungsmittel^paar gereinigt
werden. Die freie Base kann in jedes gewünschte Säureadditionssalz durch Neutralisation mit einer Säure,
z.B. einer der vorstehend aufgeführten, übergeführt werden. Die 5,6-Polymethylenisoxazolo-^_ 5»4-b_7~pyridine
der Formel Ia werden dadurch hergestellt, daß man

eine Benzoyl- oder substituierte Benzoylgruppe ist, von denen viele bekannt sind, können nach den in Ann., Bd.
650, S. 133 - 156 (1961); J.Amer.Chem. Soc., Bd. 82,
S. 2389 - 2392 (i960)? J.Org.Chem., Bd. 28, S. 379 383
(1963) beschriebenen Verfahren hergestellt werden.

Zu den bekannten 2—Benzocycloalkanonen, die bei dem neuen
Verfahren verwendbar sind, gehören 2-(p-Methylbenzoyl)-cyclopentanon,
2-(p-Methpxybenzoyl)-cyclopentanon, 2-(p-Jodbenzoyl)-cyclopentanon, 2—Benzoylcyclohexanon,
2-(m-Chlorbenzoyl)-cyclohexanon, 2-(p-Fluorbenzoyl)-cyclohexanon,
2-(p-Methoxybenzoyl)-cyclohexanon, 2-Benzoylcycloheptanon
oder 2-Benzoylcyclooctanon.
Zu den geeigneten Säurekatalysatoren gehören die gleichen,
die bei der Herstellung der Verbindungen der Formel I verwendbar sind und vorstehend aufgeführt sind.
Zur Herstellung der 5,6-Polyraethyl eni soxazol ο—[_ 5,4-pyridine
der Formel Ia und ihrer Säureadditionssalze durch Behandlung von 5-Amino-3-methylisoxazol (II) mit
einem entsprechenden 2—Benzoyl-cycloalRanon (lila) in
Gegenwart eines Säurekatalysators werden die gleichen
Verfahren unter den gleichen Bedingungen wie bei der
Synthese der vorstehend beschriebenen Verbindungen der Formel I angewendet..Der einzige Unterschied besteht darin,
daß- anstelle der Diketone der" Formel III 2—Benzoyl-

Die verfahrensgemäß herstellbaren ¥erbindungen der Formel
I und Ia in Form ihrer freien Base sowie ihrer Säure"
additionssalze mit pharmakologisch verträglichen Säuren,
beispielsweise Salzsäure, Bromwasserstoffsäure, Schwefelsäure,
Phosphorsäure, Salpetersäure, Essigsäure, Benzoesäure, Salizylsäure, Slykolsäure, Bernsteinsäure, Nikotinsäure, Weinsäure, Maleinsäure, Apfelsäure,-Milchsäure,
Methansulfonsäure und Cyclohexansulfamidsäure «eisen eine
biologische Wirksamkeit auf» Die Verbindungen der Formel
I besitzen fungizide Eigenschaften. Sie hemmen beispielsweise das Wachstum des Stammes Trichophyton rubrura und
lassen sieh zur Behandlung von Infektionen, die dieser
Mikroorganismus bei Mensehen, Säugetieren und Tieren, z.B. Katzen und Hunden, hervorruft, verwenden«. Diese Verbindungen sind ferner als Herbizide wirksam und können
bei der Bekämpfung von lluthirse, Quecken, Ackerwinden, Bogenamarant sowie Sängefuß verwendet werden. Die Verbindungen
der Formel Ia besitzen eine entzündungshemmende Wirkung; sie könnön ^ei der Behandlung von Entzündungserscheinungen
der Haut, der Augen sowie der Atemwege
von Säugetieren und Tieren, z-.B» Mäusen, Ratten und
Vpgeln, verwendet werden.
Die nachfolgenden Beispiele dienen der weiteren Erläuterung
der vorliegenden Erfindung,
3,4,6-Trimethylisoxazol-^5,4-b_7-pyridin (I),
Ein Gemisch aus 98 g (1 Mol) S-Amino-S-methylisoxazol
(II) und 110 cem (1,07 Mol) Acetylaceton (auch 2,4-Pentandion
genannt) in etwa 150 ecm Polyphosphorsäure
wurde auf 30 C erwärmt und dann schnell in einem Eisbad
gekühlt. Die Temperatur des Reaktionsgemischs stieg
trotz äußeren Kühlens schnell auf 140°G» Is wurde bei
dieser Temperatur etwa, 15 Minuten gerührt und dann auf
Raumtemperatur gekühlt., Anschließend wurden unter Kühlen 500 ecm Wasser und konzentriertes Ammoniumhydr©xpj so
lange zugegeben, bis der pH-Wert gleich 8 war. Das 0emseli
wurde auf f5OC gekühlt und filtriert. Der Feststoff

wurde aus 800 ecm eines Gemischs aus gleichen Teilen
Isopropanol und Wasser umkristallisiert und ergab 105 g (65%-ige Ausbeute) 3,4,6-TrimethylisOxazolo-/~5,4-b_7-pyridin
(i). Schmelzpunkt: 92 bis 93°C

Berechnet für C9H10N2O; C » 66,65; H - 6,22; N - 17,27.
Gefunden: 1= C = 66,62;H » 6,26; N =17,26.
Infrarot- und kernmagnetisches Resonanzspektrum bestätigten
die Struktur der so erhaltenen Verbindung.
Durch Zugabe einer Diäthylätherlösung von Salzsäure, Bromwasserstoffsäure, Schwefelsäure, Phosphorsäure, Essigsäure,
Benzoesäure, Salyzilsäure, Zitronensäure, Bernsteinsäure, Apfelsäure, Cyclohexansulfamidsäure, Thiocyansäure
oder Trichloressigsäure zu einer Äthanollösung von 3,4T6-Trimethylisoxazolo-/~5,4-b__7-pyridin und anschließende
Zugabe von etwa 4 Volumen-Teilen Diäthyläther erhält man das entsprechende Säureadditionssalz.
3,4,6-Trimethyl-5-phenylisoxazolo-/f*5,4-b_7~pyridin (I).
Ein Gemisch aus 9,8 g (0,1 Mol) 5-Amino-3-methylisoxazol (II) und 20 g (0,11 Mol) 3-Phenyl-2,4-pentandion in
etwa 25 ecm Polyphosphorsäure wurde langsam auf etwa 1000C erwärmt, wobei an diesem Punkt ein plötzlicher
Temperaturanstieg auf etwa 155 C erfolgte. Das Reaktionsgemisch wurde eine halbe Stunde lang auf I40 bis 16O0C
erhitzt und dann gekühlt. Anschließend wurden 200 ecm
Wasser und konzentriertes Ammoniumhydroxyd zugegeben, bis das Reaktionsgemisch neutral war. Der braune Feststoff
wurde abfiltriert, zweimal aus Äthanol umkristallisiert und zum Entfärben mit Holzkohle behandelt. Man
erhielt 12,5 g=(53%ige Ausbeute) 3,4,6-Trimethyl-5-phenylisoxazolo-/~5»4-b__7-pyrldin
(i). Schmelzpunkt: 208,5 bis 210°C.

nollösung von 3,4,6-Trimethyl-5—phenylisöxazolo-
^~5,4~b>_7-pyridin und anschließende Zugabe von etwa
Volumen—Teilen Diäthyläther erhält man das entsprechende Säureadditxonssalz.
'Beispiel 3
3-Methyl-4,6-diphenylisoxazolo-/^5,4-b_7~pyridin (I).
Ein Gemisch aus 9,8 g (0,1 Mol) 5-Amino-3-methylisoxazol (II) und 22,5 g (0,1 Mol) 1,3-Diphenyl-1,3-propandion
in etwa 2 5 ecm Polyphosphorsäure wurde langsam auf
140°G erhitzt. Danach stieg die Temperatur schnell auf 16O°C. Das Reaktionsgemisch wurde bei dieser Temperatur Jk
20 Minuten lang gehalten, gekühlt, und eine große Menge Wasser wurde zugegeben. Anschließend wurde konzentriertes
Ammoniumhydroxyd zugegeben, bis das Reafctionsgemisch
neutral war. Der Feststoff wurde abfiltüert und verworfen.
Das Filtrat wurde mit Äther extrahiert, die Ätherschicht getrocknet und unter Vakuum eingeengt. Der Rückstand
wurde aus' 250 ecm Isopropanol umkristallisiert
und ergab 2,7 g (10 %ige Ausbeute) 3-Methyl-4,6,-diphenylisoxazolo-/~5,4-b_7-pyridin
(I)· Schmelzpunkts 157 bis 158,5°C.

Durch Zugabe einer Diäthylätherlösung von Salzsäure, Bromwasserstoffsäure, Schwefelsäure, Phosphorsäure,

Thiocyansäure oder Trichloressigsäure zu einer Äthanollösung eines im vorstehenden Abschnitt beschriebenen
Isoxazolo-^~5,4-b__7-pyridin und anschließende Zugabe
von etwa 4 Volumen-Teilen Diäthyläther erhält man das entsprechende Säureadditionssalz.
4-(p-Methoxyphenyl)-3-methyl-5,6-tetramethylenisoxazolo-/~5,4-b_7-pyridin
(Ia).
Ein Gemisch aus 9,8 g (θ,1 Mol) S-Amino-S-methylisoxazol
(II) und 23,2 g (0,1 Mol) 2-(p-Methoxybenzoyl)-eyclohexanon in etwa 2 5 ecm Polyphoshorsäure wurde auf
etwa 70 C erhitzt, wobei an diesem Punkt ein rascher Temperaturanstieg erfolgte. Das Reaktionsgemisch wurde
gekühlt, wiederum auf etwa 120 C erhitzt, wobei die Temperatur erneut schnell anstieg. Das Reaktionsgemisch
wurde gekühlt, dann eine halbe Stunde lang auf 130° bis 1500G erhitzt, wieder gekühlt, und anschließend
wurden 2 50 ecm Wasser und konzentriertes Ammoniumhydroxyd so lange zugegeben, bis es leicht alkalisch war.
Es wurde 1 Liter Wasser zugegeben und dann dreimal mit Chloroform extrahiert. Die organische Schicht wurde getrocknet
und zur Trockne eingeengt. Der Rückstand wurde dreimal aus Isopropanol umkristallisiert (Behandlung
mit Holzkohle zum Entfärben) und ergab 6,5 g (22 %±gs
Ausbeute) weißes 4-(p-Methoxyphenyl)-3-methyl-5,6-tetramethylenisoxazolo-^~5,4-b_/-pyridin
(la). Schmelzpunkt: 160,5 bis 162,50C.
Analyse;
Berechnet für C18H18N2O2: C - 73,45? H - 6,16} N- 9,52.
Gefunden: ' C - 73,44; H - 6,49j N - 9,39.

- Patentanspruch;Verfahren zur Herstellung von Isoxazol j__ 5»4-b_7-pyridinen der allgemeinen Formel
