Description
Gegenstand des Patents 16 42 062.5 sind antimikrobielle Mittel mit einem Gehalt an 2-Brom-nitrobutyl-N-phenyl-carbamat
und 2-Brom-2-nitrobutyI-N-(3,4-dichlorphenyl)-carbamat in den Gewichtsverhältnissen
1 :4 bis 4 :1, insbesondere 1:1.
Diese synergistische Kombination stellt ein besonders wirksames Mittel sowohl gegen grampositive und
gramnegative Bakterien als auch gegen Hefen und Pilze dar, mit einem wesentlich verbesserten Wirkungsspektrum
gegenüber den einzelnen Nitroalkyl-N-phenylcarbamaten.
Wenn auch die abtötende Wirkung dieser synergistischen Kombination bereits in sehr niedrigen
Konzentrationen gewährleistet ist, so erfordert sie doch in einigen Fällen längere Zeiträume, die bei manchen
Anwendungsarten nicht vorhanden sind.
Der Erfindung liegt daher die Aufgabe zugrunde, die Abtötungswirksamkeit dieser antimikrobiellen Mittel so
zu verbessern, daß auch innerhalb kürzester Zeiten eine sichere Abtötung von Mikroorganismen gewährleistet
ist.
Diese Aufgabe wird erfindungsgemäß dadurch gelöst, daß man antimikrobielle Mittel auf Basis einer
synergistischen Kombination von 2-Brom-2-nitrobutyl-N-phenyl-carbamat
und 2-Brom-2-nitrobutyl-N-(3,4-dichlorphenyl)-carbamat in den Gewichtsverhältnissen
1 :4 bis 4 :1, insbesondere 1:1 mit einem Gehalt an Dimethylsulfoxid und Äthanol oder Isopropanol verwendet.
Die Herstellung der dem erfindungsgemäßen antimikrobiellen Mittel zugrundeliegenden substituierten
Nitroalkal-N-phenylcarbamate kann in an sich bekannter
Weise durch Anlagerung von 2-Brom-2-nitrobutanol-(3) an Phenylisocyanat bzw. 3,4-Dichlorphenylisocyanat
erfolgen. Zu diesem Zweck werden 0,1 Mol des Nitroalkohols mit 0,12 Mol des Isocyanats in wasserfreiem
Benzol 4-5 Stunden unter Rückfluß erhitzt. Nach Abdestillieren der Hauptmenge des Benzols kann das
substituierte Nitroalkyl-N-phenylcarbamat isoliert und
aus einem Lösungsmittelgemisch Benzol/Benzin 1 :1 umkristallisiert werden.
Durch die gleichzeitige Anwesenheit von Äthanol oder Isopropanol und Dimethylsulfoxid neben der
synergistischen Kombination von 2-Brom-2-nitrobutyl-N-phenylcarbamat
und 2-Brom-2-nitrobutyl-N-(3,4-dic!ilorphenyl)-carbamat in den erfindungsgemäßen
antimikrobiellen Mitteln wird deren Abtötungswirksamkeit überraschenderweise ganz wesentlich verbessert.
Es werden mit den erfindungsgemäßen Mitteln Abtötungszeiten erreicht, die den Einsatz der synergistischen
Kombination der substituierten Nitroalkyl-N-phenylcarbamate
auch in antimikrobiellen Mitteln mit nur kurzen Anwendungszeiten angezeigt erscheinen
lassen, in denen sie allein nicht ausreichend wirksam wäre.
Der Gehalt der antimikrobiellen Mittel an der Wirkstoffkombination aus 2-Brom-2-nitrobutyl-N-phenylcarbamat
und 2-Brom-2-nitrobutyl-N-(3,4-dichlorphenyl)-carbamat in den Gewichtsverhältnissen 1 :4 bis
4:1, insbesondere 1 :1 beträgt 0,05—5 Gew.-%,
vorzugsweise 0,1 — 1 Gew.-%.
Der Gehalt der antimikrobiellen Mittel an Äthanol oder Isopropanol beträgt 5—30 Gew.-°/o, vorzugsweise
10—20 Gew.-% und Dimethylsulfoxid soll in ihnen in einer Menge von 10—50 Gew.-%, vorzugsweise 20—40
Gew.-% enthalten sein.
In Präparationen, in denen ein Zusatz von Komplexbildnern angebracht ist, können die erfindungsgemäßen
antimikrobiellen Mehrstoffkombinationen auch mit Komplexbildnern, die im Hampshire-Test ein größeres
Calciumcarbonat-Bindevermögen als 230 mg je g Komplexbildner aufweisen, kombiniert werden. Hierdurch
ist eine weitere Wirkungssteigerung möglich.
Die antimikrobiellen Mittel aus einer synergistischen Kombination von 2-Brom-2-nitrobutyl-N-phenylcarbamat
und 2-Brom-2-nitrobutyl-N-(3,4-dichlorphenyl)-carbamat in den Gewichtsverhältnissen 1 :4 bis
4:1, insbesondere 1:1, Dimethylsulfoxid und Äthanol oder Isopropanol können als Reinigungs- und Desinfektionsmittel
für Hände und Instrumente sowie als antimikrobielle Tinkturen und Sprays zur Bekämpfung
4i) von Hauptpilzen und dergleichen Verwendung finden.
Durch die nachstehenden Beispiele wird der Gegenstand der Erfindung näher erläutert.
Die Abtötungskonzentrationen und -zeiten der Einzelsubstanzen sowie der Kombinationen wurden mit
Hilfe des halbquantitativen Suspensionstestes ermittelt. Dieser Test stellt eine abgewandelte Ausführungsform
des in den Richtlinien für die Prüfung chemischer Desinfektionsmittel der Deutschen Gesellschaft für
Hygiene und Mikrobiologie (1959) angegebenen Suspensionsversuches zur Feststellung der bakteriziden,
fungiziden und sporiziden Wirkung dar.
0,1 ml derKeimsuspension wurden bei Zimmertemperatur von 18—210C in kleine Petrischalen pipettiert.

Wie den vorstehenden Tabellen zu entnehmen ist, läßt sich weder durch die alkoholische Komponente
noch durch Dimethylsulfoxid (DMSO) noch durch die Kombination von 2-Brom-2-nitrobutyl-N-phenylcarbamat
und 2-Brom-2-nitrobutyl-N-(3,4-dichlorphenyl)-carbamat in den verschiedenen Gewichtsverhältnissen
bei alleiniger Verwendung eine Abtötung der
Testkeime in kurzer Zeit erreichen. Auch die Kombination zweier Komponenten bringt keine nennenswerte
Änderung der Testergebnisse. Erst bei dem erfindungsgemäßen Einsatz aller drei Komponenten, wobei die
Kombination von 2-Brom-nitrobutyl-N-phenylcarb- r>
amat und 2-Brom-2-nitrobutyi-N-(3,4-dichlorphenyl)-carbamat in den verschiedenen Gewichtsverhältnissen
als eine Komponente zu betrachten ist, werden die Abtötungszeiten überraschenderweise äußerst stark
reduziert. ι ο
Nachstehend werden Zusammensetzungen für einige antimikrobielle Mittel angegeben.

Der mit den erfindungsgemäßen Mitteln erzielbare
Vorteil besteht darin, daß man mit ihnen innerhalt
r> kürzester Zeiten eine Abtötung von grampositiven und
gramnegativen Bakterien, sowie von Hefen und Pilzen erreicht.
Claims (4)
1. Antimikrobielle Mittel nach Patent 16 42 062.5 auf Basis einer synergistischen Kombination von
2-Brom-2-nitrobutyl-N-phenylcarbamat und 2-Brom-2-nitrobutyl-N-(3,4-dichlorphenyl)-carbamat
in den Gewichtsverhältnissen 1 :4 bis 4 :1, insbesondere
1:1, gekennzeichnet durch einen Gehalt an Dimethylsulfoxid und Äthanol oder
Isopropanol.
2. Antimikrobielle Mittel nach Anspruch 1, dadurch gekennzeichnet, daß sie die Kombination
von 2-Brom-2-nitrobutyl-N-phenylcarbamat und
2-Brom-2-nitrobutyl-N-(3,4-dichlorphenyl)-carbamat in einer Menge von 0,05—5 Gew.-%,
vorzugsweise 0,1 —1,0 Gew.-% enthalten.
3. Antimikrobielle Mittel nach Anspruch 1 und 2, dadurch gekennzeichnet, daß sie Äthanol oder
Isopropanol in einer Menge von 5—30 Gew.-°/o, vorzugsweise 10—20 Gew.-% enthalten.
4. Antimikrobielle Mittel nach Anspruch 1 bis 3, dadurch gekennzeichnet, daß sie Dimethylsulfoxid in
einer Menge von 10—50 Gew.-%, vorzugsweise 20—40 Gew.-% enthalten.
