Description
Die Erfindung betrifft neue 6,14-Endoäthen-tetrahydrooripavinderivate und ihre Salze der allgemeinen
Formel I
N-CH,

HO-C — R1
in der R1 ein ,Wasserstoffatom oder einen Alkylrest
mit 1 bis 3 Kohlenstoffatomen und R2 ein Wasserstoffatom, einen Alkylrest mit 1 bis 5 Kohlenstoffatomen,
eine Phenäthyl- oder Cyclohexylgruppe bedeutet.
Die Erfindung betrifft ferner ein Verfahren zur Herstellung der Verbindungen der allgemeinen Formel
I, das dadurch gekennzeichnet ist, daß man in an sich bekannter Weise ein 6,14-Endoäthen-tetrahydrothebainderivat
der allgemeinen Formel II

in der R1 und R2 die obige Bedeutung haben, mit
einem Alkalimetallhydroxid in einem Lösungsmittel bei Temperaturen über 200° C behandelt und gegebenenfalls
die erhaltene freie Base in ein Salz überführt.
Eine bevorzugte Ausführungsform des erfindungsr. gemäßen Verfahrens besteht darin, daß man die
Umsetzung in Diäthylenglykol als Lösungsmittel durchführt.
Die Verbindungen der Erfindung zeigen blutdrucksenkende, analgetische, sedative und hustenstillende
Wirkungen.
In ihrer analgetischen Wirkung sind sie dem bekannten, analgetisch wirksamen Dihydrodesoxymorphin sowie Methyldihydromorphinon erheblich überlegen. Die Toxizitäten liegen dabei nahezu in der gleichen Größenordnung.

Die ausgezeichneten therapeutischen Eigenschaften der neuen Verbindungen im Vergleich zu der bekannten
Verbindung Dihydrodesoxymorphin ergeben sich aus der nachstehenden Tabelle. Die analgetischen
Wirkungen wurden an Ratten mittels der bekannten Schwanz-Druck-Methode (vergleiche z. B. Green
und Young, Brit. J. Pharmacol. (1951), S. 572)', bestimmt.

6,14-Endoäthen-7a-(l -hydroxy-1 -äthyl)-tetrahydrooripavin
8 Teile 6,14-Endoäthen-7a-(l-hydroxy-l-äthyl)-tetrahydrothebain
werden in eine Lösung von 60Teilen Kaliumhydroxid in 150 Teilen Diäthylenglykol eingetragen.
Die Lösung wird unter Rückfluß auf 200 bis 210°C erhitzt. Das Gemisch wird unter kräftigem
Rühren so lange unter Rückfluß gekocht, bis eine Probe nach dem Verdünnen mit der lOfachen Volümenmenge
Wasser eine homogene Lösung ergibt. Das Reaktionsgemisch wird dann mit der 5fachen
Volumenmenge Wasser verdünnt und so lange mit gesättigter Ammoniumchloridlösung versetzt, bis sich
kein Niederschlag mehr bildet. Der ausgefällte Niederschlag wird abfiltriert, gut mit Wasser gewaschen
und aus 2-Äthoxyäthanol umkristallisiert. Man erhält die freie Base in Form von weißen Prismen, F. 2980C.

Durch Behandeln der Base mit einer Lösung von Chlorwasserstoff in Äthanol und Zusatz von Diäthyläther
wird das Hydrochlorid in Form weißer Prismen (F. 2720C) erhalten.
C22H27O4N-HCl-V2H2O.
6,14-Endoäthen-7-(2-hydroxy-2-propyl)-tetrahydrooripavin
Diese Verbindung wird gemäß Beispiel 1 aus 6,14 - Endoäthen - 7 - (2 - hydroxy - 2 - propyl) - tetrahy drothebain
hergestellt. Die freie Base wird mit Äther extrahiert, der Ätherextrakt getrocknet, eingedampft
und der feste Rückstand aus wäßrigem 2-Äthoxyäthanol umkristallisiert. Man erhält die Base in Form
weißer Prismen, F. 266° C

Das Hydrochlorid schmilzt bei 288 bis 290° C (Zersetzung). Das Hydrochlorid des 3-Acetats schmilzt
bei 226° C.

Diese Verbindung wird gemäß Beispiel 1 aus 6,14-Endoäthen-7a-(2-hydroxy-2-butyl)-tetrahydrothebain
hergestellt. Nach Umkristallisation aus 2-Äthoxyäthanol werden weiße Prismen (F. 268° C) erhalten.

Diese Verbindung wird gemäß Beispiel 1 aus 6,14-Endoäthen-7a-(2-hydroxy-2-pentyl)-tetrahydrothebain
hergestellt. Die freie Base fällt 'nach Umkristallisation aus wäßrigem 2-Äthoxyäthanol in Form
weißer Prismen (F. 215° C) an

hergestellt. Die freie Base fällt nach Umkristallisation aus 2-Äthoxyäthanol in Form blaßweißer Prismen (F. 28O0C) an. ■

Diese Verbindung wird gemäß Beispiel 1 aus 6,14 - Endoäthen - 7 - (3 - hydroxy - 3 - pentyl) - tetrahydrothebain
hergestellt. Die Base fällt nach Umkristallisation aus wäßrigem 2-Äthoxyäthariöl in Form weißer
Prismen (F. 157° C) an.

Diese Verbindung wird gemäß Beispiel 1 aus 6,14-Endoäthen-7a-(2- hydroxy - 3 - methyl -2-butyl)-tetrahydrothebain
hergestellt. Nach Umkristallisation aus wäßrigem 2-Äthoxyäthanol fällt die freie Base in
Form weißer Prismen (F. 278° C) an.
Das Hydrochlorid schmilzt bei 296° C.

hergestellt. Sie fällt nach Umkristallisation aus wäßrigem 2-Äthoxyäthanol in Form weißer Prismen
(F. 1200C) an.

hergestellt. Sie fällt nach einer Zwischenreinigung über das in alkoholischer Lösung hergestellte Hydrochlorid
in Form weißer Prismen (F. 218 bis 219° C) an.
Das Hydrochlorid schmilzt bei 298° C.

Verfahren zur Herstellung der Verbindungen nach Anspruch 1, dadurch gekennzeichnet, daß man in an sich bekannter Weise ein 6,14-Endo-äthen-tetrahydrothebainderivat der allgemeinen Formel IICH3 O χN-CH1CH3O'HO —C-R1R2mit einem Alkalimetallhydroxid in einem Lösungsmittel bei Temperaturen über 2000C behandelt und gegebenenfalls die erhaltene freie Base in ein Salz überführt.
