Es ist bereits bekannt. Sultone mit primären, sekundären oder tertiären Aminen zu den entsprechenden
ω-AIkylaminoalkansulfonsäuren umzusetzen.
So erhält man aus Butylamin und Butansulton die N-Butylaminobutansulfonsäure. die als
inneres Ammoniumsalz vorliegt. Da die Aminogruppen mit den Sultonen sehr rasch reagieren,
können diese Reaktionen in Wasser oder in Alkohol als Lösungsmittel durchgeführt werden.
Es wurde nun gefunden, daß man N-(<w-Aminoalkyl)-aminoalkansulfonsäuren
erhält, wenn man aliphatische Diamine mit aliphatischen Sultonen gegebenenfalls in- einem Lösungsmittel bei Temperaturen
von —20 bis H~100°C, vorzugsweise bei —10
bis +4O0C umsetzt.
überraschenderweise reagiert bei diesem Verfahren nur eine Aminogruppe des Diamins mit dem Sulton.
während die zweite Aminogruppe unverändert bleibt.
Die für das Verfahren verwendeten aliphatischen Diamine haben die allgemeine Formel
NH2 — R — NH2
worin R für einen gegebenenfalls substituierten Polymethylenrest steht. Zum Beispiel können verwendet
werden: Äthylendiamin. Propylendiamin-1,3. Tetramethylendiamin-1.4, Hexamethylendiamin-1,6,
Octamethylendiamin-1.8 und ihre an einem oder
mehreren Kohlenstoffatomen ζ. B. durch Alkylreste. wie z. B. Methyl. Äthyl und Propyl. substituierten
Derivate. v>

Die erfindungsgemäß verwendeten aliphatischen Sultone haben die allgemeine Formel

worin R' einen gegebenenfalls substituierten Polymethylenrest bedeutet. Als aliphatische Sultone
kommen insbesondere Propansulton-1,3 und Butansulton-1,4
sowie ihre an einem oder mehreren Kohlenstoffatomen durch Alkylreste wie Methyl. Äthyl
und Propyl substituierten Derivate in Betracht. Die Reaktion erfolgt nach der allgemeinen Formel

Die Reaktion kann in einem Lösungsmittel durchgeführt werden. Als Lösungsmittel sind alle solche
geeignet, in denen sich beide Reaktionspartner lösen, ohne mit dem Lösungsmittel zu reagieren, z. B.
aromatische Kohlenwasserstoffe, wie Benzol, Toluol und Xylol. Halogenkohlenwasserstoffe, wie Methylenchlorid,
Chloroform. Tetrachlorkohlenstoff. Chloroder Dichlorbenzol.
Im allgemeinen werden die Reaktionspartner in äquimolaren Mengen umgesetzt. Man kann aber
auch mit einem größeren Überschuß einer der beiden Komponenten arbeiten, der dann gleichzeitig als
Lösungsmittel wirkt. Ein zusätzliches Lösungsmittel ist in diesem Fall nicht erforderlich. Verwendet man
überschüssiges Sulton als Lösungsmittel, dann muß die Reaktion bei niederen Temperaturen, vorzugsweise
bei etwa 15 bis 400C, durchgeführt werden.
Zur Herstellung der Additionsprodukte kann man das aliphatische Diamin in einem Kohlenwasserstoff
wie Benzol lösen und dann das Sulton auf einmal oder in mehreren Portionen zugeben. Es ist aber auch
möglich, einen größeren Überschuß an Sulton vorzulegen und dann das Diamin auf einmal oder nach
und nach zuzugeben. Das gleiche gilt, wenn man flüssige oder tiefschmelzende aliphatische Diamine
gleichzeitig als Lösungsmittel verwendet, indem man das Diamin, wie z. B. Äthylendiamin. im Überschuß
vorlegt und dann mit dem Sulton reagieren läßt.
Die so durch Additionsreaktion erhaltenen N-(tt)-Aminoalkyl)-aminoalkansulfonsäuren fallen in
ausgezeichneten Ausbeuten als weiße kristallisierte Substanzen an, die in Wasser und zum großen Teil
auch Methanol sehr leicht löslich sind. Sie lassen sich zum Teil aus Äthanol gut Umkristallisieren.

Die definierte Umsetzung nur einer Aminogruppe eines Alkylendiamins mit aliphatischen Sultonen,
auch wenn diese im Überschuß angewandt werden, ist außerordentlich überraschend, da bei vollkommen
gleichartig vorliegenden reaktionsfähigen Gruppen Additionsreaktionen selektiv kaum durchgeführt
werden konnten.
Die erfindungsgemäß erhaltenen Verbindungen sind neu und besitzen zum Teil bakterizide und
lcsungsvermittelnde Eigenschaften.
In 1600 Raumteilen Benzol werden 464 Teile Hexamethylendiamin gelöst und bei 10 bis 15°C unter
Rühren mit 560 Teilen Butansulton versetzt. Nach 40 Stunden wird der entstandene Niederschlag abgesaugt
und aus Äthanol umkristallisiert. Es werden 850S N-(6-Aminohexyl)-4-aminobutansulfonsäure erhalten.
Äquivalentgewicht: titriert 250 (Theorie: 252).

116 Teile Hexamethylendiamin und 544 Teile Butansulton
werden in 900 Raumteilen Benzol unter Kühlung und Rühren vereinigt. Nach 24 Stunden
Stehen bei Raumtemperatur wird abgesaugt und mit Benzol gut ausgewaschen. Die entstandene N-(6-Aminohexy!)-4-aminobutansulfonsäure
wird nach Trocknen aus Äthanol umkristallisiert.
Ausbeute 92%; Äquivalentgewicht: titriert 265 (Theorie: 252).
Unier Rühren und Kühlen auf 20 bis 25 0C werden
116 Teile Hexamethylendiamin und 820 Teile Butansulton zusammengegeben. Nach etwa 3 Stunden ist
die R.eaktionsmischung erstarrt. Man läßt 12 Stunden bei Raumtemperatur stehen und saugt nach Ausrühren
in Essigester ab. Durch Umkristallisieren aus Äthanol erhält man in 70%iger Ausbeute N-(6-Aminohexyl)-4-aminobutansulfonsäure.
Äquivalentgewicht: titriert 248 (Theorie: 252).
Zu einer Lösung von 116 Teilen Hexamethylendiamin in 400 Teilen Toluol werden unter Rühren
bei 00C 125 Teile Propansulton in 100 Teilen Toluol
gelöst eingetropft. Nach Beendigung der Zugabe hält man das Reaktionsgemisch 30 Stunden bei Raumtemperatur
und saugt dann ab. Aus Methanol— Wasser umkristallisiert erhält man 90% N-(6-Aminohexyl)
- 3 - aminopropansulfonsäure. Äquivalentgewicht: titriert 243 (Theorie: 238).
60 Teile Äthylendiamin werden in 600 Raumteilen Benzol gelöst und unter Rühren und Kühlen auf
100C 140 Teile Butansulton portionsweise eingetragen. Nach 30 Stunden wird der erhaltene Niederschlag
abgesaugt und aus Methanol—Wasser umkristallisiert.
N-(2-Aminoäthyl)-4-arninobutan-sulfonsäure wird in 75%iger Ausbeute erhalten. Äquivaientgewicht:
titriert 202 (Theorie: 196).
Unter Kühlen und Rühren werden bei 100C 44 Teile Tetramethylendiamin in 820 Teile Butansulton
eingetragen. Die Reaktionsmischung läßt man nach dem Erstarren noch 12 Stunden bei 25 C stehen
und rührt dann in Essigester aus. Nach dem Umfallen der abgesaugten Kristalle aus Äthanol—Essigester
erhält man 90% N-(4-Aminobutyl)-4-aminobutansulfonsäure. Äquivalentgewicht: titriert 236 (Theorie:
224).
In 700 Raumteile Äthylendiamin werden unter Rühren und Kühlung auf 10 bis 150C 136 Teile
Butansulton eingetropft. Man rührt noch 12 Stunden bei 25 0C und zieht dann das überschüssige Äthylendiamin
bei 40 bis 500C im Wasserstrahlvakuum ab. Der ölige Rückstand kristallisiert nach Versetzen
mit Äthanol durch und wird aus Methanol—Wasser
umkristallisiert. In 73%iger Ausbeute erhält man N-(2-Aminoäthyl)-4-aminobutansulfonsäure. Äquivalentgewicht:
titriert 198 (Theorie: 196).
Läßt man analog Beispiel 7 in 600 Raumteilen Äthylendiamin 122 Teile Butansulton reagieren, so
erhält man nach Umkristallisieren aus Methanol— Äthanol 68 % N-(2-Aminoäthyl)-4-aminobutansulfonsäure.
Äquivalentgewicht: titriert 182 (Theorie: 182).
Bei 35 bis 400C werden unter Rühren 136 Teile
Butansulton in 384 Teile Tetramethylendiamin eingetropft. Nach beendeter Zugabe läßt man 12 Stunden
bei 25° C stehen und rührt dann in 1500 Raumteile Benzol ein. Die entstehenden Kristalle werden
nach Absaugen aus Äthanol umkristallisiert. Es resultieren 83% N-(4-Aminobutyl-4-aminobutansuI-fonsäure.
Äquivalentgewicht: titriert 233 (Theorie: 224).
Claims (4)
1. Verfahren zur Herstellung von N-(oj-Aminoalkyl)-aminoalkansulfonsäuren.
dadurch gekennzeichnet, daß man aliphatische Diamine mit aliphatischen Sultonen gegebenenfalls
in einem Lösungsmittel bei Temperaturen von -20 bis +10O0C, vorzugsweise bei -10 bis
+400C umsetzt.
2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, daß man als inerte Lösungsmittel
aromatische Kohlenwasserstoffe und Halogenkohlenwasserstoffe verwendet.
3. Verfahren nach Anspruch 1. dadurch gekennzeichnet,
daß man einen Überschuß an Sulton als Lösungsmittel verwendet und die Reaktion bei Temperaturen von 15 bis 400C
durchführt.
4. Verfahren nach Anspruch 1, dadurch gekennzeichnet, daß man als Lösungsmittel flüssige
oder tiefschmelzende aliphatische Diamine im Überschuß verwendet.
