Abstract

通过用氢氧化钠水溶液水解4，4－二甲基氧杂环丁－2－酮，制得3－羟基－3－甲基丁酸钠溶液，并用合成硅酸吸收所制得的溶液，由此制备3－羟基－3－甲基丁酸钠固体制剂。所述制剂略微吸湿，并易于搬运。

Description

制备3-羟基-3-甲基丁酸钠固体制剂的方法
本发明涉及制备以下通式所示3-羟基-3-甲基丁酸(β-羟基-β-甲基丁酸，HMB)的钠盐的固体制剂的方法。

3-羟基-3-甲基丁酸是一种活性化合物，它用作人类和动物用的药物和食物添加剂，例如用于降低血液中的胆固醇(WO-A-94/06417)，或者用于降低氮排泄，尤其是用于患者和/或老年人(WO-A-94/14429)。通常，用于此目的的并不是游离酸，而是生理上可接受的盐，例如钙盐。出于经济原因，尤其是用于动物营养或兽用药物时，最好也能使用其钠盐。虽然这在文献中已经重复提及，但是并没有说明其固体制剂的工业制备方法，并且发现它不能稳定储存并吸湿，因此难以进行加工。
因此，本发明的目的是提供一种制备3-羟基-3-甲基丁酸钠的简单且经济的方法，所述3-羟基-3-甲基丁酸钠能稳定储存，其吸湿性低并且呈固体形式(粉状)。
通过本发明权利要求1中所述的方法可以达到这一目的。
已经发现，用氢氧化钠水溶液水解4，4-二甲基氧杂环丁-2-酮可以制得3-羟基-3-甲基丁酸钠的溶液，若浓缩后合适的话，将此溶液加入到合成二氧化硅中。如果合适的话，之后进行干燥，可以形成低吸湿性的、可自由流动并且便于搬运的形式。
特别是，合适的合成二氧化硅是市售的常规沉淀二氧化硅，例如可从Degussa以Slipernat商品名购得。
所述3-羟基-3-甲基丁酸钠溶液较好是通过在减压条件下蒸馏除去水来浓缩成至少50重量％，尤其较好是70-80重量％。在上述浓度下，所述溶液方便地保持在60-65℃的温度下，防止结晶。所述浓缩可以在实验室规模下进行，例如在常规蒸馏装置内；或者在工业规模下进行，例如在薄膜蒸发器中。但是，也可以直接使用在合成过程中出现的含量例如为20-30重量％的溶液。
施加3-羟基-3-甲基丁酸钠溶液可以在常规混合装置中进行，较好是使用较缓和的混合器类型，例如滚筒混合器、犁头混合器或者锥形混合器、喷雾混合器或者(慢速)强力混合器。令人惊奇的是，混合过程太剧烈会导致形成结块。
所述施加较好是以如下方式进行：首先加入合成二氧化硅，然后再加入3-羟基-3-甲基丁酸钠溶液。
为了降低成品中的含水量，可以以本来已知的方式干燥所述成品。但是，即使不干燥，也可以获得含水量最高30重量％的产品，它仍旧可以自由流动并且其外表是干燥的。
而且为了提高其流动性，若合适的话，可以往本发明制得的产品中加入憎水二氧化硅(如SopernatD17)或者另一种流动性改进剂。在这种情况下，例如以所述干物质计，可以在所述混合工艺结束时加入0.2-0.4重量％的憎水二氧化硅，并可在短时间(例如30分钟)内混合。
本发明方法制得的固体组合物同样是本发明的主题，该组合物包含3-羟基-3-甲基丁酸钠和合成二氧化硅。
以干物质计，本发明组合物较好包含至少25重量％，尤其更好是25-50重量％3-羟基-3-甲基丁酸钠。
以下实施例说明本发明的步骤，但决不限于此。
实施例1
制备3-羟基-3-甲基丁酸钠溶液
将氢氧化钠(127.3g，3.15mol)的水(1.5l)溶液冷却至0℃。然后，在约2.25小时剧烈搅拌过程中滴加粗制4，4-二甲基氧杂环丁-2-酮(β-异戊内酯，336.7g，3mol，通过使乙烯酮和丙酮反应来制得)，同时温度保持在1.5-2℃。在0℃下将所述反应混合物再搅拌15分钟，然后在22℃下再加热40分钟。之后，在约3小时内在300毫巴下蒸馏除去415g水。以这种方式，制得1533g包含约27.5重量％3-羟基-3-甲基丁酸钠的溶液。
实施例2
制备固体制剂
在56℃下，在50毫巴压力下将实施例1所述的溶液浓缩至57.5重量％。将50g合成二氧化硅(Sipernat50，Degussa AG)加入Eirich强力混合器(R02型)中。将盘片旋转设定在2档(旋转叶片头不动)，并在1小时内通过管子加入100g溶液。然后再继续混合0.5小时。获得不含结块、可自由流动、外表干燥的均匀产品。

1.一种制备通式所示3-羟基-3-甲基丁酸钠固体制剂的方法，

其特征在于，在第一工艺步骤中，使4，4-二甲基氧杂环丁-2-酮和氢氧化钠水溶液反应，形成3-羟基-3-甲基丁酸钠溶液，若浓缩后合适的话，在另一工艺步骤中将这种溶液加入到合成二氧化硅中，若合适的话将所得产品干燥。
2.权利要求1所述的方法，其特征在于，在减压条件下将3-羟基-3-甲基丁酸钠溶液浓缩至至少50重量％的浓度。
3.权利要求1或2所述的方法，其特征在于，加入到合成二氧化硅中的操作是在滚筒混合器、犁头混合器或者锥形混合器或者强力混合器中进行的。
4.权利要求1-3任一项所述的方法，其特征在于，加入到合成二氧化硅中的操作是通过首先加入合成二氧化硅，然后加入3-羟基-3-甲基丁酸钠溶液来进行的。
5.权利要求1-4任一项所述的方法，其特征在于，为了提高流动性，再加入憎水二氧化硅。
6.一种固体组合物，所述组合物包含3-羟基-3-甲基丁酸钠和合成二氧化硅，可由权利要求1-5任一项所述方法制得。
7.权利要求6所述的固体组合物，其特征在于，以干物质计，3-羟基-3-甲基丁酸钠的含量至少为25重量％，较好是25-50重量％。
