Description
发明背景
骨移植被广泛用于治疗骨折、骨不连合和诱导关节固定。自体松质骨——从被移植者的一个部位取出并植入被移植者的另一个部位——是目前最有效的骨移植物。自体松质骨提供了支持骨愈合反应和生成新的软骨或骨的祖代细胞的分布的平台。不过,收获自体骨会导致成本和发病率上升,包括瘢痕、血液损失、疼痛、手术与复原时间延长和感染的风险。此外,移植部位的体积可能超过可利用的自体移植物的体积。

在有些情形中,复合植入物是这样制成的,将合成移植材料与相似或较少体积的、从骨髓抽吸物获得的细胞悬液混合。不过,具有分化为软骨、骨、肌肉、纤维组织和其他结缔组织的能力的祖代细胞在骨髓中的含量非常少。存在于1ml骨髓中的祖代细胞数因患者而异,从约100个细胞至20,000个细胞。这代表了有核细胞在骨髓抽吸物中的平均比例为约1∶20,000至1∶40,000。因而,将给定体积的合成移植材料与相当体积的新鲜骨髓混合所制成的复合植入物含有相对少的祖代细胞。
因此,以前已经开发了一种技术,以增加祖代细胞在复合植入物中的相对浓度。这种技术牵涉平板接种骨髓细胞悬液到组织培养皿上,在选定的培养基中培养细胞一天或多天,以实现祖代细胞群的提高,然后从组织培养皿分离出细胞,得到增加了祖代细胞群的细胞悬液。然后将合成陶瓷载体浸渍在这种富集了祖代细胞的悬液中,制成复合植入物。不幸的是,这种制备复合植入物的方法是非常费时的。而且,如果原始的祖代培养细胞是源于从被移植者获得的骨髓抽吸物的,那么被移植者必须经历多个侵入性程序;一个程序取出他或她的骨髓,后一个程序植入复合移植物。所以,被移植者可能面临一次以上的麻醉。

发明概述

骨髓抽吸物
骨髓抽吸物含有血浆、有核祖代细胞(祖代细胞)、有核造血细胞、内皮细胞、和源于外周血的细胞,包括红细胞和血小板。因为骨髓抽吸物含有外周血,优选的是将抽吸物收集在含有抗凝剂的注射器内。适合的抗凝剂包括肝素、柠檬酸钠和EDTA。优选地,用在本发明方法中的骨髓抽吸物是从将接受移植的患者(被移植者)获得的。次优选地,骨髓抽吸物可以是从另一免疫学上可相容的供体获得的。

最优选地,所提供的微粒性骨材料是同种移植物的松质骨微粒,它们是大块、薄片或片段的形式,平均直径为1-15、优选2-8mm。最优选地,所提供的纤维性骨材料是同种移植物的去矿化密质骨纤维,长度至少5mm,更优选至少1cm,更优选至少2cm,更优选至少3cm,更优选至少4cm,最优选至少5cm。可选地,所提供的纤维性骨材料是不同长度纤维的混合物,长度范围为5mm-2cm、5mm-3cm、5mm-4cm、5mm-5cm、5mm-15cm或一些其他范围。可选地,纤维性骨材料是作为柔韧性垫而供应的,例如可从Osteotech,Inc得到的Grafton Flex。
将微粒性与纤维性骨材料按下列方式结合构成优选的复合基质。将骨纤维,优选去矿化的密质骨纤维,长度如上所述——与微粒性骨微粒按下列优选比例结合:约225、次优选200-300、次优选150-375、次优选100-450、次优选75-500、次优选25-1000mg干重的去矿化密质骨纤维与约10、次优选8-12、次优选6-14、次优选4-16、次优选2-18、次优选1-25cc(总体积)的微粒性骨微粒,后者的平均直径为1-15、优选2-8mm。可选地,去矿化的密质骨纤维可以从包含这类纤维的柔韧性垫获得。在使用这样一种垫子时,首先用等渗溶液洗去任何可能存在的有毒或高渗材料,例如甘油。然后将垫子悬浮在盐水或其他适合的等渗溶液中,以促进单个骨纤维的分离。将所分离的骨纤维与微粒性骨材料按下列比例结合,构成优选的复合基质:来自最初尺寸为2.5cm×5cm×约2.5mm(最初体积约3.1cm3)的垫子的纤维与约10cc、次优选8-12cc、次优选6-14cc、次优选4-16cc(总体积)的微粒性骨微粒,后者的平均直径为1-15、优选2-8mm。
应当注意,若不同大小的移植物是必要的,可以按照本发明制备不同大小的复合基质,以符合纤维性与微粒性骨的上述比例。例如,(假定堆积密度均匀)可以将20cc微粒性骨与450mg骨纤维结合,提供优选的复合基质。
凝集材料
凝集材料可以是由血液形成的血块,血液从被移植者(或免疫学上相容的供体)的静脉或动脉获得。更优选地,凝集材料是由无抗凝处理的骨髓抽吸物形成的骨髓凝块,骨髓抽吸物最优选地是从被移植者获得的。优选地,形成骨髓凝块的骨髓抽吸物是在移植手术期间从被移植者获得的。次优选地,凝集材料可以是本领域已知的血小板凝胶、血小板浓缩物、纤维蛋白凝集材料和/或纤维蛋白胶。
向富集了祖代细胞的移植物基质加入骨髓凝块(从无抗血处理的骨髓抽吸物获得)惊人地显著提高所得复合骨髓移植材料的功效,相对于没有凝集材料的复合移植物而言。已经观察到,向相似富集了的基质加入骨髓凝块比单独的骨髓凝块多呈递50-70%的有核细胞(包括祖代细胞数量的两倍以上),所得移植物既优于单独的富集基质,也优于与骨髓凝块结合的非富集基质。因此,向富集了祖代细胞的基质加入骨髓凝块提高了移植物的性能。
在不希望局限于任何特定理论的同时,据信包含骨髓凝块可以提高复合骨移植物的功效,原因是下列的一种或几种。首先,一些对成功的骨愈合过程而言重要的细胞有可能不附着于移植物基质,因此不会充分地集中在移植部位(甚至有可能排除在外),导致该部位愈合的效率低下或无效。由凝血级联所致纤维蛋白原聚合成纤维蛋白(进一步解释如下)可以提供有价值的补充基质,促进对移植部位愈合反应而言重要的细胞的附着和移行。这类细胞包括移行性内皮细胞,它们增殖生成管状结构,后者对经由血管生成作用生成血管而言是重要的前体。
第二种可能性是在移植部位形成凝块的生理学过程为移植在该部位的成骨细胞创造了优良的环境。具体而言,无抗凝处理的骨髓抽吸物的凝集导致其中所含有的血小板的活化,导致血小板的脱粒。血小板的脱粒继而释放生长因子和亲骨细胞因子,它们可能不存在于移植部位。在这种过程期间所释放的若干重要的生物活性因子包括血小板衍生生长因子(PDGF)、表皮生长因子(EGF)、成纤维细胞生长因子(FGF)和转化生长因子β(TGF-β)。另外,作为凝血级联的结果,从纤维蛋白原生成纤维蛋白基质可以在直接的术后期间在移植部位提供重要的稳定性。此外,存在于移植物植入之后前几天内的纤维蛋白分解活动的过程在移植物掺入的早期阶段为血管生成因子(例如本领域已知的纤维蛋白分裂产物)提供另外的来源。据信移植部位在植入之后的所得血管生成作用可以增强新血管在该部位中的生成,为新植入的祖代细胞和其他负责骨愈合与生长的细胞提供营养来源,从而加速愈合反应。
优选的实施方式和仪器
现在将描述用于提供复合骨移植物的本发明方法的优选实施方式。本文所述的仪器部件优选地是由塑料制成的,优选地是透明的或半透明的。按照本发明的第一种优选实施方式,并首先参照图1,将如上所述制备的复合的生物相容性可植入基质10置于基质容器内,该容器最优选地是柱子12(基质10可以是紧密或松散包装的,这取决于材料和它的结构)。所提供的柱子12可以具有多种内部容积,适合于容纳必要体积的特定移植物的基质。本文所用的基质的容积(或基质容积)表示非多孔性固体的排除容积,该固体的外部尺寸等于特定的基质。例如,可以提供内部容积为5、10、15、20、25或30cc或一些其他内部容积的柱子,以容纳各种基质容积。优选地,柱子12的内部直径为0.5-3.0、更优选1-2、更优选1-1.5cm。优选地,柱子12的长度比它的内部直径大至少1.5、更优选至少2、最优选至少3倍。端帽14可拆装于柱子12,经由丝线连接、按钮连接或任意其他已知的连接装置。
可选地,所提供的端帽14可以带有屏或膜15(见图8),可允许抽吸物20穿过其中,同时保留基质10的微粒。优选地,这样一种膜具有开口,直径为至少20、优选至少30、优选至少40μm。
骨髓抽吸物20(优选地含有抗凝剂)是经由已知装置、优选地从被移植者获得。然后将抽吸物20装入第一载入注射器28。最初,抽吸物20含有祖代细胞32和其他有核细胞33,比例在1∶20,000与1∶40,000之间(见图2A)。抽吸物还含有血小板、红细胞和血清(包括可溶于或悬浮在血清中的分子)。所提供的载入注射器28带有注射器连接器30,适合于与端帽连接器31匹配,在载入注射器28与柱子12的各自内部容积之间提供流体互通。所提供的第二载入注射器29类似地带有注射器连接器30,适合于与端帽连接器31匹配。如图2所示,然后将第一与第二载入注射器28与29经由上述连接器附加在与端帽14相对的柱子12末端,从而在第一载入注射器28、柱子12与第二载入注射器29的内部容积之间提供流体互通。
然后推压第一载入注射器28,投放抽吸物20至柱子12,在那里抽吸物20流经基质10或者与之接触,之后在柱子12的另一端被收集在第二载入注射器29中。使骨髓抽吸物与基质接触以提供富集基质可以这样进行,使抽吸物流经基质,在抽吸物中培育基质,或者通过本领域已知的其他手段。作为替代选择,使抽吸物20与基质10通过任意已知手段接触,以提供富集基质。祖代细胞有利地和选择性地粘附于基质10的表面,由此留在基质内,而过量的其他细胞、例如血细胞和其他源于骨髓的有核细胞相对自由地流经基质,被收集在第二载入注射器29中。为了在基质10中富集祖代细胞群,流经基质10的抽吸物20的体积优选地超过基质容积。以这种方式,存在于抽吸物体积中的祖代细胞优选地集中在基质容积中,得到富集的基质。本文所用的“富集”意味着祖代细胞与全部有核骨髓细胞之比在基质中大于在原来的骨髓抽吸物中。优选地,富集基质中祖代细胞与全部源于骨髓的有核细胞之比为至少1∶20,000,更优选为至少1∶10,000,更优选为至少1∶5,000,反映了祖代细胞的普遍性或浓度增加了至少2倍,优选至少4倍。更优选地,富集基质的祖代细胞浓度比具有骨髓凝块的非富集基质增加了至少5倍,更优选至少6倍,更优选至少8倍。优选地,抽吸物体积与基质容积之比为至少2∶1,更优选为至少3∶1,更优选为至少4∶1。例如,在使用基质容积为15cc的基质时,穿过基质的抽吸物的总体积优选为至少30cc,更优选为至少45cc,更优选为至少60cc。可选地,最初从柱子12投放至第二载入注射器29的流出物可以在继续该方法之前被弃去。
可选地,通过交替推压第一和第二载入注射器28和29,使抽吸物20在柱子12中反复流经基质10。根据细胞和祖代细胞与特定基质结合的速率,这种使抽吸物20流经基质10的程序重复至少1次,但是优选地可以重复至少2次,优选至少3次,优选至少4次。可选地,在最初的骨髓抽吸物悬液和任意流出物已经穿过基质10之后,使洗涤溶液穿过基质10。优选地,洗涤溶液包含无菌的、等渗的、缓冲的溶液,pH范围为7.3至7.5。适合的洗涤溶液包括磷酸盐缓冲盐水、Hank氏平衡盐溶液、人血清和极限必需培养基。
上述程序之后,从端帽1 4拆下载入注射器28和29。在图3中可以见到,留在基质10中的祖代细胞32均匀分布在基质10内。具体而言,在基质10内存在祖代细胞浓度梯度,由此祖代细胞32集中在柱子12两端附近的区域(见图3A),而骨髓抽吸物20在进入柱子12后首先与基质材料接触。所以,基质10的中心区域具有低得多的祖代细胞群(见图3B)。这种效应是由于祖代细胞和其他细胞在进入柱子12后迅速粘附于基质表面。因而,机械混合步骤可有效地形成祖代细胞分布更均匀的富集基质。
参照图4,从柱子12除去端帽,在柱子12上安装注射器转接器13,构成施用注射器50。注射器转接器13包含一个活塞,它适合于推压柱子12,并逐出其中的基质材料。从而从柱子12中逐出富集了祖代细胞的基质材料11,与一定体积的凝集材料18一起投放至混合碗40内。凝集材料18优选地是无抗凝处理的骨髓抽吸物,如上所述从已被允许凝血的被移植者获得。可选地,可以在凝血之前将无抗凝处理的骨髓抽吸物与富集基质结合,以便在混合过程期间和之后形成凝块。凝集材料体积与富集了祖代细胞的基质材料体积之比优选为约1∶1,次优选为大约或至少1∶5、1∶4、1∶3、1∶2、2∶1、3∶1、4∶1或5∶1。一旦在混合碗40中结合,将富集基质材料11与凝集材料18机械混合在一起,得到异质的可植入复合骨移植物混合物8(见图5),组成基本上是均匀的。组成基本上均匀意味着复合骨移植物混合物8包含复合基质的纤维性与微粒性骨材料、富集了的祖代细胞群、和凝集材料,它们都基本上均匀地分布在整个混合物内,从而基本上不表现关于任意单一组分的体相浓度梯度。这类机械混合例如可以利用刀具38在混合碗40中进行,或者经由任意其他的已知机械混合装置进行。
参照图5,将漏斗41安装于混合碗40(仍然含有复合骨髓移植材料8),将施用注射器50安装于漏斗41,如图所示。随后将如图5所示完整的仪器倒置,如图6所示,除去混合碗40的底盖42。可选地,可以将碗内容物倒入或挖入附加于施用注射器50的漏斗41内。然后将可植入复合骨髓移植材料8装填在施用注射器50中,这可以借助刀具38。可选地,刀具38在一个末端装有适合于将材料装填在施用注射器中的活塞,如图所示。一旦装填后,从漏斗41除去施用注射器50,可以用于将可植入复合骨髓移植材料8施用于患者。移植材料8有效地诱导移植部位的骨愈合或骨再生。可选地,复合骨髓移植材料可以通过本领域已知的其他机械装置从混合碗直接转移到患者。
根据本发明的复合骨髓移植材料还可以按照下列第二种优选实施方式加以制备。如上所述分别获得和制备复合基质10和骨髓抽吸物20(优选地含有抗凝剂),分别放置在混合碗40中,比例类似于上述第一种优选实施方式。然后利用刀具38或一些其他已知的机械混合装置将基质和抽吸物机械混合在一起,得到富集基质。然后如下从富集基质11排出液体抽吸物20。参照图7,连接漏斗41与混合碗40,再连接柱子12与漏斗41,如图所示,单一的端帽14连接在柱子12的对侧末端。端帽14也装有带开口的膜或屏(没有显示),开口适合于允许抽吸物穿过而留下富集基质11。开口的直径优选为至少20、更优选30、更优选40μm。经由已知的连接装置连接流出注射器45与端帽14,从而在流出注射器45与柱子12各自的内部容积之间实现流体互通。将所述完整的仪器倒置,如图7所示,如果必要的话除去底盖42。抽出或排出混合碗40内的液体,经过柱子12进入流出注射器45。液体可以被推回到柱子12中。这种反复程序可以重复若干次,如上所述。然后弃去所排放的液体。
作为替代选择,通过压缩容器壁(例如混合碗40的基底)与带开口的多孔性屏或膜(没有显示)之间的富集基质,可以从富集基质中排出液体抽吸物20,开口适合于允许抽吸物穿过其中,但是留下受容器壁压缩的富集基质。然后倾泻已经透过膜的液体抽吸物,在容器中留下被压缩在容器壁与多孔性膜之间的富集基质。优选地,容器是在其出口上游装有多孔性膜的注射器,容器壁是注射器活塞。在这种优选的实施方式中,富集基质的压缩是这样实现的,在注射器内将基质推向多孔性膜,由此通过出口逐出抽吸物,而将富集基质留在注射器内。
然后将富集基质11与一定体积的凝集材料18混合,如图4所示,并类似于上述第一种优选实施方式进行该方法,得到可植入的复合骨髓移植材料8,它有效地诱导被移植者的骨愈合或骨再生。
可选地,上述每种优选实施方式还可以有另外一个步骤。在可植入骨移植材料8的植入之前,可以在凝集前向移植材料8投放一定量的无抗凝处理的骨髓抽吸物(例如经由排放),例如移植材料8位于施用注射器50中。以这种方式,液体抽吸物将渗透移植材料8的空隙体积,最终在其中凝固。抽吸物必须在抽吸之后立即投放至移植材料8,以确保它长时间保留液体足以有效渗透材料。该步骤向可植入移植材料8提供另外的源于骨髓的有核细胞(包括另外的祖代细胞)。
制备复合骨髓移植材料的本发明方法通常仅需不到60分钟即可完成。因而,可以在骨髓供体/被移植者尚处于手术室时即可进行本发明的方法。因此,利用本发明方法能够减少被移植者为接受复合骨移植物而必须经历侵入性程序的场合的次数。
还提供了根据本发明的试剂盒,以帮助外科医生实施本发明的方法。参照图8,这样一种试剂盒将优选地能够按照上述优选实施方式之一实施该方法,并且优选地包含至少下列经过灭菌的器具,优选地由塑料制成,优选地是透明的或半透明的:基质容器(例如基质柱子12);至少一个端帽14,经由已知的连接装置可拆装于基质容器;端帽14上的端帽连接器31,用于与载入注射器实现流体连通;至少一个载入注射器28,具有注射器连接器30,用于与端帽14实现流体连通;混合碗40,带有可除去的底盖42;漏斗41,一端与混合碗40的开口端匹配,另一端与柱子12匹配;注射器转接器13,用于从柱子12转换至施用注射器;和刀具38。最优选地,试剂盒进一步包含第二载入注射器29和第二端帽14,构造类似于上述。可选地,所提供的试剂盒带有多孔的生物相容的植入底物,用作骨移植物基质,和多孔性膜15,以保留基质在基质容器内。可选地,本发明试剂盒可以另外包含肝素化骨髓抽吸注射器、抽吸针头、另外的载入注射器或其他可用于骨移植领域的器具。
下列实施例进一步阐述本发明的各个方面。
实施例1
有人已经指出,具有富集基质的骨移植材料表现比非富集移植物改进了的性能(见美国专利No.5,824,084和6,049,026)。下列实验证明了联合有富集基质与抽吸骨髓凝块(ABMC)的移植材料的其他功效。
对22只beagle犬实施脊髓后索融合术。在每只动物中,在三个独立的脊柱融合部位实施局灶性融合(L1-2,L3-4和L5-6)。使用松质骨薄片作为基质材料,制备三种类型的复合骨移植物。在11只犬中,使用去矿化的松质骨薄片,而在其余11只犬中,使用矿化的松质骨薄片。三种复合移植物是单独的富集基质、包括骨髓凝块的非富集基质和包括骨髓凝块的富集基质。将基质材料与过量体积的抽吸物结合,如上所述实现富集。
使用接合得分作为初步结果参数,比较融合效果。利用CT扫描的定量分析,还评估了每个部位的骨体积,在每个部位进行力学测试。表1和表2总结了每种复合材料在每只动物和移植部位中分别关于矿化与去矿化基质的接合得分。

上述数据表明,向骨移植物基质加入骨髓凝集材料显著增强复合移植物的功效。在使用矿化的松质骨基质时,关于富集+ABMC组的接合得分是最大的(平均=1.9)。此外,富集+ABMC组的接合得分显著大于单独的富集组(平均=0.6,p=0.008)。富集+ABMC组在统计学上也优于单独的ABMC组(平均=1.2,p=0.04)。这种程度的优越性能是惊人的和意外的。关于三种复合材料富集组+ABMC组、单独的ABMC组和单独的富集组的总体接合率分别是11分之9(81%)、11分之7(63%)和11分之4(36%)。融合得分较高的融合部位聚集在富集+ABMC组。在富集+ABMC组中,11个融合部位有4个(36%)得分3.0或更高。相反,在单独的富集组中11个部位只有1个(8.3%)、在单独的ABMC组中11个部位只有2个(18%)得分近似。
在使用去矿化的松质骨基质时,关于富集+ABMC组的接合得分也是最高的。关于三种材料富集组+ABMC组、单独的ABMC组和单独的富集组的总体接合率分别是11分之7(63%)、11分之6(54%)和11分之5(45%)。融合得分更高的融合部位仍然聚集在富集+ABMC组。在富集+ABMC组中,11个融合部位有5个(45%)得分3.0或更高。相反,在单独的富集组中11个融合部位只有2个(18%)、在单独的ABMC组中11个融合部位只有3个(27%)得分在这个水平上。ABMC与富集骨基质的结合提供了与富集骨基质相比表现更加惊人和意外优越的性能的骨移植物。
对全部样本还进行了力学测试。仅对骨接合产生有限屈服点的那些样本计算了关于最大负载、变形失效和失效能的数据。下列表3和表4分别总结了关于去矿化与矿化样本的力学测试数据。
测量了每个移植部位的强直度,将数据制表,得到关于矿化与去矿化骨基质的三种类型移植物各自的平均与标准偏差值。关于矿化的骨基质,单独的富集、单独的ABMC和富集+ABMC移植物分别表现6.9±2.4、7.9±2.3和8.2±4.2的强直度。关于去矿化的骨基质,单独的富集、单独的ABMC和富集+ABMC移植物分别表现9.1±6.0、9.4±6.2和9.6±4.8的强直度。
关于每种复合移植物,都得到有关融合体积、融合面积和融合物内平均骨密度的CT图象分析数据。表3和表4分别总结了关于矿化与去矿化松质骨基质的数据。从表3和表4可以明显看出,关于富集+ABMC组的融合体积和融合面积都是最大的。

14.根据权利要求1的复合骨髓移植材料,所述祖代细胞、所述凝集材料和所述基质都基本上均匀地分布在所述复合骨髓移植材料中。
15.根据权利要求1的复合骨髓移植材料,其中细胞粘连分子是与所述可植入基质的表面结合的。
16.根据权利要求1的复合骨髓移植材料,所述移植材料有效地诱导被移植者中的骨愈合。
17.根据权利要求1的复合骨髓移植材料,所述移植材料有效地诱导被移植者中的骨再生。
18.制备复合骨髓移植材料的方法,包含下列步骤:
a)提供骨髓抽吸物;
b)提供多孔的生物相容性可植入基质;
c)使所述骨髓抽吸物与所述基质接触,得到富集了的基质;和
d)机械混合所述富集基质,得到复合骨髓移植材料,祖代细胞
基本上均匀地分布在所述复合骨髓移植材料中。
19.根据权利要求18的方法,进一步包含混合凝集材料与所述富集基质的步骤。
20.根据权利要求18的方法,其中所述可植入基质包含微粒性骨材料和纤维性骨材料。
21.根据权利要求20的方法,其中所述微粒性骨材料包含同种移植物松质骨微粒,所述纤维性骨材料包含去矿化的密质骨纤维。
22.根据权利要求21的方法,其中所述可植入基质包含约1-25cc总体积的微粒性骨微粒和约25-1000mg干重的骨纤维。
23.根据权利要求22的方法,所述骨纤维的长度为5mm-15cm。
24.根据权利要求18的方法,进一步包含向所述富集基质加入无抗凝处理的骨髓抽吸物的步骤。
25.用于复合骨髓移植材料制备的试剂盒,所述试剂盒包含基质容器、第一端帽和第一载入注射器,所述第一端帽可拆装于所述基质容器,所述第一端帽和所述第一载入注射器适合于彼此连接,以在所述第一载入注射器与所述基质容器之间实现流体互通。
26.根据权利要求25的试剂盒,所述试剂盒进一步包含多孔的生物相容性可植入基质。
27.根据权利要求25的试剂盒,所述试剂盒进一步包含肝素化的抽吸注射器和抽吸针头。
28.根据权利要求25的试剂盒,所述试剂盒进一步包含注射器转接器,适合于匹配所述基质容器,以与之构成施用注射器。
29.根据权利要求28的试剂盒,所述试剂盒进一步包含带有可除去的底盖和漏斗的混合碗,所述漏斗适合于在其一端匹配所述混合碗的开口端,在其另一端匹配施用注射器。
30.根据权利要求25的试剂盒,所述试剂盒进一步包含刀具,所述刀具在其一端具有活塞,所述活塞适合于装填移植材料至所述基质容器内。
31.根据权利要求25的试剂盒,所述试剂盒进一步包含多孔膜,可有效保留基质材料在所述容器内。
32.根据权利要求25的试剂盒,其中所述基质容器是基质柱。
33.根据权利要求25的试剂盒,所述试剂盒进一步包含第二载入注射器和第二端帽,所述第二端帽可拆装于所述基质容器,所述第二端帽和所述第二载入注射器适合于彼此连接,以在所述第二载入注射器与所述基质容器之间实现流体互通。
