Abstract
在一种能使淋巴细胞增殖和/或活化的细胞因子和α-糖基神经酰胺存在条件下,通过培养一种可以从人外周血获得的单核细胞组分而扩增人Vα24+自然杀伤T细胞,该外周血中的造血干细胞是被粒细胞集落刺激因子动员的。利用本方法扩增的含有人Vα24+自然杀伤T细胞的细胞组分可作为一种有效的癌症治疗剂。

技术领域
本发明涉及一种用以扩增人Vα24+自然杀伤T(NKT)细胞的方法,以及由本方法获得的人Vα24+NKT细胞和一种包含人Vα24+NKT细胞的组分的应用。
背景技术
NKT细胞是成熟淋巴细胞中的一种独特亚群,其兼有NK细胞受体和T细胞受体(Annu.Rev.Immunol.,15,535-562,1997;J.Exp.Med.,182,633-638,1995)。鼠NKT细胞表达NK1.1和TCRαβ受体,并且在骨髓(J.Immunol.,145,3209-3215,1990)和肝脏(J.Exp.Med.,180,699-704,1994)中尤为密集。这种细胞表达的TCR所有组成成分非常有限(J.Exp.Med.,180,1097-1106,1994;Int.Immunol.,7,1157-1161,1995),其中包括一种恒定的α链。这些结果暗示NKT细胞的配体是非多态的和非经典的MHC I类分子,这类分子表现出一种独特的经过TAP(抗原呈递相关转运蛋白)-非依赖性途径的特异性抗原呈递过程。最近的研究确定,一种MHC Ib类分子CD1d是NKT细胞的配体。新近报道,表达发生不包括N区的规范重排的恒定Vα24 JαQ TCR的人类T细胞与鼠Vα14-Jα281+ NKT细胞类似(J.Exp.Med.,180,1097-1106,1994)。此外,研究显示,在用α-半乳糖苷神经酰胺(α-GalCer)预处理的CD1d抗原呈递细胞(APCs)的刺激作用下,鼠Vα14-Jα281+细胞和人Vα24 JαQ TCR+NKT细胞可发生增殖。据报道,活化的NKT细胞表现出一种对多种肿瘤细胞系的穿孔素依赖性NK-样细胞毒性(Cancer Res.,59,5102-5105,1999),以及抑制某些实验动物模型中的肿瘤转移(Pro.Natl.Acad.Sci.USA,95,5690-5693,1998)。
十九世纪八十年代,开展了大量的免疫疗法临床试验,包括在注射或不注射细胞因子的条件下,所进行的LAK(淋巴因子动员性杀伤)细胞疗法和TIL(肿瘤浸润性淋巴细胞)疗法试验。然而,在临床效果方面,这些临床试验的结果显示这些细胞疗法缺乏前景,因为没有发现明显的临床疗效。十九世纪九十年代,在鉴定人类癌症免疫应答的分子成分方面取得了重要进步。目前,一些不同的免疫疗法正被用于临床试验,如树突状细胞(DC)疗法和细胞毒性T淋巴细胞(CTL)疗法。
如上所述,与T细胞、B细胞和NK细胞等其他淋巴样细胞不同,NKT细胞的特征在于其共表达NK受体和一种恒定的T细胞受体。已知这种新的淋巴细胞系能产生大量的白细胞介素4(IL-4)和γ干扰素(IFN-γ),并且表现出强的杀肿瘤活性。因此,NKT细胞的这种功能对癌症的免疫疗法非常重要,并且利用这些细胞的能力期望代表一种新的免疫疗法。已开展了一些先体外后体内扩增Vα24+NKT细胞的研究(Cancer Res.,59,5102-5105,1999;Immunology,99,229-234,2000)。然而,甚至从脐带血中也很难获得足够量的这种细胞以实现免疫治疗。
发明内容
本发明的目的是提供一种方法,用以产生足够量的Vα24+NKT细胞。本发明的另一个目的是提供Vα24+NKT细胞和适用于细胞疗法的包含Vα24+NKT细胞的细胞组分,其中的细胞疗法是指一种通过使用细胞来治疗疾病的方法。
本发明人发现一种方法,该方法通过使用α-糖基神经酰胺,用以从一种单核细胞组分中获得足够量的Vα24+NKT细胞,该单核细胞组分可从外周血中获得,该外周血中的造血干细胞由粒细胞集落刺激因子(G-CSF)所动员(G-PBSCs)。
该单核细胞组分包括DCs和T细胞、单核细胞、B细胞、NK细胞、NKT细胞、粒细胞等。当α-糖基神经酰胺作用于该组分时,α-糖基神经酰胺被呈递到DC的CD1d分子上,并且由于细胞因子的分泌,使得Vα24+NKT细胞、NK细胞等发生增殖。当单核细胞组分是从外周血中获得,该外周血中的造血干细胞由G-CSF所动员时,NKT细胞会发生更为有效的增殖,并且单核细胞组分会引起更强的免疫应答。此外,与从没有G-CSF动员的外周血中获得的单核细胞组分(PBMCs)比较,在IL-2和α-糖基神经酰胺存在条件下,通过培养可以从造血干细胞由G-CSF所动员的外周血中获得的单核细胞组分(G-PBSCs),所促成的NKT细胞的扩增更为持久。
本发明基于这些发现而得以完成,并且提供一些扩增方法和产生方法(包括利用这些方法所获得的细胞和细胞组分),以及用于治疗癌症试剂,如下所述:
(1)一种用以扩增人Vα24+NKT细胞的方法,该方法包括在一种影响淋巴细胞增殖和/或活化的细胞因子以及α-糖基神经酰胺存在的条件下,培养可以从人外周血中获得的单核细胞组分(G-PBSCs),该外周血中的造血干细胞由G-CSF所动员。
(2)一种根据(1)的方法,其中影响淋巴细胞增殖和/或活化的细胞因子包括IL-2。
(3)一种用以产生人Vα24+NKT细胞的方法,该方法包括在一种影响淋巴细胞增殖和/或活化的细胞因子以及α-糖基神经酰胺存在的条件下,培养可以从人外周血中获得的单核细胞组分(G-PBSCs),该外周血中的造血干细胞由G-CSF所动员,以及从培养细胞中分离人Vα24+自然杀伤T细胞。
(4)一种根据(3)的方法,其中影响淋巴细胞增殖和/或活化的细胞因子包括IL-2。
(5)一种用以产生一种包含人Vα24+NKT细胞的细胞组分的方法,该方法包括在一种影响淋巴细胞增殖和/或活化的细胞因子以及α-糖基神经酰胺存在的条件下,培养可以从人外周血中获得的单核细胞组分(G-PBSCs),该外周血中的造血干细胞由G-CSF所动员。
(6)一种根据(5)的方法,该方法进一步包括在肿瘤抗原存在的条件下,培养那些通过在IL-2和α-糖基神经酰胺存在的条件下培养而获得的细胞。
(7)一种根据(5)或(6)的方法,其中影响淋巴细胞增殖和/或活化的细胞因子包括IL-2。
(8)一种用于治疗癌症的试剂,该试剂含有作为一种活性成分的由(3)或(4)中定义的方法所获得的人Vα24+自然杀伤T细胞。
(9) 一种用于治疗癌症的试剂,该试剂含有作为一种活性成分的由(5)-(7)中任一项定义的方法所获得的含有人Vα24+自然杀伤T细胞的一种细胞组分。
此外,本发明还提供一种治疗癌症的方法,包括将有效量的人Vα24+NKT细胞或包含人Vα24+NKT细胞的细胞组分给药于需要癌症治疗的患者,其中的人Vα24+NKT细胞是由(3)或(4)中定义的方法获得的,而包含人Vα24+NKT细胞的细胞组分则是由(5)至(7)中定义的任一方法获得的。此外,本发明提供由(3)或(4)中定义的方法所获得的人Vα24+NKT细胞或由(5)至(7)中定义的任一方法所获得的包含人Vα24+NKT细胞的细胞组分在制造用于癌症治疗的试剂中的用途。

实施本发明的最佳方式
以下将详细描述本发明。
本发明所述的扩增方法和产生方法的特征在于在一种影响淋巴细胞增殖和/或活化的细胞因子以及α-糖基神经酰胺存在的条件下,培养可以从人外周血中获得的单核细胞组分(G-PBSCs),该外周血中的造血干细胞由G-CSF所动员。
制备G-PBSCs的方法,例如,采用以下方法。
当将G-CSF以2-16μg/kg/日的剂量经皮下给药于癌症患者5-10天时,作为造血干细胞指数的CD34+细胞的数量和粒细胞-巨噬细胞集落形成单位(CFU-GM)就会增加,并且大约从第5-6天起,它们都显示出最大值。因此,起始G-CSF给药后的4-12天,通过血浆分离置换法来收集含有外周血干细胞的细胞组分(粗制G-PBSCs)。再通过进一步使用密度梯度离心等方法从其中制备一种单核细胞组分(G-PBSCs)。此外,还可以通过类似方法从健康个体中制备G-PBSCs。
从外周血中获得的单核细胞组分主要包含单核细胞和粒细胞,如单核细胞和淋巴细胞(T细胞、B细胞、NK细胞和NKT细胞),并且从其中去除了红细胞和血小板。
一般而言,来自健康个体的外周血含有0.02%的造血干细胞。与此相比,来自健康个体并且通过G-CSF的给药引起动员的外周血中,造血干细胞的含量增加至约0.5-1%,而来自癌症患者并且在化疗后通过G-CSF的给药引起动员的外周血中,造血干细胞的含量增加至约2-7%。因此,与从造血干细胞不由G-CSF动员的外周血中获得的单核细胞组分相比,从造血干细胞由G-CSF动员的外周血中获得的单核细胞组分含有更多的造血干细胞。
如果在培养基中加入能影响淋巴细胞增殖和/或活化的细胞因子以及α-糖基神经酰胺,在惯用的培养条件下就可以培养PBSCs。影响淋巴细胞增殖和/或活化的细胞因子以及α-糖基神经酰胺的加入量和培养时间应能有效地使Vα24+NKT细胞扩增。尽管依据所加入的影响淋巴细胞增殖和/或活化的细胞因子的不同会有所变化,但是通常细胞因子的浓度为50-200U/ml,α-糖基神经酰胺的浓度通常为10-200ng/ml,并且培养时间为2-40天。通过利用流式细胞仪对荧光抗体染色的细胞进行分析可以测定Vα24+NKT细胞的扩增情况。
α-糖基神经酰胺是一种神经鞘糖脂,其中由半乳糖、葡萄糖等构成的糖链通过α-键与神经酰胺结合。其实例包括WO 93/05055(1993年3月18日公开)、WO 94/02168(1994年2月3日公开)、WO 94/09020(1994年4月28日公开)、WO 94/24142(1994年10月27日公开)和WO 98/44928(1998年10月15日公布)所公开的那些内容。具体地说,优选的是(2S,3S,4R)-1-O-(α-D-吡喃半乳糖基)-N-二十六醇基-2-氨基-1,3,4-十八烷醇。
影响淋巴细胞增殖和/或活化的细胞因子的实例包括IL-2、IL-7、IL-15、IL-12和IL-18。可以仅使用一种细胞因子或将两种或多种细胞因子结合使用。优选使用的细胞因子包括IL-2,即,IL-2或IL-2与另一种细胞因子的组合。
关于用于培养的培养基,应提到补加了10%胎牛血清(FCS)的RPMI-1640,其中包含2mM L-谷氨酰胺、1%丙酮酸、2%碳酸氢盐、100U/ml青霉素,以及100U/ml链霉素。
培养单核细胞组分,其中的造血干细胞如上文所述经G-CSF动员,这种方法与采用相同方式但不经G-CSF动员的PBMCs培养情况相比,可以使Vα24+NKT细胞的数量显著增加。
在以前的使用LAK细胞和TILs的临床试验中,将10×1010-11个效应细胞给予患者。根据本发明,不同的是,例如,从经G-CSF完成动员的外周血中平均收集8-40×109个单核细胞,将所收集的细胞在IL-2和α-糖基神经酰胺存在条件下培养12天后,就可以获得1×109-10个自体Vα24+NKT细胞。所得细胞的数量与以前临床研究中所用细胞数量相当,并且可有效的用于临床免疫疗法。
因此,根据本发明的扩增方法,已知具有杀灭多种类型肿瘤能力的Vα24+NKT细胞可有效地增殖以达到实用程度。同样,使用本发明的生产方法获得的含有Vα24+NKT细胞的细胞组分也能引起强免疫应答。
在本发明的生产方法中,在培养那些通过在影响淋巴细胞增殖和/或活化的细胞因子以及α-糖基神经酰胺存在的条件下培养而获得的细胞后,优选在一种肿瘤抗原存在条件下培养细胞。
如果培养基中加入了肿瘤抗原,就可以在培养PBSCs的惯用条件下完成肿瘤抗原存在条件下所进行的培养。肿瘤抗原的加入量和培养时间当充分,使得细胞组分包含的DCs有效地诱导CTL,该细胞组分通过在影响淋巴细胞增殖和/或活化的细胞因子以及α-糖基神经酰胺存在条件下培养获得。尽管依据肿瘤抗原类型的不同会有所变化,但是通常肿瘤抗原的浓度为10-10000μg/ml,培养时间为2-14天。确定CTL诱导的方法可以有前体频率分析(Sharrock CE et al.:Immunology Toda 11:281-286,1990)、ELISPOT方法(Pass Hetal.:Cancer J Sci Am 4:316-323,1998),以及四聚物方法(RomeroP et al.,J Exp Med 188:1641-1650,1998)等。
通过在肿瘤抗原存在条件下进行培养,细胞组分中的CTL被诱导,因此,该细胞组分也可以引起对肿瘤抗原的特异性免疫应答。
实现从培养细胞中分离Vα24+NKT细胞的方法可以是一些常用方法,如磁珠法。
本发明用以治疗癌症的试剂包括,作为活性成分的通过本发明的方法获得的人Vα24+NKT细胞或一种包含人Vα24+NKT细胞的细胞组分。
通过本发明的生产方法获得的人Vα24+NKT细胞应能强效地引起肿瘤抗原特异性免疫应答,并同时清除肿瘤细胞。
在影响淋巴细胞增殖和/或活化的细胞因子以及α-糖基神经酰胺存在条件下,与通过培养不经G-CSF动员的单核细胞组分PBMCs所得到的细胞组分比较,通过本发明的生产方法获得的包含人Vα24+NKT细胞的细胞组分中,NKT细胞的扩增更为持久。
此外,在肿瘤抗原存在条件下,通过进一步实施培养所获得的包含人Vα24+NKT细胞的细胞组分能通过该组分中的DCs诱导CTL从而引起肿瘤抗原特异性免疫应答。也就是说,该细胞组分既可以通过扩增人Vα24+NKT细胞而有效地引起非肿瘤抗原特异性免疫应答,又可以引起肿瘤抗原特异性免疫应答。据报道,依据肿瘤组织部分(即,肿瘤细胞)的不同,主要组织相容性抗原(MHC)的表达水平也不同。由于能引起两种免疫应答,该细胞组分中的肿瘤抗原特异性CTL应能攻击表达MHC的肿瘤细胞,并且非肿瘤抗原特异性NKT细胞应能攻击低水平表达MHC或不表达MHC的肿瘤细胞。因此,将该实施方案的细胞组分应用于癌症治疗是很有利的。
因此,将由本发明所述产生方法获得的人Vα24+NKT细胞或含有人Vα24+NKT细胞的细胞组分作为癌症治疗剂的一种活性成分是很有利的。
在联合独立分离自另一种细胞群的经肿瘤抗原处理而活化的DC s的条件下,由本发明的生产方法获得的人Vα24+NKT细胞可以用于细胞疗法,如癌症治疗。获得活化DCs的方法有,用GM-CSF和IL-4或IL-3处理包含在PBSCs中的单核细胞,或者是用GM-CSF或IFNα处理CD34阳性造血干细胞。
本发明用于治疗癌症的试剂的给药途径可包括静脉内给药、皮下给药、皮内给药和淋巴节内给药。试剂的形式可包括一种在生理盐水或林格氏液等中的悬浮液。依据给药途径不同,给药量也不同,如果是皮下给药,剂量为0.1-5ml,或者,如果是静脉内给药,剂量可高达300ml。Vα24+NKT细胞的给药量通常以每平方米体表面积给药105-1010个细胞。所需治疗的疾病可包括多发性骨髓瘤、淋巴瘤、白血病、乳腺癌、结肠癌、肺癌、前列腺癌、黑素瘤、肾癌、肝癌,以及胰腺癌等。此外,本发明的试剂还适用于病毒性疾病,如CML和HIV。
根据本发明,通过使用α-糖基神经酰胺可先体外后体内地产生大量的自体Vα24+NKT细胞。该细胞数量之大足以使其满足用于临床领域的要求。此外,培养的Vα24+NKT细胞显示出对肿瘤具有强杀肿瘤活性,无论该肿瘤是否表达MHC。在本发明中,与PBMCs比较,通过α-糖基神经酰胺的刺激,G-PBSCs显示出在体外非常有效地扩增人Vα24+NKT细胞的能力。无论是来自健康个体还是癌症患者,G-PBMCs都能扩增至相同的程度。
实施例
下文将结合实施例以详细描述本发明。
实施例1.人Vα24+NKT细胞的扩增
(1)PBMCs的制备和G-PBSCs的制备
来自健康个体的400ml全血制备出的血沉棕黄层(粗制PBMCs)由日本红十字血液中心提供。含有PBSCs的细胞组分(粗制G-PBSCs)来自经过化疗的患者,并且这些患者在化疗后接受G-CSF以100-250μg/人/天的剂量经皮下给药6-10天(表1),方法是在最后一次注射G-CSF后2-24小时内,用COBE SpectraTM细胞分离器(LAKE WOOD,CO USA 80215)来完成血浆分离置换法(造血干细胞:生物学和治疗学应用。Levit DJ,Mertelsmann R(eds),Marcel Dekker,Inc.,New York,1995,611-630)。
使用Lymphosepal密度梯度培养基(Immuno-BiologicalLaboratories Gunm,Japan)从所获得的粗制PBMCs和粗制G-PBSCs中制备出单核细胞组分(分别称为“PBMCs”和“G-PBSCs”)。

Vα24+NKT细胞的体外活化和扩增
在100U/ml IL-2和100ng/mlα-糖基神经酰胺的存在条件下,用RPMI-1640培养基培养(1)中所得的PBMCs和G-PBSCs,该RPMI-1640中补加了2mM L-谷胺酰胺、1%丙酮酸、2%碳酸氢盐、100U/ml青霉素、100U/ml链霉素(GIBCO BRL)。37℃条件下,在含有5%CO2的水-汽饱和空气中孵育这些细胞。分别在第0天(d0)和第12天(d12)检测这些培养细胞的数量和特性。所用的α-糖基神经酰胺是由KirinBeer Kabushiki Kaisha制备的,即(2S,3S,4R)-1-O-(α-D-吡喃半乳糖基)-N-二十六醇基-2-氨基-1,3,4-十八烷醇;被称为“α-半乳糖基神经酰胺”。
按照如下方法,用FACScan血细胞计数器(Becton Dickinson,Sunyvale,CA)来进行培养细胞的流式细胞术。异硫氰酸荧光素(FITC)偶联的抗Vα24(C15)、藻红蛋白(PE)偶联的抗Vβ11(C21)、PE偶联的抗CD3、PE偶联的抗CD56、PE偶联的抗CD83和PE偶联的抗CD8来自Immunotech(Marseilelles Cedex,France)。FITC偶联的PE偶联的抗CD161、PE偶联的抗IL-2Rβ、PE偶联的抗HLA-DR和PE偶联的抗CD4来自Becton Dickinson。PE偶联的抗CD86来自Pharmingen(San Diego,CA)。PE偶联的抗CD57来自SIGMA(SaintLouis,Missouri),并且用作为同种型对照。收集培养细胞并以1×105/ml的密度重悬于10%胎牛血清(FCS)-RPMI1620培养基中。用MRX-150高速冷冻微量离心机(TOMY SEIKO CO.,LTD.Tokyo,Japan)以2000rpm的转速离心3分钟,将这些细胞重悬于50μl含有1μl上述抗体的PBS中,4℃孵育30分钟。然后用PBS清洗细胞两次,再重悬于PBS中,并且在进行分析之前4℃贮存。显示为FSC、SSC、FL-1的数据是指Vα24或CD4,显示为FL-2的数据是指Vβ11、CD8、CD3。
Vα24+NKT细胞在第0天和第12天之间的变化显示在图1中。来自PBMCs的Vα24+NKT细胞反应于IL-2和α-糖基神经酰胺而扩增。来自由五位不同供体提供的一系列PBMCs的数据显示在表2中。尽管在第0天时Vα24+NKT细胞占总细胞数量的0.03-0.14%,但这一百分比在第12天增加了1.4-6.1倍。对于来自G-PBSCs的Vα24+NKT细胞,第12天的扩增显示出32-1414倍的增加量(图1,表3)。这一扩增比在PBMCs中观测到的扩增高出至少10倍(图2)。图3显示Vα24+NKT细胞在第12天的表型。这些细胞中,超过98%的细胞表达TCR Vβ11,73.33%的细胞表达CD4或CD8,而小于5%的Vα24+NKT细胞分别表达CD57、CD83、IL-2Rβ和HLA-DR(图3)。此外,尽管源于PBMCs的Vα24+NKT细胞的数量在第12天后逐渐减少,但是源于G-PBSCs的Vα24+NKT细胞直到培养的第30天仍继续扩增(图5)。表2.由α-GalCer引起的来自正常PBMCs的Vα24+NKT细胞的扩增

此外还确证了来自健康个体的G-PBSCs能以与来自癌症患者的G-PBSCs相同的程度进行扩增。
从上述结果可清晰的看出,通过使用G-PBSCs,经过短期培养就可以产生至少108个Vα24+NKT细胞。
实施例2评定Vα24+NKT细胞对人肿瘤细胞的细胞毒活性
用FACS Vantage细胞分类系统从第12天的培养细胞中纯化出人Vα24+NKT细胞,并且对这些细胞对肿瘤细胞的细胞毒活性和这些细胞的细胞因子产生活性,均加以评定。
利用以下靶细胞系对α-GalCer活化的Vα24+NKT细胞的细胞毒性进行了三次重复测定,这些靶细胞系为:Molt-4淋巴瘤细胞系和K562髓细胞性白血病细胞系(ATCC,Pockville,MD)。以每1×106个细胞用100μl Ci铬酸钠(NEN Life Science Products,Inc.,BostonMA02118)来标记靶细胞1小时。将纯化的α-GalCer活化的Vα24+NKT细胞或Vα24-NKT细胞作为效应细胞,并按指定的效应细胞(E)/靶细胞(T)比将这些细胞接种到圆底96孔板中的51Cr标记的每个靶细胞上。在5%CO2的条件下,37℃培养6小时后,用γ计数器对裂解的靶细胞所释放的放射性进行计数。由公式(样品的cpm-本底cpm)/(最大cpm-本底cpm)×100来计算特异裂解百分比。由仅有靶细胞的上清液来计算本底cpm,并通过往靶细胞中加入1M HCl来计算最大释放量。以三次计算的平均值加上标准差的形式来表示数据(图4)。
发现超过78%的培养细胞表达恒定的Vα24+/Vβ11+TCR。如图4所示,这些细胞对K562肿瘤细胞和Molt-4肿瘤细胞表现出强细胞毒活性。
用磁性细胞分类仪从单核细胞中分离出CD14+细胞。为了获得来源于单核细胞的DCs,在50ng/ml重组人(rh)GM-CSF、1000U/ml rhIL-4和500U/ml rhTNF-α(SIGMA,St Louis,MO)存在的条件下,用RPMI-1640培养基培养这些细胞,该RPMI-1640中补加了2mM L-谷胺酰胺、1%丙酮酸、2%碳酸氢盐、100U/ml青霉素、100U/ml链霉素(GIBCO BRL,Gaithersburg)以及10%FCS(DainipponPharmaceutical Co.,Ltd.)。37℃条件下,在含有5%CO2的水-汽饱和空气中孵育这些细胞。挑选出的Vα24+NKT细胞与来自同一供体的源自单核细胞的DCs一起孵育24小时。用ELISA系统(DIACLONE,BESANCON,FRANCE)测定培养液上清中的IFN-γ和IL-4。
在自体的来自单核细胞的DCs存在条件下,进行48小时培养后,用ELISA测定Vα24+NKT细胞的细胞因子分泌。同时,还确定了Vα24+NKT细胞分泌IFN-γ,但Vα24+NKT细胞分泌IL-4却不能确定。
工业实用性
根据本发明,经过短期培养就可以产生大量的Vα24+NKT细胞。此外,还可以获得一种细胞组分,其中的Vα24+NKT细胞可进行扩增。通过使用全部组分,预期可得到一种能引起广泛免疫应答的细胞疗法,而用传统细胞疗法不能实现这个目的。
Claims (9)
1.一种用以扩增人Vα24+自然杀伤T细胞的方法,该方法包括在一种影响淋巴细胞增殖和/或活化的细胞因子以及α-糖基神经酰胺(α-GlyCer)存在的条件下,培养可以从人外周血中获得的单核细胞组分(G-PBSCs),该外周血中的造血干细胞由粒细胞集落刺激因子所动员。
2.根据权利要求1的方法,其中影响淋巴细胞增殖和/或活化的细胞因子包括白介素2。
3.一种用以产生人Vα24+自然杀伤T细胞的方法,该方法包括在一种影响淋巴细胞增殖和/或活化的细胞因子以及α-糖基神经酰胺存在的条件下,培养可以从人外周血中获得的单核细胞组分(G-PBSCs),该外周血中的造血干细胞由粒细胞集落刺激因子所动员,以及从培养细胞中分离人Vα24+自然杀伤T细胞。
4.根据权利要求3的方法,其中影响淋巴细胞增殖和/或活化的细胞因子包括白介素2。
5.一种用以产生一种包含人Vα24+自然杀伤T细胞的细胞组分的方法,该方法包括在一种影响淋巴细胞增殖和/或活化的细胞因子以及α-糖基神经酰胺存在的条件下,培养可以从人外周血中获得的单核细胞组分(G-PBSCs),该外周血中的造血干细胞由粒细胞集落刺激因子所动员。
6.根据权利要求5的方法,该方法进一步包括在肿瘤抗原存在的条件下,培养那些通过在影响淋巴细胞增殖和/或活化的细胞因子和α-糖基神经酰胺存在的条件下培养而获得的细胞。
7.根据权利要求5或6的方法,其中影响淋巴细胞增殖和/或活化的细胞因子包括白介素2。
8.一种用于治疗癌症的试剂,该试剂含有作为一种活性成分的由权利要求3或4中定义的方法所获得的人Vα24+自然杀伤T细胞。
9.一种用于治疗癌症的试剂,该试剂含有作为一种活性成分的由权利要求5-7中任一项定义的方法所获得的含有人Vα24+自然杀伤T细胞的一种细胞组分。
