Abstract

Description
游离基是活有机体的有氧代谢中固有的并且通常随生理和病理过程产生。它们有时有意地产生,从而起生物功能作用,例如吞噬细胞中的杀微生物剂,或者它们可能为化学变化,之后呈现破坏性行为。无论它们的产生机理如何,如果游离基的产生和除去得不到控制,那么它们对有机体的影响可能为破坏性的。为了对抗过度且不适宜的破坏,开发出了一种精细的抗氧化剂防御系统。
当有利于氧化剂的氧化剂与抗氧化剂之间失衡时,存在氧化应力症状,它可能导致组织损伤。
本发明尤其提供了一种克服家庭猫和狗中氧化应力问题的方法。
因此,本发明的第一个方面提供了一种增加家养猫或狗中血浆维生素E水平的方法,该方法包括步骤:向所述猫或狗给予维生素E,其量足以增加血浆维生素E水平。该增加可以是在该动物血浆中可以测定的最大/饱和点。该增加可以为该动物自身的血浆维生素E水平基线的2-3倍(大约最大生理增加)。当与给动物喂食对照饮食时的血浆维生素E水平比较时,该增加可以使单个动物的血浆维生素E水平增加高至25%,优选25%或以上(优选高至50%,或25-50%,或者甚至50-90%)。对照饮食例如是猫或狗的总维生素E消耗为10 IU/400kcal。
维生素E为几种具有相同生物活性的生物学上类似的化合物的共同术语,包括称作生育酚类和生育三烯酚类的那些。动物组织中生物学活性最大的维生素E的生物形式(也为活性最大的抗氧化剂)为α-生育酚。维生素E不能在体内合成。维生素E防止细胞膜失去完整性,细胞膜完整性的丧失有害地改变细胞和细胞器功能。
维生素E的单位可以国际单位(IU)表示,其中1 IUα-生育酚等于1mgα-生育酚。其它维生素E化合物具有通过其与α-生育酚比较的生物潜能测定的IU,如McDowell,L.R(1989)Vitamin E:In vitamins in AnimalNutrition,Chapter 4,page 96,Academic Press,UK中所述。
时至今日,足够防止家庭狗或猫中呈现维生素E缺乏症状的最小维生素E水平之上和之下的维生素E水平还未引起注意。本发明认为,狗或猫中维生素E的水平反映了它们饮食中的水平,并且这些水平提供了动物以预制宠物食品为食时没有超过的典型基线水平(参见实施例)。本发明显示了可以通过在动物饮食中加入较高水平的维生素E(并且这可以通过提供特定预制的宠物食品和/或者猫或狗补充物来实现)来增加狗和猫中的维生素E水平。
本发明的一个方面提供一种降低家庭猫和狗中氧化应力的方法。氧化应力的这种降低,特别增强了免疫反应,并提供了更健康的动物。狗或猫中氧化性损伤的标记物尤其包括:血浆羰基(蛋白质氧化的终产物)、血浆脂质过氧化氢(脂质氧化的标记物)和响应LDL氧化产生的抗-LDL抗体。任意这些降低都将显示氧化性损伤减少。
按照本发明第一个方面的维生素E可以为任意形式。它可以为生育酚或生育三烯酚。它可以为α-生育酚(d-α或dl-α)、β-生育酚(d-β或dl-β)、γ-生育酚(d-γ或dl-γ)、δ-生育酚、α-生育三烯酚、β-生育三烯酚、γ-生育三烯酚或δ-生育三烯酚。优选α-生育酚。
维生素E的来源没有限制。优选的维生素E源包括醋酸维生素E(例如醋酸生育酚)、醋酸维生素E吸附物或经喷雾干燥的醋酸维生素E。尽管可以使用天然的来源,但是优选来源为合成的。
维生素E的给予形式没有限制。可以为饮食、食品或补充物的形式。本文下面的术语“食品”包括所有食品、饮食和补充物。任意这些形式可以为固体、半固体或液体。
该补充物对未含足够高的一种或多种本发明组分的饮食或食品特别有用。这些组分在补充物中的浓度可以用于“增加至”动物饮食或食品中的水平。这可以通过在动物饮食中包含大量该补充物或者通过另外向动物喂食大量该补充物进行。补充物可以形成为具有极高量一种或多种本发明组分的食品,该组分在喂食动物之前需要稀释。补充物可以为任意形式,包括固体(例如粉末)、半固体(例如食品样稠度/凝胶)、液体或者,它可以为片剂或胶囊形式。液体可以方便地与食品混合或者直接喂给动物,例如经过匙或经过吸管样装置。该补充物可以为高含量的一种或几种本发明组分或者可以为至少两部分的组合包装,每一部分含有所需量的一种或多种组分。
优选将该维生素E加入商品化宠物食品或商品化饮食补充物中。宠物食品可以为干燥、半干燥、润湿或液体(饮料)产品。润湿产品包括在锡或箔容器中销售并具有70-90%水分含量的食品。干燥产品包括具有相似组成的食品,但具有5-15%水分和以饼干样粗粒出现。优选将该饮食、食品或补充物包装。以这种方式,消费者可以从包装上了解到食品中的组分并考虑它是否适合狗或猫。该包装可以为金属(通常为锡或柔性箔)、塑料、纸或卡片。任何产品中的水分含量都有可能影响使用或所需的包装类型。
本发明的食品包括狗或猫饮食消费的任何产品。因此,本发明包括标准食品产品,还包括宠物食品小吃(例如小吃棒、饼干和甜食)。食品优选煮制产品。可以加入肉或得自动物的物料(例如牛肉、鸡肉、火鸡肉、羊肉、血浆、髓骨等,或其两种或多种)。为了提供蛋白源,该食品或者可以没有肉(优选含有肉代用品如大豆、玉米谷蛋白或大豆产品)。产品可以含有其它蛋白源如大豆蛋白浓缩物、乳蛋白、谷蛋白等。该产品还可以含有如一种或多种谷物的淀粉源(例如小米、玉米、大米、燕麦、大麦等)或者可以没有淀粉。典型的干燥商品化狗和猫食品含有约30%粗蛋白、约10-20%脂肪,其余为碳水化合物,包括膳食纤维和灰分。典型地的润湿、或含水产品含有(以干物质计)约40%脂肪、50%蛋白质且其余为纤维和灰分。本发明特别涉及猫或狗的食品或补充物,其为固体。
本文中的术语“家庭”狗和“家庭”猫意指狗和猫,特别是Felisdomesticus和Canis domesticus。
可以容易地测定狗或猫中血浆维生素E的水平。测定血浆维生素E水平的代表性例子描述在实施例的引言部分。
可以容易地测定产品(固体或液体或任意其它形式)中维生素E的浓度。这也描述在实施例的引言部分中。
在本发明的第一个方面,对照饮食可以向动物提供15 IU/400kcal的总维生素E。

维生素C是水溶性物质。在家庭猫和家庭狗中它都是经重新合成的。由于它是在体内合成的,以前从未调查过狗和猫中的维生素C补充物的影响。特别是从未调查过猫和狗中的维生素C补充物作为潜在抗氧化剂与维生素E补充物组合时的影响。
本发明显示了通过补充可以增加猫和狗中维生素C的水平。这可以通过补充维生素C之后血浆值增加来证实。维生素C增加可以有利于减少游离基,因此降低了动物中的氧化应力。
根据本发明第一个方面的维生素C可以为任意形式。它可以为液体、半固体或固体。优选它为热稳定形式,例如磷酸钙形式。
维生素C的来源没有限制。优选的维生素C源包括结晶抗坏血酸(任选纯的)、涂布乙基纤维素的抗坏血酸、抗坏血酸的磷酸钙盐、抗坏血酸-2-一磷酸盐或抗坏血酰基-2-单磷酸盐,还有少量二磷酸盐和痕量三磷酸盐、磷酸钙,或者例如鲜肝。
可以容易地测定产品(固体、液体或任意其它形式)中维生素C的水平。这描述在本实施例的引言部分中。
关于与维生素C组合使用维生素E的其它有用的观点为其协同作用的潜能。这借助于维生素E为脂溶性而维生素C为水溶性的事实。已知α-生育酚位于脂质体膜中。抗坏血酸盐和α-生育酚例如在细胞膜或脂蛋白与水之间的界面相互作用。抗坏血酸快速减少膜中的α-生育酚游离基,从而再生α-生育酚。与动物喂食对照饮食时相比,按照本发明第一个方面的维生素C的优选浓度为优选使动物的维生素C水平增加高至约25%(优选25%或更多)的水平,这样(猫或狗)的维生素C消耗的总量为5mg/400kcal饮食。没有达到该增加的维生素C水平仍然被本发明的第一个方面覆盖。按照本发明第一个方面的维生素C的水平包括从10、12、15、17、20、22、25、27、30、32、38、40、42、48至高达约50mg/400kcal饮食。猫的优选水平为10-48mg/400kcal的上面选项,狗的优选水平为12-50mg/400kcal的上面选项。55mg/400kcal以上的水平并未赋予增加的益处,并且通常最好避免。
本发明的第一个方面包括给予一定量的牛磺酸。牛磺酸可以为除上述的补充维生素C之外的,或者代替上述的维生素C。
牛磺酸是在各种动物种中发现的不常见的氨基酸。牛磺酸为猫的必需营养素,与狗不同,猫不能由前体氨基酸合成牛磺酸。人们认为牛磺酸保护细胞膜免遭包括氧化剂的毒性组分。动物饮食中维生素牛磺酸水平增加可以有利于减少游离基,因此降低了动物中的氧化应力,特别是其与本发明的其它组分组合时。
按照本发明第一个方面的牛磺酸可以为任意形式。它可以为粉状、结晶、半固体或液体。
牛磺酸的来源没有限制。优选的牛磺酸来源包括氨基乙基磺酸(C2H7NO3S)。这些来源可以为天然或合成的。
通常产品的加工方法(例如产品是否干燥或罐装)决定了按照本发明的第一个方面给予的牛磺酸的适宜浓度。为了将猫中血浆牛磺酸水平保持在正常范围(>60μmol/l),罐装(湿)饮食每天必需补充至少39mg牛磺酸/kg体重,并且干燥饮食每天补充至少19mg/kg体重。就未经过高温法(例如罐装)的产品而言,本发明的第一个方面提供的优选水平为约80mg/400kcal饮食,以mg/400kcal饮食计的更优选约100,渐增更优选120、150、180、200、220、250、280、300、320、350、400和以上。在例如通过高温加工的产品中,按照本发明的水平优选约380mg/400kcal饮食,以mg/400kcal饮食计的更优选约400,渐增更优选420、450、480、500、520、550、580、600、620、650、700和以上。
可以容易地测定产品(固体、液体或任意其它形式)的牛磺酸浓度。代表性方法描述在实施例的引言部分中。通过膳食补充可以强化牛磺酸在猫体内的状态。通过血浆水平可以测定膳食牛磺酸含量的剂量响应效果。这也描述在实施例的引言部分中。
本发明的第一个方面还可以包括给予一定量的类胡萝卜素。该类胡萝卜素可以为除上面所述的补充维生素C和/或牛磺酸之外的,或者是代替这两者。
类胡萝卜素为一组主要在植物食品,特别是水果和蔬菜,以及以植物为食的动物组织中发现的红色、橙色和黄色颜料。它们为亲脂化合物。一些类胡萝卜素起维生素A前体的作用,一些则不能。该性能与其抗氧化剂活性无关。类胡萝卜素可以起强抗氧化剂的作用。类胡萝卜素以不同程度被不同动物种吸收。类胡萝卜素可以分成两大类:以胡萝卜素为基础的那些和以叶黄素为基础的那些(包括氧化化合物)。普通的类胡萝卜素包括:β-胡萝卜素、α-胡萝卜素、番茄红素、黄体素、玉米黄质和虾青素。未证实类胡萝卜素为猫或狗膳食中的主要营养素。与人和狗不同,猫不能将前体β-胡萝卜素转化成活性维生素A形式,这是由于在猫的肠粘膜中缺少该转化所需的酶(猫没有裂解胡萝卜素分子所需的二氧化酶。
本发明显示了类胡萝卜素可以被家庭猫和狗吸收(从而使血浆浓度增加)并且可有利于降低氧化应力。而且,本发明已证实,类胡萝卜素在加入商业化产品之后可以被吸收。如上所述,本发明第一个方面的组分可以协同作用。维生素E能够防止β-胡萝卜素氧化,可以具有保护β-胡萝卜素的效果。维生素E被认为可防止β-胡萝卜素的化学键被氧化。

该第三个方面也具有第一和二方面的优选特征。
本发明的第四个方面是将本发明第三个方面的狗或猫食品用于预防或治疗狗或猫的低抗氧化状态。
该第四个方面也具有第一至三个方面的优选特征。

在免疫攻击期间,提高的免疫反应对动物有帮助,并且抗原特定地用于年轻动物,这是由于年轻动物不可能具有发育完全的免疫系统。由于经常对年轻动物进行预防接种,因此本发明提供了一种接种时可以通过动物赋予提高的免疫反应的方式。本发明特别用于在接种瘟热、细小病毒和/或腺病毒疫苗抗原之前喂食狗。本发明特别用于向猫喂食抗猫泛白细胞减少症、猫杯状病毒和/或猫疱疹病毒的疫苗或组合疫苗时给予。在免疫攻击之前喂食的适宜时间长度取决于正在讨论的动物和该免疫攻击。可以一致地喂食食品。在免疫攻击之前的8、6、4、2和1周是适宜的。更长时间也是适宜的。

本发明的第7个方面提供了维生素E以25 IU/400kcal或以上的量加入作为狗或猫中体内抗氧化剂的食品中的用途。1-6个方面的所有优选特征在第七个方面也具有。特别是维生素E的量可以与本发明第一个方面设定的维生素E的优选量相同。

血浆维生素E

血浆中类胡萝卜素的测定

维生素C-食品产品
抗坏血酸是经酶氧化成脱氢抗坏血酸,它与邻亚苯二胺缩合成荧光喹噁啉衍生物。通过逆相HPLC将后者与干扰化合物分离,并用荧光计检测。
维生素A和E食品产品

自由牛磺酸-食品产品

通过如下分析自由牛磺酸:取出样品,加入稀盐酸。然后将其浸软并转移到定容烧瓶中。然后取出少量并加入磺基水杨酸,将蛋白质沉淀。然后将样品离心并将上清液过滤出。所得溶液与丹磺酰氯反应并使用荧光检测通过HPLC分析。
类胡萝卜素-食品产品
取出20-25g样品用于分析。在90℃、氮气以及用连苯三酚作为抗氧化剂下,样品用28%乙醇氢氧化钾皂化30分钟。冷却之后,将皂化的提取物用2×250ml混合醚(石油醚/乙醚1∶1)提取,将有机相用水冲洗直至中性。在真空下用BHT作为抗氧化剂在35℃将醚提取物蒸发,残余物再溶于HPLC移动相中。使用逆相HPLC在450nm下使用紫外线检测测定这些类胡萝卜素。
除了给出的这些试验工作之外,本发明通过给猫喂食该抗氧化合剂之后减少猫红细胞的渗透性脆性从而成为改善健康的指示剂。
测定红细胞经受渗透应力的能力。
该方法涉及将洗过的红细胞再悬浮于具有不同NaCl浓度的溶液中;它们经培养,然后分离。根据其渗透性脆性血红蛋白从这些细胞中释放。结果显示,喂食本发明抗氧化合剂的猫的红细胞显示了更大的渗透应力耐性,同时需要显著低浓度的NaCl诱导相同水平的溶血作用。红细胞耐受渗透应力情况的能力为生理状态提高的指示剂。
实施例1
维生素E
将一组8只狗供养营养上完全的饮食(见对照段的代表性饮食)持续6周,然后接受两种补充水平的维生素E(下面的α-生育酚醋酸酯(表1))之一。

实施例2
猫的抗氧化剂补充
维生素C
将4组8或9只猫口服补充4种不同水平的维生素C,同时供养营养上完全的饮食(见对照饮食段)。
基线血浆维生素C水平证实,在补充之前4组之间没有显著差异。
正如在图2中可以看到的,7天内所有4组的血浆维生素C水平明显增加至补充之前的基线之上。
尽管更高维生素C补充导致更大的血浆值,但是在4组之间的这种增加没有明显差异。
补充结束之后,血浆维生素C水平回到基线。
这些数据证实,猫的抗氧化剂状态可以通过相对低水平的饮食维生素C补充增加。
实施例3
维生素E
已评价了典型膳食维生素E水平对猫的维生素E状态的影响以及膳食补充的影响。
测定供给具有不同膳食维生素E水平的2种商业化营养完全的猫饮食(参见对照饮食段)的2组12只猫的血浆维生素E浓度。表2中所示的血浆水平(2周间隔,一式两份测定)证实,供给4周较高维生素E水平的饮食的猫具有相对增加的维生素E状态。

为了测定补充的效果,将口服维生素E补充物(α-生育酚醋酸酯)给予一组12只猫,同时供给营养完全的饮食。
补充的效果可以见图3。补充之后血浆维生素E水平显著增加,在补充约4-6周之后达到饱和。
因此,膳食维生素E补充可以强化猫的抗氧化剂状态。
实施例4
猫中总血浆抗氧化剂;正常范围和年龄的影响。
家庭猫中的总抗氧化剂状态以前从未报道过。该研究的目的是验证一种测定猫中总抗氧化剂状态的方法、测定健康成年猫中的正常总抗氧化剂的范围并研究年龄对总血浆抗氧化剂状态的影响。
用Randox Laboratories Limited生产的比色测定试剂盒(NX2332)验证猫中总抗氧化剂状态的测定。通过猫血浆样品的重复分析和商业化质量控制(QC)材料(PAR 721013和PAR 721014;Bio-stat Diagnostics)的重复分析确定测定精度,内和中间变异系数证实血浆样品的分光光度测定的可接受的变异CV<10%且QC CV<5%。猫血浆的稀释在测定中产生线性反应。
评价年龄对血浆抗氧化剂水平的影响。供给营养完全猫食品(参见饮食对照段)的134只猫参与该研究。在来自69只雄性和65只雌性年龄在6个月至14岁之间的家庭短毛猫的单个血浆样品中评价所有抗氧化剂状态。所有方式都引入±标准偏差(SD)。
结果显示,在约6岁龄时存在抗氧化剂状态的过渡期。Newman-Keuls多样范围试验显示,6岁龄猫的总血浆抗氧化剂状态显著(p<0.01)高于6.5岁龄的(参见表3)。不清楚该年龄的抗氧化剂状态衰退是否标志着对疾病的更大易感性,或者该年龄的抗氧化剂状态是否影响猫的寿命。

实施例5
狗中总血浆抗氧化剂和超氧化物歧化酶
有关家庭狗中抗氧化剂状态的了解很少。本研究的目的是验证一种狗中总血浆抗氧化剂(TPAO)和红细胞SOD(超氧化物歧化酶)活性的正常范围的测定方法。
将Randox laboratories Limited生产的比色测定试剂盒用于确定狗中TPAO状态(试剂盒号:NX2332)和SOD状态(试剂盒号:SD 125)。就TPAO而言,通过一式两份分析狗血浆样品和商业化质量控制(QC)材料(PAR721013和PAR 721014:biostat Diagnostics)确定测定精度。内和中间分析变异系数(CV)证实分光光度测定的可接受的变异,血浆样品CV<10%和QC<5%。通过一式两份分析狗红细胞样品确定SOD状态测定精度。内和中间分析变异系数(CV)是指红细胞样品的分光光度测定的可接受的变异CV<10%。
在9个月至16岁之间年龄段的30条雄性和20条雌性狗中评价TPAO和红细胞SOD状态。代表了10个品种。所有狗临床上都正常,并已长期供给各种营养完全的饮食。年龄与TPAO或SOD活性之间没有明显关系。狗不等价代表横跨年龄组(数据未显示),进一步的工作以评价观察是否得到更大样品和更好年龄扩散的支持。在性别之间平均TPAO状态(雄性:0.719±0.135,雌性:0.786±0.101)或SOD活性(雄性:1275.41±264.46,雌性:1267.61±166.34U/g Hb)没有看到显著差异。然而,Bartletts测定显示了雄性(6994.0)和雌性(27670.3)狗中SOD活性方差之间的显著差异(p<0.05)。
对具有5或更多代表性的这些品种分析显示,在与性别无关的品种之间对SOD活性和TPAO状态存在显著差异(p<0.01)。Newman-Keuls多样范围测定显示,Beagles比Labradors和Yorkshire Terriers具有明显低的平均SOD活性(p<0.05)和平均TPAO状态(p<0.01),表4。

这些数据显示,它不足以假定狗中正常SOD和TPAO范围的单个值。在这种需要两个值的情况下,一个为Beagles的,一个为两个品种之一的。Beagles中TPAO状态和红细胞SOD活性的平均范围的95%可靠间隔分别为0.569±0.067mmol/1和1084.9±97.27U/g Hb;并且两个品种中另一个为0.822±0.047 mmol/1和1316.2±88.03U/g Hb。
实施例6
使用全自动比色测定评价猫和狗中的总抗氧化剂状态
该研究的目的是验证用Randox Laboratories Ltd生产的比色测定试剂盒测定猫和狗中总抗氧化剂状态。其次,该验证测定用于评价猫和狗中总抗氧化剂状态随年龄的变化并以其它生物化学和血液参数比较这些变化。该测定将2,2’-连氮基-二-(3-乙基苄噻唑啉氨基磺酸盐)(ABTS)与过氧化物和H2O2反应产生残基阳离子ABTS+,它可以在600nm下测定。样品中存在的抗氧化剂使得该颜色的产生抑制到与其浓度成比例的程度。使用Randox Laboratories提供的方案在Cobas Mira分析器(RocheDiagnostics)上进行该测定。测定精度通过对猫和狗血浆样品和商业化质量控制(QC)材料的一式两份分析来确定。内分析和中间分析CV都低于QC材料的5%,并且对猫和狗血浆样品都低于10%。猫和狗血浆的稀释在测定中产生线性反应。对不同年龄的家庭短毛猫和几个血统品种的狗进行包括总抗氧化剂状态的常规血液学和生物化学图谱。猫中抗氧化剂状态增加高至2月龄,然后显示接下来的衰退。性别差异不明显。小于1岁的雌性狗显示抗氧化剂状态轻微降低,否则随年龄没有明显变化。多重回归分析证实,猫中抗氧化剂状态、白蛋白、天冬氨酸盐氨基转移酶和钙之间的关系,狗中抗氧化剂状态、磷酸盐和丙氨酸氨基转移酶之间的关系。确认结果是可以接受的,并且该测定适合确定猫和狗中总抗氧化剂状态。
实施例7
维生素C补充物对健康成年猫中血浆状态的影响
维生素C为体内主要水溶性抗氧化剂,它可以延迟或抑制氧化,特别是在细胞外液中很重要。然而,以前从未研究过猫(Felis domesticus)对不同维生素C膳食水平的反应。本研究的目的是确定在健康成年猫中补充维生素C对血浆状态的影响。
通过逆相高效液相色谱法测定33只猫的血浆维生素C浓度。接着通过分层随机抽样将这些猫分成4组,确信这4组的基线血浆维生素C水平之间没有显著差异(ANOVA p>0.05)。将日维生素C补充物(结晶L-抗坏血酸,ICN药剂,UK,为明胶胶囊,Analytical Supplies Limited,UK)连续21天口服给这些猫。给予这4组猫3.5、7.0、10.5或者21.0mg维生素C/天。在补充的第7、14和21天以及补充后第21天测定血浆维生素C水平。所有猫保持营养完全的罐装饮食,其中维生素C含量为11.6mg/1.6MJ。
在日补充之后的第7天在所给的所有4个膳食水平下这些猫的血浆维生素C水平都增加(成对t-test p<0.05),并且补充期间保持这些水平,表5。尽管当与较低膳食水平的那些比较时,维生素C的膳食补充水平越高,血浆值达到越大,但在膳食组之间的血浆增加没有显著差异(使用常规线性模型重复测定多个因素ANOVA p<0.05)。在补充之后第21天,所有猫的血浆水平回到基线水平。

该铜合酶,血浆铜蓝蛋白对身体具有许多功能作用。作为贮藏铜的蛋白质,血浆铜蓝蛋白帮助游离铜离子抗催化能力。游离铜可以通过单电子(基团)转移加速自动氧化反应,还与过氧化氢反应形成高反应性的氢氧根,它可以引起细胞分裂。当外伤时,血浆铜蓝蛋白还促进铁从其预氧化剂亚铁形式转化成三价铁离子。作为急性相蛋白质,血浆铜蓝蛋白可以指示发炎或感染,并因此用于与血清铁蛋白结合测定铁贮藏。
基于对苯二胺为底物的Sunderman F.W.和Nomoto,S.(1990)Clin.Chem 16,903方法验证了一种测定猫和狗中血浆铜蓝蛋白氧化酶活性的比色法。在单一运转下将两只狗和两只猫的血清样品分析10次。测定狗样品的内分析变异系数为1.94%和2.95%,猫样品的内分析变异系数为1.81%和3.94%。对10个单独天的相同样品,测定狗样品的中间分析的变异系数为8.21%和7.01%,猫样品的中间分析的变异系数为6.88%和9.35%。因此获得可接受水平的内和中间分析的变异性。之后,评价血清和血浆样品中血浆铜蓝蛋白浓度的差异。在每一种中测定血浆和血清样品之间差异不显著。因此为了降低所需样品的总体积,收集血浆样品。
为了确定正常范围,从102只健康狗(混合种、年龄和性别)和54只健康家庭短毛猫(混合年龄和性别)获得血浆样品。测定狗和猫的平均血浆铜蓝蛋白浓度分别为9.28IU/L(SD 3.03IU/L)和10.90IU/L(SD 3.34IU/L)。使用这些值,确定狗的正常范围(平均±2SD)为3.22IU/L-15.35IU/L,猫的为4.22IU/L-17.58IU/L。确定的狗的正常范围与前面报道的(Solter P.F等人(1991)Am J Vet Res 52,1738,1991)一致。以前从未确定过猫的正常范围,然而它的与狗的相似,这暗示在这两个种之间血浆铜蓝蛋白循环水平有很少的差异。在本研究期间确定的正常范围可用于评价猫和狗的健康状况。
实施例9
犬铁蛋白:分析验证和血清的正常范围
铁蛋白在身体的抗氧化剂防御系统中起着重要作用。作为铁的高亲和力贮藏蛋白,铁蛋白使铁保持安全结合形式,它防止了反应性亚铁离子参与Fenton反应,该反应可以导致氧化损伤。在正常健康铁蛋白中,种特异性蛋白质,如果在血液中发现其浓度,反映身体铁贮藏的浓度并与其它参数结合可用于评价体内铁状况。为了确定狗中循环铁蛋白水平,开发了一种酶链免疫测定(ELISA),并采用使用单克隆抗体的Weeks,B.R.等人(1988)Am J Vet Res 49,1193)(1988)方法。该测定验证之后,确定狗的正常范围。
在0-40ng/ml的范围内,铁蛋白标准为线性(最小平方回归分析,r=0.997)并加入到犬血清的纯化铁蛋白的回收率为97.7%。由测定两个血清样品的铁蛋白浓度12次获得的内分析变异系数为8.2%和6.6%。在单独天两个血清样品测定10次的中间分析变异系数为16.6%和16.2%。
为了确定正常范围,血清样品从96个测定铁蛋白用的混合性别和5个不同种的健康狗获得。每种样品分析三次或四次,将这些值的平均值用作样品的铁蛋白浓度。血清铁蛋白浓度的范围为67.20-621.07,平均值为371.62ng/ml(SD 102.85ng/ml)。这些数据正常分布。这些结果证实,可以良好重复性和再现性地测定狗的血清铁蛋白。获得的值将用作未来研究的正常范围,并且可以提供一种测定狗中铁贮藏水平的有用方法。
实施例10
补充膳食生育酚将使正常范围之外的犬血浆值增加
维生素E是8种自然存在的分子—4种生育酚和4种生育三烯酚的共同名称。不同形式的生物活性大致与其抗氧化活性有关,相对过氧化清除反应性的次序为α>β>γ>δ-生育酚。通常提供过量需求的营养物不会带来任何测量益处。成年狗中维生素E的最小需求已确定为2.5IUα-生育酚/400kcal饮食(Nutrient Requirements of Dogs(1985)NationalResearch Council(U.S.)National Academy Press Washington DCISBN;0-309-03496-5)。制备的宠物食品典型地含有高达10倍的该量,但通过宠物食品中的维生素E的正常范围,血浆浓度趋于恒定。在评价补充的维生素E对血浆反应的影响时,将6只成年给予营养完全(见饮食对照段)罐装狗食品(维生素E含量为8.2mg/400kcal(8.2IU/400kcal)的狗,提供6周的补充物,其相当于100IU维生素E/天。对照组6只狗保持在相同条件下,但是未接受补充膳食。在补充期间或补充之后的4周内监控血浆α-生育酚。
血浆α-生育酚浓度示于表6中。处理组的平均基线α-生育酚水平与对照组的没有明显差异(p=0.43)。补充2周后,与对照(p=0.002)和其基线(p<0.001)相比,处理组的平均值明显增加。在第56和70天(补充后第14和28天)进行测定,显示其水平明显高于基线。这些结果清楚地说明,典型地在制备的宠物食品中发现的水平之上的膳食维生素E可以使狗中血浆状态上升,补充之后这可以维持几周。为了使血浆饱和,很显然需要喂食比目前在制备的宠物食品中发现的那些更高的浓度。只要维生素E的抗氧化能力有效,可以审慎地增加狗中维生素E状态,以便最大化地防止自由基损伤和相关退化病的机会。

健康狗中典型血浆维生素E范围
维生素E是8种具有相同生物活性的不同生育酚和生育三烯酚的共同名称。这8种中,α-生育酚为生物和化学上活性最大形式的维生素E,1mgα-生育酚相当于1IU维生素E。维生素E为体内的有效的抗氧化剂,它主要留在生物膜中,在其中它防止膜磷脂免受过氧化破坏。维生素E还抑制维生素A氧化,因此还防止维生素A缺乏。已确定成年狗中对维生素E的最小需求为2.5IU α-生育酚/400kcal饮食(NutrientRequirements of Dogs(1985)National Research Council(U.S.)National Academy Press Washington DC ISBN;0-309-03496-5)。然而,关于健康成年狗和幼犬中的正常维生素E血浆值只有少量信息。为了确定健康成年和幼犬中典型基线血浆α-生育酚水平,在喂食一定罐装和干燥营养完全制备的宠物食品的狗中进行了一系列研究(参见饮食对照段),表7。根据体重喂食饮食(110 kcal/kg0.75)。单个血浆α-生育酚水平的范围为9-39.2mg/l,平均值为21.1mg/l。供养不同饮食的狗的组的平均基线α-生育酚水平之间没有显著差异(p=0.24)。这些数据说明,在幼犬和成年狗中典型α-生育酚水平相似,并且在制备的宠物食品中发现的常规维生素E水平内,血浆生育酚水平呈现相似范围。

健康成年狗锻练使血浆TBARS-氧化应力指示剂增加
通过预柱衍生化的HPLC测定的血浆TBARS(硫代巴比妥酸反应性物质)为体内脂质过氧化的公知标记物。本研究的目的是调查狗(Canisfamiliaris)锻练一次对氧化应力指数的影响。
在该试验之前3个月和整个该试验过程中将一组14只混合种和年龄的狗给予营养完全的商业化干饮食。在急性20分钟一次围场锻练之后立即通过测定形成的丙醛(MDA)作为TBARS定量脂质过氧化程度。该分析是按照如下所述方法测定的:Bird,R.P.和Draper,H.H.(1984),Methods inEnzymology.105:299-305(1984)。这些结果证实:锻练之后血浆TBARS(Paired TTest p<0.05)显著增加(22%)(锻练前0.74μM±0.2,锻练后0.92μM±0.2)。为了检查锻练后存在的增加是浓缩血液体积的直接结果,测定PCV(包裹的细胞体积)和血浆白蛋白。结果证实锻练前和锻练后没有明显差异(p>0.05),因此从该研究的观察暗示,体内增加的脂质过氧化以锻练诱导的氧化应力的直接结果存在。现有文献提出,氧化损伤的特定位置为细胞膜,由此在高氧化应力条件下过氧化残基(RO2)增多。这种观察推断在健康成年狗中膳食脂质相抗氧化介入的潜在作用。
下面描述动物抗氧化状态的其它例子和本发明抗氧化合剂对动物的影响。
实施例13
家庭猫中膳食类胡萝卜素吸收
现发现,将抗氧化剂如β-胡萝卜素和番茄红素加入商业化猫食中将明显增加猫对类胡萝卜素的吸收,尽管它们的代谢适应食肉。在该研究中,使用具有相同基本配方的相同批次的原料生产三种罐装猫食。对照饮食,饮食A,具有3.39MJ/kg的可代谢能量(ME)含量,且β-胡萝卜素和番茄红素含量>0.1mg/1.67MJME。饮食B补充有红色棕榈油(3.70MJME/kg),β-胡萝卜素含量为0.36mg/1.67MJME并且番茄红素含量>0.1MG/1.67MJME;饮食C补充有番茄浮石(3.54MJME/kg),且β-胡萝卜素含量>0.1MG/1.67MJME,番茄红素含量为0.9MG/1.67MJME。
该研究选择了2组各5只健康猫。组1中的猫的平均年龄为8.72岁(SD2.1岁),包括3个雄性和2个雌性。组2中的猫的平均年龄为7.4岁(SD1.7岁),包括2个雄性和3个雌性。该研究的所有猫以前都给予各种市购营养完全且制备好的猫食,然后给予几天的饮食A,使其喜欢该饮食。在本研究的第8天,测定基线血浆类胡萝卜素水平。接着,将组1猫喂食饮食B,组2猫喂食饮食C,持续5天。在本研究的第13天,以相同方式重复抽取血浆作为基线样品。通过高效液相色谱法进行分析。
给予饮食B的组1猫中β-胡萝卜素浓度明显增加(平均17.62ng/mL,SD2.50ng/mL)至基线值(平均6.35ng/mL,SD 3.23ng/mL)之上。在该组中,血浆番茄红素浓度保持在检测的限度之下。在给予饮食C的组2猫中,基线水平(平均5.30ng/mL,SD5.78ng/mL)和喂食后水平(平均6.61ng/mL,SD2.83ng/mL)的血浆β-胡萝卜素浓度没有明显改变。然而,组2中猫显示,血浆番茄红素浓度从基线水平的0ng/mL至喂食后水平的平均14.6ng/mL,SD7.25ng/mL明显增加。
因此,本研究证实了,抗氧化剂的天然来源如β-胡萝卜素和番茄红素加入到市购猫食之后将使得猫对类胡萝卜素的吸收明显增加,尽管其代谢适应食肉性。循环抗氧化剂时的这种增加将赋予猫抗氧化剂状态的有益效果。类胡萝卜素以低膳食浓度在猫中的这种增加的吸收至今还从未看到过。
实施例14
母乳可能不足以使发育中的小猫的抗氧化剂状态最大化。
就免疫功能而言,抗氧化剂的效果同样可以适用于因年龄具有受损免疫功能的动物如成长动物以及接受免疫攻击的那些动物。以下研究的结果显示,母乳可能不足以使发育中的小猫的抗氧化剂状态最大化,反而可能使小猫对氧化应力的易感性增加。因此,可以发现,以膳食给小猫补充抗氧化合剂同样显示出与接受抗氧化合剂补充物的幼犬中发现的抗体反应时间的提高。
哺乳动物抵抗氧化损害的能力既取决于其内部抗氧化剂防御系统又取决于饮食提供的整个抗氧化剂状态。测定动物抗氧化剂状态的一种方式是评价其总血浆抗氧化能力(TPAO)。已将Randox Laboratories Ltd.生产的比色测定试剂盒用于猫中测定TPAO的正常范围。在与其血液浓度成比例的试验中血样中存在的抗氧化剂将使色彩产生受到抑制。
进行本研究,以确定健康小猫中TPAO的正常范围并确定年龄与TPAO之间是否存在任意关系。适用上述方法分析样品,适用ANOVA比较这些结果。结果示于下表7中。

试验设计:

通过促细胞分裂剂诱导的淋巴细胞转化测定(MILT)评价外周血液单核细胞(PBMC)增殖反应
在Histopaque 1077(Sigma)上通过密度梯度离心将周围血液单核细胞从肝素化血液中分离。这些细胞用磷酸盐缓冲盐水(PBS)冲洗2次,用补充有10%热失活的胎牛血清、1%青霉素/链霉素和2%丙酮酸钠的RPMI-1640(Dutch modification)冲洗1次。通过台盼蓝排除试验(Sigma)评价细胞生存能力。
将细胞一式三份以1×105/井在96孔平底微量滴定平皿中在37℃下用植物凝集素(PHA)(5g/ml)(Murex)、刀豆球蛋白A(Con A)(7.5μg/ml)和美洲商陆促细胞分裂剂(PWM)(1μg/ml)(Sigma)培养96小时。在培养的最后18小时内以每分钟加入的[3H]-胸腺嘧啶核苷的数量(CPM)(0.5μCi/井)测定增殖。
通过流式细胞计分析淋巴细胞子集
CD4和CD8阳性细胞为猫免疫学中最公知特征的淋巴细胞子集,适宜全部的这些细胞说明了健康免疫系统。既使用提纯的淋巴细胞又使用全血并选择各种单克隆抗体(Mabs)进行该测定。
结果

材料和方法
动物
1)动物类型
该研究包括了40只年轻成年(0.8-3.3岁)和年长(6.5-12.5岁)、纯种(Labradors、Beagles、West Highland white terrier、Newfoundland和Golden Retriver)、以及性别混合的狗(无伤、阉割/切除卵巢)。
2)居住处
将狗成对分组并且都可以从其栅栏到外面环境。在白天和晚上所有狗都可以进入户内。来自相同处理组的狗住在一起,防止因食粪癖(粪便消耗性疾病)交叉污染。温度控制在22℃,具有自然光循环。允许狗保持其日常定期锻练程序。
3)喂养方案
在开始本研究之前所有狗都给予1周完全且平衡的干燥狗饮食。适应1周之后,2/3的狗作为对照组喂养基本饮食,而剩余1/3转为测定饮食。喂食每只狗的食品量根据保持体重进行调节。在所有时间所有狗都可以使用新鲜水。

对照饮食段
营养完全的饮食
食用的完全饮食,特别是营养完全的宠物食品(或饮食)为满足单个动物生活方式和生命的所有营养需求的一种饮食。
该饮食或食品可以按照本领域已知的任意方法制备,例如Waltham Bookof Dog and Cat Nutrition,Ed.ATB Edney,Chapter by A.Rainsbird,entitled"A Balanced Diet"in pages 57 to 74,Pergoren Press Oxford。

Claims (33)
1.一种增加猫或狗中血浆维生素E水平的方法,该方法包括步骤:向所述猫或狗给予足够增加血浆维生素E水平量的维生素E。
2.如权利要求1的方法,还包括向所述猫或狗给予一定量的维生素C。
3.如权利要求1或2的方法,还包括向所述猫或狗给邓一定量的牛磺酸。
4.如权利要求1或2的方法,还包括向所述猫或所述狗给予一定量的维生素C和一定量的牛磺酸。
5.如权利要求1-5任一的方法,还包括向所述猫或狗给予一定量的类胡萝卜素。
6.如权利要求2-6任一的方法,其中组分同时、分别或相继给药。
7.一种狗或猫食品,它向所述动物传递的成分浓度足够增加动物抗氧化剂状态。
8.一种狗或猫食品,它提供了浓度水平为25IU/400kcal饮食或之上的维生素E。
9.如权利要求10的狗或猫食品,提供了浓度水平为10mg/400kcal或之上的维生素C。
10.如权利要求10的狗或猫食品,提供了浓度水平为80mg/400kcal或之上的牛磺酸。
11.如权利要求10的狗或猫食品,提供了浓度水平为10mg/400kcal或之上的维生素C并提供了浓度水平为80mg/400kcal或之上的牛磺酸。

13.如权利要求7-12任一的狗或猫食品,用于预防或治疗狗或猫中的低抗氧化剂状态。
14.如权利要求7-12任一的狗或猫食品,用于预防或治疗具有应力成分的任意紊乱。
15.如权利要求14的狗或猫食品,其中这些紊乱为以下任意一种或多种:癌症、老化、心脏病、动脉粥样硬化、关节炎、白内障、炎症性肠病、肾病、肾衰竭、神经变性病或受损性免疫。
16.如权利要求7-14任一的狗或猫食品,用于治疗或帮助猫或狗响应免疫攻击。
17.如权利要求16的狗或猫食品,其中免疫攻击为接种。

20.维生素E在生产预防或治疗狗或猫中低抗氧化剂状态的药物中的用途。
21.维生素E在生产预防或治疗具有氧化应力成分的任意紊乱的临床饮食中的用途。
22.如权利要求21的维生素E的用途,用于在猫或狗中使响应免疫攻击的免疫反应保持、最佳化或提高。
23.维生素E加入食品内作为狗或猫的体内抗氧化剂的用途。
24.一种制备权利要求7-14或21中任一的食品的方法,该方法包括将至少两种食品组分混合在一起。
25.一种狗或猫食品,含有浓度为15mg/400kcal或之上的维生素C。
26.维生素C在生产用于预防或治疗具有应力成分的紊乱的狗或猫食品中的用途。
