根据几项研究,了解到在细胞生长和分化,创伤愈合,和细胞粘附过程中涉及βig-h3,尽管这些功能的潜在的机制还不清楚。然而,在各种组织中,在形态发生和与细胞和胞外基质蛋白的相互作用过程中,βig-h3似乎扮演着重要的角色。
通过几项研究,提供了与βig-h3在介导细胞附着和脱附过程中的角色有关的一些证据。例如,发现在无血清培养基中,纯化的βig-h3蛋白促进皮肤成纤维细胞的附着和铺展,而抑制A549,HeLa和Wi-38细胞的粘附。特别地,已知βig-h3具有抑制肿瘤细胞生长的活性,并影响群体形成和形态学。该抑制活性被一项报道所证实,即在裸鼠中,将βig-h3表达质粒转染到CHO(中国仓鼠卵巢)细胞中,导致细胞分化和这些细胞形成肿瘤细胞的能力明显降低。而且,在发现对创伤施用药物有效量的βig-h3,使得细胞,尤其是成纤维细胞覆盖并粘附在创伤部位的基础上,发展了一种创伤愈合方法。因此,在各种细胞系中由TGF-β诱导的βig-h3,一种细胞粘附分子,在细胞生长,细胞分化,创伤愈合,形态发生和细胞粘附过程中扮演非常重要的角色(Rawe,I.M.等.,眼科视觉科学研究(Invest.Ophthalmol.Vis.Sci.)38,893,1997;Lebaron,R.G.等.,J皮肤学研究(Invest.Dermatol.)104,844,1995)。
βig-h3包含四个具有内部同源性的140个氨基酸重复,即fas-1结构域。在各种物种,包括哺乳动物,昆虫,海胆,植物,酵母和细菌的分泌蛋白或膜蛋白中,发现内部重复结构域具有高度保守的序列。包含保守序列的蛋白实例有periostin,成束蛋白I,海胆HLC-2,藻类-CAM和分枝菌属MPB70。在这些蛋白中的保守结构域(下文指“fas-1”)由约110-140个氨基酸组成,该氨基酸具有两个高度保守的支链,H1和H2,每个支链约有10个氨基酸(Kawamoto,T.等.,生物化学生物物理快报(Biochem.Biophys.Acta.)1395,288,1998)。
发现在βig-h3、periostin和成束蛋白I中有四个fas-1结构域,在HLC-2中有两个fas-1结构域,在MPB70中仅有一个fas-1结构域。尽管这些蛋白的功能没有清楚地阐明,但已知它们中的一些充当细胞粘附分子。例如,报道了βig-h3,periostin和成束蛋白I分别介导成纤维细胞,成骨细胞,和神经细胞的粘附。另外,公开了藻类-CAM是存在于藻类团藻属(algaeVolvox)胚胎中的细胞粘附分子(LeBaron,R.G.,等.,J.Invest.Dermatol.104,844,1995;Horiuchi,K.等.,骨矿质研究(J.Bone Miner.Res.)14,1239,1999;Huber,O.等.,EMBO J.13,4212,1994)。
最初,认为C末端的RGD基序介导βig-h3的细胞附着活性。然而,一些研究结果揭示RGD基序不是促进软骨细胞铺展所必需的,并且通过羧基末端加工缺失RGD基序的成熟可溶βig-h3能抑制细胞粘附,由此产生了一个结论,即对于βig-h3的介导细胞附着活性而言,βig-h3的RGD基序不是必需的。另外,近来已报道了通过整联蛋白α1β1,βig-h3促进成纤维细胞的铺展,而βig-h3的RGD基序对于βig-h3的介导细胞粘附的性能不是必需的。根据最近的报道,不考虑RGD基序,βig-h3特异地结合整联蛋白以增强细胞粘附和铺展(Ohno,S.等.,Biochm.Biophys.Acta1451,196,1999)。而且,发现βig-h3的保守肽H1和H2对βig-h3介导的细胞粘附无影响。综合这些结果,表明对于βig-h3的细胞附着活性必需的氨基酸存在于除了H1和H2区域以外的某个区域。基于不仅在βig-h3的重复fas-1结构域之间,而且在其它蛋白的fas-1结构域之间存在同源性的计算机研究揭示,除了H1和H2肽,存在几个高度保守的氨基酸,提示在细胞附着活性中涉及保守的氨基酸序列的可能性。
按照本发明,已知在细胞粘附过程中扮演重要角色的βig-h3的结构域的第二或第四结构域的任何一个被鉴定为分子的细胞粘附所必需的最小结构域。基于这些发现,按照本发明,包含必需功能结构域的重组蛋白也被鉴定为对创伤愈合是有效的。
近来创伤愈合研究已被细分为细胞生物学和分子生物学,并且创伤愈合的促进作用已经越来越多地应用到各种临床领域。然而,创伤愈合的细胞生物学和分子生物学机制仍然不清楚。根据迄今公开的发现,创伤愈合是对外伤的组织反应,通过复杂的生物学过程,包括趋化性、细胞分化和复制,基质蛋白合成,血管发生,和创伤恢复,导致组织修复(Steed,D.L.,等.,Clin.Plast.Surg.25,397,1998)。
生长因子是在创伤愈合早期阶段出现的有代表性的物质,并且控制随后的创伤愈合过程。对创伤愈合的全部阶段具有强烈影响的生长因子控制细胞的生长,分化和新陈代谢,并且通过它们的趋化性能组织再生创伤环境,趋化性能吸引在炎症和组织修复,细胞增殖,刺激血管发生和胞外基质的合成和降解中涉及的各种细胞类型。PDGF(血小板衍生的生长因子)吸引成纤维细胞至创伤部位,并刺激它们增殖,并且转化生长因子-β(TGF-β)使得它们产生胶原。PDGF对在创伤愈合、刺激血管发生、重塑和收缩中涉及的大多数细胞具有趋化性,并激活创伤愈合细胞(Mustoe,T.A.等.,临床研究杂志(J.Clin.Invest.)87,694,1991;Lepisto,J.等.,外科研究杂志(J.Surg.Res.)53,596,1992)。EGF(表皮生长因子)刺激角化细胞迁移,血管发生和肉芽组织发生,并激活角化细胞和成纤维细胞的有丝分裂(Franklin,J.D.等.,Plast.Recsonst.Surg.64,766,1979;Buckly,A.等.,美国国家科学院院刊(Proc.Natl.Acad.Sci.)美国,82,7340,1985).bFGF(碱性成纤维细胞生长因子)刺激血管发生,上皮形成,和胶原纤丝沉积,并以各种形式与肝素结合以执行相关功能(Tsuboi,R.等.,实验医学杂志(J.Exp.Med.)172,245,1990;Kinsnorth,A.N.等.,英格兰外科学杂志(Br.J.Surg.)77,409,1990)。IGF(胰岛素样生长因子)增强细胞分化。VEGF(血管内皮生长因子)增加血管透性(vasopermeability)并促进内皮细胞有丝分裂(endothelial mitogenesis)。
在创伤愈合过程中涉及的生长因子和细胞因子中,TGF-β是最有代表性的。在哺乳动物中以三种形式(TGF-β1,TGF-β2和TGF-β3)存在,细胞因子在各种细胞的生长和分化过程中扮演重要的角色,并具有各种复杂的功能,包括控制细胞生长,调节免疫反应,刺激骨发生,诱导软骨特异的大分子,和促进创伤愈合(Bennett,N.T.等.,Am.J.Surg.165,728,1993)。TGF-β在创伤愈合期间在上皮细胞(ephithelium)中出现,认为在再形成上皮(re-epithelialization)期间其刺激整联蛋白在角化细胞内的表达。在近来对TGF-β表达的研究中,揭示了TGF-β3 mRNA在正常皮肤和急性和慢性创伤的上皮细胞中表达,而TGF-β1 mRNA在正常皮肤和慢性创伤的上皮细胞中不表达,但在再生自急性创伤的上皮层中表达,TGF-β2 mRNA在任何地方都不表达(Schmid,P.等.,病理学杂志(J.Pathol.)171,191,1993)。基于这些作用,即使它们的机制未被明确地确定,仍期望TGF-β在再形成上皮过程中扮演较重要的角色。
βig-h3的表达是被TGF-β正调节的,提示在TGF-β的某些信号的调节过程中涉及βig-h3。报道了用βig-h3表达质粒转染的CHO(中国仓鼠卵巢)细胞显示降低的肿瘤发生能力(Skonier,J.等.,DNA细胞生物学(DNA Cell Biol).13,571,1994)。相反,在地塞米松处理的干细胞、一些肿瘤细胞和从遭受局部肢骨纹状肥大的骨肥厚的皮肤损伤部位培养的成纤维细胞中βig-h3的表达被负调节。还报道了βig-h3作为骨发生的负调节剂(Genini,M.等.,国际癌症学杂志(Int.J.Cancer)66,571,1996;Schenker,T.等.,实验细胞研究(Exp.Cell.Res.)239,161,1998;Kim,J.等.,细胞生物化学杂志(J.Cell Biochem.)77,169,2000)。除了这些功能,已知作为一种细胞粘附分子的βig-h3促进真皮中成纤维细胞的粘附和铺展。根据研究βig-h3在眼组织中的分布,报道了在创伤愈合过程中,粘附分子在正常成年人的角膜上皮、角膜内的胎基质细胞(fetal stromalcell)和内皮和基质细胞中表达。另外,βig-h3在肾脏的球旁器和近端小管中表达,并且在糖尿病患者中它的表达被增加。另外,在正常人的冠状动脉内皮下的平滑肌中发现βig-h3,并且在动脉硬化的情况下,在子宫内膜的血管中它的量被增加。然而,还未明确地确定βig-h3在正常皮肤组织和皮肤创伤中的表达(Klintworth,G.K.等.,美国病理学杂志(Am.J.Pathol.)152,743,1998;Munier,F.L.等.,自然遗传学(Nature Genetics)15,247,1997;Streeten B.W.等.,Arch.Ophthalmol.Vis.Sci.38,893,1997)。如上述提及,还不清楚βig-h3在正常人组织中的分布和表达。特别是还没有关于βig-h3在皮肤创伤中的表达形式的报道。然而,一些研究小组已报道βig-h3促进皮肤成纤维细胞的粘附和铺展(spreading)的功能,因此期望它对促进创伤愈合有所贡献。
发明概述
由于在这种愿望的背景下,对βig-h3介导的细胞粘附进行了深入细致而详尽的研究,结果通过计算机研究分析发现,在βig-h3的fas-1结构域之间和其它肽的fas-1结构域之间,除了H1和H2基序以外,存在着高度保守的氨基酸序列,特别是检测到在H2区域附近位置的天冬氨酸和异亮氨酸残基具有高度的同源性,从而导致了本发明。另外,发现每个包含保守的氨基酸残基的βig-h3的第二和第四结构域通过α3β1整联蛋白诱导细胞粘附。而且,被设计具有βig-h3的第二和/或第四结构域的重组蛋白在细胞附着和铺展活性和创伤愈合作用方面与野生型βig-h3相一致。
因此,本发明的一个目的是提供肽,其包含对于细胞附着(attachment),铺展和脱附活性必需的保守氨基酸序列。

发明详述
在本发明中,在单独或联合使用βig-h3的第二和第四fas-1结构域的基础上制备适于细胞粘附和铺展的重组蛋白。为选择第二和第四结构域,鉴定了在细胞粘附和铺展过程中有活性的βig-h3的结构域。为此,检测已知作为几种整联蛋白配体识别序列的C-末端序列精氨酸-甘氨酸-天冬氨酸(RGD)对βig-h3的细胞粘附性能的作用。利用附着细胞的数目和表面积测量细胞附着活性。结果发现,不依赖于RGD基序,βig-h3促进细胞粘附和铺展。
基于这个发现,使用化学试剂以定位βig-h3的细胞粘附活性的特异性,并且获得关于βig-h3的细胞表面受体性质的更进一步的线索。从使用化学试剂获得的数据提示在βig-h3细胞粘附活性中涉及的βig-h3的细胞表面受体可能是依赖RGD的整联蛋白的一种,其需要二价阳离子以与βig-h3相互作用。
接着,为确定βig-h3的细胞粘附功能必需的最小结构域,对每个fas-1结构域介导细胞粘附的能力进行检测。
这个检测是基于一个事实,即在各种细胞粘附分子,例如βig-h3,periostin,成束蛋白I,HLC-2和藻类-CAM中发现fas-1结构域,并且在这类粘附分子中存在的fas-1结构域的数目各自不同。这个事实导致一个推论,即对于βig-h3的细胞粘附活性,可能不需要所有四个fas-1结构域,并且在极端情况下,仅一个结构域就能介导βig-h3的细胞粘附活性。在本发明中,揭示βig-h3的第二或第四fas-1结构域的任何一种对于βig-h3的细胞粘附功能是足够。这些结果导致一个结论,即在βig-h3的四个结构域中共同存在的H1和H2序列不是介导βig-h3的细胞粘附活性所必需的。另外,发现在第二和第四fas-1结构域内近H2区位置的两种氨基酸,即天冬氨酸和异亮氨酸是高度保守的,暗示这些氨基酸残基组成与细胞粘附有关的基序。利用βig-h3的第四fas-1结构域的取代突变型确定了这两种保守的氨基酸对细胞粘附而言是不可缺少的。
在本发明的另一个实施方案中,提供了一种创伤愈合方法,其中βig-h3的第二和第四fas-1结构域被单独或联合使用。
对仅包含细胞粘附活性结构域的突变体βig-h3蛋白和那些包含一部分结构域的野生型βig-h3(βig-h3-WT)的创伤愈合作用进行了比较。在这一点上,将包含细胞粘附活性结构域的重组蛋白施用于大鼠。
当将包含βig-h3的第四fas-1结构域的重组蛋白用作软膏的药用有效成分时,除了再形成上皮和形成胶原纤丝以外,观察到创伤收缩。归根结底,这些结果意味着存在保守的天冬氨酸和异亮氨酸的βig-h3第二或第四fas-1结构域之一对创伤愈合是有效的,并且因此可被用来开发创伤的治疗剂。
另外,使用包含βig-h3的第二fas-1结构域或第二和第四结构域的重组蛋白获得了极好的细胞粘附和创伤愈合作用。
胜过包含所有结构域的蛋白,包含部分结构域的重组蛋白具有一个优点,即由于它们以水溶形式合成,并且因此不经过变性,所以可更大量生产。
实施例
根据下列实施例可更好地理解本发明,下列实施例是用来举例说明,而不应被理解为限制本发明。
实施例1:依赖RGD的βig-h3蛋白的细胞粘附活性的鉴定
1-1:重组βig-h3蛋白的生产
在实践中,为找到具有细胞粘附和铺展活性的βig-h3的结构域,检测已知作为几种整联蛋白配体识别序列的C-末端序列精氨酸-甘氨酸-天冬氨酸(RGD)对βig-h3的细胞粘附性能的作用。在这点上,制备RGD缺失的重组βig-h3蛋白(βigh3-ΔRGD)和野生型重组βig-h3蛋白(βigh3-WT)。
首先,用NdeI和BglII消化克隆到pBluescript中的全长人βig-h3cDNA(pBsβig-h3)。将DNA片段亚克隆到pET-29b(+)(Novagen Inc.)的EcoRV-EcoRI位点中。通过将切自βig-h3 cDNA的1351bp NcoI片段引入到这个克隆的NcoI位点制备βigh3-WT。通过用AoCI和NotI切割βigh3-WT质粒的3’片段,接着通过平端化和自身连接,βigh3-ΔRGD衍生自βigh3-WT,如图1所示。
在用每个重组质粒转化后,将大肠杆菌BL 21 DE3在含50μg/ml卡那霉素的LB培养基中37℃培养,直至595nm的光密度(OD)达到0.5-0.6。使用1mM异丙基-β-D-(-)-硫代吡喃半乳糖苷(IPTG)37℃诱导重组βig-h3蛋白3小时。将如此获得的沉淀重悬于溶菌缓冲液(50mMTris-HCl(pH8.0),100mM NaCl,1mM EDTA,1%Triton X-100,1mMPMSF,0.5mM DTT)中,然后超声处理。将内含体溶解在8M包含20mM尿素的变性缓冲液中,接着用Ni-NTA树脂(Qiagen)纯化变性蛋白。用200mM咪唑溶液洗脱重组蛋白,然后在含50mM NaCl的20mMTris-HCl缓冲液中从高浓度至低浓度尿素连续透析。利用SDS-PAGE分析这些重组蛋白,如图2所示。
1-2:重组βig-h3 fas-1结构域蛋白的细胞粘附活性的分析
在5%CO2条件下,将用于此分析的人角膜上皮(HCE)细胞在补充以15%胎牛血清,庆大霉素(40μg/ml),胰岛素(5μg/ml),霍乱毒素(0.1μg/ml)和人表皮生长因子(hEGF)的DMEM(EMEM/F-12,GibcoBRL)中37℃培养。
如下进行细胞粘附分析。首先,通过37℃温育1小时使重组βig-h3或其它胞外基质蛋白粘附到96-孔微量培养板(Falcon)底部,并用含0.2%BSA的PBS封闭。包被的胞外基质蛋白是人血浆玻连蛋白(普洛麦格),纯化的人血浆纤连蛋白(pFN),鸡I型和II型胶原蛋白(ChemiconInternational Inc.),牛IV型和VI型胶原蛋白(Chemicon),鼠层粘连蛋白(Chemicon)和牛血清清蛋白(BSA)(Sigma)。用胰蛋白酶消化细胞并以2×105细胞/ml的密度悬浮于培养基中。将0.1ml细胞悬液加到用重组蛋白包被的培养板的每个孔中。
接着37℃培养1小时,通过用PBS漂洗去除未附着细胞。将附着细胞在50mM、pH5.0的柠檬酸(盐)缓冲液中37℃培养1小时,该柠檬酸(盐)缓冲液含3.75mM作为氨基己糖苷酶底物的对硝基酚-N-乙酰基1-β-D-glycosaminide和0.25%Triton X-100,接着通过加入50mMpH14.0的含5mM EDTA的甘氨酸缓冲液封闭酶活性。在MultiskanMCC/340微量培养板读出器中测量405nm处的吸光度。
为确定作为细胞粘附活性指标的细胞面积,将4×104细胞施加到48-孔培养板中的基质上。用8%戊二醛(Sigma)固定附着的细胞,然后0.25%结晶紫(Sigma)的20%甲醇溶液染色。用Image-Pro plus软件(MediaCybernetics)进行细胞面积的测量。对于每个实验,每个位点测量200或300次,重复实验三次。以具体时间点的平均面积±平均标准误差的形式记录数据。
作为利用βigh3-WT和βigh3-ΔRGD测量细胞粘附和铺展活性的结果,粘附到βigh3-WT上的HCE细胞的数目和表面积明显大于那些吸附到作为阴性对照的清蛋白上的HCE细胞数目和表面积,并可与那些粘附到纤连蛋白上的细胞的数目和表面积相匹敌,如图3所示。βig-h3的细胞粘附和铺展活性是浓度依赖型的,如图4A和4B所示。缺失RGD基序的βig-h3ΔRGD在支持细胞粘附和铺展方面几乎是等效的(图5A和5B)。综合这些结果,确定不依赖RGD基序,βig-h3支持细胞粘附和铺展。
实验实施例1:在βig-h3的细胞粘附活性中涉及的βig-h3的细胞表面受体的鉴定
1-1:使用基质肽和试剂鉴定细胞粘附活性
为鉴定在βig-h3蛋白的细胞粘附活性中涉及的细胞表面受体,使用各种试剂进行抑制分析。
最初,用10μg/ml纤连蛋白,βigh3-WT或βigh3-ΔRGD包被塑料培养皿。在包含5mM EDTA,100μg/ml βigh3-WT,100μg/ml βigh3-ΔRGD,1mM RGD,1mM RGE或100μg/ml纤连蛋白,或不含它们的培养基中预培养HCE细胞,然后,如实施例1,分析细胞粘附。
细胞对βig-h3的粘附被βig-h3本身,RGD肽和EDTA明显地抑制,被纤连蛋白和EGTA部分地抑制,而不被RGE肽所抑制。细胞对纤连蛋白的粘附也被纤连蛋白本身,RGD肽和EDTA明显地抑制,被βig-h3和EGTA部分地抑制,而不被RGE肽所抑制,如图6A所示。这些结果表明在βig-h3的细胞粘附活性中涉及的βig-h3的细胞表面受体可能是依赖RGD的整联蛋白的一种。
1-2:二价阳离子对细胞粘附活性的作用
为分析βig-h3介导的粘附的二价阳离子敏感性,以2×105细胞/ml的密度将细胞悬浮于HEPES-缓冲的盐水(HBS)(150mM NaCl,25mMHEPES,2mM EDTA,pH7.4)中,并37℃培养30分钟。然后,在HBS中洗涤细胞两次,并重悬于相同的缓冲液中。然后将等份的细胞(50μl)加到微量培养板孔中,并用50μl等份的HBS在5%CO2的湿润气氛中37℃培养30分钟,HBS包含二倍于终浓度的浓度的二价阳离子(MnCl2,MgCl2或CaCl2)。随后,将细胞铺到配体包被的平皿上进行粘附分析。
Mn2+强烈地促进细胞对βig-h3的粘附,Mg2+较小程度地促进细胞对βig-h3的粘附,但Ca2+仅勉强地促进细胞对βig-h3的粘附,如图6B所示。综合这些结果证实,βig-h3的细胞表面受体是一种需要二价阳离子以与βig-h3相互作用的依赖RGD的整联蛋白。
1-3:利用抗整联蛋白的单克隆抗体鉴定βig-h3的细胞表面受体
为鉴定βig-h3的受体,检测抗整联蛋白亚基的功能-封闭的单克隆抗体对HCE细胞粘附到用βig-h3包被的表面上的影响。在这点上,最初,在存在每种抗不同类型整联蛋白的单克隆抗体(5μg/ml)的培养液中37℃预培养HCE(3×105细胞/ml)30分钟。将预培养的细胞移至用βig-h3蛋白预包被的培养板上,然后37℃进一步培养1小时,接着进行与氨基己糖苷酶底物结合的βigh-3的定量分析。数值表示为在缺失单克隆抗体的条件下的粘附到βig-h3上的细胞数的百分比。
对βig-h3包被的表面的粘附被抗α3亚基的抗体特异地抑制。由于已知整联蛋白α3亚基与整联蛋白β1亚基相偶联,抗β1抗体明显地阻断细胞对βig-h3的粘附,如图6C所示。使用HT1080细胞也观察到了相似的结果。
作为功能封闭抗体的对照实验,使用纤连蛋白,玻连蛋白,层粘连蛋白和I型胶原蛋白作为基质。将HCE细胞与抗整联蛋白亚基的功能封闭单克隆抗体一同预培育,然后移至用10μg/ml纤连蛋白,玻连蛋白,I型胶原蛋白或层粘连蛋白包被的孔中。接着培养,分析粘附细胞的细胞数。
显示细胞对纤连蛋白的粘附被抗整联蛋白α3和α5抗体明显抑制。对玻连蛋白和I型胶原蛋白的粘附分别被抗整联蛋白αv和α2抗体阻断,而细胞对层粘连蛋白的粘附被抗整联蛋白α3和α6抗体抑制,如图6D所示。另一方面,抗β1整联蛋白抗体有效地抑制细胞对上述提及的所有配体的粘附。
对于另一个对照实验,使用了已知表达α5但不表达α3整联蛋白的K562细胞。将K562细胞接种到用βig-h3-WT,纤连蛋白,层粘连蛋白或I型胶原蛋白包被的培养板上,并培养1小时,接着进行氨基己糖苷酶分析。K562细胞不粘附到βig-h3上,但粘附到纤连蛋白和玻连蛋白上。综合这些结果,提示在HCE细胞中,整联蛋白α3β1是βig-h3的特异受体,如图6E所示。
实施例2:βig-h3的细胞粘附活性必需的结构域的鉴定
试图鉴定赋予βig-h3细胞粘附活性所必需的氨基酸,进行一个检测以确定是否每个重复结构域都能介导细胞粘附。
制备四种分别相应于四个重复结构域的重组蛋白:用PCR扩增四个分别编码氨基酸129-241,237-377,368-506,和498-637的βig-h3 cDNA片段,并克隆到pET-29b(+)的EcoRV-XhoI位点,使用得到的四个表达载体,命名为pβig-h3 D-I,pβig-h3 D-II,pβig-h3 D-III和pβig-h3 D-IV,制备重组蛋白,如图7所示。具有表达载体pβig-h3 D-II和pβig-h3 D-IV的大肠杆菌转化体表示为E.coli BL21/His β-g和E.coli BL21/His β-e,并于2000年12月4日分别保藏于韩国生物科学和生物技术研究所(KRIBB)的韩国典型培养物保藏中心,保藏号为KCTC 0905BP和KCTC 0904BP。
接着按实施例1-1中描述的程序表达并纯化重组蛋白βig-h3 D-I,βig-h3 D-II,βig-h3 D-III和βig-h3 D-IV,并通过SDS-PAGE鉴定,如图8所示。
关于细胞粘附的介导,与野生型βig-h3相比,第二和第四fas-1结构域具有相等的活性,而第一fas-1结构域具有较弱活性,第三fas-1结构域根本就不具有活性,如图9所示。
在用抗整联蛋白亚基的功能封闭抗体进行的实验中,第二和第四fas-1结构域介导的细胞粘附均几乎完全被抗α3和β1整联蛋白抗体所阻断,提示第二和第四fas-1结构域均具有与α3β1整联蛋白相互作用所必需的氨基酸,如图10所示。这些结果也支持H1和H2序列均不介导βig-h3的细胞粘附活性的结论,因为第一和第三结构域在细胞粘附方面不具有活性,尽管它们具有H1或H2序列。
实施例3:βig-h3的细胞粘附所必需的保守氨基酸序列的鉴定
3-1:通过氨基酸序列对比鉴定保守的基序
为找出在βig-h3的第二和第四fas-1结构域中负责细胞粘附的氨基酸序列,第二和第四结构域独立地显示细胞附着活性,不仅在βig-h3的重复fas-1结构域之间,而且在其它蛋白的fas-1结构域之间进行基于同源性的计算机研究。结果发现,在各个蛋白之间,在H2区域附近的两种氨基酸,天冬氨酸和异亮氨酸是高度保守的,如图11所示。另外,发现在具有高细胞附着活性的βig-h3的第二和第四fas-1结构域中天冬氨酸和异亮氨酸均是保守的,而在显示中等细胞附着活性的第一fas-1结构域中仅天冬氨酸是保守的。至于不显示细胞附着活性的第三fas-1结构域不含这两种氨基酸的任何一种。这个事实是表明H2区域附近的天冬氨酸和异亮氨酸残基对介导细胞附着和铺展活性是必不可少的更进一步的证据。
3-2:利用取代突变型鉴定保守氨基酸序列的细胞粘附活性
为进一步确定这两种氨基酸对细胞粘附是必需的,如图12所示,通过取代突变包含βig-h3第四fas-1结构域的重组蛋白。通过PCR制备βig-h3 D-IV的取代突变型,并通过碱基测序确定它的序列。如同实施例1同样地分离并纯化突变体蛋白质,并在SDS-PAGE上确定,如图13所示。
检测突变蛋白的细胞附着活性,在突变蛋白中βigh3 D-IV的Pro616,Asp617和Ile618分别被丝氨酸,丙氨酸和丝氨酸取代。具有丙氨酸取代Asp617的突变体蛋白质,命名为D617A(βigh3 D-IV-PaI),和具有丝氨酸取代Ile618的突变体蛋白质,命名为I618S(βigh3 D-IV-PDs),明显地阻断细胞粘附,而发现具有丝氨酸取代Pro616的突变体蛋白质,命名为P616S(βigh3 D-IV-sDI),对细胞粘附活性没有影响。至于三个氨基酸被突变的突变体蛋白质,命名为P616S/D617A/I618S(βigh3 DIV-sas),它同样阻断细胞粘附,如图14所示。
在Asp617和Ile618突变的第一fas-1结构域中,第一fas-1结构域介导的细胞附着活性的几乎完全缺失证明617位置的天冬氨酸和618位置的异亮氨酸在介导βigh3的细胞附着活性中是非常重要的。
实施例4:影响创伤愈合的βig-h3结构域的鉴定
4-1:重组βigh3蛋白的表达和纯化
为检测是否仅仅在细胞粘附过程中具有活性的βig-h3结构域显示与包含所有四个fas-1结构域的天然βig-h3相同的创伤愈合功能,如图15所示,制备各种重组βigh3蛋白:包含所有四个fas-1结构域的His-β-b;仅包含第四结构域的βigh3-D-IV;每个至少包含一个第四结构域的βigh3-D-IV,2X,3X和4X。显示与在实施例1中制备的βigh3-WT相同的细胞粘附活性,从重组表达载体pET-29β制备重组βig-h3蛋白His-β-b,重组表达载体在它的EcoRV-EcoRI位点锚定一个通过将βig-h3 cDNA缺失某些氨基末端区域获得的Asp718-BglII片段。如同实施例1-1和3同样表达并纯化重组蛋白His-β-b和βigh3-D-IV。
如下制备至少包含一个第四结构域的重组蛋白,例如βigh3-D-IV,2X,3X和4X。通过PCR获得编码相应于第四结构域的氨基酸498-637的DNA片段,并将PCR产物用Klenow酶平端化。将这个平端化的cDNA片段插入到包含βigh-3第四结构域的pβig-h3 D-IV的EcoRV位点,将得到的表达载体命名为pβI-h3 D-IV 2X。通过用EcoRV和XhoI消化切割pβig-h3 D-IV 2X的插入片段,并通过用Klenow处理平端化,接着将平端化的片段插入到pβig-h3 D-IV和pβig-h3 D-IV 2X的EcoRV位点。将得到的表达载体命名为pβig-h3 D-IV 3X和pβig-h3 D-IV 4X。在存在1mM IPTG的条件下诱导所有重组蛋白的表达3小时,并通过利用Ni-NTA树脂(Qiagen)分离。通过用20mM包含50mM NaCl和300mM咪唑的Tris-HCl洗脱纯化分离的重组蛋白。可大量生产βig-h3 D-IV2X,3X和4X,因为不象包含所有四个结构域的βig-h3重组蛋白,它们是以水溶形式被合成,并且不经过变性,如图16A所示。使用非变性凝胶的电泳揭示βig-h3 D-IV不形成聚合物,而2X部分地形成聚合物,3X和4X每个容易地形成聚合物,如图16B所示。
大肠杆菌BL21/Hisβ-e4X,其带有含四个βig-h3的第四结构域的表达载体pβig-h3 D-IV 4X,于2000年12月4日保藏于韩国生物科学和生物技术研究所(KRIBB)的韩国典型培养物保藏中心,保藏号为KCTC 0906BP。
通过利用明胶-琼脂糖4B的亲和层析,从柠檬酸(盐)处理的大鼠血浆纯化作为阳性对照的纤连蛋白。室温下通过未取代的琼脂糖4B过滤血浆,并将洗脱物装到用0.05M Tris-Cl平衡的明胶琼脂糖4B上,Tris-HCl包含0.05M EACA(ε-氨基己酸)、0.02M柠檬酸钠和0.02%叠氮化钠。洗脱后,用含1M氯化钠的缓冲液洗涤大部分血浆蛋白。然后,用3M尿酸等渗缓冲液洗脱吸收的纤连蛋白,随后用pH 7.2的PBS透析约48小时,以纯化纤连蛋白。通过280nm处的紫外线吸光度确定它的浓度,并在储存于-20℃之前冻干。
4-2:含第四结构域的βig-h3 D-IV的创伤愈合活性分析
为比较重组βig-h3蛋白His-β-b和重组βig-h3蛋白βigh3-D-IV之间的创伤愈合活性,其中重组βig-h3蛋白His-β-b如天然βig-h3蛋白那样包含所有四个fas-1结构域,而重组βig-h3蛋白βigh3-D-IV仅包含第四结构域,如下检验包含重组蛋白的软膏基料。
在大鼠的背部产生四个整个皮层的伤口,每个直径2cm,并按照对大鼠涂敷的软膏将大鼠分成检验组1-A,1-B,1-C和1-D。
1-A:用不结合任何物质的软膏基料以每天1gm的剂量涂敷。
1-B:用软膏以每天1gm的剂量涂敷,在软膏中以100μg/ml的浓度纤连蛋白结合基料。
1-C:用软膏以每天1g的剂量涂敷,在软膏中以100μg/ml的浓度Hisβ-b蛋白结合基料。
1-D:用软膏以每天1g的剂量涂敷,在软膏中以100μg/ml的浓度βigh3-D-IV蛋白结合基料。
削刮用乙醚麻醉的鼠的背部,接着用betadin溶液消毒削刮的区域。在检验组1中,通过利用No.15外科刀片切割每只鼠的背部,以形成四个直径2cm的贯穿整个皮层的环形伤口。将适于检验组1-A,1-B,1-C和1-D的软膏以约1g的剂量涂敷到伤口上,伤口随后用合成的敷料(Tegaderm3M)覆盖,并轻轻地包扎。每天涂一次软膏。
具有一种含水物质的基料(SamA基料),每种软膏每1g包含38mg鲸蜡,116mg十八烷醇,38mg聚乙基乙二醇,192mg conc.甘油,23mg乙醇,十二烷醇硫酸钠mg,0.87mg对氧基苯甲酸乙酯(ethylparaoxybenzoate),0.12mg对氧基苯甲酸丁酯(butyl paraoxybenzoate)和纯净水。
首先,观察伤口的形态学。在每个伤口附近定位相同的刻度,并距每个伤口相同的距离拍照。将照片扫描入计算机,并借助于NIH图象分析系统(Bio-Optics),这些照片用于测量伤口的面积。为拍照,通过乙醚麻醉大鼠完全放松肌肉。每隔一天进行一次测量直至第22天。对于比较检验组,按照ANOVA检验和Scheffer’s检验分析测量值。
在所有检验组中,在创伤形成之后,就观察到创伤面积逐渐缩小。测量涂敷纤连蛋白和重组βig-h3蛋白的检验组,其创伤面积比仅涂敷软膏基料的检验组的创伤面积减少得更快。涂敷软膏7天后,看到创伤面积显著的差异。统计学上,在检验组1-A和其它彼此不显示显著差异的检验组的创伤之间存在着显著差异(p<0.05)。结果示于下表1和图17中。

在光学显微镜下进行组织学分析。在第3,7,10,14和20天进行创伤部位的活组织检查,在10%福尔马林中固定,并用石蜡固化。在显微镜下观察前,用苏木精-曙红(H&E)和三色Masson’s染色6μm样品切片。通过再形成上皮和形成胶原纤丝评估依据时间每个检验物质的创伤愈合作用。关于再形成上皮,以这种方式半定量上皮形成,即零表示未形成上皮层,1+表示开始形成上皮,2+表示不完全的上皮层结构,3+表示完全的上皮层结构。关于各检验组之间的比较和在每个检验组内依据时间的差异,利用ANOVA检验和Scheffe’s检验对测量值进行统计学分析。至于胶原纤丝形成,它被分为1+表示当用三色染料染色观察时未明显形成胶原纤丝,2+表示疏散的(scatteringly spaced)胶原纤丝,3+表示致密的胶原纤丝。
当用光学显微镜观察时,在检验组1-B,1-C和1-D中,在第7至10天似乎开始再形成上皮,并且在第20天完全形成。另一方面,关于对照组1-A,即使在第14天也未开始再形成上皮,并且在第20天未完全形成。结果示于下表2和图18中。

形成胶原纤丝(collagenous fiber)的结果示于下表3中。从表3中可见,直至第7天在检验组1-A和1-D中未明显形成胶原纤丝,保持在等级1+,而检验组1-B和1-C为等级2+。然而,在第10天,所有的检验组为等级2+,具有相对丰富的胶原纤丝。在第14天,观察到形成致密的胶原纤丝,并且完全归为等级3+,如图19所示。当然,较致密的胶原纤丝反映更加改良的创伤愈合进展。

基于发现,即仅包含第四结构域的βigh3-D-IV对创伤愈合是有效的,制备分别含两倍,三倍和四倍第四结构域的βigh3-D-IV 2X,3X和4X以便分析创伤愈合活性。
如同实施例1-2,同样分析重组蛋白的HCE细胞粘附活性。结果示于图20中。从图20可见,发现重组蛋白βig-h3 D-IV 2X,3X和4X都能有效诱导HCE细胞的粘附。
为检验重组蛋白的创伤愈合作用,进行下列试验。
在恒定的温度和湿度下,用定额饲料喂养体重为250-300gm的成熟Spraque-Dawley谱系鼠。
在检验组2中,在每个大鼠的背部产生四个环形整个皮层伤口,并用结合以附加物质的脱乙酰壳多糖基料涂敷:
2-A:仅用脱乙酰壳多糖基料涂敷的伤口
2-B:用脱乙酰壳多糖基料结合以500μg/ml的纤连蛋白涂敷的伤口
2-E:用脱乙酰壳多糖基料结合以500μg/ml的βig-h3 D-IV 3X蛋白涂敷的伤口
2-F:用脱乙酰壳多糖基料结合以500μg/ml的βig-h3 D-IV 4X蛋白涂敷的伤口
如下制备基于脱乙酰壳多糖的组合物。将1g具有600,00Da分子量的水溶脱乙酰壳多糖(聚(1-4)2-氨基-2-脱氧-β-D-葡聚糖)溶解在100ml无菌蒸馏水中,得到的1%溶液以2ml等分到12孔培养板(Corning,美国)中,接着每孔加入100μg庆大霉素。单独加入纤连蛋白,βigh3-D-IV3X和βigh3-D-IV 4X至浓度为500μg/ml,并在-70℃下冷冻,接着在冷冻干燥机(Ilshin)中冷冻干燥12小时,以提供盘形组合物。
削刮用乙醚麻醉的鼠的背部,接着用betadin溶液消毒削刮的区域。贯穿整个皮层,在每只鼠的背部形成四个直径为7mm的环形伤口。用分别用于检验组2-A,2-B,2-E和2-F的组合物涂敷伤口,然后覆上Tegaderm(3M)并轻轻包扎。每隔两天换以新的组合物。
如实施例4-2,通过确定伤口的外观评估创伤愈合作用。
高的创始愈合作用获自包含重组蛋白βig-h3 D-IV 3X或4X的组合物。
除了检验组2-A中所有的大鼠,检验组2-B中的一只,检验组2-E和2-F每组中的两只,所有的大鼠在创伤的第12-15天完全痊愈。所有的创伤面积在创伤形成后就减少了大小。至于检验组2-A,在整个愈合期间,观察到它的创伤面积以相对慢的速率减少。在其它检验组中,前三天,创伤面积大大地减少,至第9天逐渐减少,然后又大大减少。转向创伤之间的比较,在15天的整个期间,检验组2-B,2-E和2-F比检验组2-A存在更显著的差异(p<0.05),如表4和图21所示。

因此,本发明的重组蛋白,其单独或组合、或多倍包含βig-h3的第二和第四结构域,对细胞粘附和创伤愈合是有效的,并且最终可用于开发细胞培养和创伤愈合剂。

在本发明中,提供了重组蛋白,其包含至少一个βig-h3的第二和第四结构域,在βig-h3的第二和第四结构域中,已知结合整联蛋白所必需的一个天冬氨酸和一个异亮氨酸是高度保守的。另外,重组蛋白本身有用于细胞粘附和创伤愈合的用途,对于开发细胞培养方法和创伤愈合剂有贡献。
以举例的方式已描述了本发明,应理解所使用的术语是意图描述而非限制本发明。根据上述教导,可对本发明进行很多修饰和变更。因此,可理解为除了具体的描述以外,在后附的权利要求范围内,可以其它的方式实施本发明。

Claims (14)
1.一种用重组蛋白体外附着表达α3β1整联蛋白的哺乳动物细胞的方法,该重组蛋白由一个或多个拷贝的βig-h3的第二和/或第四fas-1结构域组成。
2.按照权利要求1的方法,其中βig-h3的第二fas-1结构域为βig-h3的氨基酸序列237-377。
3.按照权利要求1的方法,其中βig-h3的第四fas-1结构域为βig-h3的氨基酸序列498-637。
4.按照权利要求1的方法,其中重组蛋白由四份βig-h3的氨基酸序列498-637组成。
5.一种表达载体,其包含一段基因,所述基因编码一种重组蛋白并能表达βig-h3的第二fas-1结构域,其中所述重组蛋白由βig-h3的氨基酸序列237-377组成。
6.一种大肠杆菌菌株,其被权利要求5的表达载体转化,命名为大肠杆菌BL21/Hisβ-g(保藏号为KCTC 0905BP)。
7.一种表达载体,其包含一段基因,所述基因编码一种重组蛋白并能表达βig-h3的第四fas-1结构域,其中所述重组蛋白由βig-h3的氨基酸序列498-637组成。
8.一种大肠杆菌菌株,其被权利要求7的表达载体转化,命名为大肠杆菌BL21/Hisβ-e(保藏号为KCTC 0904BP)。
9.一种表达载体,其包含一段基因,所述基因编码一种重组蛋白并能表达四份βig-h3的第四fas-1结构域,其中所述重组蛋白由四份βig-h3的氨基酸序列498-637组成。
10.一种大肠杆菌菌株,其被权利要求9的表达载体转化,命名为大肠杆菌BL21/Hisβ-e4X(保藏号为KCTC 0906BP)。
11.按照权利要求1的方法,其包含以下步骤:
1)通过利用一种表达载体制备一种重组蛋白,其由一个或多个拷贝的βig-h3的第二和/或第四fas-1结构域组成;
2)将该重组蛋白包被到一个固相支持体上;和
3)将细胞施加到该蛋白包被的固相支持体上。
12.一种用于愈合哺乳动物创伤的药物组合物,其包含由一个或多个拷贝的βig-h3的第二和/或第四fas-1结构域组成的重组蛋白。
13.一种重组蛋白在制备用于附着表达α3β1整联蛋白的哺乳动物细胞的药物中的应用,其中该重组蛋白由一个或多个拷贝的βig-h3的第二和/或第四fas-1结构域组成。
14.一种重组蛋白在制备用于愈合哺乳动物创伤的药物中的应用,其中该重组蛋白由一个或多个拷贝的βig-h3的第二和/或第四fas-1结构域组成。
