Abstract
本发明公开了一种植物乳杆菌LP23菌株及其应用,属于微生物技术领域,该植物乳杆菌LP23于2023年8月31保藏在中国典型培养物保藏中心,保藏编号为CCTCC NO:M 20231552。该植物乳杆菌LP23的代谢产物具有抗菌活性,经分子鉴定,其中一条胞外分泌的多肽序列如序列表SEQ ID NO:1所示,该多肽为植物乳杆菌LP23的特征多肽分子,简称乳杆菌23肽。本发明的植物乳杆菌归属于饲料添加剂目录中的益生菌,安全性好,可改善动物肠道微生物平衡,发挥益生效果;体外试验表明其代谢产物含乳杆菌23肽,且具有抑杀革兰氏阴性菌与革兰氏阳性菌等功能,在猪、禽、水产与宠物饲料中具有广阔的应用前景。

技术领域
本发明涉及微生物技术领域,具体涉及一种植物乳杆菌LP23菌株及其应用。
背景技术
抗生素为人类生命的健康与肉食品生产起到了重大促进作用,然而近年来,科研工作者从养殖动物的肠道、粪便以及相关水土环境中分离出了对杆菌肽锌、金霉素、氨苄青霉素、氯霉素、四环素、氧氟沙星等抗生素耐药的超级耐药菌,养殖业中这些超级细菌的出现给传统饲用抗生素的使用带来了极大的挑战,同时,在动物饲料中长期大量地添加抗菌药与养殖现场滥用抗生素可导致药物残留,并引起细菌抗药性日益增强,使得抗菌药对各种细菌性疾病的治疗疗效明显下降,给人类健康也带来潜在危害。目前,包括瑞典、丹麦等欧盟成员国及韩国在内的许多国家现已全面禁止抗生素在饲料中的使用,我国自从2020年7月1日起禁止饲料中使用促生长类抗生素以来,行业研究人员积极寻求安全有效的抗生素替代品用于饲料与养殖现场,这些替代品及其生产方法是目前畜禽养殖业和饲料工业的重要任务和发展趋势。
酶制剂、微量元素、有机酸、益生元、益生菌、中药提取物等不同类型抗菌生长促进剂已被广泛研究并取得了重要进展,其中饲料添加剂目录中益生菌的筛选与应用成为重要方向,主要包括芽孢杆菌类、乳酸菌类、酵母菌类等,乳酸菌类在生产应用中效果尤为突出,从传统的食品如酸奶、酸白菜、酸豆角、酸辣椒与霉豆腐等产品中筛选有益菌往往能达到事半功倍的效果。植物乳杆菌是比较容易被分离的菌株,其代谢产物往往对革兰氏阳性菌抑制效果明显,在一定pH条件下,其代谢产物对革兰氏阴性菌也有较好的抑菌效果,筛选具有广谱抑菌效果的代谢产物及其活性成分具有重要的应用价值。在我国目前只有Nisin被允许应用与食品保鲜、防腐,然而,Nisin只对革兰氏阳性菌有杀灭效果,对阴性菌等基本不起作用,限制了其在革兰氏阴性菌为主的食品中的保鲜使用,为此,从植物乳杆菌中筛选到能同时对革兰氏阴性菌与阳性菌进行抑杀的多肽显得尤为重要,对于提升我们食品安全与减少养殖现场的抗生素使用具有重大意义。
发明内容
有鉴于此,为解决上述技术问题,本发明的目的在于提出一种植物乳杆菌LP23菌株及其应用,该植物乳杆菌LP23菌株是从自然界中分离到归属于饲料添加剂目录中的益生菌,且该益生菌能产生广谱抑菌效果,可以用于饲料与饲料添加剂中提升产品的应用效果、减少抗生素的使用。
所采用的技术方案为:
本发明的植物乳杆菌(Lactobacillus plantarum)LP23保藏在中国典型培养物保藏中心,保藏编号为CCTCC NO:M 20231552。
具体的,所述植物乳杆菌(Lactobacillus plantarum)LP23已于2023年8月31日在中国菌株保藏中心进行了保藏,保藏单位为中国典型培养物保藏中心(CCTCC),地址为湖北省武汉市武昌区八一路珞珈山,保藏编号为CCTCC NO:M 20231552。
上述植物乳杆菌(Lactobacillus plantarum LP23)是从湖南永州农村家庭自制酸辣椒中分离获得。
植物乳杆菌LP23经革兰氏染色为阳性,菌落呈光滑圆形,乳白色,中间凸起,边缘整齐。
所述植物乳杆菌(Lactobacillus plantarum)LP23的代谢产物具有抗菌活性,经分子鉴定,其中一条胞外分泌的多肽序列如序列表SEQ ID NO:1所示,该多肽为植物乳杆菌(Lactobacillus plantarum)LP23的特征多肽分子。
具体的,利用SDS-PAGE方法分析上述植物乳杆菌LP23的代谢产物,通过染色后切胶,回收到一条小分子多肽,经MALDI-TOF-MS分析与多肽分子测序鉴定,其中一条胞外分泌的多肽序列为PYSLQMGATALKWWKWLFKKKGD,分子量为2780±3Da,该多肽为植物乳杆菌LP23菌株的特征多肽分子,简称乳杆菌23肽。
本发明的一种抑菌剂,其含有植物乳杆菌(Lactobacillus plantarum)LP23。
进一步地,所述抑菌剂含有的植物乳杆菌(Lactobacillus plantarum)LP23的活菌数≥1×108CFU/ml。
本发明的一种饲料添加剂,其含有植物乳杆菌(Lactobacillus plantarum)LP23。
进一步地,所述饲料添加剂为禽饲料添加剂、水产饲料添加剂或宠物饲料添加剂。

本发明的植物乳杆菌LP23菌株的代谢产物乳杆菌23,该肽由编码氨基酸组成,没有稀有氨基酸,二级结构为alpha-螺旋,具有广谱抗菌活性和广谱抑菌效果,革兰氏阴性菌大肠杆菌与沙门氏菌具有良好的抑杀作用,对革兰氏阳性菌金黄色葡萄球菌与魏氏梭菌也具有较好的抑菌作用,尤其对于大肠杆菌病原菌具有较强的抑杀作用,可用于替代养殖现场的部分抗生素。本发明筛选到的植物乳杆菌归属于饲料添加剂目录中的益生菌,安全性好,可改善动物肠道微生物平衡,发挥益生效果;体外试验表明其代谢产物含乳杆菌23肽,且具有抑杀革兰氏阴性菌与革兰氏阳性菌等功能,在猪、禽、水产与宠物饲料中具有广阔的应用前景。
综上,本发明的优点在于:
该植物乳杆菌LP23是从自然界中分离得到,属于饲料添加剂原料目录中的益生菌,且其代谢产物对于上述致病菌具有广谱抗菌效果,该植物乳杆菌的代谢产物中有一条由23个氨基酸组成的活性肽,热稳定性好,该植物乳杆菌LP23可用于发酵饲料提升发酵饲料抗氧化效果,植物乳杆菌LP23的代谢产物用于肉鸡养殖可以改善肉鸡的生产性能,显示出了良好的产业化应用潜力。

具体实施方式
为了更加凸显本发明的创新性,下面结合实施例对本技术方案做进一步说明,下述的实施例仅是本发明其中部分的例子,并不代表本发明所限定的权利保护范围。
实施例1
本发明中的植物乳杆菌LP23是从湖南省永州市农村家庭自制酸辣椒中分离获得。具体分离方法为:从不同家庭的自制酸辣椒罐中取腌制的酸辣椒汤汁10ml,并使用无菌生理盐水冲洗酸辣椒表面,收集10ml冲洗液,将10ml酸辣椒汤汁与10ml冲洗液混合均匀,再用生理盐水进行10倍梯度稀释,将梯度稀释液分别涂布与MRS琼脂培养基平板表面,37℃温度下,倒置培养24小时,挑取与植物乳杆菌菌落形态相似的单菌落接种于MRS液体培养基中,37℃温度下,静置培养36小时,8000rpm离心5分钟收集上清液,先80-90℃高温处理10分钟,在冷却至室温后,加入氢氧化钠调节pH至4.5-6.0,分别以大肠杆菌和金黄色葡萄球菌为指示菌,使用抑菌圈法测定代谢产物的抑菌效果,如图1所示,筛选出能产生明显抑菌效果的菌株。
筛选抑菌效果较好的菌株进行革兰氏染色鉴定为阳性,菌落呈光滑圆形,乳白色,中间凸起,边缘整齐,这些菌落特征与典型的植物乳杆菌菌落特征基本一致,如图2所示。从中筛选出对大肠杆菌抑菌直径12mm以上的菌株一株,通过GB4789.35-2016鉴定此菌株,将该菌株用接种针在MRS琼脂平板上进行划线培养,37℃温度下,倒置培养36-48小时,直至看到明显的单菌落长出,再用无菌接种针从MRS琼脂平板中挑取单菌落分别接种于纤维二糖、麦芽糖、水杨苷、甘露糖、蔗糖的生化管中,用酒精消毒后的封口膜封口,置于37℃温度下静置培养24小时后观察各管中的颜色变化,鉴定结果就见表1。

将该抑菌菌株在37℃进行MRS液体培养18h后,取2ml培养液于8000rpm离心5min,保留菌体沉淀,用细菌总DNA提取试剂盒并参照试剂盒说明书提取该菌株的总DNA,以该总DNA为模板,采用16S rRNA的引物[27F(5’-AGAGTTTGATCMTGGCTCAG-3’), 如SEQ ID NO:5所示;1492R(5’-GGTTACCTTGTTACGACTT-3’)],如SEQ ID NO:6所示;进行PCR扩增,扩增条件为预变性 94℃ 5min 1 cycle,变性 94℃ 30s、退火54℃ 30s、延伸72℃ 90s、35cycles,延伸 72℃ 10min 1cycle。PCR结束后并取8微升PCR产物进行琼脂糖凝胶电泳分析扩增效果,如图3所示。用纯化试剂盒回收PCR产物,交由生工生物工程(上海)有限公司进行PCR产物的单反应测序:27F与1492R引物测序结果分别如SEQ ID NO:2与SEQ ID NO:3所示,拼接后的DNA序列约1500bp,其与PCR电泳后的分子大小相符,如序列表SEQ ID NO:4所示。
经BLAST对比,该基因序列与植物乳杆菌的16s rRNA基因序列有97.6%的同源性,通过上述基因水平、生化水平与菌落特征等分析,确定该抑菌菌株为一株植物乳杆菌。
实施例2
本发明从该抑菌菌株代谢产物中分离到的一条活性多肽,具体方法如下:
1、将本发明中的抑菌菌株进行活化,按照2%的接种量接种至MRS液体培养基,37℃静置36小时,于8000rpm离心5分钟,收集上清溶液,并在80-90℃高温处理10分钟,将处理液用于Tricine-SDS-PAGE电泳,然后进行生物自显影操作,电泳后的凝胶加入1% Tirton X-100(V/V) 温和摇动20min,无菌水漂洗2次; 以0.1mol/L pH7.2磷酸缓冲液平衡30min,弃去缓冲液,将凝胶转入无菌培养皿中,再将45℃左右加金黄色葡萄糖菌的LB半固体培养基(0.5%酵母提取物,1%胰蛋白栋,0.5%氯化钠, 0.8%琼脂粉)平铺于凝胶上,置37℃培养过夜,观察抑菌结果,发现在低分子量区域具有抑菌效果,如图4所示。
2、根据本实施例步骤1的结果,用电泳回收的方法分离目的条带,具体方法如下:
将步骤1收集到的上清液进行大量的点样并进行Tricine-SDS-PAGE电泳,待电泳指示条带到达凝胶中部时停止电泳,取出凝胶,用4℃预冷的KCL染色液染色,直至看到在低分子量区域出现显色的条带,用干净刀片切下含显色条带的凝胶,将凝胶条带置于蒸馏水中浸泡,直至条带变成无色透明状,再将此无色透明凝胶从蒸馏水中取出,置于2ml离心管中,用消毒过的镊子夹碎凝胶,然后在此管中加入适量的PBS溶液,4℃放置过夜以便多肽浸出,10000rpm离心10分钟,所取上清液中则含分离的目标多肽,此溶液用于MALDI-TOF-MS进行质谱分析与多肽测序分析,分析结果如下图5与图6所示。
图6为分离多肽的测序结果,数据库检索结果的序列与MADLI-TOF-MS基本一致。结合参见如下表2所示。

经上述实施例2质谱与测序分析,从该抑菌菌株分离的多肽分子量为2780Da,多肽序列为PYSLQMGATALKWWKWLFKKKGD,如序列表SEQ ID NO:1所示,共有23个氨基酸(简称乳杆菌23肽),为区分其他植物乳杆菌,根据此抑菌菌株的代谢产物分子特征,特将此分离的植物乳杆菌抑菌菌株命名为植物乳杆菌LP23(Lactobacillus plantarum LP23)。
将此植物乳杆菌LP23于2023年8月31日在中国菌种保藏中心进行了保藏,保藏单位为中国典型培养物保藏中心(CCTCC),地址为湖北省武汉市武昌区八一路珞珈山,保藏编号为CCTCC NO:M 20231552( Lactobacillus plantarum LP23)。
实施例3
将此植物乳杆菌LP23用于生产发酵饲料,具体方法如下:
将植物乳杆菌LP23活化后,按照2%的接种量接种至MRS液体培养基,37℃静置培养36小时,使植物乳杆菌LP23密度≥1×1010CFU/ml,再选用优质玉米20%、豆粕10%、麦麸70%三种饲料原料合计500kg于1吨混合机中混合均匀,用180kg水将植物乳杆菌LP23稀释至1×108CFU/ml,再往水中分别加入酿酒酵母菌1×107CFU/ml与枯草芽孢杆菌1×107CFU/ml,将此含有三种菌剂的混合水溶液按照100kg/min的流量喷雾至上述500kg饲料原料表面,边搅拌边喷雾,直至所有水溶液喷洒完成,同时,用市面上某商品发酵菌剂做的发酵饲料一批,用呼吸袋分别将此混合的湿饲料按照30kg/袋打包,于20℃~37℃的温度发酵3天,发酵完成后,各称取10g发酵饲料置于烧杯中,加入20ml无菌水,4000rpm离心5min取上清测定抗氧化效果,即DPPH清除率,如下表3与图7所示。

实施例4
将此植物乳杆菌LP23的代谢产物用于仔猪的养殖,具体方法如下:
利用四级发酵进行液体扩大培养,具体是将活化好的植物乳杆菌LP23按照2%接种于2升MRS液体培养基中,在37℃下静置培养18小时,再把这2升植物乳杆菌LP23接种到灭菌的80升MRS液体培养基中,37℃下静置培养18小时后,把此80升菌液接种到灭菌的800升扩大培养基中(培养基配方为:蔗糖15g/L、胰蛋白胨15 g/L、牛肉膏10 g/L、酵母膏5 g/L、磷酸氢二钾2 g/L、柠檬酸三胺2 g/L、乙酸钠5 g/L、硫酸镁 0.6 g/L、硫酸镁0.1 g/L、吐温801 mL/L、pH 6.6),发酵条件为温度35℃、通气量为0,转速30rpm,培养18小时左右,再转移到10立方的四级发酵罐中,含8立方扩大培养基,配方同上述扩大培养基配方,发酵条件为35℃、通气量为0,转速20rpm,培养24小时后,菌液90℃灭菌30分钟,待温度冷却至60℃,加入800kg扶态粉载体,搅拌均匀,冷却至40℃左右,转移至储存罐,再进行喷雾干燥获得植物乳杆菌LP23的代谢产物,喷雾干燥条件为进风温度,170℃,出风温度75℃。测定植物乳杆菌23肽的含量约为1%,将此喷雾干燥的植物乳杆菌LP23代谢产物按照1吨教槽料使用1kg的用量进行拌料饲喂仔猪,选用杜长达三元仔猪80头,分成两组,试验组使用LP23代谢产物1kg/吨饲料,对照组不使用本发明所述代谢产物,在山东大益试验猪场饲喂21天,结果见下表4。

结果表明,试验组显著提高了仔猪的生长性能,降低了仔猪的料肉比。
实施例5
将此植物乳杆菌LP23的代谢产物用于肉鸡的养殖,具体方法如下:
植物乳杆菌LP23代谢产物的规模生产参考实施例4,将1%含量的植物乳杆菌23肽稀释至0.5%,并在山东大益合作鸡场进行养鸡试验,选用两栋白羽肉鸡5.2万羽,分成两组,每栋2.6万羽,其中试验组饮水500g/吨水,对照组饮水中不添加本发明所述代谢产物,试验为期42天,结果如下表5。

*注:试验组为每吨水中含0.5kg LP23代谢产物,对照组为每吨水中不含本发明所述代谢产物。
结果表明,试验组显著提高了肉鸡养殖的综合水平,降低了肉鸡的料肉比。

Claims (8)
1.植物乳杆菌(Lactobacillus plantarum)LP23,其特征在于,所述植物乳杆菌(Lactobacillus plantarum)LP23于2023年8月31保藏在中国典型培养物保藏中心,保藏编号为CCTCC NO:M 20231552。
2.根据权利要求1所述的植物乳杆菌(Lactobacillus plantarum)LP23,其特征在于,所述植物乳杆菌(Lactobacillus plantarum)LP23的代谢产物具有抗菌活性,经分子鉴定,其中一条胞外分泌的多肽序列如序列表SEQ ID NO:1所示,该多肽为植物乳杆菌(Lactobacillus plantarum)LP23的特征多肽分子。
3.一种抑菌剂,其特征在于,所述抑菌剂含有权利要求1或 2所述的植物乳杆菌(Lactobacillus plantarum)LP23。
4.根据权利要求3所述的抑菌剂,其特征在于,所述抑菌剂含有的植物乳杆菌(Lactobacillus plantarum)LP23的活菌数≥1×108CFU/ml。
5.一种饲料添加剂,其特征在于,所述饲料添加剂含有权利要求1或 2所述的植物乳杆菌(Lactobacillus plantarum)LP23。
6.根据权利要求5所述的饲料添加剂,其特征在于,所述饲料添加剂为禽饲料添加剂、水产饲料添加剂或宠物饲料添加剂。
7.权利要求1或 2所述的植物乳杆菌(Lactobacillus plantarum)LP23在发酵饲料中的应用。
8.权利要求1或 2所述的植物乳杆菌(Lactobacillus plantarum)LP23在仔猪或肉鸡养殖中的应用。
