本发明涉及贵重中药的DNA序列及利用该序列鉴定其品种和辨别其真伪的方法,更具体地说,本发明涉及中药石斛的ITS2及部份5.8S和28S核糖体部份DNA序列及利用该序列鉴定其品种或辨别其真伪的方法。
石斛为贵重中药材,应用历史悠久,在《神农本草经》中列为上品,具有滋阴消热、生津益胃、润肺止咳等功效,用于热病伤津、口干烦渴、病后虛热等多种病症。现代药理研究表明,石斛还具有抗肿瘤、抗衰老、增强人体免疫力及扩张血管的作用。因此石斛在临床上及复方中药制剂中被广泛应用。中国药典(1995年版)收载有5种石斛,即金钗石斛(D.nobile Lindl.),铁皮石斛(D.candidum Wall.ex Lindl.),束花石斛(D.chrysanthum Wall.ex.Lindl.),美花石斛(D.loddigesii Rolfe)和流苏石斛(D.fimbriatum Hook)。
石斛药材的市场需求量甚大,每年达数千吨之多,而部份商品的售价昂贵,例如一级石斛(Herba Dendrobii)每公斤的售价达3500美元。
市售石斛的来源复杂,涉及石斛属植物30多种,并时有其他属的兰花(如石仙桃属)混作石斛药材使用,直接影响到石斛的临床疗效及消費者的利益。再者,石斛药材的原植物均是濒危品种,如铁皮石斛列于中国受保护植物名录的第III类级別。
现行的石斛鉴定方法包括性状鉴別、化学分析及微观解剖。但这些方法仍未对药典所列的石斛品种作全面和客观的鉴定。
植物核糖体脱氧核糖核酸(rDNA)以順反子18S-ITS1-5.8S-ITS2-26S的形式存在。植物大多拥有该順反子的重复序列,其以头尾相连的方式聚集在染色体上。目前研究显示,18S、5.8S和26S等rDNA基因的碱基对序列非常保守,不同植物间的差別很小;而位于植物rDNA基因之间的ITS1(转录空隙区1)和ITS2(转录空隙区2)的碱基对序列则差別较大。同时由于基因转化和交换的作用,就同一品种植物而言,其rDNA序列大致相同(Hillis和Dixon,1991)。
因此,本发明的目的是提供石斛药材的ITS2及5.8S和28S核糖体部份DNA序列。
本发明的另一个目的是提供利用所述DNA序列鉴定石斛的品种及辨别其真伪的方法。
为达到本发明的目的,本发明人从香港及中国大陆的不同地点收集了46种样品,包括16种石斛和一种石仙桃。所有的样品均经过兰科专家鉴定。除了罗河石斛之外,其余每一品种都有从数个不同群体获得的样品。所测得的46个样品的ITS2及5.8S和28S核糖体部份DNA序列如下:

上述46个样品的ITS2及5.8S和28S核糖体部分DNA序列的测定方法包括下列步骤:
(1)收集植物样品,进行品种鉴定工作;
(2)从步骤(1)选出的植物中提取DNA;
(3)以步骤(2)提取的DNA为模板,以寡核苷酸作为聚合酶链反应(PCR)的引物,扩增ITS2及部份5.8S和28S的区段,所用的引物分別采自植物5.8S和28S的保守区段;
(4)应用直接测序技术对步骤(3)中所获得的PCR产物进行测序。
每一样品至少重复测序4次。而每一样品的序列也在互联网页National Center for Biotechnology Information(NCBI)和GeneticsComputer Group package(GCG)与其他兰花品种的序列作比较和验证以确保本发明的46个序列准确。
如上所示,除了罗河石斛之外,所测得的同一品种的样品之间在DNA水平的变异百份率不高于1.22%。所述16种石斛的种间DNA序列(ITS2及部份5.8S和28S)的差异为2%至17%,而石斛与细叶石仙桃的DNA序列(ITS 2及部份5.8S和28S)的差异为16%至20%。
本发明利用石斛类植物内ITS2(转录空隙区2)和部份5.8S及28S的DNA序列进行种间和种内的比较,借此鉴定不同的石斛品种并辨别其真伪。所采用的方法包括下列步骤:
(1)从植物中提取DNA;
(2)以步骤(1)中提取的DNA为模板,以寡核苷酸作为聚合酶链反应(PCR)的引物,扩增ITS2及部份5.8S和28S的区段,所用的引物分別采自植物5.8S和28S的保守区段;
(3)应用直接测序技术对步骤(2)中所获得的PCR产物进行测序;
(4)将步骤(3)中测得的植物ITS2及部份5.8S和28S的DNA序列与上述46个石斛及石仙桃样品的DNA序列比较以鉴定所测植物的石斛品种或分辩其真伪。
下面详细描述所述方法:
(1)DNA的提取
植物DNA提取方法与Draper和Scott(Draper和Scott,1988)所述大致相同,简述如下:将干燥的植物材料用70%乙醇的蒸馏水溶液冲洗,然后,与液氮混合并研磨成粉末。将所得的粉末样品与12倍体积的CTAB混勻,在56℃水浴中保温30分钟后,加入等量的氯仿-异戊醇(24∶1)混合液,离心,取上清液。向残余物中加入0.1体积的10%CTAB溶液,室温放置10分钟后再用氯仿-异戊醇萃取一次,离心,取上清液并与前次的上清液混合。向所得上清液中加入等量的CTAB产生沉淀,室温放置1小时后以13000g离心15分钟。将DNA沉淀物用乙醇纯化后溶解于TE缓冲液(10mM Tris-HCl,pH 7.5,1mM EDTA)中备用。
(2)PCR扩增反应
ITS2及部份5.8S和28S区段的DNA由PCR扩增反应得到。反应所用的引物为5.8d和28cc(Hillis和Dixon,1991):

反应溶液包括:1ng植物DNA,1Xtaq缓冲液(10mM Tris-HCl,pH8.3,0.001%明胶),0.2mM dNTPs,1μM引物(5.8d和28cc),以及1个单位的Taq DNA多聚酶。反应条件为:于94℃预处理5分钟,然后进行35个循环,循环条件是:94℃,1分钟,60℃,1分钟,72℃,2分钟;最后72℃,10分钟。
(3)直接测序技术
将步骤(2)得到的PCR产物用乙醇醇化后,取出1μL(约50ng)PCR醇化产物,与4μL反应终止混合液,0.64μL 25μM引物(5.8d和28cc),以及4.86μL蒸馏水混合。然后进行以下25个循环,反应条件为:96℃,10秒,56℃,5秒,60℃,4分钟。最后使用Perkin Elmer的序列分析软件(“Sequencing Analysis第三版”软件和Hitachi“DNASIS第七版”)分析和确定样品的DNA序列。
因此,采用本发明提供的石斛的ITS2及部份5.8S和28S区段的DNA序列及鉴定方法,通过其ITS2及部份5.8S和28S区段的DNA序列的比较,使得对贵重中药石斛的鉴定更加方便和可靠。

3.一种鉴定石斛品种或辨别其真伪的方法,该方法包括如下步骤:
a.从植物样品中提取DNA;
b.以步骤a提取的DNA为模板,以寡核苷酸作为聚合酶链反应(PCR)引物,扩增ITS2及部份5.8S和28S的区段,所用的引物分别采自植物5.8S和28S的保守区段;
c.应用直接测序技术测定步骤b所获得的PCR产物的序列;
d.将步骤c测得的DNA序列与权利要求1或2中所述的任一石斛品种的ITS2及5.8S和28S核糖体部份rDNA序列比较以确定石斛品种或辨别其真伪。
