Abstract
本发明公开了基于STYK1激酶结构的多肽及其在制备治疗癌症药物中的应用,所述多肽的序列如SEQ ID NO.1或SEQ ID NO.2所示,该多肽能用于制备治疗癌症的药物,且其至少发挥以下功能:阻断细胞内STYK1与GSK3β相互作用,抑制细胞内Wnt信号通路,抑制胰腺癌细胞的迁移能力,抑制胰腺癌细胞的增殖,抑制胰腺癌肿瘤的生长。本发明为胰腺癌的治疗提供了一种新的蛋白药物,具有重要的价值和应用前景。
Description
技术领域
本发明属于生物医药技术领域,具体涉及一种基于STYK1激酶结构的多肽及其在制备治疗癌症的药物中的应用。
背景技术
胰腺导管腺癌(PDAC)是全球最致命的癌症之一,约占所有胰腺癌的85%。它是由导管上皮细胞的恶性转化引起,导管上皮细胞形成毛细血管样导管系统,将酶和其他分泌物从胰腺中运送出去。病理生理上,恶性向侵袭性腺癌的转变是一个渐进且通常缓慢的过程,包括一些初始驱动突变,包括KRAS、CDKN2A、TP53和SMAD。尽管在过去十年中诊断和治疗取得了进步,但胰腺癌的5年生存率约为9%。
Wnt信号是调节发育和干细胞的关键级联之一,也与癌症密切相关。Wnt信号通路包括经典的Wnt信号通路与非经典的Wnt信号通路,经典的Wnt通路通常是高度保守的,并通过自分泌/旁分泌方法将细胞外Wnt配体与膜受体结合而激活。结合后的LRP5/6/Frizzle协同受体会招募GSK3β、CK1γ和Axin形成LRP6信号体,抑制GSK3β/Axin1/β-Catenin降解复合体的形成,进而抑制细胞质中β-Ctenin的磷酸化和降解,从而使其在细胞质中累积,随后入核联合Pygo2、Bcl-9以及FoxM1蛋白共同与TCF/LEF-1转录因子家族形成复合体并激活Wnt信号通路下游靶基因的转录激活。Wnt信号通过对细胞增殖、存活、分化、干性和肿瘤微环境的广泛影响,促进PDAC的发生和进展。已有研究表明,在PDAC中Wnt信号通路是被异常激活的。因此,靶向Wnt信号通路设计特异性的蛋白药物,尤其是多肽药物来治疗胰腺癌具有重要的价值和应用前景。
STYK1(丝氨酸/苏氨酸/酪氨酸激酶1)是一种新发现的致癌蛋白,属于受体酪氨酸激酶(RTK)家族成员。据报道,STYK1通过激活MEK/ERK和PI3K/AKT信号传导促进多种类型的癌症发展和转移,促进肿瘤进展过程中血管和淋巴管的发生和重塑。但STYK1与PDAC的关系未见报道。
发明内容
发明人通过多样本临床结果以及生物学手段,首次发现STYK1能够与β-Catenin降解复合体发生直接的相互作用进而促进Wnt信号通路的异常激活,基于此,发明人开发了能够发挥抑制胰腺癌肿瘤功能的多肽分子。
本发明的技术方案具体如下:
本发明首先提供了一种用于阻断细胞内STYK1与GSK3β相互作用的多肽,所述多肽的序列如SEQ ID NO.1或SEQ ID NO.2所示。
基于上述发现,发明人通过GST Pulldown实验鉴定出了STYK1与β-Catenin蛋白发生相互作用的蛋白区间,再针对STYK1与GSK3β(β-Catenin降解复合体成分,β-Catenin的磷酸化激酶)蛋白的相互作用,以STYK1为模板,设计得到了位于该区间内2段特异性的α-螺旋结构的多肽分子(可通过商业手段合成),且这两个多肽分子能够阻断细胞内STYK1与GSK3β的相互作用。
优选地,所述多肽的N端连接有穿膜短肽。
在本发明一具体的实施例中,穿膜短肽的序列如SEQ ID NO.3所示。
优选地,所述多肽的N端有乙酰化修饰且C端有酰胺化修饰。
本发明提供进一步提高了上述多肽在制备治疗癌症的药物中的应用。
优选地,所述癌症为胰腺癌;更为优选地,所述癌症为胰腺导管腺癌。
本发明的实验数据表明,上述多肽在胰腺癌的治疗中至少发挥以下功能:
a)抑制细胞内Wnt信号通路;
b)抑制胰腺癌细胞的迁移能力;
c)抑制胰腺癌细胞的增殖;
d)抑制胰腺癌肿瘤的生长。
本发明还提供了一种治疗癌症的药物组合物,该药物组合物的活性成分含有序列如SEQ ID NO.1和/或SEQ ID NO.2所示。
可以理解的是,在上述药物组合物中,还可以含有药学上可接受的辅料。
相对于现有技术,本发明的有益效果为:
基于STYK1能够与β-Catenin降解复合体发生直接的相互作用进而促进Wnt信号通路的异常激活的新发现,发明人通过鉴定STYK1与β-Catenin蛋白发生相互作用的蛋白区间,开发出了用于阻断细胞内STYK1与GSK3β相互作用的多肽分子,而且实验结果证明,该多肽分子可以发挥抑制肿瘤的功能,同时能够显著抑制细胞内Wnt信号通路的激活,对胰腺癌细胞的生长具有明显抑制作用。可见,本发明为胰腺癌的治疗提供了一种新的蛋白药物。

具体实施方式
下面将结合实施例,对本发明的技术方案进行清楚、完整地描述。应当理解的是,此处所描述的具体实施方式仅用于说明和解释本发明,并不用于限制本发明。
下述实施例中,若无特殊说明,均为常规方法;所述试剂和材料,若无特殊说明,均可从商业途径获得。
实施例1STYK1与β-Catenin相互作用区域的鉴定
基于机制中STYK1(Gene ID:55359)与β-Catenin有相互作用,本例先PCR技术将STYK1截短体构建在p3×Flag-CMV载体上。截短方式见图1,按照STKY1蛋白结构域进行截短,并将其激酶(Kinase domain)结构域隔约87个氨基酸设计一个缺失截短。然后通过将Flag-STYK1的各截短载体转染至HEK293T细胞,将GST-β-Catenin用BL21大肠杆菌原核细胞表达纯化,进而通过GST Pulldown与western blotting结果检测与鉴定STYK1各截短体与β-catenin蛋白的结合作用。
具体试验过程如下:
(1)截短载体的构建。
根据截短方式,设计获得各片段的引物。以STYK1全长为模版,采用设计的引物通过PCR获得各目标片段,其中PCR反应体系以及反应条件分别如表1和表2所示。

对PCR产物进行琼脂糖凝胶电泳的检测并切胶回收得到单一DNA。
将上步纯化回收的目的片段和需要插入的空载质粒用设计引物时的两种限制性内切酶进行双酶切,以切出相同的粘性末端便于酶连。以STYK1片段与p3×Flag-CMV为例,用设计引物时选好的HindⅢ和NotI这两种限制性内切酶进行该反应,反应条件为:37℃、45min,体系如表3和表4。

使用DNA产物纯化试剂盒对酶切产物进行纯化。在T4连接酶的催化作用下,酶切后的目的片段和质粒会在互补的粘性末端生成磷酸二酯键,从而使目的片段和质粒重组,形成一个完整的环状重组质粒。使用T4连接酶的连接体系如表5所示(实际操作中,目的片段和质粒的体积可以根据回收产物测得的浓度进行调整),酶连条件为:16℃酶连6h。

使用热击转化的方法,将酶连成功后形成的环状DNA转入大肠杆菌内。其中转化方式具体为:取出冻存在-80℃的感受态,放在冰上溶解,可以短暂离心使感受态沉在底部,减少管壁残留;在无菌操作台中,将感受态细胞加入酶连产物或质粒中,用移液枪轻轻吹打混匀,冰上静置30min;将酶连产物与感受态细胞的混合物在42℃水浴锅中热击90s,之后迅速将EP管放在冰上,静置2min;将500μL无抗性的LB液体培养基加入到EP管中,此过程在无菌操作台进行,在摇床上37℃,100rpm活化菌体1h;将活化后的菌液4,000rpm离心2min使菌体全部沉淀到管底,在无菌操作台上弃掉400μL上清培养基后,吹打混匀;将上述离心后的菌液缓慢滴加到带有相应抗性的LB的固体平板上,将菌液用提前烧好的三角涂布棒涂开;将涂布均匀的平板放在37℃培养箱中,倒置过夜培养。
由于目的片段插入的空载质粒均带有抗性基因,因此只有带有抗抗生素的质粒才可以存活,而杂菌则不会生长。转化成功后长出的菌可以在对应抗性(氨苄/卡那)的平板上生长。在无菌操作台中准备1.5mL EP管,加入500μL对应抗性的LB液体培养基,用烧过的小镊子夹取10μL小枪头挑取单个菌落到EP管中,在200rpm摇床中37℃摇菌6-8h,将得到的菌液按照表的体系进行菌液PCR(反应体系和反应程序分别见表6和表7)和测序,即可筛选出成功构建的重组质粒。将其菌液加甘油冻存或提取质粒保存(-20℃)以便进行后续实验。

(2)利用各截短载体筛选STYK1与β-Catenin的相互作用区域。
将Flag-STYK1的各截短载体转染至HEK293T细胞,并将GST-β-Catenin用BL21大肠杆菌原核细胞表达纯化,再进行GST Pulldown与western blotting检测。
GST Pulldown检测的具体步骤如下:
①将GST空载质粒与GST融合蛋白质粒用BL21感受态进行转化;
②挑取单个菌落,接种到500μL含有氨卞抗性的LB液体培养基中,37℃,200rpm摇菌6-8h至菌液浑浊;
③吸取200μL摇浑的菌液加入到25mL含有氨卞的LB液体培养基中,在37℃摇床,200rpm摇菌约2h,使菌液OD值大约在0.6-0.8时,按照1:1000的比例加入浓度为10%的IPTG(无菌水配置,0.22μm的滤膜过滤)诱导12h;
④菌液离心,离心条件为4000rpm,4℃离心10min,弃掉上清培养基,加5mL预冷的PBS重悬菌沉淀,加入终浓度为1mg/mL的溶菌酶,室温反应30min后,加入25-50μL浓度为100mM的PMSF;
⑤转移到10mL离心管中,超声破碎菌体至液体澄清,取20μL到已做好标记的1.5mLEP管中,加20μL 2×loading buffer;
⑥将超声完的液体分装至1.5mL EP管中,4℃,13000rpm,离心30min。分别取上清20μL加入20μL 2×loading buffer;沉淀中加100μL ddH2O搅匀后再加入50μL 2×loadingbuffer;与步骤⑤中样品一起98℃煮样10min;
⑦上述样品用SDS-PAGE电泳检测目的蛋白是否诱导出来;将上清合并一起后,加入GST-beads,使用前摇匀,使珠子均匀分散,其中GST空载中加入24μL,GST融合蛋白中加入30μL,4℃摇床上下颠倒摇样1.5h,后分装至1.5mL EP管中,4℃,1,000rpm离心2min;
⑧弃掉上清,用GST Washing Buffer洗珠子5-10次(1000rpm,2min离心洗),取10μL珠子,加10μL 2×loading buffer;
⑨在珠子中加入50μL浓度为10mM的谷胱甘肽还原剂,4℃摇床上下颠倒摇样30min。再4℃,1000rpm离心2min,取上清10μL,加入10μL 2×loading buffer;与步骤②样品一起98℃煮样10min。检测珠子及纯化的蛋白量。
⑩将还原后的GST融合蛋白与CO-IP的磁珠混合,4℃孵育2h,之后按照CO-IP的方法洗磁珠,加入50μL 1×loading buffer,98℃煮样10min,跑胶,检测蛋白之间是否存在直接相互作用。
免疫共沉淀技术(Co-immunoprecipitation,Co-IP)是在体内检测蛋白质之间相互作用的常用方法。其原理具体为:目的蛋白与相互作用蛋白结合,抗体会与裂解液中目的蛋白结合,蛋白磁珠会结合抗体,进而使磁珠、抗体、目的蛋白、相互作用蛋白形成一个整体共同存在于磁珠上,洗掉磁珠上杂蛋白,再将磁珠整体进行Western Blot检测蛋白质之间是否存在相互作用。
其中,免疫共沉淀的操作步骤具体如下:
①瞬时转染36~48h后的细胞弃除培养基,沿壁轻轻加入PBS润洗两遍。将培养皿置于冰上;
②在提前预冷好的RIPA裂解液中加入蛋白酶抑制剂(1:100),将其加入细胞中(10cm培养皿加1mL裂解液,6cm培养皿加500μL裂解液,6孔板每个孔加200μL裂解液),冰上静置几分钟;用细胞刮轻轻将细胞刮下,并用移液枪将其转移到已做好样品标记的1.5mL离心管中,4℃孵育裂解2h;
③裂解完成后,小型台式低温离心机提前预冷至4℃,将样品13000rpm,4℃离心30min,将上清重新吸入做好样品标记的1.5mL离心管内,弃沉淀;此时可取出50μL加入50μL2×loading buffer,再在98℃煮10min,作为全细胞裂解液(Whole Cell Lysate,WCL),来鉴定全细胞中存在的蛋白;
④向每个离心管中加入15μL提前清洗好的磁珠,4℃孵育2h,进行预吸附,吸附非特异性杂蛋白;
⑤小型台式低温离心机提前预冷,将预吸附后的样品13000rpm,4℃离心30min,取上清于做好样品标记的1.5mL离心管内,弃沉淀。按实验需要加入对应的抗体(根据抗体浓度,每1mL裂解液中加入1μg抗体),4℃孵育2h。
⑥抗体与目的蛋白充分结合后,向每管样品中加入25μL磁珠,4℃过夜孵育。
⑦用配套的磁力架吸附磁珠,弃掉上清,对着管壁轻轻加入1mL PBS,撤去磁铁,轻轻晃动,再插上磁铁弃去上清,清洗一次,用同样的方法加入高盐溶液(1×TBST)清洗磁珠3遍,最后再用PBS清洗一次。
⑧最后一遍吸尽液体,向每管样品中加入50μL 1×loading buffer,98℃煮样10min,跑胶,检测蛋白之间是否存在相互作用。
Western Blotting实验的具体操作如下:
①电泳:根据实验目的提前选好SDS-PAGE蛋白胶进行电泳,将实验前期得到的样品,根据样品浓度及实验需求取适量点入蛋白胶中,电压80V恒压进行电泳,待蛋白迁移至分离胶后,调整电压为130V继续恒压电泳至蛋白迁移至胶底部;
②转膜:将电泳后的SDS-PAGE胶取出,用甲醇浸泡PVDF膜约15s对膜进行激活,按照从负极到正极的顺序,依次摆放海绵-滤纸-胶-PVDF膜-滤纸-海绵,在摆放过程中避免气泡产生,排除空气,采用湿转法,210mA转膜2h(4℃)。
③封闭:将转膜完成的PVDF膜放入含有5%脱脂奶粉的1×TBST封闭液中,放在摇床上室温50rpm封闭2h。
④孵育一抗:封闭结束后,用1×TBST将膜上的封闭液洗干净,放入抗体孵育盒中,加入一抗,放在脱色摇床上室温50rpm孵育2h。
⑤清洗一抗:一抗孵育完成后,将一抗回收到离心管于-20℃冰箱保存,用1×TBST在脱色摇床上室温100rpm,每5min洗一次,清洗5遍。
⑥孵育二抗:提前用1×TBST按照1:20000的比例稀释对应一抗属性的二抗,于脱色摇床上室温50rpm孵育45min。
⑦清洗二抗:用1×TBST在脱色摇床上室温100rpm,每5min洗一次,清洗5遍。
⑧用ECL显色液于凝胶成像仪显影,分析结果。
最终检测结果如图1所示:缺失激酶的STYK1不能与β-catenin结合,表明STYK1激酶结构介导该相互作用。
实施例2与GSK3β具有强相互作用的多肽的获得
根据实施例1的鉴定结果,本例利用I-TASSER数据库(https://zhanggroup.org/I-TASSER/)对STYK1蛋白116-422氨基酸区间的蛋白二级结构进行预测,结合蛋白结构的模拟,发现STYK1在该区域存在9个α-螺旋结构(见图2)。
将这9段α螺旋结构的肽段为基础设计多肽序列(具体序列见图2,SkP0为对照肽),同时在设计好的多肽N端连接穿膜短肽TAT(序列为YGRKKRRQRRR,SEQ ID NO.3)。并在多肽N端进行乙酰化修饰,C端进行酰胺化修饰。
将上述9段多肽和对照肽与GFP荧光蛋白融合,构建融合表达载体(参照实施例1构建),转染至HEK293T细胞后,利用免疫共沉淀和western blotting验证其与目标蛋白GSK3β之间的结合情况(实验过程参照实施例1)。
结果如图3所示:SKP5(RADVWSFGILLYEMV,SEQ ID NO.1)和SKP8(PRELRLRLEAAIKT,SEQ ID NO.2)与GSK3β之间具有较强的相互作用,且SKP1(LHEVQDFLGRIQFHQYL,SEQ IDNO.4)较为稳定的与GSK3β相互作用弱。
实施例3多肽药物对Wnt通路活性的影响
根据实施例2的结果,本例以SPK1为对照,SKP5和SKP8这两段短肽为实验组,将三段短肽SKP1、SKP5和SKP8进行商业合成,并在其N端加入带有TAT的穿膜肽(同实施例2),使得合成的短肽能够进入细胞发挥功能,并将得到的三个短肽药物分别命名为NOKCn(SKP1),NOKP1(SKP5)和NOKP2(SKP8)。
分别使用NOKCn、NOKP1和NOKP2处理胰腺癌PANC-1细胞24小时,终浓度为5μM。收集细胞裂解液并进行western blotting实验(参照实施例1),检测Wnt信号通路靶基因AXIN2、C-myc和CyclinD1的表达量。
结果如图4所示:显示NOKP1和NOKP2能够显著下调Wnt信号靶基因的表达,说明这两段短肽可以抑制Wnt信号通路。
实施例4多肽药物对胰腺癌迁移的影响
本例通过划痕实验,将5μM的NOKCn,NOKP1和NOKP2加入到已做划痕的PANC-1细胞,检测细胞在加入短肽药物后12小时和24小时的划痕恢复能力。
其中,划痕实验的具体操作如下:
①培养板划线:首先使用马克笔和直尺在6孔板背面,画“井”字,划线时注意线不要太粗。
②铺细胞:接种细胞,生长12至24小时后细胞长满,覆盖率达到100%。
③细胞划线:第二天用枪头垂直于细胞平面,沿着马克笔的线,从六孔板一端划向另一端(划线时使用同一只枪头,控制粗细一致)。
④洗细胞:划痕完成后,吸掉旧培养基,使用无菌PBS洗细胞表面3次,去除划下的细胞,使划线后留下的间隙清晰可见,然后使用新鲜无血清培养基进行培养。
⑤细胞培养、观察:将细胞放入37℃、5% CO2培养箱培养。然后在适当的时间点,如0,6,12,24h取出六孔板,在显微镜下观察并测量划痕的宽度,并拍照。
⑥结果分析:随机划取几条不同的水平线,计算细胞间距离的均值。
结果如图5所示:NOKP1和NOKP2能显著抑制胰腺癌细胞向划痕创伤口的迁移能力。
实施例5多肽药物对胰腺癌细胞增殖的影响
本例的方法为:将5μM的NOKCn,NOKP1和NOKP2加入到胰腺癌PANC-1细胞24小时后,利用Edu染料(5-乙炔基-2’-脱氧尿苷,5-Ethynyl-2ˊ-deoxyuridine)对细胞内新合成的DNA进行染色,通过绿色染色的细胞数量来判定细胞增殖的影响。
其中,本例使用碧云天BeyoClickTMEdU-488细胞增殖检测试剂盒进行检测,且其操作步骤具体如下:
①在12孔板细胞密度达到90%左右时,将预先融化的EdU工作液与DMEM培养基按照1:500的比例稀释,此时EdU工作液终浓度为20μM;
②继续在细胞培养箱中培养细胞2h;
③培养完成后,去除12孔板中的培养基,用PBS润洗细胞3遍后,每孔加入300μL4%多聚甲醛覆盖细胞,对细胞进行固定,固定时间为15min;
④去除固定液,用PBS润洗细胞3遍,每孔加入300μL 0.2% TritonX-100进行通透,通透时间为15min;
⑤去除通透液,用PBS润洗细胞3遍,每孔加入250μL Click反应液(包含ClickAdditive Solution 25μL,CuSO4 10μL,Azide 488工作液0.5μL,Click Reaction Buffer215μL),轻晃12孔板使反应液均匀覆盖细胞;
⑥室温避光孵育30min;
⑦反应完成后吸掉反应液,用PB润洗细胞3遍,加入用PBS配置的终浓度为1mg/mL的DAPI对细胞进行染核,每孔加入300μL,室温避光染核5min,用PBS润洗细胞以去除残留的DAPI。
⑧在荧光显微镜下观察细胞增殖情况并进行图像采集。
结果如图6所示:NOKP1和NOPK2能够抑制胰腺癌PANC-1细胞的增殖。
实施例6多肽药物对裸鼠体内胰腺癌肿瘤生长的影响
本例对裸鼠进行皮下注射肿瘤细胞PANC-1,以研究多肽药物NOKP1对胰腺癌肿瘤生长的影响。本例的动物实验均按照湖北工业大学动物保护与使用委员会专门批准的实验动物使用方案进行。
本例的实验流程具体如下:
①将肿瘤细胞进行传代培养,最终每组传代至10个10cm的细胞培养皿,并保证细胞处于对数期生长。
②同时从北京“Vital River Laboratory Animal Technology”购买4周龄BALB/c裸鼠(每组5只裸鼠),保证每只裸鼠都为雌鼠,体重控制在18~22g。
③待10个细胞培养皿的细胞密度都至95%时,消化收集细胞,用DMEM重悬细胞,保证细胞密度107个/100μL。
④提前准备好注射器、酒精棉球等工具,在裸鼠尾部皮下注射3×106个胰腺癌细胞;其间每隔10天观察并记录成瘤情况(肿瘤大小=[(肿瘤长度×宽度×2)/2]),
⑤60天后,将短肽药物NOKP1和对照组NOKCn(浓度为2mg/kg/d)腹腔注射到裸鼠体内,10天后称量裸鼠体内形成的皮下瘤的体积和重量。
结果如图7所示:注射NOKP1的裸鼠形成的肿瘤较对照组显著变小,体积明显降低,说明NOKP1能够显著抑制胰腺癌细胞在裸鼠体内的生长。
综上所述,本发明提供的多肽分子可以发挥抑制肿瘤的功能,同时能够显著抑制细胞内Wnt信号通路的激活,对胰腺癌细胞的生长具有明显抑制作用,故可以被开发为治疗胰腺癌的药物。
以上所述是本发明的优选实施方式,不能以此来限定本发明之权利范围,应当指出,对于本技术领域的普通技术人员来说,凡在本发明的精神和原则之内所作的任何修改、等同替换和改进等,均应包含在本发明的保护范围之内。
Claims (9)
1.一种用于阻断细胞内STYK1与GSK3β相互作用的多肽,其特征在于,所述多肽的序列如SEQ ID NO.1或SEQ ID NO.2所示。
2.根据权利要求1所述的多肽,其特征在于,所述多肽的N端连接有穿膜短肽。
3.根据权利要求2所述的多肽,其特征在于,所述穿膜短肽的序列如SEQ ID NO.3所示。
4.根据权利要求2所述的多肽,其特征在于,所述多肽的N端有乙酰化修饰且C端有酰胺化修饰。
5.如权利要求3所述的多肽在制备治疗胰腺癌的药物中的应用。
6.根据权利要求5所述的应用,其特征在于,所述胰腺癌为胰腺导管腺癌。
7.根据权利要求6所述的应用,其特征在于,所述药物具有以下至少一种功能:
a)抑制细胞内Wnt信号通路;
b)抑制胰腺癌细胞的迁移能力;
c) 抑制胰腺癌细胞的增殖;
d)抑制胰腺癌肿瘤的生长。
8.一种治疗癌症的药物组合物,其特征在于,其活性成分含有如权利要求1~4任一项所述的多肽。
9.根据权利要求8所述的药物组合物,其特征在于,所述药物组合物中还含有药学上可接受的辅料。
