Abstract
本发明涉及生物检测领域,具体涉及一种叶酸解离剂及其应用。目前叶酸解离技术通常为变性剂加还原剂的双组分解离策略,本发明创造性的简化了现有的叶酸解离方案,首次实现了单一组分、无变性剂、温和高效的叶酸解离。同时,该检测技术应用于叶酸检测试剂盒设计,首次在化学发光平台上首次实现了三组分(解离组分+磁珠组分+发光组分)的叶酸检测试剂盒。该检测试剂盒反应流程简单、检测时间短、灵敏度更高、线性范围更宽,方法学和精密度良好。
Description
技术领域
本发明属于生物检测领域,具体涉及一种叶酸解离剂及其应用。
背景技术
叶酸(Folate),由蝶啶、对氨基苯甲酸和谷氨酸盐构成,是一种核酸、胸苷酸、神经递质、磷脂以及激素合成必不可少的维生素类物质,以8种辅酶形式存在于生物体内,为一碳单位的载体,参与嘌呤、嘧啶等重要物质的合成。因此叶酸在蛋白质合成及细胞分裂与生长过程中具有重要作用,对正常红细胞的形成有促进作用。人体长期缺乏叶酸可导致巨幼红细胞贫血,孕妇缺乏叶酸可使胎儿神经管畸形、先兆子痫、胎盘早剥的发生率增高。另外,叶酸缺乏还可引起高同型半胱氨酸血症,高浓度同型半胱氨酸对血管内皮细胞产生损害,并可激活血小板的黏附和聚集,因而增加心血管疾病的风险。因此,准确评估人体的叶酸水平具有重要的临床诊断价值和预防价值。
目前,叶酸的检测方法包括微生物法、比色法、高效液相色谱法、质谱法、电化学分析法和化学发光法等。其中化学发光法具有自动化程度高、灵敏度高、检测范围宽、操作简便快速、标记物稳定性好,无污染等优点,逐渐成为发展趋势。进一步地,磁微粒化学发光分析法将化学发光分析技术和免疫磁微粒技术相结合,大大提高了检测灵敏度、准确性和自动化程度。
叶酸在血液中以叶酸和蛋白质结合的结合态形式存在,检测时需先通过解离步骤将叶酸从结合态转变为游离态。在目前基于蛋白结合法的叶酸检测试剂盒中,叶酸的解离几乎都采用变性剂加还原剂的策略。其中变性剂可以是强酸、强碱、强表活、强电解质中的一种或多种。其原理是,通常单纯的还原剂只能作用于蛋白表面,而无法还原蛋白结构内部的二硫键,解离效果有限。因此,传统解离技术,均是先使用变性剂使目标蛋白结构展开,暴露出蛋白内部的二硫键,然后还原剂与之作用,彻底破坏蛋白结构,达到更好的解离效果。但是,由于变性剂的破坏性作用,通常容易导致样本凝集,反应体系中的pH、离子、表面张力等微观环境处于极端状态,影响后续游离叶酸与叶酸结合蛋白的结合活性,导致叶酸检测试剂盒的灵敏度、线性范围、精密度等受限;同时,由于变性剂与空气接触会有反应或消耗,导致基于此类解离方案的试剂盒开瓶稳定性不佳;另外,考虑到还原剂的存储稳定性,变性剂和还原剂不能溶解于同一组分中,通常单独存在于两个组分中,导致试剂盒的组分增多,反应流程复杂,检测时间长。
发明内容
为了解决上述变性剂加还原剂解离方案带来的试剂组分多,反应不稳定,检测效率不高,开瓶稳定性不佳等问题,本发明创新性的提出一种叶酸解离剂及其应用。该方案放弃了变性剂的应用,通过还原剂和缓冲体系的筛选、组合、优化,强化了还原剂对样本中叶酸蛋白结合态构象的解构作用,从而实现在温和的反应条件下高效释放出游离叶酸的目的。为叶酸检测试剂盒的实现提供了一种温和的、高效的解离方案。
该解离方案应用于化学发光平台的叶酸检测试剂盒开发,首次实现了三组分叶酸检测试剂盒设计,简化了反应流程,缩短了检测时间(20min),提高了检测灵敏度、精密度和线性范围,改善了试剂开瓶稳定性。同时,该试剂盒与罗氏叶酸检测试剂盒方法学比对良好,具备良好的临床应用价值。
具体如下:
一种叶酸解离剂,所述解离剂含有还原剂和缓冲体系,所述还原剂的浓度为5-50g/L,所述缓冲体系的浓度pH=3.5-8.5,所述缓冲体系选自硼酸、醋酸、三(羟甲基)氨基甲烷、三羟甲基氨基甲烷盐酸盐、磷酸、磷酸盐、柠檬酸、柠檬酸盐、4-吗啉乙磺酸、4-羟乙基哌嗪乙磺酸或哌嗪-1,4-二乙磺酸缓冲液,所述解离剂不含有破坏性的变性剂。
本发明所述“破坏性的变性剂”主要指强酸或强碱性化合物,包括但不限于盐酸、硫酸、氢氧化钠、氢氧化钾等化学物质。
优选的,所述缓冲体系的浓度为:20~200mmol/L
优选的,所述还原剂选自二硫代苏糖醇、2,3-二巯基丁二酸、2,3-丁二硫醇、1,4-丁二硫醇、磷酸三(2-氯乙基)酯、2-巯基乙烷磺酸钠、2,3-二巯基丙磺酸钠、硫代甘油、3-巯基-1-丙磺酸钠、3-巯基-1-丙醇,巯基乙醇、2-巯基乙醇、1,2-乙二硫醇、1-硫代甘油、巯基乙酸中的一种或多种。
更优选的,所述还原剂的浓度为20-50g/L。
优选的,所述还原剂为两种以上,更优选的,所述还原剂为2种。
本发明创造性的发现,解离剂仅含有还原剂就能够实现解离的效果,且多种还原剂共用能够相对单一还原剂减少用量,当还原剂是2种时,2种还原剂的用量相加是一种用量的一半,用更低的用量实现了相同的效果,节约成本,且解离剂成分简单。
另一方面,一种含单解离剂的叶酸检测试剂盒,其特征在于,所述试剂盒包括所述的叶酸解离剂。
本发明“单解离剂”指,检叶酸检测试剂盒中仅含有一种解离剂且采用一步解离,不存在多步解离或者不同组分的多种解离剂。
本发明叶酸解离剂能够解离并释放出叶酸后,应用到各种免疫检测中,优选的,所述试剂盒为化学发光检测试剂盒。
优选的,所述试剂盒为所述解离剂、磁微粒组分和发光物标记的叶酸抗原三组分组成。
更优选的,所述磁微粒组分选自叶酸结合蛋白包被的磁微粒,叶酸抗体包被的磁微粒。
本发明所述“包被”包括直接包被和间接包被,所述直接包被是指包被物与被包被物通过化学键直接偶联,间接包被是指包被物与被包被物之间通过各自连接物间的分子间作用力结合,例如在本发明中所述直接包被是通过共价键偶连的方式将叶酸结合蛋白或叶酸抗体结合到磁珠表面;所述间接包被指叶酸结合蛋白或叶酸抗体通过连接物(例如生物素与链霉亲和素、二抗等)结合到磁珠表面。
优选的,所述磁微粒组分选自叶酸结合蛋白包被的磁微粒,所述包被为直接包被。
本发明试剂盒采用包被叶酸结合蛋白的磁微粒,使释放的叶酸能与包被于磁珠上的叶酸结合蛋白有效结合,提高抗原-结合蛋白的结合效率和所形成复合物的稳定性,从而达到提高试剂的检测灵敏度的目的。
优选的,所述发光物选自碱性磷酸酶、辣根过氧化酶、吖啶酯、鲁米诺、异鲁米洛、三联吡啶钌。
所述发光物搭配相应的底物可以产生化学发光。通过检测光值信号值,可以计算出样本中叶酸的含量。
另一方面,本发明公开了一种叶酸前处理方法,其特征在于,包括以下步骤:
(1)样本与所述叶酸解离剂混合。
另一方面,本发明公开了叶酸检测方法,其特征在于,包括以下步骤:
(1)样本与权利要求1-2任意一项解离剂混合。
(2)将步骤(1)处理后的样本与包被叶酸结合蛋白的磁微粒混合;
(3)将发光物标记的叶酸抗原添加入步骤(2)反应后的溶液中;
(4)洗涤步骤(3)反应后的样本;
(5)进行检测。
有益效果
(1)本发明去除了变性剂(强酸、强碱、强表活、强电解质等)组分,避免了该组分与空气的接触,导致酸碱环境变化对试剂稳定性的影响,从而改善试剂的开瓶稳定性;同时避免了生产过程中高浓度变性剂对操作人员的潜在危害和试剂长期储存对包材的腐蚀。同时,因为反应体系中没有变性组分导致的极端pH、离子、表面张力环境的影响,整个叶酸解离过程和检测反应过程处于温和且均相状态,解离效果更充分和稳定,试剂的精密度、灵敏度、线性范围得到显著提升。
(2)本发明创造性的利用单一组分解离剂,甚至是只需酸性还原剂就能够实现充分的叶酸解离反应,打破了常规复杂的解离剂配方,发现了叶酸解离过程中最核心的成分,简化了配方和反应流程,提高了测试通量,降低了检测成本。
(3)本发明在化学发光平台上首次实现了三组分(解离剂、叶酸结合蛋白包被的磁微粒和酶标记的叶酸抗原)的叶酸检测试剂盒设计。该试剂盒采用单一组分解离方案,实现了对生物样本中叶酸分子温和而高效的解离释放,由于没有变性剂存在,检测反应过程始终保持温和其稳定的反应环境。解离后,游离叶酸分子可迅速与包被于磁微粒上的叶酸结合蛋白高效、充分的结合,使检测试剂盒的精密度、灵敏度、线性范围和开瓶稳定性大大提高。

以下结合实施例对本发明的特征和性能作进一步的详细描述。
A、试剂盒制备
本发明解离方案适用于免疫检测方法学,包括但不限于酶免、放免、荧光免疫、化学发光等技术,由于化学发光对于检测灵敏度、精密度的要求较高,相应的对于叶酸解离的要求更高,所以实施例中选择碱性磷酸酶发光技术进行验证。
(1)试剂准备
解离组分:浓度为5-100g/L的解离剂,将还原剂溶解于缓冲体系中。
磁微粒组分:浓度为0.1-1mg/mL的包被叶酸结合蛋白的磁微粒:在磁场中用PBS缓冲液清洗并用EDC、NHS活化磁珠,按150:1比例将叶酸结合蛋白与磁珠混合后反应过夜,用Tris缓冲液在磁场作用下清洗去除未反应的蛋白,加入稀释液配制成磁珠母液。
发光组分:浓度为0.1-1μg/mL的酶标记的叶酸抗原:用置换碱性缓冲液的脱盐柱纯化碱性磷酸酶,并调整浓度至0.5-5.0mg/mL;称取1.0-5.0mg的叶酸-NHS,用DMSO溶解为1.0-5.0mg/mL的溶液,碱性磷酸酶与叶酸-NHS溶液按1:5摩尔比加入混合,35-37℃偶联2-4小时,并用含伯胺的溶液封闭20-50分钟;反应结束后利用脱盐柱纯化,加入保存液后制成酶标物浓缩液。
在一些实施例中,所述样本可以选自尿液、血液、血清、血浆、唾液、骨髓、脑脊液、胸水、腹水、心包液、关节液、鞘膜积液或胆汁等穿刺液。
B.叶酸检测方法
(1)样本与解离剂混合;
(2)将步骤(1)处理后的样本与包被叶酸结合蛋白包被的磁微粒混合;
(3)将酶标记的叶酸抗原添加入步骤(2)反应后的溶液中;
(4)洗涤步骤(3)反应后的样本;
(5)进行检测。
实施例1本发明三组分试剂盒性能验证
本实施例的样本为血清。
反应步骤如下:
1、向反应杯中加入50μL样本与60μL解离组分(柠檬酸缓冲液+DTT,DTT浓度20g/L),37℃孵育5min,样本中的叶酸从蛋白结合态转变为游离态;2、继续向反应杯中加入20μL磁微粒组分(包被叶酸结合蛋白的超顺磁性微粒,浓度为0.8mg/mL)混合,37℃孵育5min,样本中的游离叶酸和磁珠上的叶酸结合蛋白结合;
3、将20μL酶标组分(碱性磷酸酶标记的叶酸抗原,0.8μg/mL)继续加入反应杯中,37℃孵育3min。通过洗涤磁微粒,与叶酸结合蛋白的剩余位点结合,去除未参与反应的物质及其他物质。加入全自动免疫检验系统用底物液(AMPPD),与反应杯中的碱性磷酸酶发生酶促化学发光反应,产生光子,测量相对发光强度(RLU);通过预先设定的曲线计算得到样本中叶酸浓度的检测结果。
检测结果如下表1-3及图1和图2所示,表格中浓度单位为ng/ml,其中图1是叶酸检测的特征曲线,可以看出检测的浓度范围为0-50ng/ml;图2为本发明三组分试剂盒与市面上购买的罗氏试剂盒的相关性实验,可以看出相关性良好,性能优良,能够满足检测需求;表1为精密度实验,表2为灵敏度实验,可以看出本发明三组分试剂盒的精密度小于5%,最低检测限为0.17ng/ml,精密度和灵敏度均较高,叶酸解离的效果好,保证里整个试剂盒的检测效果好;表3为线性实验,表明在0~45.12ng/ml的范围内,线性良好。

实施例2解离剂成分影响
A、更改还原剂的种类
操作步骤与实施例1一致,只更改还原剂的种类,通过比较信噪比来判断不同种类解离剂的解离效果。本发明信噪比指在叶酸检测中,含0ng/mL叶酸生物样品检测信号和含4ng/mL叶酸生物样品检测信号之比。当信噪比高于4.0时,表明对叶酸生物样本的解离充分,检测能力强。

可以看出,更改还原剂的种类对解离效果影响不显著;还原剂单独或联合使用,都能够实现高效的解离,且还原剂的联合使用能在相对更低的工作浓度下实现同等的解离效果。
B、还原剂浓度的影响
选择2-巯基乙烷磺酸钠作为还原剂,改变还原剂的浓度,所有操作步骤与实施例1相同;通过线性范围、信噪比、相关性、最低检出限和信噪比,来判断还原剂浓度对叶酸检测试剂性能的影响。

还原剂浓度为5-50g/L的范围内检测效果好,在20-50g/L的范围内检测效果最好;浓度过高,可能影响后续反应的活性。
C、缓冲体系的影响
操作步骤与实施例1一致,只更改缓冲体系的种类,通过比较信噪比来判断不同缓冲体系对解离效果的影响。

可以看出缓冲体系的种类和浓度对于解离效果影响不大。
D、解离剂pH的影响

可以看出pH在3.5-8.5效果最好,10.0时解离效果差,影响后续检测效果,因此后续实施例的pH选择3.5-8.5。

可以看出,在解离剂中添加表活、防腐剂、保护剂、曾敏剂、蛋白水解酶对检测效果并不起决定性的作用,而添加强碱或强酸物质,会导致检测效果降低。

可以看出不添加还原剂,添加其他多种添加物(配方1),添加酶(配方3),均不能达到效果,且有多种添加物,与单纯的缓冲液相比效果无明显差别(配方1-3),都不能实现本发明的效果。
实施例4传统解离方案和本发明解离方案效果对比
本发明:操作方法与实施例相同,单步三组分解离,叶酸解离剂:50g/L的2-巯基乙烷磺酸钠和50mM的醋酸盐缓冲液。
反应流程:
(1)样本与解离剂(组分1)混合孵育;
(2)将步骤(1)处理后的样本与包被叶酸结合蛋白的磁微粒混合(组分2);
(3)将碱性磷酸酶酶标记的叶酸抗原(组分3)添加入步骤(2)反应后的溶液中孵育;
(4)磁分离洗涤步骤(3)反应混合物,加入底物孵育测光。
对比方案1:双步5组分解离,变性剂加还原剂解离(传统解离方案,双组分两步解离):以市售罗氏叶酸检测试剂盒(电化学发光法)为例(其解离剂为两组分,分别是氢氧化钠和2-巯基乙磺酸钠)。
反应流程:
(1)样本与变性剂氢氧化钠混合;
(2)向步骤(1)的混合物中加入还原剂2-巯基乙磺酸钠孵育;
(3)将步骤(2)处理后的样本与钌标记的叶酸结合蛋白共同孵育;
(4)向步骤(3)的混合物中加入链霉亲和素包被的磁微粒(组分4)
(5)向步骤(4)的混合物中加入生物素标记的叶酸(组分5)并孵育;
(6)将步骤(5)的混合物吸入测量池,洗涤和电化学反应测光。

结果显示:相比于传统变性剂加还原剂的解离方案(以罗氏为例),单一组分解离方案对各浓度段样本表现出更高的信噪比区分度,从而实现更高的灵敏度和更宽的检测范围。同时从反应流程来看,单一组分反应流程步骤更少,反应时间更短。
对比方案2:二步解离,解离剂1:50g/L的2-巯基乙烷磺酸钠和50mM的醋酸盐缓冲液,解离剂2:20g/L的2-巯基乙醇和100mM的硼酸盐缓冲液。
反应流程:
(1)样本与解离剂1(组分1)混合孵育;
(2)样本与解离剂2(组分2)混合孵育;
(2)将步骤(1)处理后的样本与包被叶酸结合蛋白的磁微粒混合(组分3);
(3)将碱性磷酸酶酶标记的叶酸抗原(组分4)添加入步骤(2)反应后的溶液中孵育;
(4)磁分离洗涤步骤(3)反应混合物,加入底物孵育测光。
最终的检测限均为0.2ng/mL,检测范围均为0.5-40ng/mL,参考区间附近样本信噪比4.5。
可以看出多次解离并不会带来更优异的结果,只是让操作更为复杂.
本发明通过对试剂盒中各组分的优化,改善了变性剂(强酸或强碱)的影响、反应步骤复杂、反应时间长等弊端,提高了灵敏度和检测范围;同时样本处理试剂和检测方法,使血清中叶酸的释放更加迅速和完全,且与包被结合蛋白的磁珠结合力增强,提高了试剂盒的准确度并降低了成本。

1.一种叶酸解离剂,其特征在于,所述解离剂含有还原剂和缓冲体系,所述还原剂的浓度为5-50g/L,所述缓冲体系的浓度pH=3.5-8.5,所述缓冲体系选自硼酸、醋酸、三(羟甲基)氨基甲烷、三羟甲基氨基甲烷盐酸盐、磷酸、磷酸盐、柠檬酸、柠檬酸盐、4-吗啉乙磺酸、4-羟乙基哌嗪乙磺酸或哌嗪-1,4-二乙磺酸缓冲液;所述叶酸解离剂不含有破坏性的变性剂。
2.如权利要求1所述的叶酸解离剂,其特征在于,所述还原剂选自二硫代苏糖醇、2,3-二巯基丁二酸、2,3-丁二硫醇、1,4-丁二硫醇、磷酸三(2-氯乙基)酯、2-巯基乙烷磺酸钠、2,3-二巯基丙磺酸钠、硫代甘油、3-巯基-1-丙磺酸钠、3-巯基-1-丙醇,巯基乙醇、2-巯基乙醇、1,2-乙二硫醇、1-硫代甘油、巯基乙酸中的一种或多种,优选的,所述还原剂的浓度为20-50g/L。
3.如权利要求1或2所述的叶酸解离剂,其特征在于,所述还原剂为两种以上。
4.一种含单解离剂的叶酸检测试剂盒,其特征在于,所述试剂盒包括权利要求1-3任意一项所述的叶酸解离剂。
5.如权利要求4所述的叶酸检测试剂盒,其特征在于,所述叶酸检测试剂盒为化学发光检测试剂盒。
6.如权利要求5所述的叶酸检测试剂盒,其特征在于,所述叶酸检测试剂盒由所述叶酸解离剂、磁微粒组分和发光物标记的叶酸抗原三组分组成。
7.如权利要求6所述的叶酸检测试剂盒,其特征在于,所述发光物选自碱性磷酸酶、辣根过氧化酶、吖啶酯、鲁米诺、异鲁米洛或三联吡啶钌。
8.如权利要求6或7所述的叶酸检测试剂盒,其特征在于,所述磁微粒组分选自叶酸结合蛋白包被的磁微粒或叶酸抗体包被的磁微粒。
9.叶酸前处理方法,其特征在于,包括以下步骤:
(1)样本与权利要求1-3任意一项所述叶酸解离剂混合。
10.叶酸检测方法,其特征在于包括以下步骤:
(1)样本与权利要求1-3任意一项所述叶酸解离剂混合;
(2)将步骤(1)处理后的样本与包被叶酸结合蛋白的磁微粒混合;
(3)将发光物标记的叶酸抗原添加入步骤(2)反应后的溶液中;
(4)洗涤步骤(3)反应后的样本;
(5)进行检测。
