技术领域
本发明涉及一种粒径受控的β-葡聚糖与核酸形成的复合物。
背景技术
CpG寡核苷酸(CpG ODN)是含有免疫刺激性的CpG基序的约20个碱基的单链合成DNA。Toll样受体9(TLR9)被鉴定为CpG ODN的宿主受体,经由TLR9激活自然免疫,诱导I型干扰素(以下,记作I型IFNs)和炎性细胞因子(非专利文献1、2)。TLR9在浆细胞样树突细胞(plasmacytoid dendritic cells。以下,记作pDCs)和B细胞的内体(endosome)内表达,pDCs或B细胞摄入的CpG ODN与存在于内体内的TLR9相互作用,激活自然免疫(非专利文献2)。
希望利用CpG ODN所诱导的I型IFNs或炎性细胞因子,开发用于预防或治疗感染症、癌症、哮喘和花粉症等疾病的新的免疫疗法剂(非专利文献2、3)。作为已实用化的包含CpG ODN的药品,可列举:作为乙型肝炎疫苗的Heplisav-B(R)。Heplisav-B(R)是预防乙型肝炎病毒(以下,记作HBV)感染的疫苗,CpG ODN作为疫苗佐剂来添加。与现有的作为添加了铝佐剂的乙型肝炎疫苗的Engerix-B(R)相比,发现Heplisav-B(R)有可能显示高的药效,由此暗示了:与疫苗中最常使用的铝佐剂相比,CpG ODN有可能具有高的疫苗佐剂活性(非专利文献4)。另外,利用了CpG ODN的抗癌活性的临床试验也在进行,作为针对转移性黑色素瘤的治疗药的开发也在进行(非专利文献5)。
存在骨架序列和免疫刺激特性各不相同的至少4种类型的CpG ODN(非专利文献6)。被称为A型或D型的CpG ODN典型的是包含1个回文结构的CpG基序连同磷酸二酯(PO)骨架和硫代磷酸酯(PS)聚G尾,激活pDCs以产生大量的IFN-α,但无法诱导pDCs的成熟或B细胞激活(非专利文献7、8)。被称为B型或K型的CpG ODN由PS骨架构成,其中多数含有非回文结构的多个CpG基序,激活B细胞以产生IL-6等炎性细胞因子,激活pDCs以使其成熟,但不会诱导IFN-α产生(非专利文献8、9)。专利文献1中记载了多个K型CpG ODN。作为除此之外的CpGODN的类型的C型和P型分别含有1个和2个回文结构CpG序列,两者均可像K型那样激活B细胞,而且还可像D型那样激活pDCs,但与P型CpG ODN相比,C型CpG ODN更弱地诱导IFN-α产生(非专利文献10-12)。
D型和P型CpG ODN形成以下高级结构:形成被称为G-四联体(G-tetrads)的平行四链结构的Hoogsteen碱基对和顺式回文结构部位与反式回文结构部位之间的Watson-Crick碱基对的高级结构,这些结构对于pDCs产生强效的IFN-α是必需的(非专利文献12-14)。认为这些高级结构对于初期在内体内的局部化或由TLR9介导的信息传递是必需的,但认为缺点是成为CpG ODN的多型性和聚集的原因,其结果,妨碍D型和P型CpG ODN的临床应用(非专利文献15)。另一方面,K型和C型CpG ODN未被发现聚集等制造中的障碍,认为可用作人用的免疫疗法剂和疫苗佐剂(非专利文献16和17)。另外,报道了K型CpG ODN在人临床试验中提高以感染症和癌症为目标的疫苗的免疫原性的结果(非专利文献6、16)。
作为来自裂褶菌(Schizophyllum commune)的可溶性β(1→3)葡聚糖的裂褶菌多糖(schizophyllan)(以下,称为SPG)作为宫颈癌患者的放射疗法的刺激剂在日本有临床使用的历史记录(非专利文献18)。同样,作为来自香菇的可溶性β(1→3)葡聚糖的香菇多糖(lentinan)(以下,记作LNT)是1985年批准的药品,对于不能手术和复发的胃癌患者通过与氟嘧啶类药剂并用来使用(非专利文献19、20)。研究显示:β(1→3)葡聚糖与聚脱氧腺苷(以下,称为聚(dA))形成三重螺旋结构的复合物(非专利文献21)。
专利文献2~4中公开了:包含SPG的β(1→3)葡聚糖与核酸(基因)的水溶性复合物作为基因载体的应用。这些文献中记载了:通过形成该复合物,基因的反义作用和对核酸分解酶(核酸酶)的耐性作用提高。
专利文献5中公开了:通过使用具有β(1→3)键的多糖类作为载体(转染剂),包含CpG序列、且将磷酸二酯键取代成了硫代磷酸酯键或二硫代磷酸酯键的免疫刺激性寡核苷酸的作用得到提高。
专利文献6中记载了免疫刺激性复合物,其特征在于:由免疫刺激性寡核苷酸和具有长链的β(1→6)糖苷键侧链的β(1→3)葡聚糖构成。
研究显示:在与SPG形成了复合物的CpG ODN的3’末端连接有具有磷酸二酯键的聚(dA)的小鼠和人源化的CpG ODN会增强细胞因子的产生,作为流感疫苗佐剂或辅助性T细胞2型(以下,记作Th2)相关疾病的预防或治疗药起作用(非专利文献22、23、专利文献7)。另外,研究显示:若在CpG ODN的3’末端连接具有硫代磷酸酯键的聚(dA),则复合物的形成几乎上升至100%(非专利文献24)。
若使在K型CpG ODN(K3)的3’末端连接有40个碱基dA的CpG ODN(以下,记作K3-dA40)与SPG的复合物(以下,将该复合物记作K3-SPG)与人外周血单核细胞(humanperipheral blood mononuclear cells。以下,称作hPBMCs)作用,则观察到在单独的K3中没有观察到的IFN-α的产生,在核酸处理浓度低的条件下,与D型、P型、C型的CpG ODN相比为高IFN-α产生水平(非专利文献25)。另外,还判明:在小鼠模型中,由K3-SPG诱导的IFN-α产生和使用K3-SPG作为疫苗佐剂时的细胞毒性T细胞(CTL)或辅助性T细胞1型(以下,记作Th1)诱导增强活性依赖于由TLR9介导的I型IFNs(非专利文献25)。而且,使用各种小鼠癌移植模型来评价K3-SPG单独的抗癌活性的结果判明了:静脉内给药的K3-SPG的抗癌活性比K3高(非专利文献26)。暗示了小鼠癌移植模型中的K3-SPG的抗癌活性也依赖于I型IFNs,而且,还暗示了:有可能还依赖于对CTL诱导较为重要的IL-12(非专利文献26)。
专利文献8中公开了抗原/CpG寡核苷酸/β(1→3)葡聚糖类三元复合物的制造方法。
β(1→3)葡聚糖类(BG)与CpG ODN核酸形成的复合物(以下,记作BG-CpG)在生物体内激活自然免疫,与单独的CpG ODN相比具有强炎症应答诱导等生理活性(非专利文献25)。即,认为BG-CpG是有可能用于作为期待CpG ODN单独临床应用的目标疾病的感染症、癌症、作为过敏性疾病的哮喘和花粉症的免疫疗法剂(非专利文献2、3、25、26)。
最近,CpG ODN在癌免疫疗法中的应用受到关注(非专利文献27)。作为临床应用的报告例,在修饰了黑色素瘤癌抗原的CTL表位而得的病毒样纳米粒子(virus-likenanoparticles。以下。记作VLPs)中封入了CpG ODN的核酸制剂在22名II~IV期黑色素瘤患者中的14名中诱导了黑色素瘤癌抗原特异性CTL(非专利文献27)。另外,在非临床模型中,也报道了纳米粒子化的CpG ODN对T细胞淋巴瘤、肝癌、大肠癌、神经胶质瘤的抗癌活性(非专利文献28-31)。由以上的报道暗示了:CpG ODN制剂的粒子大小有可能与抗癌活性相关,认为形成纳米粒子形态的BG-CpG对癌免疫疗法有效。
作为BG-CpG的重要的生理活性指标,可列举:诱导作为I型IFNs的IFN-α的产生(非专利文献25)。特别是,与使用非临床癌移植模型的生物体的药效评价系统相比,使用暗示BG-CpG在临床中的药效的hPBMCs来测定IFN-α产生诱导水平的体外评价系统的定量性或数据稳定性优异(非专利文献25、26)。另外,在非临床小鼠模型中的体外药效评价中,认为通过使用小鼠生物体测定与抗癌活性相关的IFN-α和IFN-γ的产生,可预测在使用了小鼠生物体的癌移植模型中的抗癌活性(非专利文献25、26)。另一方面,认为BG-CpG的癌增殖抑制活性需要使用小鼠移植模型进行评价来明确(非专利文献26)。作为代表性的小鼠癌移植模型,可列举:作为黑色素瘤模型的B16癌皮下移植模型、作为胰腺癌的腹膜接种模型的Pan-02癌腹腔移植模型(非专利文献26)。
如上所述,BG-CpG在医药领域、特别是在癌免疫疗法或疫苗佐剂中的有效性已得到确认。为了面向实用化,希望通过伴随BG-CpG活性的提高而减少给药量来降低制造成本、确立详细的制造方法、品质评价法。作为确定BG-CpG的有效性的要素之一,本发明人着眼于粒径。如本领域技术人员所周知,确认了供本用途的BG-CpG在水系介质中具有数十nm~数百nm的平均粒径(专利文献9-14)。具有这样的大小的药品被称为纳米药物,作为具有与化学合成药品或利用细胞培养等制造的生物药品不同的特性或功能的药品形态受到关注,近年来相继公布了有关开发的指南等(非专利文献32-34)。这些之中,为了将纳米药物的有效性最大化、同时得到药品所应具备的品质,阐述了需要将各种特性最优化。其中,关于粒径,认为也是决定纳米药物的特性的特别重要的要素之一。
关于BG-CpG的粒径与药理作用的相关性,有如下所述的报道。在专利文献10中公开了:作为该用途中的通常的平均粒径为10nm~100nm、作为优选的平均粒径为20nm~50nm的范围。另外,还公开了:实际供试验的BG-CpG(K3-SPG)的平均粒径为30nm。另一方面,在以体外的细胞刺激性为基础指标研究了BG-CpG的粒径与药理作用的相关性的专利文献11中,公开了BG-CpG(核酸为D35-dA40、且β-葡聚糖为SPG的复合物)的适合的回转半径范围是20nm~200nm。详细而言,作为以回转半径计具有5nm~200nm的大小的该复合物的药理作用的比较结果,显示回转半径20nm~150nm作为合适的范围。更详细而言,作为该复合物所具有的生理活性的回转半径依赖性,公开了以下的1)-3)的事实。1)回转半径为5nm~10nm的该复合物的生理活性明显低;2)回转半径为150nm~200nm的该复合物的生理活性稍低;以及3)在回转半径为20nm~150nmm的范围内该复合物的生理活性没有大的差别。
这里,作为回转半径小的复合物的活性低的原因,可列举:在小的复合物中实际上复合物形成效率不充分;以及小的分子具有不易被细胞摄入的性质。这里,公开了:尽管测定原理存在不同,但若将回转半径为2倍的粒径换算成大概的平均粒径,则该复合物的平均粒径在40nm~300nm的范围适合,在该范围内该复合物的药理作用没有显著差异。另一方面,关于作为BG-CpG的构成成分的β-葡聚糖的分子量和BG-CpG的粒径控制法,在上述的专利文献11中公开了:在核酸为D35-dA40时,通过与各种分子量的SPG进行复合化,可得到在以回转半径计为10nm以下~200nm左右的范围内平均粒径不同的一系列BG-CpG。另外,在专利文献12中公开了分子量为45万(以三聚体计)的SPG的制造方法,但关于SPG的分子量对该复合物的粒径产生的影响则没有讨论。在专利文献13中显示:作为β-葡聚糖所应具备的重均分子量为2000以上、以及分子量为45万(以三聚体计)的SPG被用作BG-CpG的构成成分,但关于SPG的分子量对该复合物的粒径产生的影响则没有讨论。
根据上文,关于BG-CpG的粒径与药理作用的相关性,在数nm~数百nm的平均粒径范围内进行了研究,公开了:1)与平均粒径为20nm~300nm的BG-CpG相比,平均粒径为20nm以下或300nm以上的BG-CpG的活性低;2)虽然在平均粒径为20nm~300nm的范围内BG-CpG的生理活性的粒径依赖性并不明确,但通常适用的范围是10nm~100nm,优选的平均粒径的范围是20nm~50nm。另外,判明平均粒径为30nm的BG-CpG被积极地用作抗癌药或疫苗佐剂。

发明内容
发明所要解决的课题
BG-CpG具有β-葡聚糖与核酸通过非共价键缓慢地聚集的特征性结构,但关于BG-CpG的粒径与药理作用的相关性的见解还不充分。因而,为了得到由BG-CpG等β-葡聚糖与核酸的复合物构成的药品,需要详细研究粒径对药理作用产生的影响。本发明的课题为:对于β-葡聚糖与核酸的复合物,找到以往未知的适合的粒径范围。而且,本发明的课题还在于:确立用于制造粒径被控制在适当范围的BG-CpG等β-葡聚糖与核酸的复合物的稳健性高的制造方法和分析方法。
用于解决课题的手段
本发明人进行深入研究,制造了平均粒径阶段性地不同的一系列的BG-CpG。其结果,本发明人发现了:可制造平均粒径被严格控制在约20nm~180nm、优选约20nm~130nm或约40nm~180nm、更优选约80nm~130nm或约130nm~180nm的范围的BG-CpG。随后研究了平均粒径处于20nm~130nm或40nm~180nm的范围的BG-CpG的抗癌作用,发现平均粒径越大则其作用也越大。
因而,本发明包含以下内容。

发明效果
根据本发明,提供BG-CpG,其中,通过在实用性高的制造方法、制造条件下选择或控制β-葡聚糖的分子量特性,在20nm~180nm、优选20nm~130nm或40nm~180nm、更优选80nm~130nm或130nm~180nm的范围严格地控制平均粒径。因而,作为癌免疫疗法等的治疗药或预防药或它们的构成成分之一,可选定最适的粒径。与以往的BG-CpG相比,本发明的BG-CpG以较少的给药量得到显著的有效性,因此有可能可适用于利用以往的BG-CpG难以治疗或预防的疾病。

[图9]图9显示K3-LNT的药理作用的体内评价。在C57BL/6小鼠的皮下移植2.5×105个B16肿瘤细胞(将移植日作为第0天),在7天、9天和11天后总计3次向肿瘤内给予K3-LNT。作为比较,给予生理盐水(saline)。横轴显示进行B16的皮下移植后的天数,纵轴显示K3-LNT和生理盐水给药组的肿瘤大小。图9中使用的受试物质的批号如下。KALNT0706:20201106-07、KALNT0710:20201106-04、KALNT0709:20201106-03、KALNT0712:20201106-06。

具体实施方式
1.粒径受控的β(1→3)葡聚糖与核酸的复合物
本发明提供粒径受控的β(1→3)葡聚糖与核酸的复合物。本发明人通过进行深入研究,发现了在β(1→3)葡聚糖与核酸的复合物的平均粒径为20nm~180nm的范围内、优选为20nm~130nm或40nm~180nm、更优选为80nm~130nm或约130nm~180nm的范围内的情况下,该复合物的抗癌活性明显高,从而完成了本发明。因而,可使用平均粒径被控制在这种范围的BG-CpG作为癌免疫疗法等的治疗药或预防药。
本发明中使用的β(1→3)葡聚糖只要具有与核酸形成复合物的性质即可,没有特别限定,可列举:裂褶菌多糖(SPG)、香菇多糖(LNT)、凝胶多糖(curdlan)、硬葡聚糖(scleroglucan)、茯苓多糖(pachyman)、灰树花多糖(grifolan)、昆布多糖(laminaran)等β(1→3)葡聚糖类。已经报道了它们具有与核酸形成复合物的性质(WO2004/100965和日本专利第4850512号、免疫刺激剂)。作为可在本发明中使用的特别适合的β(1→3)葡聚糖,可列举:SPG和LNT。
本发明中的β(1→3)葡聚糖与核酸的复合物并不限定于此,优选为β(1→3)葡聚糖(BG)与包含CpG ODN的核酸形成的复合物(BG-CpG)。进一步优选地,本发明中的复合物是SPG与在K型CpG ODN(K3)的3’末端连接有40个碱基dA的CpG ODN(K3-dA40)的复合物(K3-SPG)或LNT与在K型CpG ODN(K3)的3’末端连接有40个碱基dA的CpG ODN(K3-dA40)的复合物(K3-LNT)。
通过筛选SPG产生能力高的裂褶菌菌株,在适当的培养条件下培养,使在培养基中高效率地分泌SPG,纯化其培养基,从而可得到SPG,这在下述4.中详细阐述。也可直接使用SPG,但根据需要在控制为适当的分子量后使用。关于进行β-葡聚糖的分子量控制的方法,也在下述2.中详细阐述。
在本发明中,“粒径受控的复合物”是指,为了使β-葡聚糖与核酸的复合物的平均粒径达到目标范围,使用分子量适当的β-葡聚糖作为构成成分而制造的复合物。这里,β-葡聚糖的分子量可不被特定。而且,在本发明中,“严格的控制”是指,以大概10nm~50nm或较其窄的间隔控制该复合物的平均粒径。通过这样的严格控制,可判明各种医药用途的β-葡聚糖与核酸的复合物的最适的平均粒径范围。而且,通过这样的严格控制,可制造具有发挥最大药理效果的平均粒径的β-葡聚糖与核酸的复合物。
在WO2016/098832中,使用在大概1万~540万的范围内分子量不同的一系列SPG,制造与被称为d(A)40-D35的核酸的复合物,公开了其回转半径与生理活性的相关性。基于此,回转半径的控制范围是5nm~8nm、8nm~10nm、20nm~70nm、60nm~100nm、100nm~150nm、150~200nm,回转半径20nm以上的控制幅度为50nm以上。这意味着,若转换成粒径,则其控制幅度为大概100nm以上,不能说是严格的控制。另外,这些复合物的药理效果的强度仅限于3个等级的半定性的评价,同时在回转半径8nm~150nm的范围没有发现药理效果的不同。
另一方面,上述现有技术(Miyamoto N.等人,Bioconjugate Chemistry 2017,28,565-573)中,在以SPG和CpG-dA40为构成成分的β-葡聚糖与核酸的复合物(记作CpG-SPG)中,尝试着增大其粒径。这里,准备具有与CpG互补的序列的核酸(cCpG-dA40),制造了其与SPG的复合物(cCpG-SPG)。接下来,通过混合CpG-SPG和cCpG-SPG,两种复合物的CpG和cCpG通过非共价键结合,从而各复合物之间交联,生成粒径大的结构体(记作CL-CpG纳米凝胶)。CpG-SPG和cCpG-SPG的回转半径大概为10nm,相对于此,CL-CpG纳米凝胶的回转半径大概为150nm。CL-CpG纳米凝胶的回转半径以平均粒径计大概对应于300nm,与CpG-SPG相比,确认了具有强效的佐剂效果。
如上所述,在该现有技术的CL-CpG纳米凝胶中,虽然与CpG-SPG相比成功地使粒径增大,但还不能说可严格地控制粒径。即,无法严格地控制300nm左右的平均粒径,不能说可使β-葡聚糖与核酸的复合物的平均粒径最优化。
如上所述,在现有技术中,虽然有平均粒径成功地增大至大概300nm的例子,但还无法进行严格的控制、即无法以大概10nm~50nm或小于此的间隔控制该复合物的平均粒径。发明人认为:由于这种缺乏严格性的粒径控制,而有可能无法发挥β-葡聚糖与核酸的复合物本来具有的显著的药理效果。即,认为开发以下的技术较为重要:通过粒径的严格控制,在特定发挥最大药理效果的平均粒径的同时,可制造具有该平均粒径的β-葡聚糖与核酸的复合物。
根据菌株或品种,所产生、含有的β-葡聚糖的分子量可不同。因而,可通过最适的菌株的种类或培养、生长条件制造大分子量的β-葡聚糖。另外,作为天然多糖类的β-葡聚糖由于分子量分布幅度广,所以即使是假设平均分子量小的情况,也包含大的分子量成分。通过对其进行分馏、纯化,可得到分子量大的β-葡聚糖。而且,还已知通过将具有规定分子量的多糖类进行化学或物理交联,可使分子量增大(Basu A.等人,Bioconjugate Chemistry2015,26,1396-1412;Pawar H.A.等人,Biology and Medicine(Aligarh)2015,6,224)。
利用这些方法,多糖类的分子量可无限大,即,可大到作为凝胶而不溶化。即,通过适当地应用这些方法,可使β-葡聚糖的分子量增大至无限大。而且,根据本发明中公开的分子量控制条件等,一旦取得、制造高分子量的β-葡聚糖后,可进行低分子量化。如已经阐述的那样,在现有技术(WO2016/098832和Miyamoto N.等人,Bioconjugate Chemistry 2017,28,565-573)中,公开了回转半径为150nm的β-葡聚糖与核酸的复合物的有用性。结果显示:通过按照上述的任何方法准备高分子量的β-葡聚糖,以平均粒径计为300nm左右的β-葡聚糖与核酸的复合物在药学上有用。然而,在该现有技术中,由于无法严格地控制粒径、即无法以大概10nm~50nm或小于此的间隔控制平均粒径,因此不能说300nm的平均粒径是最适的。
可制造由如此操作而得到的SPG和核酸(优选确认到作为癌免疫疗法剂的有用性的K3-dA40)构成、且粒径得到严格控制的BG-CpG。令人惊讶的是,本发明人发现了:在设想为癌免疫疗法的药理评价系统中,平均粒径处于80nm~130nm或130nm~180nm的范围的BG-CpG的药理效果明显高。如已经叙述的那样,在以往的研究中历史记录丰富的BG-CpG的平均粒径为30nm附近或20nm~50nm,本发明人发现的平均粒径为其范围外。
在本说明书的实施例中,对于上述的粒径得到严格控制的BG-CpG,使用充分考虑了是否适合临床使用的最新的体外和体内的药理评价系统,对作为癌免疫疗法剂的效果进行了评价。其结果,本发明人发现了:BG-CpG的粒径与药理活性存在强的相关性,平均粒径处于80nm~130nm或130nm~180nm的范围的BG-CpG具有明显高的药理作用。
如下述所示的实施例中所示,使用利用hPBMCs来评价BG-CpG的IFN-α诱导能力的体外评价系统和利用小鼠脾脏细胞来评价BG-CpG的IFN-γ诱导能力的体外评价系统,确定了最适合药理活性(抗癌活性)的BG-CpG的粒径。认为这些体外评价系统与实验间的偏差多的小鼠癌移植模型相比数据的稳定性或定量性高。另外,BG-CpG的IFN-α诱导水平在中等用量下显示了最大效果(Emax)(所谓钟形)。为了提高定量性,在下述的实施例中,通过曲线下面积(area under curve。以下,称为AUC)算出了BG-CpG的IFN-α诱导能力。同样,对于IFN-γ,也将BG-CpG的添加量设为2μg/mL和10μg/mL这2个水平,分别测定培养24小时后的IFN-γ产生量,将它们的总计值作为药理作用的强度指标。其结果,在20nm~130nm或40nm~180nm的范围,平均粒径越大,BG-CpG就显示越强的药理作用。通过本发明的粒径的严格控制,首次发现了具有最大药理作用的BG-CpG的平均粒径。
在下述实施例中,进一步使用向小鼠中移植了B16黑色素瘤癌症的模型(Kitahata,Y.等人.Circulating nano-particulate TLR9 agonist scouts out tumormicroenvironment to release immunogenic dead tumor cells.Oncotarget 7,48860-48869(2016)),对通过体外评价锁定的BG-CpG的抗癌活性进行评价。其结果,在体内观察到了BG-CpG的抗癌活性。如以上所述,发现了粒径受到严格控制的本发明的BG-CpG发挥高的抗癌活性。
2.包含粒径受控的β(1→3)葡聚糖与核酸的复合物的药物组合物
可将本发明的BG-CpG根据其作用机制用作各种疾病的治疗药或预防药、疫苗。本发明提供药物组合物,其包含上述本发明的复合物。确定对特定疾病的预防或治疗的适应性,通过探索研究、非临床试验、临床试验预测或验证其有效性和安全性后进行实用化。在该药品开发过程中的任何阶段,将本发明的BG-CpG的粒径最优化、或者确定可接受的范围。通常,在探索研究阶段,以其体外或体内的活性为指标,将粒径最优化、或者确定可接受的范围。另外,在非临床试验中的安全性试验中,从安全性方面验证了作为最优化的粒径或容许范围而设定的粒径范围的妥当性。
本发明的药物组合物可通过按照常规方法将上述本发明的复合物制成制剂而得到。本发明的药物组合物包含本发明的复合物和药理学上可接受的载体。另外,本药物组合物可进一步包含抗原。这样的药物组合物以适合于口服或胃肠外给药的剂型的形式提供。
作为用于胃肠外给药的组合物,例如使用注射剂、栓剂等,注射剂可包含静脉注射剂、皮下注射剂、皮内注射剂、肌肉注射剂、点滴注射剂等剂型。这样的注射剂可按照已知的方法调制。作为注射剂的调制方法,例如可通过将上述本发明的复合物溶解或悬浮在用于普通注射剂的无菌水性溶剂中来调制。作为注射用水性溶剂,例如可使用:蒸馏水;生理盐水;磷酸缓冲液、碳酸缓冲液、Tris缓冲液、乙酸缓冲液等缓冲液等。这样的水性溶剂的pH可列举5~10,优选为6~8。所调制的注射液优选填充在适当的安瓿中。
另外,通过对本发明的复合物的悬浮液进行真空干燥、冷冻干燥等处理,还可调制本发明的复合物的粉末制剂。将本发明的复合物以粉末状态保存,在使用时将该粉末用注射用水性溶剂分散,从而可供使用。
药物组合物中的本发明的复合物的含量通常为药物组合物整体的约0.1~100重量%、优选为约1~99重量%、进一步优选为约10~90重量%左右。
本发明的药物组合物可单独含有本发明的复合物作为有效成分,也可将本发明的复合物与其他有效成分组合而含有。
3.医药用途
本发明的复合物具有优异的免疫刺激活性,因此本发明的复合物和药物组合物可用作免疫刺激剂。通过对哺乳动物(人等灵长类、小鼠等啮齿类等)给予本发明的复合物或药物组合物,可在该哺乳动物中引起免疫反应。特别是,本发明的复合物具有D型CpG ODN的活性特性,刺激外周血单核细胞,大量地产生I型干扰素(Pan-IFN-α、IFN-α2等)和II型干扰素(IFN-γ)两者,因此可作为I型干扰素产生诱导剂、II型干扰素产生诱导剂、I型和II型干扰素产生诱导剂。由于诱导I型和II型干扰素两者的产生,所以本发明的复合物和含有其的药物组合物对I型和II型干扰素中的任何一方或双方有效的疾病的预防或治疗有用。作为I型干扰素有效的疾病,可列举:病毒感染症(例如,丙型肝炎病毒(HCV)、疱疹病毒、乳头瘤病毒、RS病毒、流感病毒等)、癌症等。作为II型干扰素有效的疾病,可列举:过敏性疾病、细胞内寄生性原虫(利什曼原虫等)或细菌(李斯特菌、结核菌等)等的感染症等。对于包含RS病毒或流感病毒的急性病毒感染症,I型干扰素和II型干扰素均提高参与病毒排除的免疫应答,因此本发明的复合物和含有其的药物组合物可期待对急性病毒感染症有效。
对可通过本发明的复合物和含有其的药物组合物来治疗的肿瘤没有特别限定,优选列举:胰腺癌、肝癌、胃癌、大肠癌、肾癌、乳腺癌、子宫癌(例如,宫颈癌)、卵巢癌、肺癌、甲状腺癌、皮肤癌(例如,黑色素瘤(黒肉瘤))、头颈部癌、肉瘤、前列腺癌、膀胱癌、脑瘤、胃肠道间质瘤(GIST)、白血病(例如,急性骨髓性白血病(AML)、慢性骨髓性白血病(CML)、或慢性淋巴性白血病(CLL)或急性淋巴性白血病(ALL))、淋巴瘤或恶性淋巴瘤(例如,T淋巴瘤(包括成人T细胞白血病)、B细胞淋巴瘤、非霍奇金淋巴瘤或弥漫性大B细胞淋巴瘤(diffuselarge B-cell lymphoma、DLBCL)),更优选列举:认为是对由本发明的复合物和含有其的药物组合物强效诱导的细胞因子即IFN-α和IFN-γ的敏感性高的癌症(非专利文献26和Martin-Hijaro,L.和SainZ Jr.,B.2020:The interactions between cancer stem cellsand the innate interferon signaling pathway.Frontiers in immunology 11,Article 526.)的黑色素瘤、T淋巴瘤、大肠癌、胰腺癌、慢性骨髓性白血病、肝癌、宫颈癌、卵巢癌、膀胱癌、成人T细胞白血病,进一步优选列举:黑色素瘤、T淋巴瘤(包括成人T细胞白血病)、大肠癌、胰腺癌。
另外,本发明的复合物具有强效的疫苗佐剂活性,若将本发明的复合物与抗原一起给予至哺乳动物,则可强效地诱导针对该抗原的免疫反应。因此,本发明还提供:用于诱导针对抗原的免疫反应的组合物,其包含(a)本发明的复合物和(b)该抗原。本发明的复合物强效地诱导针对抗原的体液免疫反应(产生抗原特异性抗体)和细胞免疫反应(诱导抗原特异性CTL)两者。因此,本发明的复合物和含有其的药物组合物可用作疫苗佐剂。
在本说明书中,佐剂是促进免疫应答的辅助剂,是指与抗原一起给予至生物体时非特异性地增强针对该抗原的免疫应答的物质。
作为抗原,只要对给药对象的哺乳动物(人等灵长类、小鼠等啮齿类等)而言具有抗原性、且抗体或细胞毒性T淋巴细胞(CTL、CD8+T细胞)可识别为抗原即可,没有特别限定,可使用成为抗原的所有物质(蛋白、肽、核酸、脂质、糖、以及上述物质的修饰物(例如,引入了一个或多个氨基酸的缺失、取代和/或添加等(以下,记作突变等)的修饰物等)。作为抗原,还可使用来自原虫、真菌、细菌、病毒等病原体的抗原、与癌症或特定的疾病有关的抗原。
在本说明书中,“抗原A来自病原体X”是指抗原A作为构成因子包含在该病原体X中。例如,在抗原A为多肽的情况下,是指该多肽的氨基酸序列存在于由病原体X的基因组编码的蛋白的氨基酸序列中。
作为来自病原体的抗原,可列举:病原体本身或其一部分、灭活或减毒的病原体本身或其一部分、或它们的引入了突变等的修饰物等。
若使用来自病原体的抗原作为抗原,则引发针对该抗原的免疫反应,构建在免疫学上将包含该抗原的病原体排除到生物体外的机制。因此,包含(a)本发明的复合物和(b)来自病原体的抗原的、用于诱导针对该抗原的免疫反应的组合物可用于该病原体的预防或治疗。
本发明的复合物强效地同时诱导针对抗原的体液免疫反应(产生抗原特异性抗体)和细胞免疫反应(诱导抗原特异性CTL)两者。因此,已知由细胞毒性T淋巴细胞识别的来自细胞内感染性病原体(病毒、原虫、真菌、细菌等)的抗原或与癌化的细胞相关的抗原(例如,肿瘤抗原)等适合用作抗原。
对细胞内感染性病毒没有特别限定,可列举:RS病毒、流感病毒、副流感病毒、丙型肝炎病毒(HCV)、甲型肝炎病毒(HAV)、乙型肝炎病毒(HBV)、埃博拉病毒、巨细胞病毒、腺病毒、脊髓灰质炎病毒、日本脑炎病毒、麻疹病毒、腮腺炎病毒、风疹病毒、狂犬病病毒、黄热病毒、水痘带状疱疹病毒、汉坦病毒、登革热病毒、诺如病毒、轮状病毒、细小病毒、冠状病毒、瘟热病毒、成人T细胞白血病病毒(HTLV-1)、人免疫缺陷病毒(HIV)、疱疹病毒、乳头瘤病毒等。作为细胞内感染性细菌,可列举:支原体等。作为细胞内感染性原虫,可列举:疟原虫、血吸虫等。细胞内感染性病原体优选为病毒(具体而言,RS病毒、或流感病毒等)。
作为与癌化的细胞相关的抗原,可列举:在癌化的细胞中特异性地表达的蛋白、糖链、肽、以及上述物质的突变体(缺失、取代和/或添加)或其修饰物等。
本发明的复合物强效地诱导I型和II型干扰素两者,因此在一个方案中,作为病毒,选择引起I型干扰素和II型干扰素均有效的急性病毒感染症的病毒(例如,RS病毒、流感病毒)。
例如,通过将包含(a)本发明的复合物和(b)来自病原体或癌症的抗原的、用于诱导针对该抗原的免疫反应的组合物给予至该病原体感染症或癌症的患者、有可能罹患该病原体感染症或癌症的人,抗原特异性地激活接受了该给予的对象中的细胞毒性T淋巴细胞(CTL),诱导该抗原特异性抗体的产生,即诱导恒温动物(优选人)的防御免疫反应,从而可预防、治疗该感染症或癌症。即,该组合物可用作用于预防或治疗上述感染症、癌症等疾病的疫苗。
另外,本发明的复合物可强效地诱导针对抗原的体液免疫反应(产生抗原特异性抗体)和细胞免疫反应(诱导抗原特异性CTL)两者,因此也可使用病原体或癌细胞的表面抗原和内部抗原中的任一种作为抗原,还希望混合表面抗原和内部抗原进行使用。
包含(a)本发明的复合物和(b)抗原的、用于诱导针对该抗原的免疫反应的组合物可依据上述本发明的药物组合物来调制。
本发明的复合物或本发明的药物组合物可与其他药物同时或分别给药。例如,可在给予其他药物之后给予本发明的复合物或本发明的药物组合物、或者在给予这种复合物或药物组合物之后给予其他药物、或者同时给予该复合物或药物组合物和其他药物。作为其他药物,可列举:化学疗法剂、放射疗法等各种抗癌药等,除本发明的复合物或药物组合物以外还包含其他药剂的药物组合物也包含在本发明中。
TLR-9激动剂通过与免疫检查点抑制剂并用而发挥显著的抗肿瘤效果(Adamus,T.和Kortylewski,M.,The revival of CpG pligonucleotide-based cancerimmunotherapies,Contenp.Oncol.(PoZn.)21(1A),56-60(2017).)。作为免疫检查点抑制剂,有PD1抑制剂、PD-L1抑制剂和CTLA-4抑制剂,例如分别已知抗PD-1抗体、抗PD-L1抗体和抗CTLA-4抗体(Bagchi,S.,Yuan,R.和Engleman,E.G.,Immune checkpoint inhibitorsfor the treatment of cancer:clinical impact and mechanism of response andresistance,Ann.Rev.Pathol.16,223-249(2021).)。被称为IMO-2125的TLR-9激动剂与抗PD-1抗体的并用效果通过胰腺癌的动物模型得到验证(Carbone C.等人,Intratumoralinjection of TLR-9 agonist promotes an immunopermissive microenvironmenttransition and causes cooperative antitumor activity in combination withanti-PD1 in pancreatic cancer,J.Immunother.Cancer 9 2021)。因此,作为TLR-9激动剂的本发明的复合物或本发明的药物组合物与免疫检查点抑制剂的并用可成为有用的治疗方法。
4.具有特定范围的平均粒径的β(1→3)葡聚糖与核酸的复合物的制造方法
本发明还提供具有特定范围的平均粒径的、β(1→3)葡聚糖与核酸的复合物的制造方法。在本发明中,使用供给源明确的原材料,通过可实现的制造方法成功地制造了粒径得到严格控制的BG-CpG。
如已经阐述的那样,SPG是裂褶菌(SchiZophyllum commune)的菌株产生的β(1→3)葡聚糖,已知其菌株有多个。本发明人深入研究了不同的裂褶菌菌株的SPG的生产量,结果发现了:在特定的菌株中生产能力特别高。因而,本发明的适合方案为:使用SPG的生产能力高的裂褶菌菌株来得到SPG。同样,本发明的适合方案为:香菇多糖(LNT)是香菇子实体中所含的β(1→3)葡聚糖,从香菇子实体中提取LNT。
(1)使用了裂褶菌菌株的β-葡聚糖的制造方法
在使用裂褶菌菌株等制造β-葡聚糖时,只要按照本领域技术人员所周知的方法(菊本昭一等人.裂褶菌の生産する多糖に関する研究.農化44,337-342(1970);水野卓·川合正允编.キノコの化学·生化学.学会出版センター(1992)),将裂褶菌菌株在该菌株可生长的培养基中培养,使在菌体外的培养基中生产SPG即可。作为其培养基,可使用:含有属于裂褶菌的菌株通常可利用的营养源(碳源、氮源、无机盐类)、根据需要还包含有机营养源的普通培养基,作为碳源,优选含有糖类的培养液。作为这些培养基,各种合成培养基、半合成培养基、天然培养基等均可利用。
作为上述碳源,优选糖类,作为该糖类,可列举:葡萄糖、果糖、甘露糖、半乳糖、木糖、阿拉伯糖等单糖类;蔗糖、麦芽糖、乳糖、海藻糖等二糖类;果寡糖、木寡糖等寡糖类;糊精或淀粉等多糖类,这些可单独或组合使用。这些之中,优选使用葡萄糖、果糖等6炭糖、蔗糖、乳糖等二糖类、淀粉或糊精、或这些碳水化合物的水解物等多糖类作为主碳源。另外,还可使用甜菜汁、甘蔗汁、以柑橘类为代表的水果汁、或在这些榨汁中添加了糖的榨汁等。此外,可适当使用甘油、乙二醇等醇类、甘露糖醇、山梨糖醇、赤藓糖醇等糖醇类、有机酸等其他碳源。这些碳源可在培养过程中随时添加,例如,若在培养基中适当加入葡萄糖等糖类,则可使β-葡聚糖的生产速度、生成量相对增大。
作为上述氮源,可将蛋白胨、肉提取物、大豆粉、酪蛋白、氨基酸、麦芽提取物、玉米浆(corn steep liquor)、酪蛋白分解物、酵母提取物、尿素等有机氮源、硝酸钠、硫酸铵、氨气、氨水等无机氮源单独或组合使用。
作为上述无机盐类,例如可使用氯化钠、氯化钾、碳酸钙、硫酸镁、磷酸钠、磷酸钾、氯化钴等盐类、重金属类盐等,根据需要,还可添加使用维生素类。需要说明的是,在培养中起泡的情况下,也可在培养基中适当添加已知的各种消泡剂。
对本发明的菌株的培养条件没有特别限定,可在该菌株可良好地生长的范围内适当选择。通常,可在pH5.0~pH8.5、20℃~35℃下培养5天~14天左右,这些培养条件可根据使用的菌株的种类或特性、外部条件等适当变更,可选择最适条件。另外,关于培养基中的菌株的接种量,在烧瓶培养的情况下为1%,在铂环、规模扩大的情况下,优选添加正式培养液的1%(v/v)~10%(v/v)的种培养液,只要实质上可进行培养即可,不受此限制。
本发明的菌株的培养在通过通气搅拌、振荡等形成的需氧条件下进行。培养时间优选进行至达到目标β-葡聚糖的生成浓度,通常进行5天~14天。另外,可连续地添加作为β-葡聚糖的底物的糖类或培养基成分,连续地生产β-葡聚糖。
在如上所述进行培养后,按照常规方法从培养液中分离、采集在菌体外分泌生产的β-葡聚糖。具体而言,可选择、组合通过离心、过滤等从培养液中分离去除菌体等固态物、利用活性炭、离子交换树脂等去除杂质或盐类等各种已知的纯化方法来进行。而且,例如可将吸附于疏水性树脂或从该树脂洗脱、使用了乙醇、甲醇、乙酸乙酯、正丁醇等的溶剂沉淀、利用了硅胶等的柱法或薄层色谱法、使用了反相柱的分离用高效液相色谱法等单独或适当组合,根据情况反复使用来进行分离纯化。
另外,在利用上述方法分离在菌体外分泌生产的β-葡聚糖的前后,可对菌体实施灭菌。灭菌温度只要是菌体死亡的温度即可,没有特别限定,例如可在90℃以上、或加压下、121℃下实施。灭菌时间可设为任意的时间,优选为15分钟~60分钟。
(2)使用了香菇子实体的β-葡聚糖的制造方法
香菇多糖是香菇子实体中所含的β-葡聚糖。因而,可通过从香菇子实体中提取、纯化而得到香菇多糖。在本发明的香菇多糖的制造中,可按照本领域技术人员周知的方法(大森清寿·小出博志.キノコ栽培全科.農山漁村文化協会(2001);最新バイオテクノロジー全書編集委員会編.きのこの増殖と育種.農業図書(1992))或新的方法,通过原木栽培或菌床栽培等方法栽培香菇子实体,从该子实体中提取香菇多糖。
原木栽培主要是使用麻栎、枹栎、白栎栽培木材腐朽菌的蘑菇的方法,是在采伐并干燥后的原木上栽种菌的方法。进行已接种作业的产生管理,收获产生的香菇。
菌床栽培可列举:在1种以上的阔叶树锯屑、木屑或木片等木材粉碎物中混合1种以上的麦麸、米糠等谷类、豆类的粉碎物,加入水调整含水率,将所得培养基按照常规方法进行加压整形装袋、灭菌、冷却,制作菌床。在其上接种香菇种菌,将进行了一定期间的培养管理的菌床在通常栽培(从袋中取出菌床,由培养基整体产生蘑菇的栽培方法)或上面栽培(仅露出菌床的上部,仅从培养基上部产生蘑菇的栽培)中进行产生管理,收获产生的香菇。
可按照本领域技术人员周知的方法(Chihara等人.Fractionation andpurification of the polysaccharides with marked antitumor activity,especiallylentinan,from Lentinus edodes(Berk.)Sing.Cancer Research 30,2776-2781(1970);千原呉郎.癌と免疫增强.講談社(1980))进行香菇多糖的提取、纯化。或者,也可按照还未知的新方法进行香菇多糖的提取、纯化。
香菇子实体中所含的β-葡聚糖按照常规方法由子实体来制造。具体而言,可选择对剪断的子实体进行热水提取,利用活性炭、离子交换树脂等去除杂质或盐类等各种已知的纯化方法组合进行。而且,例如可将使用了乙醇、甲醇、乙酸乙酯、正丁醇等的溶剂沉淀、吸附于疏水性树脂或从该树脂洗脱、利用硅胶等进行的柱法或薄层色谱法、使用了反相柱的分离用高效液相色谱法等单独或适当组合,根据情况反复使用,从而进行分离纯化。
在这些制造工序的过程中或最终工序中,根据需要利用有机溶剂、水系介质进行洗涤,按照本领域技术人员周知的方法进行冷冻干燥、减压干燥等,得到目标物。
(3)β-葡聚糖的分子量控制方法
作为进行β-葡聚糖的分子量控制的方法,可列举:酶分解、碱水解、酸水解、利用高压均质器的剪切力、超声波的能量进行的分解,但并不限于此。可从这些方法中选择适当的方法进行适当利用(宫崎利夫编.多糖類の構造と生理活性.朝仓书店(1990))。
(4)确定β-葡聚糖的结构的方法
关于所得到的β-葡聚糖的结构,与普通的有机化合物一样,可通过从1H-NMR、13C-NMR、红外吸收光谱、元素分析等方法中选择或组合来确定。
在β-葡聚糖的核磁共振波谱(以下,称为NMR)的测定中使用作为通用的测定溶剂的二甲基亚砜-d6(DMSO-d6)的情况下,在1H-NMR中还观测到了羟基的信号,提供非常宽的波谱。另一方面,0.25M的氘化氢氧化钠/重水较DMSO-d6更易溶解β-葡聚糖。另外,由于在1HNMR中没有观察到羟基的信号,所以明确地判定葡聚糖构成单元的特征。在测定溶剂使用0.25M的氘化氢氧化钠/重水的情况下,β-葡聚糖在室温下处于分解的倾向,因此通过将NMR波谱测定温度条件、所需样品量最优化,可得到适合分析的波谱。
关于分子量和分子大小,可通过多角度光散射法、尺寸排阻色谱(SEC)、动态光散射法等本领域技术人员周知的方法,作为绝对数值、作为相当于分子量已知的标准物质的相对数值、或作为具有一定相关性的尺度来确定。β(1→3)葡聚糖(BG)是链状分子,在中性水溶液中以3条链络合的状态存在,而在碱性溶液中以解离成单链的状态存在(WO2015/041318、免疫賦活活性を有するオリゴヌクレオチド含有複合体及びその用途;日本专利第5605793号、β-1,3-グルカン/核酸複合体の調製方法(特愿2010-043592);田之畑大二郎、真田雄介、望月慎一、宫本宽子、樱井和朗.核酸/β-1,3-グルカン集合体の溶液物性.高分子論文集72:37-47(2015))。使用与BG一样具有链状结构的茁霉多糖作为分子量标准品,通过碱性条件下的尺寸排阻色谱法(SEC法)可推断相对分子量。再并用多角度光散射检测器(MALLS)作为色谱检测器,从而可利用静态光散射法推断绝对分子量和分子大小(Wyatt,P.J.Anal Chim Acta 1993,272,1;PodZimek,S.J Appl Polym Sci 1994,54,91)。
(5)核酸的制造方法
关于核酸,可根据本领域技术人员周知的合成技术和核酸化学制造K3-dA40等。这样的制造方法例如记载于(1)Gait,M.J.(1985).Oligonucleotide Synthesis:APractical Approach,IRL Press、(2)Gait,M.J.(1990).Oligonucleotide Synthesis:APractical Approach,IRL Press、(3)Eckstein,F.(1991).Oligonucleotides andAnalogues:A Practical Approach,IRL Press、(4)Adams,R.L.等人.(1992).TheBiochemistry of the Nucleic Acids,Chapman&Hall、(5)Shabarova,Z.等人.(1994).Advanced Organic Chemistry of Nucleic Acids,Weinheim;Blackburn,G.M.等人.(1996).Nucleic Acids in Chemistry and Biology,Oxford University Press、(6)Hermanson,G.T.(I996).Bioconjugate Techniques,Academic Press,这些文献的与本说明书相关的部分作为参考而引用。而且,还可按照Stein等人,1988,Nucl.Acids Res.16:3209中记载的方法合成硫代磷酸酯寡核苷酸。需要说明的是,也可进一步利用Sarin等人,1988,Proc.Natl.Acad.Sci.USA 85:7448-7451中记载的方法,使用调节孔玻璃聚合物支撑体等,调制甲基膦酸酯寡核苷酸。
(6)BG-CpG的制造方法
以如上所述进行操作而得到的β-葡聚糖和核酸为原料,按照本领域技术人员周知的方法,可制造BG-CpG((1)WO2016/103531和PCT/JP2014/084772、免疫賦活活性を有する核酸多糖複合体の抗腫瘍薬としての応用、(2)WO2015/041318、免疫賦活活性を有するオリゴヌクレオチド含有複合体及びその用途、(3)日本专利第5605793号、β-1,3-グルカン/核酸複合体の調製方法(特愿2010-043592))。
即,考虑到构成β-葡聚糖的主链的糖单元数(以下,记作mG)与核酸的碱基数(使用脱氧腺苷作为与β-葡聚糖进行相互作用的核酸时,以下,记作dA)的化学计量比为2:1、即mG/dA比为2,根据目的适当地增减其中一方的构成成分的混合比例。通常β-葡聚糖形成螺旋结构或聚集体,因此在复合前施行碱处理或溶解于包含N,N-二甲基亚砜的水系介质中形成单体后进行复合物的形成。或者使两种构成成分在这些介质中共存,之后将体系中和或稀释或去除有机溶剂而形成复合物。实施BG-CpG的特性分析,以确认或保证其结构特征和药品所应具备的一般品质特性。
BG与CpG寡核苷酸的复合物(BG-CpG)在中性水溶液中稳定地存在,溶解性也良好,因此可利用动态光散射法(DLS法)进行粒径测定。若对在溶液或悬浮液中进行布朗运动的粒子照射激光,则在来自粒子的散射光中产生与扩散系数对应的波动。大的粒子运动慢,故散射光强度的波动稳定,另一方面,小的粒子运动快,故散射光的波动急剧地变化。在DLS法中,检测反映该扩散系数的散射光的波动,利用Stokes-Einstein式测定粒径。通过该方法求出的平均粒径和粒径分布是以胶体分散系制剂为中心显示其特性的重要因素之一(第17版修订日本药典参考信息“動的光散乱法による液体中の粒子径測定法”第2346-2348页)。在DLS法中,根据已检测的信号的分析方法的不同,有光子相关光谱法和频率分析法,适用于粒径为数nm~约1μm或直至确认到沉降的影响为止的粒子。在本说明书的实施例中,使用光子相关光谱法,由散射光强度的自相关函数,通过累积分析,求出基于散射光强度基准的平均粒径(第17版修订日本药典参考信息“動的光散乱法による液体中の粒子径測定法”第2346-2348页;ISO 22412:2008Particle size analysis-Dynamic light scattering(DLS))。这里,在受试物质的粒径分布为单峰型(单峰)的情况下,其峰顶的粒径为平均粒径,在本发明中,研究了该平均粒径与BG-CpG的药理作用的相关性。另一方面,在具有双峰型(双峰)或多峰型(多峰)的粒径分布的情况下,选择显示最大光散射强度的峰,研究了其峰顶的粒径即该峰的平均粒径与BG-CpG的药理作用的相关性。
接下来,给出实施例、试验例和药物组合物的例子,进一步详细地说明本发明,但本发明并不限定于这些。
实施例
实施例1通过菌体培养制造SPG
从本发明人收集并保有的、由在日本境内采集的样品分离的33株裂褶菌菌株中选出具有SPG高生产能力的裂褶菌菌株。使裂褶菌菌株在马铃薯葡萄糖琼脂(PDA)斜面上充分生长,将从该PDA斜面在表面生长的菌体的1/4用5mL灭菌水进行均质化,接种在1个预先灭菌的包含80mL SPG培养基(3.0%葡萄糖、0.3%酵母抽提物、0.15%磷酸二氢铵、0.1%磷酸二氢钾、0.05%硫酸镁7水合物、(灭菌前pH4.8))的500mL容量的三角烧瓶中。将接种了菌体的500mL容量的三角烧瓶用旋转振荡器在210rpm、28℃下振荡7天(168小时),进行培养。
培养结束后,将所得到的45mL或15mL培养液离心(10000rpm、10分钟),将培养液分离成培养上清和菌体。在得到的30mL或10mL培养上清中加入甲醇,调制成35%的甲醇溶液。对通过添加甲醇而析出的沉淀物进行离心(10000rpm、10分钟),回收其沉淀物。在回收的沉淀物中加入10mL或5mL无水甲醇进行洗涤,之后再次进行离心。在沉淀物中加入1mL~3mL灭菌水,静置约1小时左右,之后进行冷冻干燥,得到了SPG提取物。
试验例1裂褶菌的各菌株的SPG生产能力的评价
称量实施例1中得到的SPG提取物的量。以由该SPG提取物量和培养上清液量算出的培养上清中的SPG提取物浓度为指标,对各菌株的SPG提取物的生产能力进行评价。
如表1所示,确认到F-19065、F-30213、F-82046、F-33504、F-15977、F-15537的菌株具有高的SPG生产能力。另外,对于F-19065、F-30213、F-82046、F-33504的菌株,也确认到具有非常高的SPG生产能力。

实施例2通过SPG高产株的菌体培养制造SPG
选出SPG的生产能力高的裂褶菌菌株F-82046和F-30213这2株,用于制造SPG。
与实施例1同样地操作,将在PDA斜面的表面生长的菌体的1/4用3mL灭菌水进行均质化,接种在1个预先灭菌的包含80mL SPG培养基的500mL容量的三角烧瓶中。接种了菌体的500mL容量的三角烧瓶用旋转振荡器在210rpm、28℃下振荡4天,进行预培养。将5mL预培养液均质化,接种在包含80mL SPG培养基的500mL容量的三角烧瓶中。将接种了预培养液的500mL容量的三角烧瓶用旋转振荡器在210rpm、28℃下振荡10天,进行正式培养。
培养结束后,将全部培养液分别注入到离心管中,进行离心(10,000rpm、10分钟)。因所得上清的粘度高,故加入1.5~1倍量的灭菌水后,进行吸引过滤以去除游离菌体。在滤液中添加甲醇使达到35%,搅拌后静置1夜。对析出的沉淀物进行离心(10,000rpm、10分钟),用甲醇洗涤该沉淀物。接下来,在沉淀物中加入灭菌水使其浸润,之后进行冷冻干燥。将该冷冻干燥物作为SPG提取物。
将SPG提取物以1mg/mL的浓度溶解在1M的氢氧化钠水溶液中,在5℃下缓慢地振荡24小时。之后,用孔径5μm、接着是孔径1μm的Omnipore薄膜滤器(Merck Millipore制造)进行过滤,在所得到的滤液中加入2倍量的甲醇,使SPG析出。将已析出的SPG用甲醇:水=3:1的混合液洗涤数次。反复进行该洗涤直至溶液为中性,再用甲醇和丙酮各洗涤1次。最后,在40℃干燥,得到了已纯化的分子量控制前的SPG(块状SPG。以下,记作mSPG)。
以使用裂褶菌菌株F-82046制造的mSPG作为mSPG-1a、以来自菌株F-30213的SPG作为mSPG-2a。另外,使用作为试剂购入的2批SPG(制造商的批号SCZ-37-01A在本发明中是iSPG-1、批号SCZ-36-1在本发明中是iSPG-2)作为标准样品,获取这些mSPG的1H-NMR和13C-NMR波谱,进行比较。这里,溶剂为0.25M的氘化氢氧化钠/重水,发现了兼顾高S/N比和化合物的稳定性的最适测定温度为15℃,在该温度下进行了测定。如图1和图2所示,mSPG-1a和mSPG-2a的1H-NMR波谱和13C-NMR波谱均与SPG标准样品的波谱一致,同时没有观测到来自杂质的明显的信号。由以上结果确认到:本实施例中制造的2批mSPG均具有高纯度。
实施例3使用大型烧瓶通过菌体培养制造SPG
为了制造大量的SPG,使用2.5L的Ultra Yield Flask(Thomson InstrumentCompany制造)进行培养。作为裂褶菌菌株,使用F-82046株。培养设为前预培养、预培养、正式培养这3个阶段。在菌的接种中,使用1支裂褶菌菌株F-82046的冷冻种子。
为了制作裂褶菌菌株F-82046的冷冻种子,将在PDA斜面的表面生长的裂褶菌菌体的1/4用3mL灭菌水进行均质化,接种在1个预先灭菌的包含80mL SPG培养基的500mL容量的三角烧瓶中。将接种了菌体的500mL容量的三角烧瓶用旋转振荡器在210rpm、28℃下振荡3天,进行培养。向63mL培养液中添加7mL预先已灭菌的甘油进行混合,之后向Cryotube(Thermo Fisher Scientific制造)中分别注入1.8mL,在-80℃的冷库中冷冻保存。
在前预培养中,将1支冷冻种子在室温下解冻,全部接种在1个预先灭菌的包含80mL SPG培养基的500mL容量的三角烧瓶中。接种了菌体的500mL容量的三角烧瓶用旋转振荡器在210rpm、28℃下振荡3天,进行前预培养。在预培养中,将20mL前预培养液均质化,全部接种在2.5L预先灭菌的包含1000mL SPG培养基的Ultra Yield Flask中。将2.5L烧瓶用旋转振荡器在130rpm、28℃下振荡3天,进行了预培养。在正式培养中,将10mL预培养液接种在2.5L预先灭菌的包含1000mL SPG培养基的Ultra Yield Flask中。将该2.5L烧瓶用旋转振荡器在130rpm、28℃下振荡11天,进行了正式培养。在多个烧瓶间得到了稳定的SPG生产量,培养基中的SPG浓度平均为约2,000μg/mL。培养结束后,添加蒸馏水使各烧瓶的液量为1.5L,进行高压釜灭菌(121℃、20分钟)。将灭菌后的培养液再用等量的蒸馏水稀释,用尼龙袋(孔径150μm)去除菌体后,用滤纸(No.2.Advantec公司制造)进行吸引过滤。使用平流泵(Q系列、株式会社TACMINA)将所得到的处理滤液与甲醇混合,将甲醇浓度设为30%~35%后静置一夜,得到了析出物。使用镊子和柄勺、尼龙袋回收由该析出物构成的浮游组分和液体中的凝胶状浮游物。对回收的析出物进行离心(8,000rpm、30分钟),用甲醇洗涤该沉淀物。接下来,在沉淀物中加入灭菌水使其浸润后,进行冷冻干燥,以冷冻干燥体的形式得到了SPG提取物(SPG-247:收量10.4g)。
将作为该SPG提取物(SPG-247)的一部分的151mg以1mg/mL的浓度溶解在1M的氢氧化钠水溶液中,在5℃下缓慢地振荡24小时。之后,用孔径5μm、接着是孔径1μm的Omnipore薄膜滤器过滤,在所得到的滤液中加入2倍量的甲醇,使SPG析出。对析出的SPG用甲醇:水=3:1的混合液洗涤数次。反复进行该洗涤直至溶液为中性,再用甲醇和丙酮各洗涤1次。最后在40℃下干燥,得到了mSPG(mSPG-19a:收量125mg)。
实施例4通过碱处理控制SPG的分子量
以使用实施例2中记载的裂褶菌菌株F-82046制造的SPG提取物为原料,与mSPG-1a同样地操作,制造了mSPG-1b。以这里得到的mSPG-1b或mSPG-1a为原料,如下所述,制造了分子量受控的SPG。将mSPG以1mg/mL的浓度溶解在5M的氢氧化钠水溶液中。将该溶液的温度设为28℃~50℃,缓慢地振荡8小时~72小时。加入振荡后的溶液的2倍量的甲醇,使SPG析出。对析出的SPG用甲醇:水=3:1的混合液洗涤数次。反复进行该洗涤直至溶液为中性,再用甲醇和丙酮各洗涤1次。最后在40℃下干燥、纯化,得到了分子量受控的SPG。通过在这些范围内变更温度和时间,制造了具有各种分子大小的SPG。
制造的SPG见表2。

此外,还确认到:通过使用浓度为1M~5M的氢氧化钠水溶液,在温度17℃~55℃、48小时~96小时的范围的各种条件下进行振荡处理,可制造分子量受控的SPG。
实施例5使用高压均质器控制SPG的分子量
作为高压均质器,使用压力式均质器(Avestin公司制造的Emulsiflex C-5型或Powrex公司制造的微流化器M-110EH型)。将通过裂褶菌菌株F-82046的培养得到的SPG提取物溶解于30mL蒸馏水使达到1mg/mL~3mg/mL的浓度,使其浸润。将该SPG溶液加入到压力式均质器的流路中,边慢慢地升高乳化压力直至最大乳化压力为15,000psi~25,000psi,边反复将SPG溶液均质化。在处理后的SPG溶液中加入等量的甲醇,静置数小时,对析出的沉淀物进行离心、回收。用甲醇洗涤后,在沉淀物中加入少量的水使其浸润,之后进行冷冻干燥。如此操作,可通过不依赖于碱处理的方法制造分子量受控的SPG(SPG-85:收量44mg、SPG-86:收量49mg、SPG-87:收量38mg)。
试验例2分子量的测定
SPG的分子量是按照以下步骤,以用作标准品的茁霉多糖换算值的形式测定。在作为受试物质的SPG中添加0.25M的氢氧化钠水溶液,在5℃下搅拌1小时,制成测定样品。将作为分子量标准品的茁霉多糖标准品(Shodex公司。分子量分别为2350kD、1220kD、642kD、337kD、194kD和107kD)溶解于纯水,制成测定样品。在尺寸排阻色谱装置(SEC)上连接差示折射检测器(RI),对这些测定样品进行分析(SEC-RI法)。根据茁霉多糖标准品的保留时间制作分子量校正曲线,推断样品的分子量。以下记录SEC-RI法的分析条件。
高效液相色谱系统(注射器、泵、柱加热器、RI检测器):A1200(Agilent公司)、流动相:约10mM的氢氧化钠水溶液(pH12)、流速:0.4mL/分钟、柱:将Asahipak GS-2G 7B(Shodex)、Asahipak GS-620HQ(Shodex)、Asahipak GS-320HQ(Shodex)依次串联、柱温:40℃、样品浓度:约2mg/mL、样品注入量:100μL、分析时间:75分钟。
分子量校正曲线见图3。确认到:在茁霉多糖换算的分子量107kD~2350kD的范围(以“Log(分子量)”计为5.03~6.37的范围)内可测定分子量。
接下来,由上述的分子量校正曲线和分子量测定样品的保留时间算出的SPG的分子量见表3和表4。根据碱处理条件判明:可在50kD左右~1000kD以上的宽范围内控制SPG的分子量。另外,对于分子量控制前的SPG(mSPG),由于一部分峰超出了分子量校正曲线的分子量上限,所以无法算出重均分子量。然而,通过该测定确认到:这些mSPG具有比分子量受控的SPG更大的分子量、即大概大于1000kD的巨大分子量。另一方面,对于一部分分子量受控的SPG和购入的SPG(SPG-95、SPG-72、SPG-76和iSPG-1),虽然一部分峰小于分子量校正曲线的分子量下限,但可见分布,因此算出峰顶分子量和重均分子量作为参考。确认到:这些SPG具有比其他SPG小的分子量、即大概小于100kD的分子量。

实施例6β(1→3)葡聚糖与核酸的复合物的制造
选择SPG(mSPG、iSPG或分子量受控的SPG)作为β(1→3)葡聚糖,选择K3-dA40作为核酸,如下操作,制造了β(1→3)葡聚糖与核酸的复合物。称量数mg的SPG,溶解于0.25M的氢氧化钠水溶液使达到10mg/mL,在4℃下边时常搅拌边静置24小时。在K3-dA40(59钠盐)中加入蒸馏水,调制了100μM的水溶液。接下来,主要以mG/dA比为2.5的规定比率在该K3-dA40水溶液中添加上述的SPG碱性溶液进行搅拌,从而将SPG和K3-dA40复合化。之后,加入与SPG碱性溶液等量的330mM的磷酸二氢钠水溶液进行中和,在4℃下静置一夜,从而得到了K3-SPG。
需要说明的是,作为SPG,除实施例2或表2记载的物质以外,还使用表5所示的物质。

由β-葡聚糖和核酸构成的复合物的制造例

试验例3β(1→3)葡聚糖与核酸的复合物形成效率的评价
通过SEC对实施例6中制造的β(1→3)葡聚糖与核酸的复合物形成效率进行评价。作为测定样品,使用实施例6中制造的、以溶液的形式得到的K3-SPG。另外,作为对照来分析的K3-dA40单独的水溶液是以与复合物样品中的总K3-dA40的理论浓度相同的方式,使用未溶解SPG的0.25M的氢氧化钠水溶液,按照实施例6所示的工序进行制造。SEC的条件如下所示。
高效液相色谱系统(注射器、泵、柱加热器、UV检测器):A1100(Agilent)、流动相:100mM的磷酸缓冲液(pH7.4、nacalai tesuque、目录号:37244-35)、流速:0.5mL/分钟、柱:将Asahipak GF-1G 7B(Shodex)、Asahipak GF-7M HQ(Shodex)、Asahipak GF-1G 7M HQ(Shodex)依次串联、柱温:40℃、检测波长:260nm、样品注入量:10μL。
由相对于复合化前的K3-dA40量的复合化后的K3-dA40的残留量计算复合化效率。在K3-dA40全部被复合化而残留量为0的情况下,复合物形成效率为100%。
实施例6中制造的K3-SPG的复合物形成效率的测定结果见表7。确认到:形成效率均为96%以上,所有的纯化SPG(分子量控制前)、分子量受控的SPG和购入的SPG均通过本发明的K3-SPG的制造方法以非常高的效率与核酸之间形成复合物。

试验例4通过DLS评价复合物粒径
在DLS法中,利用光子相关光谱法测定实施例6中制造的复合物(K3-SPG)的粒径作为以散射光强度为基准的平均粒径。此时,用磷酸缓冲液(8g/L的NaCl、200mg/L的磷酸二氢钾、1150mg/L的无水磷酸氢二钠)适当地稀释,使达到获得可检测的散射光强度的最低限的K3-SPG浓度。
实施例6中制造的K3-SPG的平均粒径的测定结果见表8。确认到:可在平均粒径为21.9nm~122.2nm的范围内严格地控制。

试验例5 K3-SPG的药理作用的体外评价
使用由人PBMCs(LonZa公司。目录号:Cat#CC-2702Lot#31468)和C57BL/6小鼠调制的脾脏细胞,评价各自的IFN-α产生诱导能力和IFN-γ产生诱导能力。在96孔板上以每孔1×106个细胞数接种人PBMCs和小鼠脾脏细胞。向细胞中添加K3-SPG使以K3-dA40(59钠盐)换算浓度计为0、0.75、2、6、20μg/mL,24小时后的培养上清中的hIFN-α用细胞因子ELISA试剂盒(PBL公司)进行测定。关于K3-SPG的hIFN-α诱导能力,算出总K3-SPG浓度的hIFN-α诱导水平的曲线下面积(以下,称为AUC)的值,进行评价。同样,评价了K3-SPG的IFN-γ诱导能力。其结果见图4。如图4右侧的表所示,KAmSP001a(平均粒径:121.1nm)和KAmSP002a(平均粒径:122.2nm)的粒径大,KAInv3701(平均粒径:31.3nm)和KASPG0095(平均粒径:21.9nm)的粒径小。如图4所示,无论是在人冷冻PBMC中还是在小鼠脾脏细胞中,与粒径小的K3-SPG相比,粒径大的K3-SPG的IFN-α和IFN-γ的产生诱导能力高。
试验例6复合物的药理作用的体外评价
对用K3-SPG复合物处理后的人PBMCs(LonZa公司、Cat#CC-2702Lot#31468)的IFN-α(hIFN-α)产生水平与K3-SPG的平均粒径的相关关系进行了评价。在96孔板上以每孔1×106个细胞数接种人PBMCs,将各BG-CpG添加到细胞中使CpG ODN浓度为0、0.75、2、6、20μg/ml,使用细胞因子ELISA试剂盒(PBL公司)测定24小时后的培养上清中的hIFN-α产生水平。算出全部CpG浓度的hIFN-α诱导水平的AUC值,评价BG-CpG的hIFN-α诱导能力。利用Spearman检验探讨了hIFN-α产生水平与平均粒径的相关关系。其结果,相关系数为0.7789,显示正相关(图5)。即,确认到:在平均粒径为20nm~130nm的范围内,粒径越大,就具有越强的药理作用。
试验例7 K3-SPG的药理作用的体内评价
对C57BL/6小鼠皮下接种2×105个来自C57BL/6小鼠的黑色素瘤细胞株即B16癌细胞(第0天),第9天以30μg/只和100μg/只的量静脉内给予K3(Gene Design公司)或实施例3中制造的K3-SPG(复合物名称:KAmSP001a)。每隔1天给予K3或K3-SPG,总计给予3次,随时间测定肿瘤大小。其结果,如图6所示,在给予了K3-SPG的组(三角形)中,与K3给药组(四边形)相比,观测到了显著的抗肿瘤效果。即,结果如下:将给药量以摩尔数换算时,与K3相比有约3倍重量差的K3-SPG的药效较K3具有优越性。
实施例7β-葡聚糖为香菇多糖的BG-CpG的制造
如上述的具体实施方式项下所述,按照常规方法制造了香菇多糖。这里,作为香菇子实体的栽培法,应用菌床栽培,通过选择、组合已知的纯化方法进行了纯化。而且,在实施例4所示的方法中,控制其分子量,进行了纯化。使用如此操作而得到的香菇多糖,利用实施例6所示的方法制造了BG-CpG。以下,将以香菇多糖为构成成分的BG-CpG记作K3-LNT。表9显示制造的K3-LNT的平均粒径。这里,表9中还一并记载了用于制造各K3-LNT的香菇多糖的分子量控制条件。确认到:通过变更相对于香菇多糖的规定条件的碱处理的应用时间,可控制形成K3-LNT时的粒径。由显示香菇多糖的碱处理时间与K3-LNT的平均粒径的关系的图7可知:通过本发明,可制造在40nm~180nm的范围内严格地控制了平均粒径的K3-LNT。

试验例8 K3-LNT的药理作用的体外评价
与上述的试验例5同样地操作,对K3-LNT的IFN-γ产生诱导能力进行了评价。其中,作为细胞种类,选择小鼠脾脏细胞,在96孔板的每孔中接种的细胞数为1×107。另外,K3-LNT的添加量设为2μg/mL和10μg/mL这2个水平,分别测定24小时培养后的IFN-γ产生量,以它们的总计值作为K3-LNT的药理作用的强度指标。其结果,如图8所示,在平均粒径为约40nm~180nm的范围内相关系数为0.9081,确认到:K3-LNT的粒径越大,就具有越强的药理作用。
试验例9 K3-LNT的药理作用的体内评价
与上述的试验例7同样地操作,评价了K3-LNT的抗肿瘤效果。其中,将评价的第0天对C57BL/6小鼠进行皮下移植的B16癌细胞设为2.5×105个或5.0×105个,从第7天起每隔1天给予K3-LNT,总计给予3次。这里,K3-LNT的给药途径为肿瘤内,其给药量设为50μg/只。作为比较用核酸的K3的给药量设为16.7μg/只。如图9所示,确认到平均粒径为70nm~175nm的K3-LNT具有强效的抗肿瘤效果。另外,与试验例7同样地将一部分样本与K3进行比较时,如图10所示,得到了以下的结果:平均粒径为149nm和175nm的K3-LNT较K3具有优越性。
产业实用性
作为癌免疫疗法的治疗药或预防药的成分,选择BG-CpG的最适粒径。本发明的BG-CpG的平均粒径被严格控制在20nm~180nm、优选20nm~130nm或40nm~180nm、更优选80nm~130nm或130nm~180nm的范围。而且,与以往的BG-CpG相比,本发明的BG-CpG能以较少的给药量得到显著的效果,因此通过对以往的BG-CpG难以治疗或预防的疾病应用,可较以往的BG-CpG更廉价地进行制造,从而可有助于产业上的利益。

Claims (33)
1.β(1→3)葡聚糖与核酸的复合物,所述复合物是粒径受控的。
2.根据权利要求1所述的复合物,其特征在于:平均粒径为80nm~130nm。
3.根据权利要求1所述的复合物,其特征在于:平均粒径为130nm~180nm。
4.根据权利要求1或2所述的复合物,其特征在于:上述β(1→3)葡聚糖为裂褶菌多糖。
5.根据权利要求1或3所述的复合物,其特征在于:上述β(1→3)葡聚糖为香菇多糖。
6.根据权利要求1~5中任一项所述的复合物,其特征在于:上述核酸为包含K型CpG寡核苷酸的寡核苷酸。
7.根据权利要求1~6中任一项所述的复合物,其特征在于:上述核酸是在K型CpG寡核苷酸的3’末端连接有聚脱氧腺苷的核酸。
8.根据权利要求1~7中任一项所述的复合物,其中,上述核酸为寡脱氧核苷酸,其包含由SEQ ID NO:1所表示的核苷酸序列构成的人源化K型CpG寡脱氧核苷酸和聚脱氧腺苷酸,其中聚脱氧腺苷连接在人源化K型CpG寡脱氧核苷酸的3’末端,且寡脱氧核苷酸的一部分或全部的磷酸二酯键被硫代磷酸酯键取代。
9.药物组合物,其包含权利要求1~8中任一项所述的复合物。
10.根据权利要求9所述的药物组合物,其用于预防或治疗病毒感染症、癌症、过敏性疾病、细胞内寄生性原虫或细菌感染症。
11.根据权利要求10所述的药物组合物,其用于预防或治疗病毒感染症。
12.根据权利要求11所述的药物组合物,其中,病毒感染症为RS病毒或流感病毒感染症。
13.免疫刺激剂,其包含权利要求1~8中任一项所述的复合物。
14.根据权利要求13所述的免疫刺激剂,其为疫苗佐剂。
15.药物组合物,其包含:
(a)权利要求1~8中任一项所述的复合物、或者权利要求13或14所述的免疫刺激剂;以及
(b)抗原。
16.根据权利要求15所述的组合物,其用于诱导针对该抗原的免疫反应。
17.根据权利要求16所述的组合物,其中,抗原为来自病原体的抗原。
18.根据权利要求17所述的组合物,其用于预防或治疗病原体的感染症。
19.根据权利要求18所述的组合物,其中,病原体为病毒。
20.根据权利要求19所述的组合物,其中,病毒为RS病毒或流感病毒。
21.根据权利要求16所述的组合物,其中,抗原为来自癌症的抗原。

23.药物组合物,其包含:
(a)权利要求1~8中任一项所述的复合物、或者权利要求9、10、15、16、21、22中任一项所述的药物组合物;以及
(b)抗癌药。
24.根据权利要求23所述的组合物,其中,抗癌药为免疫检查点抑制剂。
25.根据权利要求24所述的组合物,其中,免疫检查点抑制剂为PD1抑制剂、PDL-1抑制剂和CTLA-4抑制剂中的任一者。
26.权利要求1~8中任一项所述的复合物的制造方法,其利用了β(1→3)葡聚糖的分子量特性。
27.病毒感染症、癌症、过敏性疾病、细胞内寄生性原虫或细菌感染症的治疗或预防方法,其包括:对受试者给予有效量的药物组合物,该药物组合物包含权利要求9所述的药物组合物。
28.根据权利要求27所述的治疗或预防方法,其中,病毒感染症为RS病毒或流感病毒感染症。
29.病毒感染症、癌症、过敏性疾病、细胞内寄生性原虫或细菌感染症的治疗或预防方法,其包括:对受试者给予有效量的药物组合物,该药物组合物包含权利要求15所述的药物组合物。
30.根据权利要求29所述的治疗或预防方法,其中,病毒感染症为RS病毒或流感病毒感染症。
31.根据权利要求27或29所述的癌症的治疗或预防方法,其特征在于:与其他抗癌药并用。
32.根据权利要求31所述的治疗或预防方法,其中,抗癌药为免疫检查点抑制剂。
33.根据权利要求32所述的治疗或预防方法,其中,免疫检查点抑制剂为PD1抑制剂、PDL-1抑制剂和CTLA-4抑制剂中的任一者。
