Abstract
本发明公开了一种重组胶原蛋白抗衰配方及其制备方法,涉及重组胶原蛋白领域,解决了现有对胶原蛋白活性物质的提取采用零下40摄氏度真空冻干技术,该技术对于动物和生物的活性成分的长期保存效果明显不足,且致活性物质的提取率低下的问题,现提出如下方案,其包括以下物质组成:水、丙二醇、羟乙基纤维素、海藻糖、甘露糖醇、透明质酸钠、甘草酸二钾、寡肽、神经酰胺、血清蛋白、烟酰胺、己二醇、甘油、可溶性胶原、可溶性蛋白多糖、芦荟提取物、燕窝提取物、积雪草提取物、牛奶提取物、光果甘草根提取物。本配方使得制得的胶原蛋白具有更好的抗衰效果,以及制备方法可以对活性提取物的活性进行更好的保留,进而进一步提高抗衰效果的特点。
Description
技术领域
本发明涉及重组胶原蛋白领域,尤其涉及一种重组胶原蛋白抗衰配方及其制备方法。
背景技术
胶原蛋白(也称胶原)是动物体中含量最丰富的一类蛋白质家族。胶原蛋白具有很强的生物活性及生物功能,能参与细胞的迁移、分化和繁殖,使结缔组织具有机械强度,同时胶原蛋白还能促进细胞生长,具有止血、生物相容性和生物降解性能。基于这些特性,胶原蛋白在烧伤、创伤、眼角膜疾病、保健、美容、矫形、硬组织修复、创面止血、药物传递、缓释技术等医药卫生领域有着广泛的应用。另外,胶原蛋白也可作为食品添加剂、饲料添加剂、絮凝剂、照相乳胶材料、表面活性剂和固定化酶载体材料等应用于各个工业和实验室领域。
而芦荟、燕窝、积雪草、牛奶、光果甘草根内均含有丰富的胶原蛋白,其均有有效的抵抗皮肤的衰老,而现有对活性物质的提取一般采用冻干技术,其他产品用的是零下40摄氏度真空冻干技术,一般用于冻干,植物,花果等一般产品,对于动物和生物的活性成分的长期保存效果明显不足,导致活性物质的提取率低下。因此提出一种重组胶原蛋白抗衰配方及其制备方法。
发明内容
本发明的目的在于提供一种重组胶原蛋白抗衰配方及其制备方法,解决了现有对胶原蛋白活性物质的提取采用零下40摄氏度真空冻干技术,该技术对于动物和生物的活性成分的长期保存效果明显不足,且致活性物质的提取率低下的问题。
为实现上述目的,本发明提供如下技术方案:一种重组胶原蛋白抗衰配方,包括以下物质组成:水、丙二醇、羟乙基纤维素、海藻糖、甘露糖醇、透明质酸钠、甘草酸二钾、寡肽、神经酰胺、血清蛋白、烟酰胺、己二醇、甘油、可溶性胶原、可溶性蛋白多糖、芦荟提取物、燕窝提取物、积雪草提取物、牛奶提取物、光果甘草根提取物。
优选的,所述各原料组分含量比为:水40-60、丙二醇20-30、羟乙基纤维素2-6、海藻糖2-5、甘露糖醇10-15、透明质酸钠10-15、甘草酸二钾8-12、寡肽1-4、神经酰胺10-24、血清蛋白2-6、烟酰胺2-6、己二醇3-6、甘油4-6、可溶性胶原3-6、可溶性蛋白多糖6-10、芦荟提取物5-10、燕窝提取物5-10、积雪草提取物风5-10、牛奶提取物5-10、光果甘草根提取物5-10。
优选的,所述各原料组分含量比为:水50、丙二醇25、羟乙基纤维素4、海藻糖4、甘露糖醇12、透明质酸钠12、甘草酸二钾10、寡肽3、神经酰胺17、血清蛋白4、烟酰胺4、己二醇5、甘油4、可溶性胶原5、可溶性蛋白多糖8、芦荟提取物8、燕窝提取物8、积雪草提取物风8、牛奶提取物8、光果甘草根提取物8。
一种重组胶原蛋白抗衰配方的制备方法,包括以下步骤:
S1:提取物制备:
(1)将芦荟组织细胞在培养瓶内培养,并在细胞消化终止后,在室温(15-25℃)以300g离心5分钟,然后用-196℃以下温度冻存细胞聚集体;
(2)复苏逐步降温解冻后,应该立即铺板在预先包被好的冻存管中;
(3)在37℃水浴中快速解冻,持续温和的在水中晃动冻存管,直到剩余少量细胞还处于冻存状态,取出冻存管,用75%的酒精或异丙醇擦D拭除菌;
(4)用2mL的血清移液管把细胞悬液转移到一个15mL的锥形试管,逐滴加入5-7mL的预热的培养基到15mL的离心管中,加入的同时轻轻混匀;
(5)离心5分钟,吸出培养基,使细胞沉淀完好无损;
(6)使用2mL血清移液管轻轻将细胞沉淀重悬于1mL的培养基中,将细胞混合物转移到含有完全培养基的预包被的培养瓶中,在二氧化碳培养箱中培养,即可制得芦荟提取物;
S2:胶原蛋白制备:
(1)将乳化锅清洗干净,相加热至85度溶解均匀后加入除提取物之外的成分搅拌混合均匀;
(2)在混合液中加入芦荟提取物、燕窝提取物、积雪草提取物、牛奶提取物、光果甘草根提取物依次加入相搅拌分散均匀;
(3)取样,pH测定,半制品取样检测符合标准后,出料;
(4)半制品检验合格后,灌装,包装,成品检验合格。
优选的,所述S1中(5)在室温(15-25℃)以300g离心。
优选的,所述S1中(6)二氧化碳培养箱的环境条件为:37℃、5%CO2、95%湿度。
优选的,所述S2中(2)各个提取物均冷却至零下45度,间隔2min依次加入。
优选的,所述燕窝提取物、积雪草提取物、牛奶提取物、光果甘草根提取物的制备与S1中芦荟提取物的制备方法相同。
与相关技术相比较,本发明提供的一种重组胶原蛋白抗衰配方及其制备方法具有如下有益效果:
本发明提供一种重组胶原蛋白抗衰配方及其制备方法,通过超低温进行冻干提取,以及皮肤表面脂质、角质细胞、细胞间脂质和天然保湿因子这4部分共同参与构成了完整的皮肤屏障。由于脂质的疏水性和细胞间的紧密连接,水和水溶性分子,加上我们超低温下提取物及培养后的活性成分更高效果更佳稳定,超低温冻干后的内容物更容易长时间保存,也便利了对提取物活性的保护。

各原料组分含量比为:水60、丙二醇30、羟乙基纤维素6、海藻糖5、甘露糖醇15、透明质酸钠15、甘草酸二钾12、寡肽4、神经酰胺24、血清蛋白6、烟酰胺6、己二醇6、甘油6、可溶性胶原6、可溶性蛋白多糖10、芦荟提取物10、燕窝提取物10、积雪草提取物风10、牛奶提取物10、光果甘草根提取物10。
实施例三:
请参阅图1-2,本发明提供一种技术方案:一种重组胶原蛋白抗衰配方,包括以下物质组成:水、丙二醇、羟乙基纤维素、海藻糖、甘露糖醇、透明质酸钠、甘草酸二钾、寡肽、神经酰胺、血清蛋白、烟酰胺、己二醇、甘油、可溶性胶原、可溶性蛋白多糖、芦荟提取物、燕窝提取物、积雪草提取物、牛奶提取物、光果甘草根提取物。
各原料组分含量比为:水50、丙二醇25、羟乙基纤维素4、海藻糖4、甘露糖醇12、透明质酸钠12、甘草酸二钾10、寡肽3、神经酰胺17、血清蛋白4、烟酰胺4、己二醇5、甘油4、可溶性胶原5、可溶性蛋白多糖8、芦荟提取物8、燕窝提取物8、积雪草提取物风8、牛奶提取物8、光果甘草根提取物8。
综上实施例一-三:
由于牛奶提取物:是从新鲜的牛奶中分离提纯获得,直径在30nm-100nm左右,有比较丰富与细胞膜相同的磷脂双分子层薄膜结构,紧密包裹细胞,活性因子特异蛋白质、脂质、miRNA、DNA片段,可迅速精准打通细胞生物膜输屏障;
燕窝提取物:含有丰富的表皮生长因子和水提取物质,能够强烈刺激细胞再生、分裂、促使组织重建,使皮肤滑嫩、红润、富有光泽、不易衰老;
积雪草提取物:对皮肤具有抗氧化、促进皮肤新陈代谢,治疗皮肤溃疡,使皮肤紧致光滑,具有镇静安定的作用;
以及还包括光果甘草根提取物、芦荟提取物、以及血清蛋白,进而使得通过本配方制得的重组胶原蛋白具有更好的抗衰防护效果。
实施例四:
请参阅图1-2所示,在实施例一的基础上,本发明提供一种技术方案:一种重组胶原蛋白抗衰配方的制备方法,包括以下步骤:
S1:提取物制备:
(1)将芦荟组织细胞在培养瓶内培养,并在细胞消化终止后,在室温(15-25℃)以300g离心5分钟,然后用-196℃以下温度冻存细胞聚集体;
(2)复苏逐步降温解冻后,应该立即铺板在预先包被好的冻存管中;
(3)在37℃水浴中快速解冻,持续温和的在水中晃动冻存管,直到剩余少量细胞还处于冻存状态,取出冻存管,用75%的酒精或异丙醇擦D拭除菌;
(4)用2mL的血清移液管把细胞悬液转移到一个15mL的锥形试管,逐滴加入5-7mL的预热的培养基到15mL的离心管中,加入的同时轻轻混匀;
(5)离心5分钟,吸出培养基,使细胞沉淀完好无损;
(6)使用2mL血清移液管轻轻将细胞沉淀重悬于1mL的培养基中,将细胞混合物转移到含有完全培养基的预包被的培养瓶中,在二氧化碳培养箱中培养,在前7天内(7x24小时),严禁移动。7天后补充10ml完全培养基(融化添加物,按10%比例加入基础培养基中,充分混合均匀,自7天后,应2天观察一次,若有组织块周围已爬出细胞,且长至半径0.5cm(即组织)镜下标记,用枪头或移液管轻轻将组织块拨离,令其从培养表面脱离,漂浮于培养瓶中,可收获更多有效内容物即可制得芦荟提取物;
S2:胶原蛋白制备:
(1)将乳化锅清洗干净,相加热至85度溶解均匀后加入除提取物之外的成分搅拌混合均匀;
(2)在混合液中加入芦荟提取物、燕窝提取物、积雪草提取物、牛奶提取物、光果甘草根提取物依次加入相搅拌分散均匀;
(3)取样,pH测定,半制品取样检测符合标准后,出料;
(4)半制品检验合格后,灌装,包装,成品检验合格。
5、根据权利要求4的一种重组胶原蛋白抗衰配方的制备方法,其特征在于,S1中(5)在室温(15-25℃)以300g离心。
S1中(6)二氧化碳培养箱的环境条件为:37℃、5%CO2、95%湿度。
S2中(2)各个提取物均冷却至零下45度,间隔2min依次加入。
燕窝提取物、积雪草提取物、牛奶提取物、光果甘草根提取物的制备与S1中芦荟提取物的制备方法相同。
本实施例中,通过超低温进行冻干提取,以及皮肤表面脂质、角质细胞、细胞间脂质和天然保湿因子这4部分共同参与构成了完整的皮肤屏障。由于脂质的疏水性和细胞间的紧密连接,水和水溶性分子,加上我们超低温下提取物及培养后的活性成分更高效果更佳稳定,超低温冻干后的内容物更容易长时间保存,也便利了对提取物活性的保护。

1.一种重组胶原蛋白抗衰配方,其特征在于,包括以下物质组成:水、丙二醇、羟乙基纤维素、海藻糖、甘露糖醇、透明质酸钠、甘草酸二钾、寡肽、神经酰胺、血清蛋白、烟酰胺、己二醇、甘油、可溶性胶原、可溶性蛋白多糖、芦荟提取物、燕窝提取物、积雪草提取物、牛奶提取物、光果甘草根提取物。
2.根据权利要求1所述的一种重组胶原蛋白抗衰配方,其特征在于,所述各原料组分含量比为:水40-60、丙二醇20-30、羟乙基纤维素2-6、海藻糖2-5、甘露糖醇10-15、透明质酸钠10-15、甘草酸二钾8-12、寡肽1-4、神经酰胺10-24、血清蛋白2-6、烟酰胺2-6、己二醇3-6、甘油4-6、可溶性胶原3-6、可溶性蛋白多糖6-10、芦荟提取物5-10、燕窝提取物5-10、积雪草提取物风5-10、牛奶提取物5-10、光果甘草根提取物5-10。

4.一种重组胶原蛋白抗衰配方的制备方法,其特征在于,包括以下步骤:
