Description

Anti-dandruff itching-relieving composition, preparation method and application thereof, shampoo and preparation method
Technical Field
The invention relates to the technical field of daily chemicals, in particular to a composition for removing dandruff and relieving itching, a preparation method and application thereof, shampoo and a preparation method thereof.
Background
In 2021, people's scalp health white books and CBNData consumption data can know that the most common scalp problems of consumers are mainly including more dandruff, greasiness and flat and collapsed shape.
Dandruff is the exfoliating keratinocytes of the scalp. Under normal conditions, keratinocytes fall off little, very slowly, and are not visibly apparent. However, under the influence of various factors, immature keratinocytes are produced in large amounts, and are agglomerated and shed to form dandruff. During dandruff formation, two substances play an important role: oil secreted from the scalp, and fungi that feed on the oil. Under certain conditions, such as when the secretion of grease is vigorous in puberty, fungi can be greatly propagated on the surface of the scalp to stimulate scalp cells to secrete more grease; the large amount of grease provides nutrients for further expansion of fungi. Eventually, the normal metabolism of scalp keratinocytes becomes unstable, and immature keratinocytes accumulate and fall off, and dandruff is generated.
Scalp is one of the thinnest parts of human skin, and the skin appendages such as hair follicles sebaceous glands and the like are distributed at high density, so that ultraviolet rays (UV), pollutants (PM 2.5) and the like are easy to damage skin structures, and skin inflammation such as dandruff, erythema, pimples, burning stink, itching and the like is caused. Thus, scalp is often subjected to the problems of dandruff, scalp oil, scalp itching, redness and the like.
With the understanding of natural plants becoming deeper and more comprehensive, natural Chinese herbal medicine components with good curative effects, hair care, dandruff removal and relieving itching gradually become the favored direction of consumers, so a shampoo product with natural extract combination is needed.

Efficacy verification experiment:
in vitro bacteriostasis test:
antibacterial effect detection is carried out by adopting a bacteriostasis ring method: the culture medium is agar culture medium, and the test strain is Malassezia furfur standard strain.
Test sample: examples 1, 4, 7 and comparative examples 4, 8.
The experimental contents are as follows:
1. malassezia furfur is prepared into bacterial suspension with the concentration of about 105 cfu.
2. Uniformly coating on a Leemung & Notman culture medium flat plate, and then placing in a sterile operation table to blow dry the bacterial liquid on the flat plate.
3. And (3) quantitatively adding each shampoo composition sample into the flat plate, culturing in a constant temperature incubator at 30 ℃ for 5-7 days, observing results, and recording and measuring the condition of the inhibition ring.
Experimental results:
recording the results as the diameter of the inhibition ring, unit: millimeter.
Results determinations refer to the national clinical trial standardization committee (NCCLS) standard: the diameter of the inhibition zone is less than 8mm, 8-13 mm is low-sensitivity, 13-19 mm is medium-sensitivity, and 19mm is high-sensitivity.
Fig. 1 is a data comparison chart of the diameter of a bacteriostasis ring obtained by performing a bacteriostasis test on malassezia by using shampoo prepared under the condition that a single composition or a combination of two components is taken as a control group.
From fig. 1, it can be seen that example 4, which is a compound containing a high content of ginger extract, green pricklyash peel extract and jujube bark extract, has an obvious inhibition effect on malassezia, up to 19.01mm, and worse is comparative example 4, wherein volatile oil and ethanol extract in the ginger extract both have inhibition effect on malassezia, and antibacterial components 4-quinolinone alkaloid in the green pricklyash peel extract and antibacterial components such as terpenes, alkaloids, glycosides, lignans, esters, phenols, aldehydes, alcohols, steroids and the like contained in the jujube bark extract are compound and combined. Comparative example 4 does not contain ginger extract, its inhibitory effect is 12.83mm, wherein example 1 containing green pricklyash peel extract and jujube bark extract has a bacteriostatic effect of 16.54mm, and comparative example 8 containing no green pricklyash peel extract and jujube bark extract has the worst bacteriostatic effect of only 8.24mm. Therefore, the high-content ginger extract and the green pricklyash peel extract and the jujube bark extract are compounded at the same time, so that the printing effect on malassezia is obvious.
Histamine soothing antipruritic test
40 healthy volunteer subjects were organized and randomized into 4 groups, and the test results were arithmetically averaged.
Test measurement parameters: skin redness (visual score), itching (subjective feeling of the subject).
Test sample: examples 2 and 4 and comparative examples 6 and 7.
The testing method comprises the following steps: histamine hydrochloride (0.01 g/ml) was added dropwise to the skin test site with a specific test blade, and after 5 minutes, the skin was red, swollen, scratched and itchy, and the test piece was used in the example (0.01 g/cm) 2 ) Comparative example product (0.01 g/cm) 2 ) And the redness, swelling degree, itching feeling of the test site were measured after 0 minutes, 30 minutes, 60 minutes, 90 minutes, 120 minutes, 240 minutes without using the product (blank), and the specific test results are shown in tables 5 and 6.
TABLE 5 redness and swelling test results
TABLE 6 test results of itching feeling
The evaluation results of tables 5 and 6 show that: the synergistic effect of the two substances of gentian extract and white willow bark extract in the anti-dandruff compositions of examples 2 and 4, wherein the direct acting target of gentian is COX-2 when the biological evaluation method is analyzed, based on the verification of the activity inhibition experiment of recombinant human COX-2 protease, the gentian can inhibit the activity of COX-2 enzyme in a dose-dependent manner, and COX-2 is a key enzyme for catalyzing the conversion of arachidonic acid into prostaglandin, is one of key enzymes for causing inflammatory reaction and can promote tissue damage caused by inflammatory reaction; the white willow bark extract has the capability of scavenging free radicals and down regulating inflammatory mediators, such as TNF-alpha and NF-kappa B. Among them, the nuclear factor- κb molecule is a transcription factor that controls transcription of DNA to continue inflammatory immune response, and serves as a switch to turn on and off inflammation in vivo. The combination of gentian extract and white willow bark extract has obviously better effects on eliminating and resisting skin redness, itching, skin repair and the like than other combinations of comparative examples 6 and 7. This demonstrates that the anti-dandruff and antipruritic composition of the present invention has good effect of relieving itching due to the inclusion of gentian extract and white willow bark extract.
Human body efficacy evaluation
Test sample: comparative examples 1, 6, 8, shampoos used in examples 4, 7 and a commercial anti-dandruff shampoo.
The test method comprises the following steps: 60 subjects with severe dandruff symptoms and no hair diseases such as psoriasis are selected between 18 and 50 years of age. The shampoo of the above examples, comparative examples and commercial products was tested for 30 days and every two days for hair washing.
Test results
Subjective efficacy assessment
Subjects self-assessed, and subjective evaluation of shampoo effect was performed based on semi-shampoo conditions. The subjects were allowed to self-evaluate the improvement degree of the dandruff amount on both sides, the scalp itching degree, the scalp fat amount, the hair glossiness, the overall hair amount, etc. by means of a questionnaire, and to divide the mixture into 0 to 5 portions, wherein 0 portion=no improvement; score 1 = little improvement; 2 min = slightly improved; 3 minutes = improvement evident; score 4 = significant improvement; score 5 = complete improvement or substantial recovery.
The average scores of the shampoo of the 6 experimental groups after the subjects continued to use are shown in the following table 7.
TABLE 7 results of human efficacy test
The above results demonstrate that the compositions without green pricklyash extract, white willow bark extract and replaced by centella asiatica extract and chamomile extract, comparative examples 1, 6, 8, all obtained average scores below 16 score, most scalp problems were not significantly ameliorated; the average scores obtained in examples 4 and 7 were all greater than 17 and above, thus demonstrating that the compositions prepared by the invention have good compatibility, have remarkable anti-dandruff, relieving and itching-relieving functions, and have less added or replaced effects. The composition component of example 4 contains high content of ginger extract and gentian extract, and the final average score of the composition component is 22.2, which is higher than that of the commercial anti-dandruff product, and the advantages are mainly represented by the remarkable improvement of scalp quantity and scalp itching. The above optimal effect shows that each extract in the anti-dandruff and antipruritic composition of the invention is indispensable.
The above examples are only for illustrating the detailed method of the present invention, and the present invention is not limited to the above detailed method, i.e., it does not mean that the present invention must be implemented depending on the above detailed method. It should be apparent to those skilled in the art that any modification of the present invention, equivalent substitution of raw materials for the product of the present invention, addition of auxiliary components, selection of specific modes, etc., falls within the scope of the present invention and the scope of disclosure.
