Description
Technical Field
The invention relates to the field of daily chemicals, in particular to an anti-dandruff composition containing a synergistic antibacterial component and application thereof.
Background
Dandruff is a common problem, and typical dandruff is flaky dandruff on or in the scalp and is mostly accompanied by marked itching. Various intrinsic and extrinsic environmental factors, such as sebum secretion, malassezia colonization, individual susceptibility, and the interaction between these factors, are responsible for the development of dandruff.
Malassezia is a lipotrophic fungus that parasitizes on the skin surfaces of humans and animals and secretes a variety of lipases with broad-spectrum activity that hydrolyze triglycerides in sebum to fatty acids. Malassezia has been considered to be a causative bacteria of dandruff, with malassezia restricta being the most prominent pathogenic bacterium, since it metabolizes saturated fatty acids and thereby produces an accumulation of proinflammatory unsaturated fatty acids, stimulating the skin barrier and causing the stratum corneum to shed.
The traditional dandruff removing agents for inhibiting malassezia are ketoconazole, ZPT, OCT and climbazole (climbazole), however, the chemical substances have certain side effects and are large in irritation, and scalp microorganisms are easy to generate drug resistance. For example, ZPT is easy to deposit on scalp, so that hair is withered and yellow and easy to break; therefore, many plant extract raw materials with anti-dandruff activity are emerging in the field of daily chemicals, but most of the plant extract raw materials have no ideal using effect.

Quinine, also known as cinchona alkaloid, is a main alkaloid in the bark of madder plant cinchona, has the effects of inhibiting the reproduction of or killing plasmodium falciparum erythrocytic endophytic protozoa, is an important antimalarial drug, also has the anti-inflammatory effect, and can be used for treating rheumatoid arthritis.
In order to verify the synergistic antibacterial effect of the anti-dandruff composition containing the synergistic antibacterial component, the following experiments are set, and the steps are as follows:
(1) preparing a restriction malassezia suspension;
(2) leeming & Notman medium was incubated at 50 ℃ for future use. Marking 16 sterile culture dishes on a dish cover by using a marker pen, wherein 2 sterile culture dishes are A, B, C, A + B, A + C, B + C, A + B + C, and 2 negative control dishes are adopted;
(3)1 sterile test tube is taken, numbered 10-2 in sequence, and 4.5mL of sterile normal saline is added into the test tube. 0.5mL of the bacterial suspension was taken with 1-mL sterile pipette, added to a 10-1 tube, and shaken well. Another 1-1 mL sterile pipette is taken to suck 0.5mL deionized water sample from the 10-1 test tube to the 10-2 test tube and is mixed uniformly for standby;
(4) 1mL of the diluted sample in a 10-2 test tube was added to each sterile petri dish labeled A, B, C, A + B, A + C, B + C, A + B + C and negative controls, and 15g of Leeming & Notman medium incubated at 50 ℃ was immediately added, shaken up quickly and gently to avoid shaking the medium out of the petri dish. Wherein the dish labeled A is added with Leeming & Notman medium mixed with 0.075g of quinine and its salts, similarly, B is added with 0.75g of clove extract, C is added with 0.3g of purslane extract, A + B is a mixture of 0.075g of quinine and its salts and 0.75g of clove extract, A + C is a mixture of 0.075g of quinine and its salts and 0.3g of purslane extract, B + C is a mixture of 0.75g of clove extract and 0.3g of purslane extract, and A + B + C is a mixture of 0.075g of quinine and its salts, 0.75g of clove extract and 0.3g of purslane extract. The Leeming & Notman culture medium without the composition components is used as a negative control, and after the flat plate is well mixed, the flat plate is immediately laid on the table top until the flat plate is completely solidified;
(5) the completely solidified measuring plate is inverted and cultured for 72h at 28 ℃;

(7) the data obtained are reported in table 2 below:
item | A | B | C | A+B | A+C | B+C | A+B+C |
Bacteriostatic ratio (%) | 63.8 | 45.6 | 38.1 | 85.2 | 76.7 | 68.4 | 96.1 |

in the formula:
e represents a measure of the interaction of the composition, the interaction being synergistic when E < -0.5 and antagonistic when E >0.5, a simple addition when E >0.5 < 0.5.
ω D represents the growth rate of malassezia restricta in the dishes of each group relative to the negative control after addition of D, where D ═ (a, B, C, AB, AC, BC, ABC), i.e. single material or mixture of several materials. That is, ω is the colony count of each group of petri dishes/the colony count of the negative control is 1-bacteriostasis rate.
min () represents the group with the smallest growth rate in parentheses, and the calculation formula based on the value of E yields E-0.85 < -0.5, indicating that the combination of three components, quinine and its salts (A), clove extract (B), and purslane extract (C), has synergistic inhibitory effect on Maraxella restricta.
