背景技术
有资料显示,木糖醇、山梨醇、甘露糖等小肠吸收消化率不足50%,大肠吸收消化率超过50%,甚至达到75%以上。由于木糖醇、山梨醇、甘露糖等这类物质其高渗透性的特性,当大量的进食后,这些高渗物质本身的渗透效应阻碍了肠壁对水和电解质的重吸收,导致肠腔中有过量的不被吸收的水溶性溶质,造成渗透性腹泻的发生,此为糖醇不耐受现象。在公开号CN109493974A的专利文献中公开了一种计算人体对糖醇及功能糖耐受量的方法,其中阐述了木糖醇存在高含量摄入会导致腹泻的问题。而通常在代糖产品开发当中为了避免这一情况发生,会选择高倍甜味剂来增加甜度,减少糖醇的使用量。如在公开号CN111436495A的专利文献中公开了一种用于减脂瘦身奶昔固体饮料及其制备方法,其中木糖醇的添加量仅1.3%左右,而更多的甜度是由三氯蔗糖等高倍甜味剂来提供,总的木糖醇添加量远远不足30g。这也是行业内为了避免过高木糖醇添加导致腹泻情况发生的常规规避手段。
发明内容
本发明所要解决的技术问题在于,提供一种调节肠道菌群组成结构的甜味剂组合物及其制备方法,利用如甲鱼肽/多肽或其他益生元物质复配糖醇的配方,通过调节肠道菌群组成结构定向刺激拟杆菌属、梭菌属、毛螺菌科分泌消化酶等方式增加木糖醇的代谢分解,从而改善木糖醇过量食用可能导致的腹泻问题。
本发明是这样实现的,提供一种调节肠道菌群组成结构的甜味剂组合物,所述甜味剂组合物包括:甲鱼肽4~10份,聚葡萄糖2~5份,低聚木糖5~10份,木糖醇30~60份,麦芽糖醇5~15份,山梨糖醇1~10份。

与现有技术相比,本发明的调节肠道菌群组成结构的甜味剂组合物及其制备方法,在所述甜味剂组合物中含有甲鱼肽、聚葡萄糖、低聚木糖、木糖醇、麦芽糖醇和山梨醇,可减少因过量食用木糖醇而导致的腹泻情况(即糖醇不耐受现象),提升木糖醇在产品应用中的安全性与实用性的有益效果,可在甜味与填充均有一定要求的产品中使用(特别是烘焙、乳制品类)。在高糖比例产品(即产品中木糖醇含量总体超过30g以上)中使用本发明的调节肠道菌群组成结构的甜味剂组合物后,能有效地降低食用者的腹泻发生率。

甜味剂组合物配方1溶液:将甲鱼肽4g、聚葡萄糖2g、低聚木糖5g、木糖醇30g、麦芽糖醇5g和山梨糖醇1g分别过80目筛后置于固定料斗混料机内混合30分钟,至颜色均匀一致,分别取出装入不同的容器,加入适量水补足至100ml即可。
甜味剂组合物配方2溶液:方法与制备甜味剂组合物配方1溶液相同,在配方2中,含有甲鱼肽5g、聚葡萄糖5g、低聚木糖5g、木糖醇40g、麦芽糖醇5g和山梨糖醇1g。

步骤3、选用6~8周龄SD雄性大鼠,根据体重均匀分为8组,每组10只,共80只。每组分别适应性喂养四种配方的甜味剂组合物溶液和四种对照组液体各1周。每天9:00am,按照人和大鼠的体表面积换算公式计算喂养量分别对8组大鼠进行灌胃。大鼠灌胃后,自由饮水和喂食,观察并记录各组大鼠腹泻情况。腹泻记录时间为灌胃后4~8小时两次,如早上9:00灌胃,下午13:30、16:30观察记录两次。可通过观察大鼠肛门口、底托或尾部牵拉促进排泄等方式判断是否腹泻。根据大鼠腹泻的记录计算大鼠的腹泻率,得到下表一所示各组大鼠腹泻率统计表和图1的各组大鼠腹泻率统计示意图。

对大鼠肠道内容物样本进行16S rDNA肠道菌群测序后,采用PICRUSt软件进行宏基因组预测,得到表一所示的消化酶总量的统计结果以及如图3所示的木糖醇代谢消化酶含量分析图。
从结合表一与图1可知,对照组1(单纯水)和对照组2(少量的甲鱼肽、聚葡萄糖和低聚木糖)不含糖醇成分不会造成大鼠腹泻的情况。配方1~4随着木糖醇的用量比例从30份逐渐增加到60份时,其腹泻率随着木糖醇用量的增加而上升。但在同样的糖醇复配用量之下,如配方1与对照组3,配方4与对照组4,因其组合物中添加了一定量甲鱼肽、聚葡萄糖及低聚木糖,使得其大鼠的腹泻率均有所降低,分别由35%降至15%,由85%降至65%。同时,通过木糖醇代谢消化酶总量汇总可知,腹泻率降低组其相关代谢酶含量均显著升高。
以上说明,通过在糖醇配方内添加一定量的甲鱼肽、聚葡萄糖、低聚木糖等,可以有效降低木糖醇食用量过高引起的腹泻,并且其发挥主要是由于增加代谢木糖醇的相关消化酶量,促进木糖醇消化分解从而缓解腹泻的作用。
另外,图2是木糖醇的代谢过程图,由图2可知,根据KEGG查询,木糖醇在肠道菌群中可能的代谢途径有三条,分别包括柠檬酸-丙酮酸循环途径、核黄素代谢途径、磷酸戊糖途径,其中有许多涉及到代谢消化酶,常见的五种代谢消化酶包括:EC1.1.1.21木糖醇还原酶、EC5.3.1.5木糖异构酶、EC1.1.1.14木糖醇脱氢酶、EC2.7.1.17木酮糖激酶和EC5.1.3.1磷酸木酮糖异构酶。根据图3统计所得,实验中的五种消化酶含量存在显著差异,说明该甜味剂组合物在配合甲鱼肽、聚葡萄糖与低聚木糖等这类益生元与多肽类后,可以有效提升体内分泌相关消化酶的肠道菌群丰度,从而加快木糖醇在体内消化代谢率,使得体内能在相同时间内消化代谢更多含量的木糖醇,一定程度上缓解体内因短时间聚集木糖醇而引起渗透性腹泻的情况,提高了大鼠对高含量木糖醇混合物的耐受程度,提升了糖醇类甜味剂在产品应用中的安全性与实用性。

Claims (6)
1.一种调节肠道菌群组成结构的甜味剂组合物,其特征在于,所述甜味剂组合物包括:甲鱼肽4~10份,聚葡萄糖2~5份,低聚木糖5~10份,木糖醇30~60份,麦芽糖醇5~15份,山梨糖醇1~10份。
2.如权利要求1所述的调节肠道菌群组成结构的甜味剂组合物,其特征在于,所述甜味剂组合物包括:甲鱼肽4份,聚葡萄糖2份,低聚木糖5份,木糖醇30份,麦芽糖醇5份,山梨糖醇1份。
3.如权利要求1所述的调节肠道菌群组成结构的甜味剂组合物,其特征在于,所述甜味剂组合物包括:甲鱼肽5份,聚葡萄糖5份,低聚木糖5份,木糖醇40份,麦芽糖醇5份,山梨糖醇1份。
4.如权利要求1所述的调节肠道菌群组成结构的甜味剂组合物,其特征在于,所述甜味剂组合物包括:甲鱼肽5份,聚葡萄糖4份,低聚木糖8份,木糖醇50份,麦芽糖醇15份,山梨糖醇10份。
5.如权利要求1所述的调节肠道菌群组成结构的甜味剂组合物,其特征在于,所述甜味剂组合物包括:甲鱼肽10份,聚葡萄糖5份,低聚木糖10份,木糖醇60份,麦芽糖醇15份,山梨糖醇10份。
6.一种如权利要求1至5中任意一项所述的调节肠道菌群组成结构的甜味剂组合物的制备方法,其特征在于,包括如下步骤:分别称取足量的甲鱼肽、聚葡萄糖、低聚木糖、木糖醇、麦芽糖醇和山梨糖醇物料,各物料分别过80目筛后置于固定料斗混料机内混合30分钟,至颜色均匀一致后即可。
