背景技术
随着人们的生活方式和工作条件的改变,用眼的时间越来越多,比如长时间看电脑、看电视、玩电子游戏等等,很容易患视觉疲劳症和干眼症,特别是经常戴框架眼镜盒和隐形眼镜的人需要更加注重眼部的护理。眼部护理液是近年来很流行的眼部护理保健品,它具有冲洗眼睛、润滑、清洁等功能,能够在一定程度舒缓眼睛疲劳,特别是对于一些长期看电脑或是带有隐形眼镜的人群很适用。
现有的眼部护理液只能暂时缓解眼部干涩,无法进行维护视网膜和提高视力,同时含有刺激性,在使用的过程中容易出现不适的症状,且为药物,无法进行缓解视疲劳、抑制眼部的细菌,不具备明目的功能,实用性低,不方便人们进行使用。
发明内容
针对相关技术中的问题,本发明提出的一种酵素蒸馏液的制备方法及其作为眼部护理液的应用,以克服现有相关技术所存在的上述技术问题,本发明中的眼部护理液没有刺激性,不是药物,且具备缓解视疲劳、抑制眼部的细菌的功能;酵素通过蒸馏得到的液体,具有维护视网膜、提高视力的功能。
为实现上述目的,本发明提供如下技术方案:
本发明提供一种酵素蒸馏液的制备方法,包括以下步骤:
(1)菌种培养
①、菌种:异常威克汉姆酵母CICC 1312、巴氏醋酸菌CICC 20002、嗜酸乳杆菌CICC6096、青春双歧杆菌CICC6180;
②、种子培养基:麦芽汁培养基、葡萄糖琼脂培养基、MRS肉汤培养基;
所述异常威克汉姆酵母CICC 1312采用麦芽汁培养基在28℃的条件下有氧发酵培养36h,巴氏醋酸菌CICC 20002采用葡萄糖琼脂培养基在30℃的条件下有氧发酵培养24h,嗜酸乳杆菌和青春双歧杆菌均采用MRS肉汤培养基在37℃的条件下无氧发酵24h;
(2)接种发酵
将培养好的菌种按照比例(异常威克汉姆酵母0.5%、巴氏醋酸菌1%、嗜酸乳杆菌、青春双歧杆菌1.5%)接种到发酵培养基中;发酵条件为:35℃、有氧发酵3d,接着转为30℃无氧发酵3d;
(3)陈酿
发酵完后转到储存罐中常温无氧发酵180d,然后用蒸馏设备进行蒸馏,制得酵素蒸馏液。
优选的,所述步骤(2)中的发酵培养基的各成分均为药食同源原料,各成分及其含量包括:金银花1-3.5g/L、菊花1-3g/L、黄精0.5-2.5g/L、刺梨3-5g/L、胡萝卜1-4g/L、余甘子1-4g/L、木瓜2-4g/L、蓝莓2-5g/L、决明子0.5-2g/L、蔗糖15-25g/L。
本发明提供一种眼部护理液,所述眼部护理液的成分及其质量百分比包括:酵素蒸馏液80%、生理盐水15%、矿物能量水5%。
优选的,所述矿物能量水包括钾离子、镁离子、钙离子、钠离子和偏硅酸。
本发明提供一种如上所述的眼部护理液的制备方法,包括以下步骤:

b、搅拌;
c、超声破碎;
d、微孔滤膜过滤;
e、灭菌。
优选的,所述步骤d中,膜的规格为0.22um。
优选的,所述步骤e中,灭菌的温度为105℃,所述灭菌的时长为15min。

(3)本发明为一种酵素蒸馏液的制备方法及其作为眼部护理液的应用,通过抑菌实验检测,使所制备的眼部护理液具有抑制眼部细菌的效果,缓解视疲劳,对相关的眼部疾病有辅助作用。

下面将结合本发明实施例,对本发明实施例中的技术方案进行清楚、完整地描述,显然,所描述的实施例仅仅是本发明一部分实施例,而不是全部的实施例。基于本发明中的实施例,本领域普通技术人员在没有做出创造性劳动前提下所获得的所有其它实施例,都属于本发明保护的范围。
实施例
一种酵素蒸馏液的制备方法,包括以下步骤:
(1)菌种培养
①、菌种:异常威克汉姆酵母CICC 1312、巴氏醋酸菌CICC 20002、嗜酸乳杆菌CICC6096、青春双歧杆菌CICC6180;
②、种子培养基:麦芽汁培养基、葡萄糖琼脂培养基、MRS肉汤培养基;
所述异常威克汉姆酵母CICC 1312采用麦芽汁培养基在28℃的条件下有氧发酵培养36h,巴氏醋酸菌CICC 20002采用葡萄糖琼脂培养基在30℃的条件下有氧发酵培养24h,嗜酸乳杆菌和青春双歧杆菌均采用MRS肉汤培养基在37℃的条件下无氧发酵24h;
(2)接种发酵
将培养好的菌种按照比例(异常威克汉姆酵母0.5%、巴氏醋酸菌1%、嗜酸乳杆菌、青春双歧杆菌1.5%)接种到发酵培养基中;发酵条件为:35℃、有氧发酵3d,接着转为30℃无氧发酵3d;
(3)陈酿
(4)发酵完后转到储存罐中常温无氧发酵180d,然后用蒸馏设备进行蒸馏,制得酵素蒸馏液。
在本实施例中,经GC-MS数据分析利用峰面积相对归一法,选择匹配度大于80度的化合物;可测出酵素蒸馏液67种挥发性成分,其中的醇类化合物为10种、酯类15类、酸类9种、其他类5种、烷烃类8种、烯烃类7种、醛类7种、芳香族9种。
优选的,所述步骤(2)中的发酵培养基的各成分均为药食同源原料,各成分及其含量包括:金银花1-3.5g/L、菊花1-3g/L、黄精0.5-2.5g/L、刺梨3-5g/L、胡萝卜1-4g/L、余甘子1-4g/L、木瓜2-4g/L、蓝莓2-5g/L、决明子0.5-2g/L、蔗糖15-25g/L,,更优选的,金银花2g/L、菊花2g/L、黄精1g/L、刺梨4.5g/L、胡萝卜2.5g/L、余甘子2g/L、木瓜2.5g/L、蓝莓3g/L、决明子1g/L、蔗糖20g/L。
在本实施例中,所述步骤(2)中的发酵培养基的各成分均为药食同源原料,各成分及其含量包括:金银花2g/L、菊花2g/L、黄精1g/L、刺梨4.5g/L、胡萝卜2.5g/L、余甘子2g/L、木瓜2.5g/L、蓝莓3g/L、决明子1g/L、蔗糖20g/L。
本发明提供一种眼部护理液,所述眼部护理液的成分及其质量百分比包括:酵素蒸馏液80%、生理盐水15%、矿物能量水5%。
在本实施例中,酵素蒸馏液通过如上所述的一种酵素蒸馏液的制备方法所制成。
优选的,所述矿物能量水包括钾离子、镁离子、钙离子、钠离子和偏硅酸。
本发明提供一种如上所述的眼部护理液的制备方法,包括以下步骤:
a、配料;
b、搅拌;
c、超声破碎;
d、微孔滤膜过滤;
e、灭菌。
优选的,所述步骤d中,膜的规格为0.22um。
优选的,所述步骤e中,灭菌的温度为105℃,所述灭菌的时长为15min。
优选的,所述步骤c中,超声破碎的频率为15-20KhZ,超声的时长为3-6min。
酵素蒸馏液对于大肠杆菌、金黄色葡萄球菌、白色念珠菌的悬液定量抑菌试验
一、器材
1.样品信息:酵素蒸馏液原液作为受试物
2.试验菌种:
大肠杆菌(8099),由广东省食品微生物安全工程技术研究开发中心提供,培养第3代;
金黄色葡萄球菌(ATCC6538),由北纳创联生物科技有限公司提供,培养第3代;
白色念珠菌(ATCC10231),由北纳创联生物科技有限公司提供,培养第3代。
3.培养基:NA/20220815、SDA/20220812
4.仪器设备:生物安全柜BSC-1604B2(WPE-RH0311)、电热恒温培养箱DHP-9272(WPE-RH0501),水浴锅DC0510(WPE-RHO327)
二、方法
1.检验依据:WS/T 650-2019《抗菌和抑菌效果评价方法》5.1.1悬液定量抑菌试验
2.检测环境:温度:25.0℃,湿度;48%RH(大肠杆菌、金黄色葡萄球菌)温度:24.7℃,湿度:49%RH(白色念珠菌)
3.抑菌效果:受试物作用20min,试验重复3次
三、结果
对大肠杆菌、金黄色葡萄球菌、白色念珠菌的悬液定量抑菌试验检测结果如下表1、表2、表3所示:

经3次重复试验,该样品作用20min对大肠杆菌、金黄色葡萄球菌、白色念珠菌抑菌率均≥50%~90%,产品有抑菌作用,符合WS/T 650-2019《抗菌和抑菌效果评价方法》5.1.1.5的要求。

Claims (8)
1.一种酵素蒸馏液的制备方法,其特征在于,包括以下步骤:
(1)菌种培养
①、菌种:异常威克汉姆酵母CICC 1312、巴氏醋酸菌CICC 20002、嗜酸乳杆菌CICC6096、青春双歧杆菌CICC6180;
②、种子培养基:麦芽汁培养基、葡萄糖琼脂培养基、MRS肉汤培养基;
所述异常威克汉姆酵母CICC 1312采用麦芽汁培养基在28℃的条件下有氧发酵培养36h,巴氏醋酸菌CICC 20002采用葡萄糖琼脂培养基在30℃的条件下有氧发酵培养24h,嗜酸乳杆菌和青春双歧杆菌均采用MRS肉汤培养基在37℃的条件下无氧发酵24h;
(2)接种发酵
将培养好的菌种按照比例(异常威克汉姆酵母0.5%、巴氏醋酸菌1%、嗜酸乳杆菌、青春双歧杆菌1.5%)接种到发酵培养基中;发酵条件为:35℃、有氧发酵3d,接着转为30℃无氧发酵3d;
(3)陈酿
发酵完后转到储存罐中常温无氧发酵180d,然后用蒸馏设备进行蒸馏,制得酵素蒸馏液。
2.根据权利要求1所述的一种酵素蒸馏液的制备方法,其特征在于,所述步骤(2)中的发酵培养基的各成分均为药食同源原料,各成分及其含量包括:金银花1-3.5g/L、菊花1-3g/L、黄精0.5-2.5g/L、刺梨3-5g/L、胡萝卜1-4g/L、余甘子1-4g/L、木瓜2-4g/L、蓝莓2-5g/L、决明子0.5-2g/L、蔗糖15-25g/L。
3.一种眼部护理液,其特征在于,所述眼部护理液的成分及其质量百分比包括:酵素蒸馏液80%、生理盐水15%、矿物能量水5%。
4.根据权利要求3所述的一种眼部护理液,其特征在于,所述矿物能量水包括钾离子、镁离子、钙离子、钠离子和偏硅酸。
5.一种如权利要求3所述的眼部护理液的制备方法,其特征在于,包括以下步骤:
a、配料;
b、搅拌;
c、超声破碎;
d、微孔滤膜过滤;
e、灭菌。
6.根据权利要求5所述的一种眼部护理液的制备方法,其特征在于,所述步骤d中,膜的规格为0.22um。
7.根据权利要求5所述的一种眼部护理液的制备方法,其特征在于,所述步骤e中,灭菌的温度为105℃,所述灭菌的时长为15min。
8.根据权利要求5所述的一种眼部护理液的制备方法,其特征在于,所述步骤c中,超声破碎的频率为15-20KhZ,超声的时长为3-6min。
