chick embryo chorioallantoic membrane irritation test:
chick embryo chorioallantoic membrane (CAM) test is performed to determine whether the product is irritating at different concentration ratios. The egg shell of the air chamber part of a nine-day-old chick embryo is stripped, a white egg membrane is exposed, the egg membrane is moistened by 0.9% sodium chloride (NaCl) solution, the intima is removed by forceps, the vascular membrane is guaranteed not to be damaged, and a preliminary experiment and a formal experiment are carried out after the damaged blood vessel is confirmed not to be damaged.
The experimental steps are as follows:
s1, grouping tests, wherein each group contains at least 6 chick embryos, and a negative control, a positive control and a blank control are required to be set in each test;
s2, preparing CAM, namely performing egg lighting inspection on 9-day-old chick embryos, peeling off eggshell parts of air chambers, dripping physiological saline on the surfaces of eggshell membranes to fully wet the eggshell membranes, and then removing the eggshell membranes;
s3, placing the test ring on the CAM, and sucking 200 mu L of test substance to drip into the test ring;
s4, immediately observing the change degree of each toxic effect after acting for 3min, and giving a stimulation score (ES).
And (4) observing results:
bleeding: blood flows from the blood vessels or capillaries of the CAM. No bleeding, mild bleeding, moderate bleeding, and severe bleeding were scored 0, 1,2, and 3, respectively.
Blood coagulation: it refers to the condition of protein denaturation inside and outside blood vessels, such as thrombus, closed swelling, blood coagulation spots inside and outside blood vessels, opacification and turbidity outside blood vessels, etc. The blood coagulation was judged to be 0, 1,2 and 3 points for non-coagulation, mild coagulation, moderate coagulation and moderate coagulation, respectively.
Vascular lysis: refers to ablation of blood vessels on the CAM membrane. The scores of 0, 1,2 and 3 are respectively judged according to avascular ablation, mild vascular ablation, moderate vascular ablation and severe vascular ablation.
Results calculation and evaluation: the average score of ES was calculated according to the following formula, and ES = (sum of degrees of bleeding, coagulation, and vascular lysis observed in 6 chick embryos)/3 was judged for stimulation classification according to Table 1
Endpoint scoring Irritation classification
ES≤12 No/light irritation
12<ES<16 Moderate irritation
ES≥16 Severe irritation property
TABLE 1 evaluation of ES results
Figure BDA0003812217640000081
