背景技术
植物类病斑是指在没有机械损伤、病原菌入侵且没有逆境胁迫下,植物体在自然生长下自发地产生类似病原菌侵染的坏死斑,该过程涉及细胞程序性死亡,其中斑点面积可以反应细胞死亡程度。根据类病斑突变体的表型可以将类病斑分为起始型和扩散型;起始型指的是植株没有受到病原物侵害,自发并随机在叶片或者植株上产生坏死斑,但随着植物体的生长不会扩散;扩散型又可称为蔓延型,即病斑会从起始发生的部位向周围随机扩散甚至蔓延到整个植株。
水稻产生类病斑的机制十分复杂,目前研究表明,大部分类病斑突变体对于稻瘟病菌或白叶枯病菌都表现出了增强的抗病性,因此,亟待对类斑病突变体进行深入研究,进而为水稻病原体广谱抗性的探索以及水稻的选育提供新的方向。

下面对本发明实施例中的技术方案进行清楚、完整地描述,显然,所描述的实施例仅仅是本发明一部分实施例,而不是全部的实施例。基于本发明中的实施例,本领域普通技术人员在没有做出创造性劳动前提下所获得的所有其他实施例,都属于本发明保护的范围。
实施例1突变材料的获得
以粳稻品种秀水11为实验材料进行EMS化学诱变:
清水浸种8小时,然后沥出水分,倒入已经配比好的1%EMS溶液浸泡诱变8小时,期间用小木棍搅拌多次确保混合均匀。
诱变完成后用流动水冲洗净有毒的EMS溶液,25℃催芽后播种,单株插秧单株收种,生长时期按常规大田管理。
水稻材料种植于浙江省金华市浙江师范大学生化学院试验田,常规管理;在大田条件下观察发现2株在分蘖盛期起始在叶尖出现病斑,从分蘖盛期到抽穗期,病斑越发严重,逐渐蔓延到整个叶片(图1),将这两种类病斑突变材料分别记为spl88-1、spl88-2。
将秀水11与2种突变材料分别接白叶枯病病菌,15天后观察剑叶表型,测量白叶枯病斑长度,并qRT-PCR检测防御相关基因表达情况,结果如图2所示,突变材料对于白叶枯病菌的抗性明显增强。
2种水稻类病斑突变材料分别自交,获得稳定遗传类病斑表型的后代,用于以下实验:
对比野生型与突变材料的主要农艺性状,发现突变材料的株高、分蘖数、分支数、每穗粒数、千粒重和结实率均显著性下降(图3)。
突变材料从分蘖盛期到成熟期叶片上都存在病斑,在分蘖盛期采取野生型与突变材料叶片进行叶绿素含量测定,结果如图4所示,突变材料叶绿素a、叶绿素b都显著低于野生型。
此外,对分蘖盛期植株的光合速率进行测定,发现突变材料的最大光合速率也发生了显著性的降低。
为了进一步探究突变材料叶绿素含量降低的原因,在分蘖盛期对野生型和突变材料进行了透射电镜观察。结果如图5所示,突变材料的叶绿体发生皱缩,内部片层结构变得紊乱。更为明显的是,突变材料叶绿体内的嗜锇颗粒明显增多。
气孔孔径是影响光合作用的重要因素,利用扫描电镜对分蘖盛期野生型与突变材料的叶片进行观察,结果如图6所示,突变材料的气孔孔径大于野生型。
以上结果表明,突变材料叶绿体结构的改变导致了其类病斑表型的出现,从而负调控植株的生长发育。
实施例2群体构建和遗传分析
将突变材料spl88-1、spl88-2分别与和常规籼稻南京6号进行正反交,F1植株均表现出正常野生型表型,说明突变材料表型受隐性核基因控制。统计F2分离群体分离比(表1、表2),结果表明,正常表型的植株和突变材料表型的植株的分离比经过卡方检验接近3:1分离,这表明突变材料类病斑、株高、分蘖数等表型是由一对单隐性核基因控制。

实施例3spl88基因的精细定位
设计均匀分布于水稻12条染色体的SSR引物,对突变材料与秀水11、南京6号进行多态性筛选;然后用21个spl88-1/南京6号、spl88-2/南京6号中F2类病斑单株进行连锁分析,初步确认目标基因所在的染色体位置;基因组DNA采用CTAB法提取,具体步骤如下:
①、称取0.1g的水稻叶片用液氮研磨成粉状,然后加入600μL的CTAB溶液配制的DNA提取缓冲液(2%(m/V)CTAB,100mmol/L Tris-Cl,20mmol/L EDTA,1.4mol/LNaCl;pH8.0),65℃水浴40分钟。再加600μL的氯仿:异戊醇(24:1的体积比),并混匀。10,000rpm离心5分钟,将上清液转移到新的离心管中。
②、在上述步骤①离心后所得的上清液中加2/3~1倍体积预冷(至4℃)的异丙醇,轻轻混匀至DNA沉淀。13,000rpm离心8分钟,倒出上清液。
③、再用70%(体积浓度)的已醇200μL洗涤上述步骤②所得的DNA沉淀物。
④、将上述洗涤后的DNA晾干并溶于100μLTE缓冲液或纯水中。
⑤、紫外分光光度法检测上述步骤④所得的DNA样品的浓度,0.7%的琼脂糖凝胶电泳检测DNA的完整性。完整合适的DNA用于PCR扩增,不完整的DNA则重新提取,直至获得完整的DNA。
PCR反应体系采用10μL体系:DNA模板1μL,10×PCR缓冲液1μL,正反向引物(10μmol/L)各0.5μL,dNTPs 1μL,rTaq酶0.2μL,加ddH2O补足10μL。PCR扩增程序如下:94℃下预变性4min;94℃下变性30s,55℃~60℃下退火30s(温度因引物不同而异),72℃下延伸30s,40个循环;最后72℃下延伸10min。
PCR产物用4%琼脂糖凝胶电泳,电泳结束后在凝胶成像仪拍照并读胶。利用筛选的120对SSR引物进行spl88基因连锁分析,发现在第2号染色体的末尾附近处表现出连锁现象。
在连锁标记上下游设计新的Indel标记,用上述21个单株将目的基因区间锁定在分子标记M1与M10之间。
在M1-M10区间再次设计新的分子标记,用579个F2单株最终将该基因定位在M5和M6之间大约84kb的区间内,引物序列见表3。

通过网站(http://rice.uga.edu/cgi-bin/gbrowse/rice/#search)查询发现,该区域内包含14个ORF。如图3所示,通过测序比对发现两突变材料均在基因LOC_Os02g51180发生突变(对应秀水11中该基因编码区序列如SEQ ID NO.1所示);其中,spl88-1突变材料该基因编码区的第132个碱基C发生缺失,使得组氨酸变成苏氨酸,导致提前终止,其突变核苷酸序列如SEQ ID NO.3所示,氨基酸序列如图4所示;spl88-2突变材料该基因编码区的第381位碱基T缺失,从而导致提前终止,其突变核苷酸序列如SEQ ID NO.4所示,氨基酸序列如图5所示。
综上,本发明所获得的spl88基因上的碱基缺失导致的蛋白质翻译提前终止造成了水稻植株变矮,类病斑表型的出现。
