Abstract
The invention relates to A61K, in particular to a blue copper peptide double agent and a using method and application thereof. The method comprises the following steps: a peptide agent comprising tripeptide-1; a copper agent, the copper agent comprising a copper salt. The tripeptide-1 and the copper salt are respectively prepared into the peptide agent and the copper agent which are mixed when in use, so that the activity of the blue copper peptide obtained from the tripeptide-1 and the copper salt can be prevented from being reduced, and therefore, when in use, the activity of the blue copper peptide is not reduced before the essence of the double agents is mixed, the repairing and whitening effects of the double agents on the skin can be fully exerted, the wrinkle reduction is promoted, the glossiness and the whiteness of the skin are improved, the conventional preservative component can not be added into the double agents provided by the invention, the preservative effect of the double agents is adjusted by a small amount of skin conditioning agents such as polyhydric alcohol and p-hydroxyacetophenone, and the double agents provided by the invention can be used for sensitive muscles, the repairing and the redness of the damaged skin are promoted, and the elasticity and the tightness of the skin are improved.
Description
Technical Field
The invention relates to A61K, in particular to a blue copper peptide double agent and a using method and application thereof.
Background
GHK tripeptide-1 is a natural substance of a human body, has small molecular weight, can effectively combine copper ions to obtain the blue copper peptide which acts on cells, promotes the regeneration of skin and the generation of protein, improves the elasticity of the skin and repairs injuries.
CN110237007A discloses a cosmetic composition containing copper peptides, a preparation method and an application thereof, and the copper peptides, the purslane extract, the centella asiatica extract and VB5 have good effects of relieving and resisting allergy under combined action.
However, tripeptide-1 and the copper peptide have poor stability, hydrolysis is easily found after long-term storage or illumination, and the problems of decomplexing and the like occur in an acidic environment, so that the activity of the copper peptide is influenced.
Disclosure of Invention
In order to solve the above problems, a first aspect of the present invention provides a bluecopper peptide double agent comprising:
a peptide agent comprising tripeptide-1;
a copper agent, the copper agent comprising a copper salt;
the peptide agent and the copper agent are prepared separately.
Tripeptide-1 has good antioxidant property, is beneficial to the expression and generation of protein, copper ions also have positive effects on the construction of collagen and elastin in human bodies, the generation of hyaluronic acid and the like, when the tripeptide-1 is contacted with the copper ions, a small molecular tripeptide-1 and copper complex can be quickly obtained, and the complex has high activity, but is easy to inactivate after long-term storage.
In a preferred embodiment of the present invention, the tripeptide-1 is in an amount of 0.2% or more by weight based on the peptide agent.
In a preferred embodiment of the present invention, the tripeptide-1 is 0.2 to 0.6% of the peptide agent, and may be 0.2%, 0.3%, 0.4%, 0.5%, 0.6%; preferably 0.4 to 0.5%.
As a preferable technical scheme of the invention, the copper salt is selected from one or more of copper acetate, copper aspartate, copper acetate, copper acetylacetonate, copper chloride, copper pyrrolidone carboxylate (PCA-Cu), copper sulfate, copper gluconate, copper acetomethionine, malachite extract, malachite, copper chlorophyll and polypeptide copper hydrochloride.
As a preferable embodiment of the present invention, the copper salt accounts for 0.6% or more by weight of the copper agent, and may be 0.6%, 0.7%, 0.75%, 0.8%, 0.85%, 0.9%, 0.95%, 1%, 1.05%, 1.1%; preferably 0.9 to 1%.
As a preferable technical scheme of the invention, the solvent of the peptide agent comprises yeast fermentation product filtrate and water, and the solvent of the copper agent comprises yeast fermentation product filtrate and water.
As a preferred technical solution of the present invention, the yeast fermentation product filtrate is one or more selected from a group consisting of a rice fermentation product filtrate, a secondary split yeast fermentation product filtrate, a galactose yeast-like bacteria fermentation product filtrate, a yeast fermentation product filtrate, a fission yeast mycelium fermentation product filtrate, a lactobacillus/soybean milk fermentation product filtrate, and a bacillus/glutamic acid fermentation product filtrate, and is preferably a galactose yeast-like bacteria fermentation product filtrate.
In addition, the inventors have also found that peptide substances such as tripeptide-1, which have poor stability, also affect the stability of tripeptide-1 and further cause subsequent reaction with copper agents to reduce the activity of the resulting bluecupreotide, and have found that, when the peptide agents are prepared, decomposition and the like of tripeptide-1 can be avoided without using preservatives by using yeast fermentation product filtrate and water together as solvents during long-term storage or exposure to heat, light and the like, so that when stored for a long time and used, the reaction with copper agents can be rapidly achieved to obtain a blue preparation for improvement of wrinkles and the like, and have also found that it is advantageous in promoting the achievement of skin lightening and whitening.
As a preferable technical scheme of the invention, the weight percentage of the yeast fermentation product filtrate in the peptide agent is 25-35%, and can be enumerated by 25%, 28%, 30%, 32% and 35%; the weight percentage of the yeast fermentation product filtrate in the copper agent is 25-35%, and can be enumerated by 25%, 28%, 30%, 32% and 35%.
As a preferable technical scheme, the copper agent further comprises an adsorbent, and the adsorbent is selected from one or more of cyclodextrin, sodium alginate and quaternary ammonium salt. In one embodiment, the weight percentage of the adsorbent in the copper adsorbent is 1 to 2%, and more preferably 1 to 1.5%.
The inventor finds that in the complexing process of the tripeptide-1 and the copper salt, if the concentration of the copper salt is higher, the copper salt can generate catalytic action on substances in the copper agent, particularly, under the action of illumination and the like, hydroxyl radicals and the like can be generated, and the stability of the copper agent is influenced.
In a preferred embodiment of the present invention, the thickener is one or more selected from the group consisting of alcaligenes polysaccharides, xanthan gum, carbomer, acrylic acid (ester)/C10-30 alkanol acrylate crosspolymer, and hydroxyethyl cellulose, and is not particularly limited.
As a preferable technical scheme of the invention, the weight percentage of the thickening agent in the peptide agent is 0.1-1%, the weight percentage of the thickening agent in the copper agent is 0.1-1%, wherein the thickening agents in the peptide agent and the copper agent can be the same or different, and one, two or more can be used. As an example, the peptide agent comprises 0.2-0.5% by weight of xanthan gum and 0.1-0.5% by weight of Alcaligenes polysaccharide, and the copper agent comprises 0.1-0.5% by weight of xanthan gum and 0.1-0.5% by weight of Alcaligenes polysaccharide.
In a preferred embodiment of the present invention, the humectant includes one or more of a polyhydric alcohol humectant, a polyether humectant, a polysaccharide humectant, and an amino acid humectant. Examples of the polyhydric alcohol-based moisturizer include, but are not limited to, butylene glycol, glycerol, 1, 2-pentanediol, polypropylene glycol, glyceryl polyether-26, sorbitol, 2-methyl-1, 3-propanediol, isoprene glycol, and the like, examples of the polysaccharide-based moisturizer include, but are not limited to, pullulan, sodium hyaluronate, tremella polysaccharide, xylitol-based glucoside, xylitol dehydrate, xylitol, and the like, and examples of the polyether-based moisturizer include, but are not limited to, glyceryl polyoxyethylene ether, polyoxypropylene ether, polyoxyethylene-polyoxypropylene copolymer, and the like.
As a preferable technical scheme, the weight percentage of the moisturizing agent in the peptide agent is 10-25%, and the weight percentage of the moisturizing agent in the copper agent is 10-25%.
In a preferred embodiment of the present invention, the humectant includes a polyhydric alcohol-based humectant and a polysaccharide-based humectant. In a preferable technical scheme of the invention, the weight percentage of the polyalcohol humectant in the peptide agent is 5-20%, the weight percentage of the polyether humectant is 1-3%, the weight percentage of the polysaccharide humectant is 0.1-1%, the weight percentage of the polyalcohol humectant in the copper agent is 5-20%, and the weight percentage of the polysaccharide humectant is 0.5-2%.
As a preferable technical scheme, the weight percentage of the skin conditioner in the peptide agent is 1-4%, and the weight percentage of the skin conditioner in the copper agent is 1-4%.
Examples of skin conditioning agents include, but are not limited to, niacinamide, panthenol, centella asiatica (CENTELLA ASIATICA) extract, pullulanase, nonapeptide-1, allantoin, hydrogenated lecithin, shea butter, ceramide 3, oat (AVENA SATIVA) kernel oil, 1, 2-hexanediol, p-hydroxyacetophenone, and the like. The skin conditioning agent in the peptide agent and the copper agent of the present invention may be the same or different, and may be one or more selected, and is not particularly limited.
As a preferable technical scheme, the peptide agent further comprises at least one of an emollient, an antioxidant, a pH regulator and an emulsifier; the copper agent also comprises at least one of an emollient, an antioxidant, a pH regulator and an emulsifier.
Examples of emollients include, but are not limited to, shea butter, cetostearyl alcohol, isononyl isononanoate, caprylic/capric triglyceride, olive oil, jojoba seed oil, squalane, jojoba oil, shea butter, diethoxyethyl succinate, and the like. In one embodiment, the weight percentage of the emollient in the peptide agent is 0.5-1%, and the weight percentage of the emollient in the copper agent is 1-1.5%.
Examples of the pH adjuster include, but are not limited to, triethanolamine, laurylamine, sodium hydroxide, aminomethyl propanol, ethylenediamine, sodium hydrogencarbonate, and the like. In one embodiment, the weight percentage of the pH regulator in the copper agent is 0.1-0.4%.
The peptide agent and the copper agent provided by the invention are prepared by mixing and homogenizing which are well known in the field, and are not particularly limited.
The second aspect of the invention provides a use method of the blue copper peptide double agent, which comprises the following steps: the copper agent and the peptide agent are mixed to generate color change reaction. In the mixing, the weight ratio of the copper agent to the peptide agent is 1: 1 to 3, are not particularly limited.
The third aspect of the invention provides an application of a blue copper peptide double agent in skin care products.
Compared with the prior art, the invention has the following beneficial effects:
the tripeptide-1 and the copper salt are respectively prepared into the peptide agent and the copper agent, and the peptide agent and the copper agent are mixed when the composition is used, so that the activity of the blue copper peptide obtained from the tripeptide-1 and the copper salt can be prevented from being reduced, and therefore, when the composition is used, the activity of the blue copper peptide is not reduced before the essence of the two agents is mixed, the repairing and whitening effects of the two agents on the skin can be fully exerted, wrinkles are promoted to be reduced, the glossiness and the whiteness of the skin are improved, the conventional antiseptic components are not added into the two agents provided by the invention, the antiseptic effect of the two agents is adjusted through a small amount of skin conditioning agents such as polyhydric alcohol and p-hydroxyacetophenone, and the two agents provided by the invention can be used for sensitive muscles, the repairing and the reddening of damaged skin are promoted, and the elasticity and the compactness of the skin are promoted to be improved. In addition, the copper agent and the peptide agent provided by the invention have a color change reaction when being mixed, a solution after color change is quickly obtained, the synthesis of the blue copper peptide can be more visually indicated, and the copper agent and the peptide agent provided by the invention have good storage stability and further play a role of the blue copper peptide.

Detailed Description
Examples
Examples 1 to 3 provide a double agent in which copper salt is copper gluconate
The present example provides a bronopol, comprising: the copper agent and the peptide agent are prepared from the raw materials in percentage by weight as shown in table 1, and the raw materials in percentage by weight as shown in table 2.

Evaluation of Performance
1. And (3) testing the efficacy: the dual agents provided in example 1 were added to two separate chambers of an essence capsule, respectively, and tested, as shown in fig. 1 to 16.
2. Testing the spot paste: the dual dose provided in example 1 was subjected to patch testing as shown in figure 17.
3. Stability: the stability tests were performed on the double agents provided in examples 1 to 3, and the peptide agents and the copper agents of examples 1 to 3 satisfied the results shown in Table 5 and Table 6, respectively.

In addition, when the single dose of the copper-peptide provided by the control group is compared, it can be seen that the stability of the double doses is a common problem, for example, the raw materials slightly change the taste at high temperature and slightly change the color under the light, which is a normal active substance change phenomenon, and the effect and the use of the product are not influenced, and when the same amount of the copper-peptide is directly added into one dose of the essence, the stability is much worse than that of the former dose, and the color and the taste change of different degrees are generated in about one week. The stability of the product then goes on, the cold resistance even at the last eighth week gives rise to precipitation, and the discoloration and off-taste, which are otherwise different, become stronger and more severe. The stability can be effectively improved by the double-agent structure provided by the invention, and the structure is specifically shown in table 7.

4. Physical and chemical properties of the copper agent: the copper agent and the peptide agent provided by the embodiment 1 and the embodiments 4-8 are mixed according to the weight ratio of 1: 2, and observing whether discoloration occurs, the results are shown in table 8.

The results of the table tests show that the double preparations provided by the invention have good stability, can be beneficial to the activity expression of the mixed blue copper peptide, and can be used for promoting the repair of damaged sensitive muscle skin, reducing wrinkles, improving the brightness and whiteness of the skin, reducing the dehydration of epidermis and promoting the elasticity and tightness of the skin.
Claims (10)
1. A blueprint peptide double agent, which is characterized by comprising:
a peptide agent comprising tripeptide-1;
a copper agent, the copper agent comprising a copper salt;
the peptide agent and the copper agent are prepared separately.
2. The bluecopper peptide double agent according to claim 1, wherein the tripeptide-1 is in an amount of 0.2% or more by weight of the peptide agent.
3. The dual agent of claim 1, wherein the copper salt is present in an amount of 0.6% or greater by weight of the copper agent.
4. The dual agent according to claim 1, wherein the copper salt is selected from one or more of copper acetate, copper aspartate, copper acetate, copper acetylacetonate, copper chloride, copper pyrrolidone carboxylate, copper sulfate, copper gluconate, copper acetomethionate, malachite extract, malachite, copper chlorophyll, and copper polypeptide hydrochloride.
5. The bluecopper peptide double agent according to any one of claims 1-4, wherein the solvent of the peptide agent comprises yeast fermentation product filtrate and water, and the solvent of the copper agent comprises yeast fermentation product filtrate and water.
6. The dual agent according to claim 5, wherein the yeast fermentation product filtrate is selected from one or more of rice fermentation product filtrate, yeast secondary split fermentation product filtrate, galactose yeast-like fermentation product filtrate, yeast fermentation product filtrate, fission yeast mycelium fermentation product filtrate, lactobacillus/soybean milk fermentation product filtrate, bacillus/glutamic acid fermentation product filtrate.
7. The bluecopper peptide doublet according to claim 1, wherein the peptide agent further comprises one or more of a humectant, a thickener, a skin conditioner.
8. The bluecopper peptide doublet according to claim 1, wherein the copper agent further comprises one or more of a humectant, a thickener, a skin conditioner.
9. The use method of the bluecopper peptide double agent according to any one of claims 1-8, characterized by comprising the following steps: the copper agent and the peptide agent are mixed to generate color change reaction.
10. The use of the dual agent of claim 1 to 8 for skin care.
