Abstract
The invention relates to A61K, in particular to a blue copper peptide double agent, and a use method and application thereof. Comprising the following steps: a peptide agent comprising tripeptide-1; a copper agent, the copper agent comprising a copper salt. The tripeptide-1 and the copper salt are respectively prepared into the peptide agent and the copper agent, and are mixed when in use, so that the activity of the blue copper peptide obtained by the tripeptide-1 and the copper salt is prevented from being reduced, the activity of the blue copper peptide is not reduced before the double-agent essence is mixed when in use, the repairing and whitening effects of the double-agent on skin can be fully exerted, the wrinkle reduction is promoted, the skin glossiness and whiteness are improved, the double-agent provided by the invention can be used for adjusting the anti-corrosion effect of the double-agent by a small amount of polyalcohol, the p-hydroxyacetophenone and other skin conditioning agents without adding conventional anti-corrosion components, and the double-agent provided by the invention can be used for sensitive muscles, promote the repairing and reddening of damaged skin and promote the improvement of skin elasticity and compactness.
Description
Technical Field
The invention relates to A61K, in particular to a blue copper peptide double agent, and a use method and application thereof.
Background
GHK tripeptide-1 is a natural substance of human body, has small molecular weight, can effectively combine copper ions, and can obtain blue copper peptide which acts on cells, promote skin regeneration and protein generation, improve skin elasticity and repair damage.
CN110237007a discloses a cosmetic composition containing blue copper peptide, a preparation method and application thereof, and the cosmetic composition has good effects of relieving and resisting sensitization through the combined action of blue copper peptide, purslane extract, centella asiatica extract and VB 5.
However, tripeptide-1 and blue copper peptide have poor stability, hydrolysis is easy to be found in long-term storage or illumination, and problems such as decomplexing and the like can occur in an acidic environment, so that the activity of the blue copper peptide is affected, and at present, the storage performance is generally improved by adding more preservative, but the problem of sensitivity and the like is easy to occur due to the addition of the preservative, so that the preparation and the use method of the blue copper peptide are needed to be provided for improving the storage and use performance.
Disclosure of Invention
In order to solve the above problems, a first aspect of the present invention provides a blue copper peptide dual agent comprising:
a peptide agent comprising tripeptide-1;
a copper agent comprising a copper salt;
peptide and copper agents are prepared separately.
The tripeptide-1 has good antioxidant property, is favorable for the expression and generation of protein, and copper ions have positive effects on the construction of collagen and elastin in a human body, the generation of hyaluronic acid and the like, when the tripeptide-1 contacts with the copper ions, small-molecule tripeptide-1 and copper complex can be quickly obtained, and the tripeptide-1 has high activity, but is easy to inactivate after long-term storage.
As a preferable technical scheme of the invention, the tripeptide-1 accounts for more than or equal to 0.2% of the weight of the peptide agent.
In a preferred embodiment of the present invention, the tripeptide-1 may be present in an amount of 0.2 to 0.6% of the peptide agent, for example, 0.2%, 0.3%, 0.4%, 0.5% and 0.6%; preferably 0.4 to 0.5%.
As a preferred embodiment of the present invention, the copper salt is selected from one or more of copper acetate, copper aspartate, copper acetate, copper acetylacetonate, copper chloride, copper pyrrolidone carboxylate (PCA-Cu), copper sulfate, copper gluconate, copper acetylmethionine, malachite extract, malachite, copper chlorophyll, and copper salt of polypeptide.
In a preferred embodiment of the present invention, the copper salt is 0.6% by weight or more of the copper agent, and examples thereof include 0.6%, 0.7%, 0.75%, 0.8%, 0.85%, 0.9%, 0.95%, 1%, 1.05%, and 1.1%; preferably 0.9 to 1%.
As a preferred embodiment of the present invention, the solvent of the peptide agent comprises yeast fermentation product filtrate and water, and the solvent of the copper agent comprises yeast fermentation product filtrate and water.
As a preferred embodiment of the present invention, the yeast fermentation product filtrate is selected from one or more of rice fermentation product filtrate, yeast fermentation product filtrate, galactose yeast-like fermentation product filtrate, yeast fermentation product filtrate, schizosaccharomyces mycelium fermentation product filtrate, lactobacillus/soybean milk fermentation product filtrate, bacillus/glutamic acid fermentation product filtrate, preferably galactose yeast-like fermentation product filtrate.
In addition, the inventors have found that the stability of peptide substances such as tripeptide-1 is poor, which also affects the stability of tripeptide-1, and further causes the subsequent reaction with a copper agent to decrease the activity of blue copper peptide, and have found that when a peptide agent is prepared, by using a yeast fermentation product filtrate together with water as a solvent, decomposition and the like of tripeptide-1 can be avoided while no preservative is used or subjected to heat, light and the like, so that when stored for a long period of time, the tripeptide-1 can still react with the copper agent rapidly to obtain a blue preparation for improving wrinkles and the like, and have found that it is also advantageous to promote the achievement of skin lightening and whitening.
In a preferred embodiment of the present invention, the weight percentage of the yeast fermentation product filtrate in the peptide agent is 25 to 35%, and examples thereof include 25%, 28%, 30%, 32%, and 35%; the weight percentage of the yeast fermentation product filtrate in the copper agent is 25-35%, and examples thereof include 25%, 28%, 30%, 32% and 35%.
As a preferable technical scheme of the invention, the copper agent further comprises an adsorbent, and the adsorbent is one or more selected from cyclodextrin, sodium alginate and quaternary ammonium salt. In one embodiment, the weight percentage of the adsorbent in the copper agent is 1 to 2%, more preferably 1 to 1.5%.
The inventor finds that in the complexing process of tripeptide-1 and copper salt, if the concentration of copper salt is higher, the copper salt can generate catalysis effect on substances in the copper agent, especially the copper salt is subjected to the effects of illumination and the like, oxyhydrogen free radicals and the like can be generated, so that the stability of the copper agent is influenced.
As a preferable technical scheme of the invention, the thickening agent is one or more selected from alcaligenes polysaccharides, xanthan gum, carbomer, acrylic acid (esters)/C10-30 alkanol acrylate cross-linked polymer and hydroxyethyl cellulose, and is not particularly limited.
As a preferable technical scheme of the invention, the weight percentage of the thickener in the peptide agent is 0.1-1%, and the weight percentage of the thickener in the copper agent is 0.1-1%, wherein the thickener in the peptide agent and the thickener in the copper agent can be the same or different, and one, two or more kinds can be used. As an example, the peptide agent comprises 0.2-0.5% of xanthan gum and 0.1-0.5% of alcaligenes polysaccharide by weight percent, and the copper agent comprises 0.1-0.5% of xanthan gum and 0.1-0.5% of alcaligenes polysaccharide by weight percent.
As a preferable technical scheme of the invention, the humectant comprises one or more of polyalcohol humectant, polyether humectant, polysaccharide humectant and amino acid humectant. Examples of the polyhydric alcohol type humectant include, but are not limited to, butylene glycol, glycerin, 1, 2-pentanediol, polypropylene glycol, glycerin polyether-26, sorbitol, 2-methyl-1, 3-propanediol, isopentane diol, etc., examples of the polysaccharide type humectant include, but are not limited to, pullulanase polysaccharide, sodium hyaluronate, tremella polysaccharide, xylitol-based glucoside, dehydrated xylitol, etc., and examples of the polyether type humectant include, but are not limited to, glycerin polyoxyethylene ether, polyoxypropylene ether, polyoxyethylene-polyoxypropylene copolymer, etc.
As a preferable technical scheme of the invention, the weight percentage of the humectant in the peptide agent is 10-25%, and the weight percentage of the humectant in the copper agent is 10-25%.
As a preferable technical scheme of the invention, the humectant comprises a polyol humectant and a polysaccharide humectant. As a preferable technical scheme of the invention, the weight percentage of the polyol humectant in the peptide agent is 5-20%, the weight percentage of the polyether humectant is 1-3%, the weight percentage of the polysaccharide humectant is 0.1-1%, the weight percentage of the polyol humectant in the copper agent is 5-20%, and the weight percentage of the polysaccharide humectant is 0.5-2%.
As a preferable technical scheme of the invention, the weight percentage of the skin conditioning agent in the peptide agent is 1-4%, and the weight percentage of the skin conditioning agent in the copper agent is 1-4%.
Examples of skin conditioning agents include, but are not limited to, niacinamide, panthenol, centella asiatica (CENTELLA ASIATICA) extracts, pullulanase polysaccharides, nonapeptide-1, allantoin, hydrogenated lecithin, shea butter, ceramide 3, oat (AVENA SATIVA) kernel oil, 1, 2-hexanediol, p-hydroxyacetophenone, and the like. The skin conditioning agents in the peptide agent and the copper agent of the present invention may be the same or different, and one or more may be selected without specific limitation.
As a preferred technical scheme of the invention, the peptide agent further comprises at least one of an emollient, an antioxidant, a pH regulator and an emulsifier; the copper agent also comprises at least one of an emollient, an antioxidant, a pH regulator and an emulsifier.
Examples of emollients include, but are not limited to, shea butter, cetostearyl alcohol, isononyl isononanoate, caprylic/capric triglyceride, olive oil, jojoba seed oil, squalane, jojoba oil, shea butter, diethoxyethyl succinate, and the like. In one embodiment, the weight percent of the emollient in the peptide agent is 0.5 to 1% and the weight percent of the emollient in the copper agent is 1 to 1.5%.
Examples of pH adjusting agents include, but are not limited to, triethanolamine, laurylamine, sodium hydroxide, aminomethylpropanol, ethylenediamine, sodium bicarbonate, and the like. In one embodiment, the weight percentage of the pH regulator in the copper agent is 0.1-0.4%.
The peptide agent and the copper agent provided by the invention are prepared by mixing and homogenizing well known in the art, and are not particularly limited.
The second aspect of the invention provides a use method of the blue copper peptide double agent, which comprises the following steps: mixing copper agent and peptide agent to generate color change reaction. In the mixing, the weight ratio of the copper agent to the peptide agent is 1:1 to 3, and is not particularly limited.
The third aspect of the invention provides an application of the blue copper peptide double agent in skin care products.
Compared with the prior art, the invention has the following beneficial effects:
the tripeptide-1 and the copper salt are respectively prepared into the peptide agent and the copper agent, and are mixed when in use, so that the activity of the blue copper peptide obtained by the tripeptide-1 and the copper salt is prevented from being reduced, the activity of the blue copper peptide is not reduced before the double-agent essence is mixed when in use, the repairing and whitening effects of the double-agent on skin can be fully exerted, the wrinkle reduction is promoted, the skin glossiness and whiteness are improved, the double-agent provided by the invention can be used for adjusting the anti-corrosion effect of the double-agent by a small amount of polyalcohol, the p-hydroxyacetophenone and other skin conditioning agents without adding conventional anti-corrosion components, and the double-agent provided by the invention can be used for sensitive muscles, promote the repairing and reddening of damaged skin and promote the improvement of skin elasticity and compactness. In addition, the copper agent and the peptide agent provided by the invention undergo a color change reaction when being mixed, a solution after color change is quickly obtained, blue copper peptide synthesis can be more visually indicated, and the copper agent and the peptide agent provided by the invention have good storage stability and further play a role of blue copper peptide.

Evaluation of Performance
1. Efficacy test: the two doses provided in example 1 were separately added to separate two chambers of the essence capsule and tested as shown in fig. 1-16.
2. Patch test: the patch test was performed on the two doses provided in example 1, as shown in fig. 17.
3. Stability: the stability test was performed on the two agents provided in examples 1 to 3, respectively, and the peptide agents in examples 1 to 3 satisfy the results in Table 5 and the copper agent satisfies the results in Table 6.

In addition, when comparing the single blue copper peptide provided by the control group, it can be seen that when the stability of the double agents is a common problem, such as slight off-flavor of the raw materials at high temperature, slight color change under illumination, normal active matter change phenomenon, no influence on the efficacy and use of the product, and when the same amount of blue copper peptide is directly added into a preparation of Jinghua, the stability is much worse than before, and the color change and off-flavor of different degrees are generated in about one week. The stability of the product then becomes increasingly cold-resistant and precipitates even at the last eighth week, and the other differently colored and off-flavored products become more intense and heavier. The stability can be effectively improved through the double-agent structure provided by the invention, and the stability is particularly shown in a table 7.

4. Physical and chemical properties of the copper agent: the copper agent and the peptide agent provided in example 1 and examples 4 to 8 are mixed according to the weight ratio of 1:2, and observing whether color change occurs, and the result is shown in table 8.

As shown by the test results of the surface, the double-agent provided by the invention has good stability, can be beneficial to the activity expression of the blue copper peptide after mixing, can reduce wrinkles, improve the brightness and whiteness of skin, reduce the water loss of epidermis and promote the elasticity and compactness of skin while promoting the repair of sensitive muscle damaged skin.
Claims (6)
1. A blue copper peptide dual agent, comprising:
a peptide agent comprising tripeptide-1;
a copper agent comprising a copper salt;
respectively preparing a peptide agent and a copper agent;
the tripeptide-1 accounts for 0.2 to 0.6 percent of the peptide agent; the copper salt accounts for 0.9-1% of the copper agent by weight; the copper salt comprises at least one of copper gluconate, malachite extract, copper chloride, copper chlorophyll, copper sulfate and PCA-Cu; the solvent of the peptide agent comprises yeast fermentation product filtrate and water, and the solvent of the copper agent comprises yeast fermentation product filtrate and water; the copper agent also comprises an adsorbent, and the adsorbent is one or more selected from cyclodextrin, sodium alginate and quaternary ammonium salt.
2. The blue copper peptide dual agent according to claim 1, wherein the yeast fermentation product filtrate is selected from one or more of rice fermentation product filtrate, saccharomyces cerevisiae fermentation product filtrate, galactosylyeast fermentation product filtrate, saccharomyces cerevisiae fermentation product filtrate, schizosaccharomyces cerevisiae mycelium fermentation product filtrate, lactobacillus/soybean milk fermentation product filtrate, bacillus/glutamic acid fermentation product filtrate.
3. The blue copper peptide dual agent according to claim 1, wherein the peptide agent further comprises one or more of a humectant, a thickener, and a skin conditioning agent.
4. The blue copper peptide dual agent according to claim 1, wherein the copper agent further comprises one or more of a humectant, a thickener, and a skin conditioning agent.
5. A method of using the blue copper peptide dual agent according to any one of claims 1 to 4, comprising: mixing copper agent and peptide agent to generate color change reaction.
6. Use of a blue copper peptide dual agent according to any one of claims 1 to 4 for skin care products.
