Description
Technical Field
The invention belongs to the field of cosmetics, and in particular relates to a wrinkle-removing and anti-aging skin care product, and a preparation method and application thereof.
Background
The skin is affected by many factors such as external environment or self metabolism, etc., free radicals are formed, and when the free radicals destroy collagen and active substances in normal cell membrane tissues, wrinkles are formed. With age, metabolism is slowed down, moisture and subcutaneous fat are reduced, elastic fibers become thicker gradually, and skin becomes wrinkled. Therefore, effective scavenging of free radicals and promotion of collagen synthesis or effective replenishment of collagen are important pathways for anti-wrinkle.
CN115517999a discloses an anti-aging composition and its application in beauty and skin care, the anti-aging composition comprises 0.1-5 parts of antioxidant, 0.0001-0.05 parts of compound polypeptide, 1-10 parts of panthenol, 0.3-2 parts of glycoside compound; the composite polypeptide used in the technical proposal has a plurality of categories, the polypeptide itself has the function of stimulating the growth of collagen, and the cosmetics prepared from the anti-aging composition need to be matched with a radio frequency beauty instrument for use so as to achieve the technical effect. The anti-aging composition has dependence on a radio frequency beauty instrument when in use, is inconvenient to operate, needs special technicians to operate, and severely limits the application of the anti-aging composition. CN114224768A discloses a repairing and anti-aging composition and a preparation method thereof, wherein the composition comprises 0.3% -1.5% of glyceroglycosides, 0.03% -0.15% of recombinant human collagen and 0.0003% -0.0015% of arginine/lysine polypeptides. At present, 5 or more than 5 components are added into an anti-aging composition in the prior art, the efficacy composition is relatively complex, the mechanism is undefined, the final efficacy of the skin care product is greatly affected, and meanwhile, the existing anti-wrinkle product generally has the problems of poor permeability, difficult absorption and unobvious anti-wrinkle repair effect.
Disclosure of Invention
The invention uses the excellent oxidation resistance of lycopene and the combination of hydrolyzed collagen and conopeptide which has good anti-wrinkle performance and is easy to be absorbed, and realizes the anti-wrinkle and anti-aging effects by controlling three different anti-wrinkle mechanisms of scavenging free radicals, promoting the absorption and secretion of collagen and affecting the release of neurotransmitters so as to weaken muscle contraction and smooth fine wrinkles. In addition, by utilizing the synergistic anti-wrinkle performance of three components of lycopene, hydrolyzed collagen and conopeptide under different action mechanisms and synergistic and easily permeable and absorbable small-molecule sodium hyaluronate, the synergistic effect of the four components is realized by compounding the four components according to a specific proportion, so that the effects of better moisturizing, wrinkle removing and the like are realized.
In order to overcome the problems of the prior art, one of the purposes of the present invention is to provide a wrinkle-removing and anti-aging composition. The wrinkle-removing and anti-aging composition can improve skin wrinkles, promote fibroblast proliferation, increase secretion of collagen, reduce or eliminate free radicals in cells, promote absorption of exogenous collagen by skin, and has obvious effects of resisting wrinkles, recovering skin elasticity and improving skin firmness.
The second purpose of the invention is to provide a wrinkle-removing and anti-aging skin care product.
The invention further aims to provide a preparation method of the wrinkle-removing and anti-aging skin care product.
The fourth object of the invention is to provide an application of the wrinkle-removing and anti-aging composition in the field of cosmetics.
In order to achieve the above purpose, the technical scheme adopted by the invention is as follows:
the first aspect of the invention provides a wrinkle-removing and anti-aging composition, which comprises the following components in parts by weight: 0.5-5.0 parts of conopeptide solution, 0.001-0.02 parts of lycopene, 0.02-0.05 parts of sodium hyaluronate and 0.01-0.2 parts of hydrolyzed collagen; the molecular weight of the sodium hyaluronate is not more than 10000Da.
Preferably, the lycopene is 0.005-0.02 parts by weight.
Preferably, the weight part of the hydrolyzed collagen is 0.1 to 0.2 part.
Preferably, the content of conopeptide in the conopeptide solution is 25+ -2 ppm.
The skin aging mechanism is as follows: skin aging is a prominent manifestation of aging of the whole organism, wherein the skin aging of the face is particularly obvious, the number of fibroblasts in the human body is gradually reduced, the secreted collagen and elastin are gradually reduced and broken due to oxidization, the supporting effect of the epidermis is eliminated, uneven collapse is caused, the dermis layer begins to be thinned, the skin elasticity is poor, and wrinkles begin to be gradually generated. Therefore, effective scavenging of free radicals and promotion of collagen synthesis or effective replenishment of collagen are important pathways for anti-wrinkle. Lycopene is a carotenoid existing in vegetable food, has 11 conjugated double bonds and 2 non-conjugated double bonds in molecular structure, and has strong antioxidant activity. In the invention, lycopene can quench free radicals in epidermal cells, and has obvious fading effect on senile plaques; lycopene also has effects in reducing skin injury caused by radiation or Ultraviolet (UV), and can combine with free radicals generated by UV to protect skin tissue from damage when UV irradiates skin. Lycopene can remove free radicals, reduce natural loss of human fibroblasts, promote accumulation of collagen, and thereby improve skin firmness and elasticity. Conotoxin (concoxin) is a mixed toxin secreted by the venom duct and the poison gland on the inner wall of the poison sac of the marine gastropoda mollusc Conus, and consists of a plurality of single toxin peptides, and the main components are active polypeptide compounds with high specificity to different ion channels and nerve receptors. Compared with other natural peptide toxins, the conotoxin has the outstanding advantages of small relative molecular mass, stable structure, high activity, high selectivity, easy synthesis and the like. The conopeptide is simulated natural mu-conotoxin, and has the advantages of high folding structure and strong permeability. The conopeptide has the effect of strongly inhibiting muscle tension, and achieves the effect of quickly smoothing wrinkles. The small molecular sodium hyaluronate is a moisturizing active substance which has excellent moisturizing performance and is easy to be absorbed by skin, and the moisturizing active substance acts on epidermal cells through the ways of smoothing, blocking, absorbing water, actively stimulating the epidermal process, strengthening the epidermal barrier function and the like. Ideal hydration can make the skin more shiny, smooth, comfortable and shiny. Properly increasing the hydration degree of the skin also contributes to the absorption of collagen by the skin, thereby reducing the generation of skin wrinkles.

Table 3 the amounts (mass%) of the wrinkle-removing and anti-aging compositions in examples 1 to 20 and comparative examples 1 to 6 were added
Performance test:
(1) Fibroblast proliferation promoting rate
The fibroblast proliferation promotion rate of the wrinkle-removing anti-aging compositions in examples 1-20 and comparative examples 1-6 were tested according to the methods described in XTT colorimetry for detecting human fibroblast proliferation (Shen Yan, liu Xilin, tang Yi, zhong Cancan, liang Peigong, li Siming. Biochemical and biophysical advances, 2002, (04): 659-662). The specific test method comprises the following steps: the absorbance was measured at 450nm on a microplate reader with reference wavelength of 655nm, and the acceleration was calculated. Acceleration rate 1 = (sample)The results of the promotion rate of the wrinkle-removing anti-aging composition samples of examples 1 to 20 and comparative examples 1 to 6 on the proliferation of fibroblasts calculated according to the formula of group absorbance-blank absorbance)/blank absorbance x 100% are shown in table 4.
TABLE 4 fibroblast proliferation promoting rate
As is clear from Table 4, the wrinkle-removing and anti-aging compositions in examples 1 to 20 had a rate of promotion of proliferation of fibroblasts of 65.2 to 92.9%, which is significantly higher than that of the wrinkle-removing and anti-aging compositions in comparative examples 1 to 16 (35.5 to 57.1%), indicating that the wrinkle-removing and anti-aging compositions of the present invention have a significant proliferation promoting effect on fibroblasts. As is clear from comparative examples 2 and comparative examples 1 to 16, comparative examples 1 to 4 omit any one of the components of the wrinkle-removing and anti-aging composition of example 2, and their promotion effect on fibroblast proliferation is significantly reduced, and comparative examples 5 to 14 omit any 2 or even 3 of the components of the wrinkle-removing and anti-aging composition of example 2, and their promotion effect on fibroblast proliferation is significantly reduced. In example 2 of the present invention, sodium hyaluronate was used, and the effect of promoting fibroblast proliferation was remarkably improved as compared with comparative examples 15 to 16. Therefore, the lycopene, the sodium hyaluronate (small molecule), the hydrolyzed collagen and the conopeptide in the wrinkle-removing and anti-aging composition have synergistic effect, and the wrinkle-removing and anti-aging composition obviously reduces the promotion effect of the wrinkle-removing and anti-aging composition on fibroblast proliferation due to the lack of any component.
(2) Collagen synthesis promotion rate
Examples 1 to 20 and comparative examples were tested respectively according to the following methods1-6, the specific test method is as follows: human fibroblasts cultured in DMEM medium containing 10% fbs were seeded in 24-well plates, and after cell attachment, medium exchange was performed with DMEM medium containing 0.25% fbs and 250 μmol/L magnesium ascorbyl phosphate, and the wrinkle-removing anti-aging compositions of examples 1 to 20 and comparative examples 1 to 6 were added, respectively. After three days of culture, the culture supernatant was recovered, centrifuged, and the type III collagen in the supernatant was measured. The biosynthesis ability of type III collagen in cells was evaluated by measuring the content of terminal peptide (abbreviated as P III P) of type III collagen secreted in the supernatant using "RIA-gnost P III P (P III P) measuring kit". The strength of the moisturizing effect of these samples was evaluated by the synthesis acceleration rate of type iii collagen. The calculation formula is as follows: acceleration rate 2 = (a-B)/b×100%, where a: type III collagen production after adding the test sample; b: the results of the collagen synthesis acceleration rate of the samples of the wrinkle-removing and anti-aging compositions of examples 1 to 20 and comparative examples 1 to 6, which were calculated according to the formula, without adding the test sample, are shown in Table 5.
TABLE 5 promotion of collagen III synthesis
As is clear from Table 5, the wrinkle-removing and anti-aging compositions in examples 1 to 20 had a synthesis promotion rate of 70.7 to 96.0% for type III collagen, which is significantly higher than that of the wrinkle-removing and anti-aging compositions in comparative examples 1 to 16 (37.6 to 56%), indicating that the wrinkle-removing and anti-aging composition of the present invention has a significant promotion effect on type III collagen synthesis. As is clear from comparative examples 2 and comparative examples 1 to 16, comparative examples 1 to 4 omit any one of the components in the wrinkle-removing and anti-aging composition of example 2, and their promotion effect on the synthesis of type iii collagen is significantly reduced, and comparative examples 5 to 14 omit any 2 or even 3 of the components in the wrinkle-removing and anti-aging composition of example 2, and their promotion effect on the synthesis of type iii collagen is significantly reduced. Compared with comparative examples 15 to 16, the small molecule sodium hyaluronate used in example 2 of the present invention has significantly improved effect of promoting the synthesis of type III collagen. Therefore, the lycopene, the sodium hyaluronate (small molecule), the hydrolyzed collagen and the conopeptide in the wrinkle-removing and anti-aging composition have synergistic effect, and the wrinkle-removing and anti-aging composition obviously reduces the promoting effect of the wrinkle-removing and anti-aging composition on the synthesis of the III type collagen due to the lack of any component.
In conclusion, lycopene, sodium hyaluronate (small molecule), hydrolyzed collagen and conopeptide in the wrinkle-removing and anti-aging composition have synergistic effects, and the lack of any one, two or three components can obviously reduce the promotion effects on fibroblast proliferation and III type collagen synthesis.
(3) Consumer test evaluation
Selecting 54 volunteers, one group of two persons, each group of persons using the essence prepared in the corresponding application example 1 in table 6, and the subjects used 1 time a day for 14 days according to the product usage method; the skin state of the sample after use is evaluated by a questionnaire mode, and the use effect of the product is analyzed.
The main dimensions for collecting feedback include: skin elasticity, skin tightening sense, fine lines reduction and wrinkle lightening effect. The questionnaire firstly collects preliminary conclusions, and is divided into effective, general effect and ineffective. In a scoring manner, 1-10 points, the higher the score, the better the effect, the 1-3 points were regarded as no effect, 4-6 points were regarded as general effect, more than 6 points represent subjects approving the effect, and scores were recorded, the average score for each group of people is shown in table 6.
TABLE 6 results of tightening efficacy analysis of essence
/>
From table 6 above, the essence added with the wrinkle-removing and anti-aging composition in the embodiment of the invention has obvious wrinkle-removing and anti-aging effects, the skin elasticity improvement evaluation is 6.5 to 10 minutes, the skin tightening sense improvement evaluation is 7 to 10 minutes, the fine line reduction evaluation is 6.5 to 10 minutes, the fine line effect evaluation is 6.5 to 10 minutes, and the result of the tightening efficacy score of the essence added with the wrinkle-removing and anti-aging composition in the comparative examples 1 to 14 is higher than that of the essence added with the wrinkle-removing and anti-aging composition in the comparative examples 1 to 14, so that the wrinkle-removing and anti-aging composition has obvious synergistic effects among the components. In addition, by comparing the essence samples No. 1, no. 2 and No. 5, the essence samples No. 6 and No. 10, the essence samples No. 11 and No. 14 and the essence samples No. 15 and No. 18, the anti-wrinkling and anti-aging effects of the essence can be obviously improved by increasing the content of any one of the components of lycopene, conopeptide, sodium hyaluronate and hydrolyzed collagen. The No. 19 and No. 20 essence samples have the best wrinkle-removing and anti-aging effects, and the scores of the skin elasticity improvement evaluation, the skin tightening sense improvement evaluation, the fine grain reduction evaluation and the fine grain effect evaluation all reach 9.5-10 minutes. The firmness of the No. 35 and No. 36 essence samples is obvious, and probably because the sodium hyaluronate with a certain film forming property is adopted in the No. 35 and No. 36 essence samples, when the sodium hyaluronate with a certain film forming property is not absorbed on the skin, a certain tensile force is generated on the skin, so that the firmness is generated, the firmness is only subjective and is not the actual firmness of the skin, and the molecular weight of the sodium hyaluronate in the No. 36 essence samples is higher, so the firmness is obviously improved, but is different from the firmness after the collagen content of the skin is increased. The wrinkle-removing and anti-aging composition provided by the invention can promote the synthesis of collagen or promote the absorption of collagen under the condition that the composition is fully absorbed by skin so as to achieve the effect of tightening the skin, and the effect can be represented by the promotion effect data of fibroblast proliferation and III type collagen synthesis.
(4) Skin elasticity test
84 volunteers were selected, and the mask liquid in application example 3, to which examples 1,2, 3, 5, 6, 10, 11, 14, 15, 18, 19, 20 and comparative examples 1 to 16 were added, was used in combination with a common fibrous membrane cloth, and as a mask, a sheet of mask was used 3 times per week for 15 to 20 minutes each time. The skin of the cheekbones and the corners of the eyes of the face was taken as the test site. According to the group standard T/ZHCA 005-2019 'cosmetic influencing skin elasticity test method', the skin elasticity characterization values R of 14d and 28d before trial and after trial are respectively measured, wherein the larger the R value change rate (namely V) is, the better the skin elasticity improvement condition is, and the calculation formula of the characterization value R is as follows: r=u1/U2. Wherein U1 is rebound displacement (mm) of skin after negative pressure is eliminated, and U2 is maximum stretching displacement (mm) of skin when negative pressure is loaded. Rate of change v= (R 2 -R 1 /R 1 ) X 100%; wherein R is 1 Is the skin elasticity characterization value before trial, R 2 The skin elasticity characterization value after the test is shown in Table 7 as the average value of the test of the skin elasticity change rate of each group.
TABLE 7 skin elasticity change Rate
As is clear from table 7 above, the skin elasticity change rate after 28 days using the mask solution added with the wrinkle-removing and anti-aging composition in the examples of the present invention is 17.55 to 31.22%, which is higher than the skin elasticity change rate (3.76 to 9.22%) after 28 days using the mask solution added with the wrinkle-removing and anti-aging composition in the comparative examples 1 to 16, further showing that the components of the wrinkle-removing and anti-aging composition of the present invention have remarkable synergistic effects in increasing skin elasticity. In addition, it is known that the skin elasticity change rate after the use of the mask liquid can be remarkably improved by comparing the mask liquid samples added with the examples 1,2, 3 and 5, the mask liquid samples of the comparative examples 6 and 10, the mask liquid samples of the comparative examples 11 and 14, and the mask liquid samples of the comparative examples 15 and 18, and further, the synergistic effect between the components of the wrinkle-removing and anti-aging composition of the present invention is demonstrated. The results of the skin elasticity change rate tested by the instrument in the experiment are basically consistent with the results of the tightening efficacy analysis of the consumer self-evaluation.
(5) Skin wrinkle area test
The wrinkle-removing anti-aging eye creams of application example 2 to which the compositions of examples 1,2, 5, 6, 11, 15, 19, 20 and comparative examples 1 to 16 were added were used 1 time per day, and the left face was used as a blank sample (blank sample was prepared according to the cream base formulation in table 2 without the wrinkle-removing anti-aging compositions of examples and comparative examples) for 4 weeks; after 30min, 2 weeks and 4 weeks, respectively, the product was analyzed for its effect, expressed as skin wrinkle area reduction rate, which indicated that the greater the skin wrinkle area reduction rate, the better the effect. The test results calculated according to this formula are shown in table 8, with the skin wrinkle mask reduction rate = (total number of wrinkles before the specified area test-total number of wrinkles after the specified area test)/total number of wrinkles before the specified area test x 100%.
TABLE 8 skin wrinkle area reduction Rate/%
/>
As is clear from Table 8 above, the reduction rate of the skin wrinkle area after 4 weeks of use of the wrinkle-removing and anti-aging eye cream to which the composition of the example was added was 25.2 to 38.9%, which is significantly higher than the reduction rate of the skin wrinkle area after 4 weeks of use of the eye cream to which the composition of the comparative example was added (7.6 to 17.4%). The subjects produced a certain immediate effect after 30 minutes of using the eye creams added with the compositions of examples and comparative examples, wherein the reduction rate of the skin wrinkle area after using the eye creams added with the compositions of comparative examples 15 and 16 was higher than that of the eye creams added with the compositions of comparative examples 1 to 14, because: although the large and medium molecular sodium hyaluronate has good permeation effect without small molecular sodium hyaluronate, the sodium hyaluronate also has obvious effect due to the existence of a certain amount of hydrolyzed collagen, lycopene and conopeptide. From comparative examples 1,2 and 5, comparative examples 2 and 6, and comparative examples 5 and 15, it is clear that an increase in the content of lycopene, conopeptide, hydrolyzed collagen, and sodium hyaluronate increases the wrinkle-removing and anti-aging effects of the sample.
The lycopene in the invention is helpful for improving the antioxidation and anti-aging effects, can remove free radicals harmful to skin, and can block the nerve from giving out signals to the muscle, so that the muscle is kept calm, and the skin is prevented from forming wrinkles; meanwhile, the absorption of hydrolyzed collagen can improve the collagen content in the skin, so that the skin is effectively supported, and the generation of wrinkles is reduced; the improvement of the skin hydration degree also helps to improve the skin tension, thereby achieving the effect of smoothing fine wrinkles. The lycopene, conopeptide, hydrolyzed collagen and sodium hyaluronate are 4 components, and the lack of any wrinkle-removing and anti-aging effect is obviously reduced, so that each component in the wrinkle-removing and anti-aging composition has a synergistic effect. In addition, the invention adopts the small molecular sodium hyaluronate (molecular weight is less than 10000 Da), the permeation effect is better, the skin is easier to absorb, the wrinkle-removing and anti-aging effects are more obvious, and compared with the large and medium molecular weight sodium hyaluronate, the wrinkle-removing and anti-aging effects are obviously improved.
While the embodiments of the present invention have been described in detail, the present invention is not limited to the above embodiments, and various changes may be made without departing from the spirit of the present invention within the knowledge of those skilled in the art. Furthermore, embodiments of the invention and features of the embodiments may be combined with each other without conflict.
