Abstract
The invention discloses a high-efficiency skin tightening polypeptide composition and application of a product thereof, and relates to the technical field of cosmetics. The high-efficiency skin tightening polypeptide composition is compounded by utilizing different polypeptides, solves the problems of skin aging and relaxation and tightening skin in a multi-target way, and meets the functional requirements of consumers. The high-efficiency skin tightening polypeptide composition provided by the invention comprises at least one of dipeptide diamino Ding Xianbian-based amide diacetate, acetyl tetrapeptide-11, acetyl tetrapeptide-9, hexapeptide-9 and oligopeptide-1, and the prepared high-efficiency skin tightening polypeptide composition has good antiseptic effect, safety performance, anti-aging performance, ultraviolet ray stain performance improvement and pore performance improvement; in addition, the ethyl vanillin propylene glycol acetal is added into the high-efficiency skin tightening polypeptide composition, so that the prepared high-efficiency skin tightening polypeptide composition has better anti-corrosion effect, anti-aging performance, ultraviolet color spot performance improvement and pore performance improvement.

Technical Field
The invention belongs to the technical field of cosmetics, and particularly relates to a high-efficiency skin tightening polypeptide composition and application of a product thereof.
Background
In recent years, many cosmetics or skin care products are favored for their delicate skin and rejuvenation effects. Early days, many animal gland and serum extracts have obvious cosmetic and cosmetology effects, but are less and less advocated by people. Over the past few years, more knowledge of the structure and function of the skin has increased the ability of the cosmetic industry to determine whether a formulation is beneficial to the skin.
The skin is compact and elastic, and people can look much younger. The lack of elasticity is mainly due to the reduced synthesis of immobilized proteins and loss of collagen fibers, resulting in collapse of the dermis and epidermis interface, and the formation of fine wrinkles. Wrinkles around the forehead, eyes, orbit and lips of a human face are evidence of aging, which is also a major problem in skin tightening against aging. These fine lines can occur around the age of 30, mainly due to positional pressure and repetitive facial movements.
The peptides are important physiological regulators of human bodies, can comprehensively regulate physiological functions of the human bodies, excite new generation of cells and collagen, and effectively resist aging, but currently, the domestic applicable cosmetic peptides for tightening and anti-wrinkle are limited, only few products focus on tightening and pulling, the applicable choices are less, and the tightening and anti-aging of the compound combination is lack of efficient and target innovation peptide combination. According to the invention, multiple polypeptide compositions are precisely combined through multiple targets, so that the skin tightening and anti-aging effects are precisely achieved, the optimal skin tightening effect can be achieved, and the requirements of consumers are met.

the polypeptide is at least one of dipeptide diamino Ding Xianbian-base amide diacetate, acetyl tetrapeptide-11, acetyl tetrapeptide-9, hexapeptide-9 or oligopeptide-1;
the preservative comprises a p-hydroxyacetophenone derivative.
Dipeptide diamino Ding Xianbian-yl amide diacetate: a neurotransmitter-inhibiting polypeptide that mimics the activity of the snake venom Waglein I, binds to the epsilon subunit of nmAChR to block acetylcholine binding to receptors and to block neuronal excitation transmission.
Acetyl tetrapeptide-11: can effectively stimulate the generation of the Syndecan-1 and XVII collagen, support the cohesion between the cells of the Syndecan and the XVII collagen and the adhesion with DEJ, and is an active peptide specially aiming at epidermis, and the skin restores the cohesion, the resistance and the delicate skin texture.
Acetyl tetrapeptide-9: effectively stimulates the synthesis of the type I collagen and the basal lamina glycan, and participates in the formation and the diameter growth of collagen fibers. It can make collagen exert good function and effectively delay skin aging and relaxation.
Hexapeptide-9: has the same sequence as human collagen IV and XVII, and has multiple repair promoting effects on epidermis, dermis and DEJ.
Oligopeptide-1: oligopeptide-1 is a synthetic peptide consisting of three amino acids glycine, histidine and lysine. Promoting epidermis regeneration, stimulating skin collagen synthesis, and repairing skin barrier.
Specifically, the preparation method of the high-efficiency skin tightening polypeptide composition comprises the following steps: adding polypeptide into deionized water, stirring, mixing, adding antiseptic, and stirring to obtain high-efficiency skin tightening polypeptide composition.
The invention aims to provide a polypeptide high-efficiency skin tightening polypeptide composition and a product application thereof, which utilize different polypeptide compounding to realize multi-target solution of skin aging relaxation and skin tightening, and meet the functional requirements of consumers. HAS3: hyaluronan synthase 3; the addition of GlcNAc or glcna monosaccharides to nascent hyaluronic acid polymers is catalyzed. Therefore, it is essential for the synthesis of hyaluronic acid, which is a major component of most extracellular matrices, has a structural role in tissue structure, and regulates cell adhesion, migration and differentiation. This is one of the isozymes catalyzing the reaction (by similarity); belonging to the NodC/HAS family.
COL4A1: collagen alpha-1 (IV) chain; type IV collagen is the major structural component of Glomerular Basement Membrane (GBM), forming a "chicken wire" network with laminin, proteoglycans, and entactin/entactin.
ITGA1: integrin alpha-1; integrin alpha-1/beta-1 is a receptor for laminin and collagen. It recognizes the proline hydroxylation sequence G-F-P-G-E-R in collagen. Anchorage-dependent down-regulation of EGF-stimulated cell growth is involved.
Preferably, the above polypeptides are used in an amount of 0.01 to 0.1wt% in the high-potency skin tightening polypeptide composition.
Preferably, the preservative is present in the high-potency skin tightening polypeptide composition in an amount of 0.5-1.2wt%.
Preferably, the preservative further comprises 1,2 hexanediol.
Preferably, the mass ratio of the p-hydroxyacetophenone derivative to 1,2 hexanediol is as follows: 1:1-1.5.
Preferably, the p-hydroxyacetophenone derivative comprises a product of sulfathiazole modification of alpha-bromo-p-hydroxyacetophenone.
The invention also discloses a preparation method of the p-hydroxyacetophenone derivative, which comprises the following steps: the alpha-bromo-p-hydroxyacetophenone and sulfathiazole are adopted to carry out substitution reaction to prepare the p-hydroxyacetophenone derivative.
The invention provides a preparation method of a p-hydroxyacetophenone derivative, which is characterized in that the p-hydroxyacetophenone derivative prepared by substitution reaction of sulfathiazole and alpha-bromo-p-hydroxyacetophenone is used for preparing a high-efficiency skin tightening polypeptide composition, so that the prepared high-efficiency skin tightening polypeptide composition has good anti-corrosion effect, safety performance, anti-aging performance, ultraviolet ray color spot performance improvement and pore performance improvement.
Specifically, the preparation method of the p-hydroxyacetophenone derivative comprises the following steps:
adding sulfathiazole (the mass volume ratio of sulfathiazole to tetrahydrofuran is 1g:25-40 mL) into tetrahydrofuran, stirring and mixing uniformly, adding sodium carbonate, stirring and mixing for 5-10min, then slowly adding a tetrahydrofuran solution of alpha-bromo-p-hydroxyacetophenone with the concentration of 0.45-0.55g/mL, judging the end of the reaction by TLC, filtering, and separating by column chromatography to obtain the p-hydroxyacetophenone derivative.
Preferably, the molar ratio of the sulfathiazole to the sodium carbonate is as follows: 1:0.8-1.5.
Preferably, the molar ratio of the alpha-bromo-hydroxyacetophenone to the sulfathiazole is as follows: 1:0.8-1.3.
The invention also discloses application of the high-efficiency skin tightening polypeptide composition in preparing cosmetics.
The invention also discloses application of the p-hydroxyacetophenone derivative prepared by the preparation method in preparation of cosmetics.
To further enhance the performance of the high potency skin tightening polypeptide composition, the present invention also incorporates ethyl vanillin propylene glycol acetal into the composition.
The invention also discloses a preparation method of the high-efficiency skin tightening polypeptide composition, wherein the ethyl vanillin propylene glycol acetal is added into the high-efficiency skin tightening polypeptide composition, and the ethyl vanillin propylene glycol acetal and the preservative are added simultaneously, so that the prepared high-efficiency skin tightening polypeptide composition has better anti-corrosion effect, anti-aging performance, ultraviolet color spot performance improvement and pore performance improvement.
Preferably, in the high-efficiency skin tightening polypeptide composition, the mass ratio of the polypeptide to the ethyl vanillin propylene glycol acetal is as follows: 1:0.3-0.8.
The invention also discloses application of the ethyl vanillin propylene glycol acetal in preparing a high-efficiency skin tightening polypeptide composition.
The invention also discloses application of the ethyl vanillin propylene glycol acetal in preparing cosmetics.

The sample was analyzed and tested by potassium bromide tabletting.
The above test was performed on the p-hydroxyacetophenone derivative and p-hydroxyacetophenone prepared in example 1, and the results are shown in FIG. 1. As can be seen from FIG. 1, the infrared spectrum of the p-hydroxyacetophenone derivative is 1493cm compared with the infrared spectrum of the p-hydroxyacetophenone -1 、1335cm -1 The existence of an infrared characteristic absorption peak of thiazole ring indicates that sulfathiazole participates in the generation reaction of the p-hydroxyacetophenone derivative.
Test example 2:
security test
The method comprises the steps of performing skin irritation test on New Zealand rabbits, dividing the rabbits into 12 groups, 4 rabbits in each group, performing dehairing treatment on the left side and the right side of each rabbit vertebra, coating 0.5g of high-efficiency skin tightening polypeptide composition sample on the dehairing area on the left side, covering with gauze and cellophane, fixing with adhesive tape, wherein the adhesive tape has no irritation, coating no sample on the right side, covering with gauze and cellophane, fixing with adhesive tape, taking as a control, sealing for 4 hours, removing the gauze, the cellophane and the adhesive tape, and flushing with warm water to remove the sample; continuous experiment 14d was performed 1 time a day, and dehairing treatment was performed before each experiment, and red spots and edema of the experimental part of the rabbit after each experiment were recorded and scored. The scoring criteria are shown in table 1.

The above test was performed on the high-potency skin tightening polypeptide composition samples prepared in example 1-example 12, and the results are shown in Table 2. As shown in Table 2, the high-efficiency skin tightening polypeptide composition prepared by the invention is safe and has no stimulation to rabbits.
Test example 3:
anti-aging Performance test
120 female volunteers with dark and dry face, dry and rough eyes and skin ages between 30 and 60 years are selected and divided into 12 groups of 10 people, the selected volunteers do not have any history of skin and systemic diseases, the volunteers do not use immunosuppressants and hormone medicines in nearly one month, the volunteers are fixedly smeared with high-efficiency skin tightening polypeptide composition samples on half faces every day, the smearing amount is 1g each time, the volunteers are used once in the morning and evening, the use is continued for 42 days, the volunteers cannot use other skin care products during the period, and then the detection results are expressed in percentage values through a VISIA professional facial image analyzer.

The above test was performed on the high-potency skin tightening polypeptide composition samples prepared in example 1-example 12, and the results are shown in Table 3. As can be seen from table 3, the improvement rate of wrinkles is improved in example 1 compared with example 10 and example 11 compared with example 12, which indicates that the p-hydroxyacetophenone derivative prepared from sulfathiazole is used for preparing the high-efficiency skin tightening polypeptide composition, so that the high-efficiency skin tightening polypeptide composition has good anti-aging performance; example 11 significantly improves wrinkle improvement compared to example 1, example 12 compared to example 10, demonstrating that the use of ethylvanillin propylene glycol acetal for the preparation of a high-efficiency skin tightening polypeptide composition results in a high-efficiency skin tightening polypeptide composition having good anti-aging properties; example 1 showed an improvement in wrinkle improvement rate as compared with examples 2 to 9, indicating that dipeptide diamino Ding Xianbian-based amide diacetate, acetyl tetrapeptide-11, acetyl tetrapeptide-9, hexapeptide-9 and oligopeptide-1 had a synergistic effect in anti-aging.
Test example 4:

The test method was the same as in test example 3.

The above test was performed on the high-potency skin tightening polypeptide composition samples prepared in example 1-example 12, and the results are shown in Table 4. As can be seen from table 4, the improvement rate of ultraviolet spots is improved in example 1 compared with example 10 and example 11 compared with example 12, which indicates that the p-hydroxyacetophenone derivative prepared from sulfathiazole is used for preparing the high-efficiency skin tightening polypeptide composition, so that the high-efficiency skin tightening polypeptide composition has good improvement performance of ultraviolet spots; compared with example 1 and example 12 and example 10, the ultraviolet stain improvement rate is obviously improved, which indicates that when the ethyl vanillin propylene glycol acetal is used for preparing the high-efficiency skin tightening polypeptide composition, the high-efficiency skin tightening polypeptide composition has good ultraviolet stain improvement performance; example 1 showed an improvement in the improvement rate of ultraviolet ray spots as compared with examples 2 to 9, and demonstrated that the dipeptide diamino Ding Xianbian-based amide diacetate, acetyl tetrapeptide-11, acetyl tetrapeptide-9, hexapeptide-9 and oligopeptide-1 had a synergistic effect in improving ultraviolet ray spots.
Test example 5:

The above test was performed on the high-potency skin tightening polypeptide composition samples prepared in example 1-example 12, and the results are shown in Table 5. As can be seen from table 5, the pore improvement rate of example 1 is improved compared with example 10 and example 11 compared with example 12, which indicates that the p-hydroxyacetophenone derivative prepared from sulfathiazole is used for preparing the high-efficiency skin tightening polypeptide composition, so that the high-efficiency skin tightening polypeptide composition has good pore improvement performance; compared with example 1 and example 12 and example 10, the pore improvement rate is obviously improved, which shows that when the ethyl vanillin propylene glycol acetal is used for preparing the high-efficiency skin tightening polypeptide composition, the high-efficiency skin tightening polypeptide composition has good pore improvement performance; example 1 showed an improvement in pore improvement over examples 2 to 9, indicating that dipeptide diamino Ding Xianbian-based amide diacetate, acetyl tetrapeptide-11, acetyl tetrapeptide-9, hexapeptide-9 and oligopeptide-1 had a synergistic effect in pore improvement.
Test example 6:
corrosion resistance test
Adding 20g of high-efficiency skin tightening polypeptide composition sample into 0.2g of staphylococcus aureus bacterial suspension, stirring and mixing uniformly, and inoculating staphylococcus aureusIs 5X 10 in initial concentration 6 CFU/mL, after 7d incubation at 25℃the microorganism concentration was measured by plate counting.

The above test was performed on the high-potency skin tightening polypeptide composition samples prepared in example 1 and examples 10-12, and the results are shown in Table 6. As can be seen from Table 6, the total number of colonies is significantly reduced in example 1 compared with example 10 and example 11 compared with example 12, which indicates that the p-hydroxyacetophenone derivative prepared from sulfathiazole has good antiseptic effect when used for preparing the high-efficiency skin tightening polypeptide composition; the decrease in colony count in example 11 compared with example 1 and example 12 compared with example 10 suggests that the addition of ethyl vanillin propylene glycol acetal contributes to the improvement of the preservative effect of the high-efficiency skin tightening polypeptide composition.
The conventional technology in the above embodiments is known to those skilled in the art, and thus is not described in detail herein.

Claims (9)
1. A high-potency skin tightening polypeptide composition, characterized in that:
the high-potency skin tightening polypeptide composition comprises a polypeptide and a preservative;
the polypeptide is at least one of dipeptide diamino Ding Xianbian-base amide diacetate, acetyl tetrapeptide-11, acetyl tetrapeptide-9, hexapeptide-9 or oligopeptide-1;
the preservative comprises a p-hydroxyacetophenone derivative.
2. A high potency skin tightening polypeptide composition according to claim 1, characterized in that: the dosage of the polypeptide in the high-efficiency skin tightening polypeptide composition is 0.01-0.1wt%.
3. A high potency skin tightening polypeptide composition according to claim 1, characterized in that: the preservative is used in an amount of 0.5 to 1.2wt% of the high-potency skin tightening polypeptide composition.
4. A high potency skin tightening polypeptide composition according to claim 1, characterized in that: the preservative further comprises 1,2 hexanediol.
5. The high potency skin tightening polypeptide composition according to claim 4, wherein: the mass ratio of the p-hydroxyacetophenone derivative to the 1,2 hexanediol is as follows: 1:1-1.5.
6. A high potency skin tightening polypeptide composition according to claim 1, characterized in that: the p-hydroxyacetophenone derivative comprises a product of sulfathiazole modified alpha-bromo-p-hydroxyacetophenone.
7. The high potency skin tightening polypeptide composition according to claim 6, wherein: the molar ratio of the alpha-bromo-hydroxyacetophenone to the sulfathiazole is as follows: 1:0.8-1.3.
8. Use of the high-potency skin tightening polypeptide composition as claimed in any one of claims 1-5 in the preparation of a cosmetic.
9. Use of a p-hydroxyacetophenone derivative according to any one of claims 6 to 7 for the preparation of cosmetics.
