Description
Technical Field
The invention relates to a composition and essence milk with blue light pollution resistance and repair effects, and also relates to a preparation method of the essence milk, belonging to the technical field of cosmetics.
Background
The 'blue light' is commonly called 'high-energy short-wave blue light', and is light with a wavelength of 400-480 nm and relatively high energy. Blue light pollution refers to exposure to artificial blue light of "hyperlinked lifestyle", which is mainly from computers, projectors, movie screens, LED displays, mobile phones, laser pens, fluorescent lamps, outdoor light boxes, LED lights and bathroom heaters. Blue light not only affects the secretion of melatonin, but also has a significant inhibitory effect on melatonin, which is a hormone assisting sleep. People not only disturb the normal biological clock of the human body when contacting blue light for a long time or before sleeping, but also cause a series of health problems such as insomnia and the like caused by the reduction of melatonin in the human body. In daily life, people are exposed to blue light more and more frequently, so that skin is excessively exposed to the blue light, and the self-barrier repair capacity of the skin is reduced, and aging is accelerated. For example, exposure of the skin to visible light or high-energy visible light KEV causes skin cells to exert a bad set of effects: increased reactive oxygen species, mitochondrial DNA damage, delayed epidermal barrier repair, increased inflammatory cascade, carotenoid depletion in the dermis, langerhans cell immunosuppression. Therefore, just as well as protection against UV or air pollution, it is also necessary to protect the skin against blue light pollution, which helps to slow down premature aging and damage to the skin.
The chlorella extract is obtained from chlorella rich in various nutrients such as protein, lipid, carbohydrate, vitamins, minerals, etc. Chlorella is belonging to Chlorella, Chlorophyceae, and contains biologically active substances with effect of accelerating growth of bacteria, animals and plants. The chlorella extract mainly contains chlorella growth factor which contains abundant nucleic acid and nucleoprotein, has the functions of repairing human body cells, resisting aging and activating immunocytes, has strong binding force with toxins and can expel the toxins out of a body. Therefore, the chlorella growth factor has good effect on the skin.
Crambe extract is derived from the robust saline-soil plant crambe which is living in special ecological environments. Contains various vitamins, phytosterol, sulfat and carotenoid, and has effects of stimulating renewal of skin basal layer cells, protecting stem cells from damage of UVB, stimulating synthesis of ceramide II, and reducing degradation of collagen and GAC caused by UVA, thereby having antiaging, free radical scavenging, and photoaging relieving effects.
The Zingiber officinale root extract is a unique extract from the root of Iris sieboldii Madagasca, is rich in saccharides, has anti-blue light and anti-UVA effects by regulating autophagy, and has anti-oxidation, anti-pollution and cell detoxification effects by protecting the mitochondrial network and energy metabolism of UVA changes.
The Ulmus pumila extract is extracted from root bark of Ulmus pumila, and is rich in natural polysaccharides such as galactose, rhamnose, glucose, glucosamine, and trehalose. The Ulmus pumila extract can inhibit hyaluronidase activity and reduce transdermal water loss (TEWL), and is suitable for strong moisturizing and antiaging formulation. Meanwhile, the composition can inhibit the release of IL-8, IL-6 and PEG2 (prostaglandin), has good anti-inflammatory effect, and can repair photodamage caused by UVA irradiation and improve the survival capability of cells.
With the pursuit of beauty, the medical and beauty industry is developed vigorously, and more people who love beauty receive medical beauty treatment. However, the skin after medical beauty is very fragile, and the people cannot avoid the invasion of blue light. Therefore, a skin care product which can resist blue light and has a repairing effect is urgently needed, the types of cosmetics in the market are various, most of essence products mainly have whitening, wrinkle resistance, oxidation resistance and the like, and the skin care product not only has a single function effect, but also has a poor effect. The invention provides the essence milk with the main functional components including the chlorella extract, the crambe extract, the ginger flower root extract and the elm extract, which not only can resist blue light pollution, but also has the function of repairing efficacy, and is beneficial to improving the adverse effect of blue light on the skin of people.

Product evaluation
Firstly, evaluating the safety of products
The blue light-resistant repair essence milks of examples 1 to 5 and comparative examples 1 to 5 were evaluated for the irritation of cosmetics.
The tested population is female, 90 people and between 16 and 65 years of age. The skin of the subject is healthy, has no allergic history of the skin disease, and meets the voluntary entry standard of the subject. Randomly grouped into 5 groups of 18 people each.
The test method is a human body patch test. Selecting a qualified spot tester, placing 0.020-0.025g of a tested object in the spot tester, sticking the spot tester with the tested object on the back or the bent side of the forearm of the tested object by using a non-irritating adhesive tape, and lightly pressing with a palm to uniformly stick the spot tester on the skin for 24 hours. And (5) removing the tested spot tester for 30min, and observing skin reaction after the indentation disappears. If the result is negative, the test is observed once more at 24h and 48h after the patch test. The results were recorded according to the reference standards of "standards for hygiene of cosmetics" in 2007 and "standards for safety of cosmetics" in 2015.
Evaluation criteria: level 0: negative reaction; level 1: suspicious reactions, only faint erythema; and 2, stage: weak positive reaction, erythema, infiltration, edema, and possibly pimple; and 3, level: strong positive reaction, erythema, infiltration, edema, pimple and reaction beyond the tested area; 4, level: very strong positive reaction, obvious erythema, severe infiltration, edema, herpes fusiformis and reaction beyond the tested area.
The results of the patch test on the skin of a human body show that: the skin reactions of all the test subjects are observed 24h and 48h after the test by the patch test of the essence milk of the examples 1-5 and the comparative examples 1-5, and the skin reactions of all the test subjects are negative, so that the blue light resistant repair essence milk prepared by the invention is safe to use.
Second, product functionality evaluation
1. Anti-blue light performance detection
Blue light resistance performance detection is carried out on the blue light resistance repair essence emulsions of examples 1-5 and comparative examples 1-5
The essence milks of examples 1 to 5 and comparative examples 1 to 5 were dissolved in dimethyl sulfoxide, respectively, to form corresponding sample solutions. A certain amount of Human Epidermal Keratinocytes (HEK) and Human Dermal Fibroblasts (HDF) were cultured in 96-microwell plates, the number of cells in each microwell plate being the same. The 96-well plates of cultured cells were covered with flat-bottom sterilized plates containing the sample solutions of examples 1-5 and comparative examples 1-5, and LED (476nm, 1900lux) blue light irradiation was performed for 9 hours above the sterilized plates at a distance of 30cm, followed by cell viability assay with CellTiter96(MTS), each treatment being repeated three times.
The detection method comprises the following steps: the degree of damage of blue light to Human Epidermal Keratinocytes (HEK) and Human Dermal Fibroblasts (HDF) under the serum protective conditions of examples 1 to 5 and comparative examples 1 to 5 was investigated by measuring the survival rate and proliferation rate of metabolically active cells using CellTiter96(MTS), and the results are shown in table 1.
Blank control: cells were not exposed to blue light at room temperature (37 ℃).
Table 1 blue light resistance test of each group of essence milks
Sample (I) | Human epidermal keratinocyte activity (%) | Human dermal fibroblast Activity (%) |
Example 1 | 86 | 88 |
Example 2 | 92 | 93 |
Example 3 | 98 | 99 |
Example 4 | 98 | 99 |
Example 5 | 98 | 98 |
Comparative example 1 | 62 | 59 |
Comparative example 2 | 58 | 57 |
Comparative example 3 | 59 | 58 |
Comparative example 4 | 51 | 54 |
Comparative example 5 | 23 | 21 |
Blank control | 100 | 100 |
From the results in table 1, after the LED (476nm, 1900lux) blue light is irradiated for 9 hours, the activity of human epidermal keratinocytes and the activity of human dermal fibroblasts of examples 1 to 5 are both higher than those of comparative examples 1 to 5, which indicates that the capability of the essence milk of examples 1 to 5 for resisting blue light is higher than that of comparative examples 1 to 5, and further indicates that the efficacy composition added in the essence milk of examples 1 to 5 has stronger blue light filtering capability and obvious synergistic effect, and can block the damage of blue light to the skin, therefore, the essence milk formula of examples 1 to 5 is better than that of comparative examples 1 to 5; wherein, the human epidermal keratinocyte activity and the human dermal fibroblast activity of the examples 4 and 3 are higher than those of the examples 1, 2 and 5, which shows that the weight ratio of the chlorella extract, the crambe extract, the ginger flower root extract and the ulmus pumila extract is optimal in the efficacy composition of the examples 3 and 4.
2. Inhibiting cellular reactive oxygen species production
Cell-inhibiting active oxygen production test was performed on the blue-light-resistant repair essence milks of examples 1 to 5 and comparative examples 1 to 5
The essence milks of examples 1 to 5 and comparative examples 1 to 5 were dissolved in dimethyl sulfoxide, respectively, to form corresponding sample solutions. 100 healthy mice with the weight of 20-30g are selected and randomly divided into 10 groups, each group comprises 10 mice, and 5 test groups and 5 control groups are arranged. The test samples of examples 1-5 and comparative examples 1-5 were applied to the exposed skin of the above 10 groups of mice one by one under the condition of 425nm of LED blue light (3mW/cm2 for 18min), wherein, the mice of the groups of the test groups of examples 1-5 and the mice of the groups of the control groups of comparative examples 1-5 were applied at a dose of 1ml/cm, and after 24h, the results were quantitatively analyzed by CellROX fluorescence intensity staining and Volocity image software, the higher the value of the cellular active oxygen indicates the poorer the blue light resistance of the test sample, and the cellular active oxygen changes of the mice were observed at 7 th, 14 th, 21 th and 30 th days after the treatment within 1 month, respectively, and the results are shown in Table 2.
TABLE 2 Effect of each group of essence milks on mouse skin cell reactive oxygen species
The results in table 2 show that the levels of reactive oxygen species in skin cells of mice are equivalent before the application of the essence milks of examples 1 to 5 and comparative examples 1 to 5, that is, on day 0, but after the application of each group of essence milks, the corresponding levels of reactive oxygen species in skin cells of mice are significantly lower than those of comparative examples 1 to 5, which indicates that the blue light pollution resistance repair essence milks of examples 1 to 5 can effectively inhibit the increase of the levels of reactive oxygen species in skin cells of mice under blue light pressure, and the effect is better than that of 1 to 5 groups with a large proportion, wherein the inhibition effect of example 3 is the best, so as to further illustrate that the blue light pollution resistance repair essence milks of examples 1 to 5 of the present invention have reasonable and obvious synergy components and have better blue light resistance.
3. Evaluation of repair Effect
Evaluation of repair Effect on blue light-resistant repair essence milks of examples 1 to 5 and comparative examples 1 to 5
The repairing effect evaluation test is carried out on 60 volunteers, namely 30 men and 30 women, which are 25-45 years old and have different degrees of damage to facial skin (such as dry skin, desquamation, pruritus and the like). The test time was 28 days, and each group was divided into 3 test groups and 3 control groups. During the test, each volunteer of 10 test groups was applied with the essence milks prepared in examples 1 to 5 and comparative examples 1 to 5, respectively, once a day in the morning and at the evening after washing the face, and each volunteer of the control group was not applied with any other cosmetics. The test and control groups were normally exposed to the LED blue light environment for 5 hours per day and evaluated 28 days after testing.
Evaluation criteria: more than 80% of the skin damage repairs were effective, and the evaluation results are shown in table 3.
TABLE 3 evaluation of the repair Effect of the essential milks of each group
The test results in table 3 show that the effective rate of skin repair of each volunteer in the test group is higher than that of the control group, and the skin damage repair of examples 2 to 5 is more than 80%, wherein the effective rate of skin repair of examples 3 to 5 reaches 90%, and the skin damage repair of comparative examples 1 to 5 is less than 80%, which indicates that the blue light resistant repair essence emulsion of the present invention has a good repair effect, and the repair effect is the best with the formulas of example 3, example 4 and example 5.
In conclusion, the essence milk provided by the invention can resist blue light pollution, has a repairing effect, integrates product functionality evaluation, and takes the formula of the embodiment 3 as the optimal essence milk formula of the invention.
