Abstract
The invention discloses a light protection and repair composition, a preparation method and application thereof, and a skin care product. The composition has the advantages that the synergistic effect of the villus villosus extract and at least two of the three light protection extracts is adopted, so that the protection range of the composition can cover the wavelength regions of ultraviolet light, blue light and red light, a large amount of active DPPH free radicals generated under the induction of light can be eliminated, and the resistance to external harmful light is improved by improving the resistance and the repairing capacity of the skin; meanwhile, mitochondrial DNA can be repaired, and ultraviolet light, blue light and infrared light can be comprehensively protected. Description
Technical Field
The invention relates to the technical field of cosmetics, and particularly relates to a light protection and repair composition, a preparation method and application thereof, and a skin care product.
Background
The light pollution is a new environmental pollution source following the pollution of waste gas, waste water, waste residue, noise and the like, and generally comprises the pollution of white light, artificial daytime pollution and color light pollution; it can be classified into infrared light pollution, ultraviolet light pollution, laser pollution, visible light pollution, and the like according to the wavelength of light.
The skin is the first line of defense of a human body, ultraviolet rays in sunlight are mainly divided into UVA, UVB and UVC, most of the UVC is radiated and scattered by an ozone layer and an atmospheric layer, the UVA can directly permeate into a deeper dermis layer of the skin to damage elastic fibers and collagen fibers, the skin is damaged after long-term accumulation, particularly, the skin is aged and relaxed, wrinkles are deepened, the elasticity is lost, melanin is precipitated and the like, the UVB level is higher, the skin can be subjected to erythema, inflammation and aging after long-term or excessive irradiation, so the problems of photoaging, photodamage and the like of the skin can be caused by excessive ultraviolet rays, if the skin is not prevented and treated in time, cell DNA can be damaged, protooncogenes are activated, and skin tumors are caused; the popularity of electronic products increases the risk of people suffering from blue light damage. The blue light is used as a wave band with the shortest wavelength and the highest energy in a visible spectrum, and the penetration capability of the blue light is stronger than that of ultraviolet rays, so the blue light can easily penetrate into the hypodermis of skin, a large amount of active oxygen can be generated under the radiation of the blue light, the excessive active oxygen can degrade collagen, the synthesis of matrix metalloproteinase MMP is increased, the skin aging process is accelerated, oxidative damage is caused, the skin is aged, and the skin is seriously damaged. The invention is realized by the following steps:
in a first aspect, the present invention provides a photoprotective repair composition comprising a photoprotective extract comprising at least two of an artemia salina extract, a spirulina maxima extract, and a chlorella vulgaris extract, and a therapeutic wound villus flower extract.
In a second aspect, the present invention also provides a method for preparing the above-mentioned photoprotective repair composition, which comprises: mixing said photoprotective extract with said herb extract.
In a third aspect, the invention also provides application of the light protection repairing composition in preparing a skin care product.
In a fourth aspect, the invention also provides a skin care product, and the skin care product contains the light protection repair composition.

Detailed Description
In order to make the objects, technical solutions and advantages of the embodiments of the present invention clearer, the technical solutions in the embodiments of the present invention will be clearly and completely described below. The inventor finds that the existing ultraviolet-resistant, blue-resistant and infrared-resistant cosmetics generally have the defects of unobvious efficacy, allergy-causing phenomenon and no universality, most of the existing cosmetics comprise artificially synthesized chemical sunscreen structures, and the ingredients generally only can protect ultraviolet rays in UVB (ultraviolet B) band and UVA (ultraviolet A) band and do not relate to the protection of near infrared band and visible blue light band, so that the inventor carries out a great deal of research and practice on the basis of the above and provides the following photoprotective repair composition without the artificially synthesized sunscreen structures.
Some embodiments of the present invention provide a photoprotective repair composition including a photoprotective extract including at least two of an artemia salina extract, a spirulina maxima extract, and a chlorella extract, and a velveteen extract.

Further, in order to better generate coordination effect among the extracts, the extract comprises 0.01-15 parts by weight of photoprotective extract and 0.01-10 parts by weight of therapeutic downy rosea extract.
In some preferred embodiments, the photoprotective extract comprises 0.1-10 parts by weight of photoprotective extract and 0.1-5 parts by weight of therapeutic villus extract.

Some embodiments of the present invention also provide a skin care product, i.e. a photoprotective repair product, the ingredients of which comprise a photoprotective repair composition as in the above embodiments. The skin care product has the corresponding effect of the light protection and repair composition. The photoprotective repair composition is 0.1-10 wt%, preferably 0.5-5 wt% in the photoprotective repair skin care product, and the balance is common matrix of cosmetic external preparations and deionized water or purified water or distilled water, such as matrix components of thickening agent, antioxidant, emulsifier, humectant, emollient oil component and thickening agent.
In some embodiments, the skin care product includes, but is not limited to, water, lotion, essence, cream, eye cream, or mask. For example, skin care products such as photoprotective facial cleanser, photoprotective facial cream, photoprotective spray, photoprotective cream, photoprotective lotion, photoprotective water, photoprotective mask, photoprotective gel, photoprotective ampoules, etc. can be prepared by conventional processes.
The features and properties of the present invention are described in further detail below with reference to examples.

Test example 1
The test principle is as follows: researches show that ultraviolet and blue light irradiation with the wavelength of 280 nm-500 nm can cause skin cells to generate excessive active oxygen, and the excessive active oxygen can press the cells to cause cell damage and damage to the skin, so that the damage of light to the skin can be reduced by inhibiting the generation of the active oxygen or eliminating the excessive active oxygen. This experiment employed dilution of human fibroblasts to 0.1% volume in a specified medium, pretreatment with 0.1% mass fraction of the compositions of examples and comparative examples for 24h, while the control cultures were maintained in an untreated condition, then the cells were irradiated with artificial UVA \ UVB, pictures were taken using a fluorescence microscope, the intracellular reactive oxygen ROS were quantified, and the percent reduction in the amount of reactive oxygen species generated by human fibroblasts under artificial UVA \ UVB was tested for the photoprotective repair compositions of examples and comparative examples, respectively. The results are shown in Table 1.

As can be seen from Table 1, the percentage of reduction in the amount of active oxygen produced by the protective products of comparative examples 1 to 9 is much lower than that of the photoprotective repair compositions of examples 1 to 10, which shows that the ingredients of the photoprotective repair compositions are synergistic with each other, so that the active oxygen generated under UVA \ UVB radiation can be effectively reduced, and the damage of UVA \ UVB light to the skin can be reduced.
Test example 2
And (3) detecting the blue light resistance: the specific detection method comprises the following steps: the damage of blue light to Human Epidermal Keratinocytes (HEK) and Human Dermal Fibroblasts (HDF) and the protective effect of the skin care cosmetic of the present invention on the damage were investigated by measuring the survival rate and proliferation rate of metabolically active cells using CellTiter96 (MTS). Cells were cultured in 96-microwell plates, with the same number of cells in each plate.
The microwells containing human epidermal cells and human dermal fibroblasts were covered with a flat-bottomed sterilized plate containing a sample solution, irradiated with LED (476nm, 1900lux) blue light for 9 hours above a culture dish at a distance of 30cm, and then subjected to cell viability assay using CellTiter96 (MTS).
Blank control: cells were not exposed to blue light at room temperature (25 ℃).
The sample solutions were obtained by dissolving the photoprotective repair compositions of examples 1 to 10 and the photoprotective repair compositions prepared in comparative examples 1 to 8 in dimethyl sulfoxide, respectively, and the concentration of the sample solutions was 1%.
The average of three measurements was taken and the results are shown in Table 2.

The results in table 2 show that the activity of human epidermal keratinocytes and the activity of human dermal fibroblasts in examples 6 to 10 containing the photoprotective repair composition are more than 98%, which indicates that the photoprotective repair composition has a very strong blue light resistance and can effectively resist the damage of blue light to the skin. The activity was also more than 90% for the cells of examples 4-5. Comparative examples 1 to 9, in which the activity of human epidermal keratinocytes and the activity of human dermal fibroblasts are relatively low, show that each active ingredient in the photoprotective repair composition according to the embodiment of the present invention not only exerts its own efficacy, but also each active ingredient supplements each other to synergistically enhance the efficacy, so that the composition can achieve the effect of repairing skin damaged by blue light while resisting the skin damaged by blue light, and improve the resistance of skin to blue light.
Test example 3
Radical scavenging DPPH assay: the specific detection method comprises the following steps: ultraviolet, blue and red light irradiation can cause organisms to generate a large amount of free radicals, the free radicals are active chemicals with extremely strong oxidizing capability, and the large amount of free radicals can cause damage to skin, such as acceleration of the formation of color spots and melanin. DPPH is a stable free radical whose light absorption is reduced by pairing with its single electron in the presence of a free radical scavenger, thereby evaluating the ability of a substance to scavenge free radicals and thus assess its antioxidant capacity. The absorbance is measured by DPPH free radical scavenging test.
The results of the measurement are shown in table 3 below:

Through free radical scavenging experiments, the DPPH free radical scavenging rate of examples 4-10 is more than 90%, and compared with comparative examples 1-9, the light protection repairing composition disclosed by the embodiment of the invention has the advantages that each active ingredient not only exerts the self effect, but also supplements each other, is synergistic and can effectively scavenge free radicals.
Test example 4
Repair mitochondrial DNA damage test: in order to test the effect of photoprotection repairing mitochondrial DNA damage, the test judges through cell activity, and the larger the cell activity is, the smaller the mitochondrial DNA damage degree is. Succinate dehydrogenase in mitochondria of living cells can reduce MTT to formazan, dead cells do not have the function, formazan in the cells can be dissolved by dimethyl sulfoxide, and the absorbance (OD) value of the formazan is measured at 490nm wavelength by an enzyme labeling instrument, wherein the larger the number of living cells is, the larger the OD value is, the stronger the cell activity is.

Then 200. mu.l MTT (1mg/ml) solution was added per well and the cells were incubated for an additional 3 hours at 37 ℃; add 200. mu.l DMSO per well, dissolve with shaking, and measure OD at 490nm in a microplate reader. Cell activity ═ OD experimental group/OD control group, and the calculation results are shown in table 4.

From table 4, it can be seen that the cell activities of examples 1-10 are improved after light irradiation compared with the control group after light irradiation (control group + light irradiation), and the cell activity of example 6 is the highest, 95.84%, indicating that the photoprotective repair composition of the present invention can repair mitochondrial DNA damage caused by light; as is clear from comparison of comparative examples 1 to 9 and examples 1 to 10, only three of at least four extracts were selected to effectively repair mitochondrial DNA cellular activity, and the absence of any two components resulted in a decrease in cellular activity and failure to maximally repair mitochondrial DNA cellular activity.
Application example 1: light protection repairing mask
The formula of the facial mask comprises the following components in percentage by mass: 5% of sodium hyaluronate, 3% of butanediol, 3% of a photoprotective repair composition (example 6), 2% of a purslane extract, 1.5% of propylene glycol, 1% of a chamomile extract, 0.5% of folic acid, 0.3% of a bacillus/soybean fermentation product extract, 0.3% of 1, 3-propanediol, 0.2% of 1, 2-hexanediol, 0.1% of caprylhydroxamic acid, 0.05% of disodium EDTA, 0.03% of beta-glucan, and up to 100% of deionized water.
The preparation method comprises the steps of uniformly mixing the components according to the formula amount until the components are completely dissolved.
Application example 2: light protection repair ampoule
The formula of the spray comprises the following components by mass: 1, 3-propanediol 7%, photoprotective repair composition (example 6) 02%, niacinamide 1.5%, peppermint leaf extract 1.2%, geranium extract 0.8%, chamomile extract 0.5%, sophora flavescens root extract 0.5%, purslane extract 0.3%, aloe barbadensis leaf extract 0.3%, butylene glycol 0.5%, glycerol 0.3%, sodium benzoate 0.2%, potassium sorbate 0.25, 1, 2-hexanediol 0.1%, caprylyl hydroxamic acid 0.1%, lactobacillus/pumpkin fermentation product extract 0.3%, allantoin 0.2%, sodium hyaluronate 0.1%, deionized water to make up to 100%.
The preparation method comprises the steps of uniformly mixing the components according to the formula amount until the components are completely dissolved.
First, physicochemical test
The results of the physicochemical tests performed on the products of the respective application examples 1 and 2 were as follows:
1. pH value: the pH ranges of both application example 1 and application example 2 were between 5.5 and 6.8.
2. Heat resistance stability test (48 ℃ C.. times.72 h): the heat resistance stability is qualified.
3. Cold resistance stability test (-15 ℃ C.. times.72 h): the cold resistance and the stability are all qualified.
Second, safety and efficacy testing
(1) Testing the human body safety patch:
the experimental steps are as follows: the selected area is not more than 50mm2And a qualified patch device with the depth of about 1mm is used for performing a patch test on the backs of 30 volunteers, samples prepared in application examples 1 and 2 and a blank control sample (purified water) are respectively placed on the patch device in sequence, 0.02mL of the samples prepared in application examples 1 and 2 are selected, the purified water is placed in a control hole to serve as a blank control sample, then a hypoallergenic tape is used for being attached to the back of each tester for testing, and the sample is uniformly applied to the skin by lightly pressing the palm for 24 hours.
The experimental results are as follows: after the test is carried out for 24 hours, 30 minutes (after the pressure mark disappears) after the spot-pasting device on the back of the subject is removed, the back skin of the subject is observed to have no red or stabbing pain phenomenon or suspected erythema reaction, which indicates that the product containing the light protection repairing composition is safe and has no adverse reaction phenomenon.
(2) Efficacy testing
1) Repair mitochondrial DNA damage test
Experimental methods the mitochondrial DNA damage test method of test example four above was referenced and the test results are shown in table 5.

From table 5, it can be seen that the cell activities of application example 1 and application example 2 were improved after light irradiation compared with the control group after light irradiation (control group + light irradiation), which shows that the photoprotective repair product of the present embodiment can repair mitochondrial DNA damage caused by light.
2) Skin elasticity test
Selecting 10 persons with wrinkles in the age of 30-45, randomly dividing the persons into 2 groups, and carrying out subject experience test on application example 1 and application example 2 by 10 persons in each group. The test period was 4 weeks, each application was made once a day, morning and evening. The test is carried out by using a skin elasticity tester Cutomer MPA580 respectively at the non-use time and the 4 th week, the closer the test value is to 1, the better the elasticity of the skin is shown, the larger the change value is, the more obvious the effect of improving the elasticity of the skin is shown, and the experimental result is shown in Table 6.

As can be seen from the experimental results of table 6, the skin elasticity was improved after 4 weeks using the products of application examples 1 and 2, which shows that the photoprotective repair product of the present embodiment can improve the skin aging phenomenon.
3) Skin complexion test experiment
The photoprotective repair skin care products of application examples 1 and 2 were subjected to a test for subject experience. Each group was evaluated on 12 female subjects aged 18-45 years, with a test period of 4 weeks, each morning and evening paint.
And measuring the change of the cheek skin color by using a Lab colorimetric system spectrophotometer, and judging the shade of the skin color according to the magnitude of the L value of the skin color, wherein the lighter the skin color is indicated by the larger the L value. Therefore, before and after the test, the L value is increased, and the corresponding whitening effect is achieved; if the value of L is not changed, the whitening effect is not achieved. As can be seen from table 7, the skin pigmentation problem can be improved after four weeks using the skin care product of application examples 1 and 2, illustrating that the skin care product containing the photoprotective repair of the present embodiment can improve the skin pigmentation problem caused by light irradiation.
In summary, the photoprotective repair composition of the embodiments of the present invention utilizes the synergistic effect of at least three substances of the artemia salina extract, the chlorella extract, the villus vulgaris extract and the spirulina maxima extract to cover the protection range in the wavelength region of ultraviolet light, blue light and red light, performs photoprotection by absorbing ultraviolet light, can also remove a large amount of active DPPH radicals generated under light induction, and improves the resistance to external harmful light by improving the resistance and repair ability of the skin itself; meanwhile, mitochondrial DNA can be repaired, and ultraviolet light, blue light and infrared light can be comprehensively protected. The light protection restoration composition is safe, free of side effects, simple in preparation method, wide in application range and suitable for large-scale production. In addition, the skin care product containing the light protection and repair composition provided by the embodiment of the invention not only can repair mitochondrial DNA, but also can solve skin problems such as skin aging and pigmentation caused by light.

Claims (10)
1. A photoprotective repair composition comprising a photoprotective extract and a herb villus extract, wherein the photoprotective extract comprises at least two of a artemia salina extract, a Spirulina maxima extract, and a Chlorella extract.
2. The photoprotective repair composition of claim 1, comprising, by weight, 0.01 to 15 parts of a photoprotective extract and 0.01 to 10 parts of a therapeutic velveteen extract.
3. The photoprotective repair composition of claim 1, comprising, by weight, 0.1 to 10 parts of a photoprotective extract and 0.1 to 5 parts of a velvetplant extract.
4. The photoprotective repair composition of any one of claims 1 to 3, wherein the photoprotective extract includes artemia extract, Spirulina maxima extract, and Chlorella extract;
preferably, the mass ratio of the artemia extract, the spirulina maxima extract and the chlorella extract is 1-3: 1-4: 1-2.
5. The photoprotective repair composition of any one of claims 1 to 3, wherein the artemia extract is an aqueous extract; the spirulina maxima extract is water extract; the chlorella extract is water extract.
6. A process for the preparation of a photoprotective repair composition according to any of claims 1 to 5, comprising: mixing said photoprotective extract with said herb extract.
7. Use of a photoprotective repair composition as claimed in any one of claims 1 to 5 in the preparation of a skin care product.
8. Use according to claim 7, wherein the skin care product is a skin care product that is simultaneously protected against UV, IR and blue radiation or a skin care product that scavenges free radicals from the body.
9. A skin care product, characterized in that the components of the skin care product comprise the light-protection repairing composition as claimed in any one of claims 1 to 5, preferably, the light-protection repairing composition accounts for 0.1-10% by mass of the skin care product, preferably, the light-protection repairing composition accounts for 0.5-5% by mass of the skin care product.
10. The skin care product of claim 9, wherein the skin care product is water, lotion, essence, eye cream, face cream, or face mask.
