The effect of the compound Chinese herbal medicine hair tonic is verified:
1. establishing an alopecia animal model:
30 male C57BL/6 mice were acclimatized for 1 week, then divided into 5 groups (blank group, MNX 2%, example 1, example 2 and example 3) by weight and using Excel software, and the initial color of each back of each group was photographed and recorded in detail, and depilatory cream was used to depilate the back of each group about 3X 4cm 2 The skin was pink and unbroken, confirming that the hair follicles in the mice were in resting stage. The feed is fed regularly and quantitatively every day, and each mouse can drink water freely by 6g of common feed per day on average.
2. Use of group mouse drugs
Minoxidil is currently used as an externally applied medicine for treating alopecia in clinic, and is used as a positive control of an externally applied hair growth promoting effect experiment in the experiment. Specific administration methods are shown in table 2:
table 2 use of group mouse drugs
3. Observation of Hair growth
The skin and hair growth conditions of the hair removal area of each group of mice were observed every day, and the time when the skin color of the hair removal area of each mouse changed from pink to black and the time when hair growth began were recorded, and the effect of the drug on the hair growth cycle of the mice was evaluated. At the same time, the mice in each group were photographed at day 1, 14, 21 and 28, and the mice were subjected to the periodic hair growth as shown in fig. 1.
As can be seen from fig. 1, after 1d, there was no significant change in the mice of each group, and the skin remained pink; after 14d of the experiment, the skin color of the shaved area on the back of the mouse becomes black, wherein in the treatment group of the example 3, the skin color of the mouse is relatively uniformly restored, and black villi grow in a small area; after 21d of the experiment, the basic skin of each group of mice completely turns black, but the hair growth of the compound Chinese herbal medicine hair-restorer treatment group is more uniform than that of the blank group and the minol treatment group; after 28 days of the experiment, the hair of each group of mice is visible, wherein the hair length of individual mice in the treatment group of the example 3 has no obvious difference from other parts, and the hair of the mice in the treatment group of the example 2 grows more uniformly. In appearance, the new hair of the mice treated by the compound Chinese herbal medicine hair restorer is darker and brighter than the hair of other parts.
Statistics for the observation of hair growth status in mice are shown in table 3.
TABLE 3 time table for skin discoloration and hair growth
The hair growth of each group of mice was scored on days 1, 7, 14, 21, and 28. Grading standard: 0 point (the skin color of the depilatory area is pink and no hair grows); score 1 (skin color gray and hair growth less than 20% in the depilated area); 2 points (the skin color of the depilated area is gray black and the hair growth degree is more than 20 percent and less than 40 percent); 3 points (the skin color of the depilated area is light black and the hair growth degree is more than 40 percent and less than 60 percent); 4, dividing (the skin color of the depilatory area is black and the hair growth degree is more than 60 percent and less than 80 percent); 5 points (the hair in the depilating area is basically full and the hair growth degree is more than 80 percent).
As shown in fig. 2, the mouse score was linear with time. It is clear that the difference in scores among the groups of mice is not very large over the first 7 days of growth, whereas the scores among the groups of mice are clearly divided from day 14 onwards.
The score of example 2 was significantly higher than the other groups, while the blank group was also significantly lower than the other groups starting at day 14.
4. Determination of mouse hair length and hair weight and calculation of hair density
The compound Chinese herbal medicine hair-restorer and minoxidil are topically applied to dorsal skin of mice 28 d. After the experiment, the mice were sacrificed by cervical dislocation, and the skin of the mouse depilation region was cut off at the same position of each group of mice by 3X 4cm 2 Scraping off the hair on the skin patch by using a scalpel, weighing the skin patch on an analytical balance, and calculating the average value of the hair weight of the new hair of the mouse; measuring the length of the hair by a digital display caliper and calculating a mean value; and the wool density per unit area was calculated as wool weight/wool length. And meanwhile, analyzing by using a statistical method, and checking to judge whether significant differences exist, wherein the data are expressed by mean standard deviation (S.D.).
The results of the collection and measurement of the hair weight and the hair length of the obtained mice are shown in Table 4.
it can be seen that the hair growth of mice smeared with the compound Chinese herbal hair-restorer and minoxidil was significantly increased compared to the blank group. Among them, the data of the hair length, the hair weight and the hair density of the mice in the treatment group of example 2 are better.
5. Histological observation
The compound Chinese herbal medicine hair-restorer is smeared on the back of a mouse for 28 days by using a method of a table 2, the mouse is killed by a cervical vertebra dislocation method after the experiment is finished, long strip-shaped unhaired skin with the width of 1cm is cut parallel to the spine of the mouse and is lightly pasted on a hard board, wrinkles and clamp marks of tweezers are avoided, and at the moment, the paraformaldehyde fixing liquid is rapidly used for fixing for 48 hours. Then, washing with water, trimming, dehydrating, waxing, embedding, and slicing along the direction of hair growth (6 μm); then, the prepared roast slices were subjected to hematoxylin and eosin staining and mounting. Finally, histological changes of the hair follicles of the skin of the mice were observed under an optical microscope, recorded by photography, counted and measured in diameter. The hair growth and the histological appearance of the follicles of the mice under the light microscope are shown in fig. 3.
As can be seen from FIG. 3, the histological appearance of all the mice in the drug group is obviously different from that of the mice in the blank group, and the hair follicles of the mice using the compound Chinese herbal medicine extract and minoxidil are obviously enlarged and increased, so that the melanin in hair bulbs is also accumulated more. The blank group had thinner skin and the lowest thickness of subcutaneous tissue, almost all hair follicles grew in the dermis, and the color in the hair bulb was lighter, indicating that the hair of the mice in the blank group was still in the resting stage and had less melanin accumulation. The hair of the hair restorer group of compound Chinese herbal medicine has thicker fat cell layer around the hair, especially the fat cell layer of the embodiment 2 is thickest, the size of the vacuolated fat cells is uniform, the shape is clear and visible, the cells are arranged compactly and tidily along the hair growth direction, the fat cell layer extends to the dermis layer, the hair of the group grows uniformly and ideally, and meanwhile, the epidermis layer, the dermis layer and the muscle layer are arranged neatly and tightly.
In conclusion, the compound Chinese herbal medicine hair growth promoting liquid has the effect of promoting hair growth, can improve the hair regeneration speed, and enables the hair to be more smooth and glossy.
