Verification of the effect of the compound Chinese herbal medicine hair tonic solution:
1. building an alopecia animal model:
after 30 male C57BL/6 mice were adaptively bred for 1 week, they were numbered by mass and randomly divided into 5 groups (blank group, MNX2%, example 1, example 2 and example 3, respectively) according to mass by Excel software, and each group of mice was photographed and recorded in detail with initial color of each back, and dehairing was performed on the backs of the mice by about 3X 4cm using dehairing paste, and the skin was pink and unbroken, confirming that the hair follicles of the mice were in resting stage. The mice are fed with the common feed at regular time and quantity every day, and each mouse can drink water freely every day with 6g of common feed.
2. Use of group mouse drugs
Minoxidil is an external medicine which is clinically used for treating alopecia at present, and the minoxidil is adopted as a positive control of an external hair growth promotion effect experiment. The specific administration methods are shown in table 2:
TABLE 2 grouping use of mouse drugs
3. Observation of Hair growth Condition
The skin and hair growth conditions of the hair removal sites of each group of mice were observed daily, and the time for changing the skin color of the hair removal site of each mouse from pink to black and the time for starting hair growth were recorded, and the effect of the drug on the hair growth cycle of the mice was evaluated. At the same time, a stepwise photograph was taken of each group of mice on days 1, 14, 21, and 28, and the stepwise hair growth of the mice was shown in fig. 1.
As can be seen from fig. 1, after 1d, the mice in each group had no significant change and the skin was still pink; after experiment 14d, the skin color of the shaved area on the back of the mice became black, wherein in the treatment group of example 3, the skin color recovery of the mice was relatively uniform, and black fluff had grown in a part of the small area; after experiment 21d, the basic skin of each group of mice is completely blackened, but the hair growth of the compound Chinese herbal medicine hair growth liquid treatment group is more uniform than that of the blank group and the minox treatment group; after 28 days of the experiment, the hair of each group of mice was visible, wherein the hair length of the individual mice in the treatment group of example 3 was not significantly different from other parts, and the hair of the mice in the treatment group of example 2 was uniform in overall growth. From the appearance, the newly grown hair of the mice treated by the compound Chinese herbal medicine hair growth liquid is darker and brighter than the hair of other parts.
The statistics of observation of the hair growth status of the mice are shown in Table 3:
TABLE 3 skin discoloration time, long hair time schedule
The hair growth of each group of mice was scored on day 1, day 7, day 14, day 21, and day 28. Scoring criteria: 0 minutes (depilatory area skin color pink and no hair growth); 1 score (depilatory area skin color grey and hair growth less than 20%); 2 minutes (the depilatory area skin color appears gray black and the hair growth level is greater than 20% and less than 40%); 3 minutes (the skin of the dehairing area is light black and the hair growth degree is more than 40% and less than 60%); 4 minutes (the depilatory area skin color is black and the hair growth level is greater than 60% and less than 80%); 5 minutes (the hair is substantially full length in the depilation region and the extent of hair growth is greater than 80%).
As shown in fig. 2, the mouse score was linear with time. It can be clearly seen that the differences in scores of the mice in each group were not very large over the first 7 days of growth, but the scores of the mice in each group were clearly divided from day 14. The score of example 2 was significantly higher than the other groups, while the blank group was also significantly lower from day 14.
4. Determination of mouse hair length and weight and calculation of hair density
The compound Chinese herbal medicine hair tonic and minoxidil are topically applied to the back skin 28d of the mice. After the experiment is finished, killing the mice by a cervical dislocation method, taking the skin of the dehairing area of the mice, cutting 3X 4cm pieces of skin at the same position of each group of mice, scraping the hairs on the skin pieces by using a scalpel, weighing on an analytical balance, and calculating the average value of the weight of the newly-grown hairs of the mice; measuring the length of the hair by using a digital caliper and calculating the average value; and the hair density per unit area was calculated as the hair weight/hair length. At the same time, analysis is performed by a statistical method, whether significant differences exist or not is judged by inspection, and the data are expressed as mean + -standard deviation (S.D.).
The obtained collection and measurement results of the weight and length of the mouse are shown in table 4:
it can be seen that the hair growth of mice coated with the composite chinese herbal hair tonic and minoxidil was significantly increased compared to the blank group. Among them, the mice of the treatment group of example 2 had better hair length, hair weight and hair density data.
5. Histological observations
The compound Chinese herbal medicine hair growth liquid is smeared on the back of a mouse for 28 days by the method of Table 2, the cervical dislocation method of the mouse is killed after the experiment is finished, the long strip-shaped dehaired skin with the width of 1cm is cut parallel to the spinal column of the mouse, the long strip-shaped dehaired skin is lightly stuck on a cardboard, wrinkles and tweezer clamp marks are avoided, and the rat is quickly fixed by using paraformaldehyde fixing liquid for 48 hours. Subsequently, washing with water, trimming, dehydrating, immersing in wax, embedding, and slicing (6 μm) along the hair growth direction; then, the prepared baked sheet was subjected to hematoxylin eosin staining and sealing. Finally, histological changes, photographic recordings, counts and diameter measurements of the skin follicles of the mice were observed under an optical microscope. The histological appearance of the hair growth and hair follicle of the mice under light microscopy is shown in fig. 3.
As can be seen from fig. 3, the histological appearance of all drug group mice was significantly different from that of the blank group mice, and the hair follicles of mice using the compound chinese herbal medicine extract and minoxidil were significantly increased, and melanin accumulation in hair bulb was also more. The blank group has thinner skin, the subcutaneous tissue has the thinnest thickness, almost all hair follicles grow in dermis, the color in hair bulb is lighter, which indicates that the blank group mice hair is still in resting stage and melanin accumulation is less. The hair of the compound Chinese herbal medicine hair tonic treatment group is provided with a thicker fat cell layer, especially the fat cell layer in the embodiment 2 is thickest, the size of the vacuolated fat cells is uniform, the shape is clear and visible, the cells are compactly and orderly arranged along the hair growth direction, the fat cell layer extends to the dermis layer all the time, the hair growth of the group is more uniform and ideal, and meanwhile, the epidermis layer, the dermis layer and the muscle layer are orderly and compact.
In conclusion, the compound Chinese herbal medicine hair-growing liquid has the effect of promoting hair growth, can promote the hair regeneration speed, and enables the hair to be more smooth and glossy.
