Description
Technical Field
The invention relates to the technical field of cosmetic moisturizing, in particular to a skin moisturizing composition, a cosmetic containing the composition and a preparation method.
Background
At present, the moisturizing cosmetics are basically prepared by adding polyalcohol moisturizing agents, plant extracts, macromolecular polysaccharides and collagen raw materials as effective moisturizing components.
The polyhydric alcohol humectant such as glycerin has a small molecular polyhydric alcohol structure which absorbs hydroxyl groups and water molecules in a water molecular structure from the environment to form hydrogen bonds, effectively prevents TEWL (water vapor potential) reduction caused by water evaporation, and has a good effect on skin percutaneous water reduction in short-term use. The research on the external efficacy of the plant extract is not sufficient, some components of the plant extract have poor solubility in solvents for external use on the skin, and most plant extract is yellow or brown, so that the smell, texture, color and appearance stability of the cosmetic are influenced; in addition, the ionic concentration of the extract also influences the stability of the product system, and the addition amount of the extract also considers the influence on the preservative stability of the system. Macromolecular polysaccharide such as sodium hyaluronate has certain viscidity, easily forms a membrane, can form one deck natural moisture retention membrane on skin surface, consequently, macromolecular polysaccharide can effectively prevent skin moisture loss, and then reaches the effect of keeping moisture for a long time, and polysaccharide polymer sodium hyaluronate is the internationally recognized ideal natural humectant, is the best material of discovery nature moisture retention at present, uses extensively in cosmetics, and its molecular weight is bigger, and the film forming nature is better, and moisture retention, but the permeability is not good enough, and the skin that the content is too high feels sticky more easily, causes the product experience to feel not good.
The recombinant collagen obtained by introducing pichia pastoris and fermenting by taking human collagen gene as a template, which is added in Chinese patent CN110934766A, is added with 0.1-1 percent of the addition amount and 0.1-0.5 percent of sodium hyaluronate. Chinese patent CN112043643A is added with type III human collagen, the addition amount is 0.5-1 part, 1-4 parts hyaluronic acid. The two prior art formulas are added with sodium hyaluronate and recombinant collagen simultaneously, and the collagen and the sodium hyaluronate are adopted for synergistic moisturizing, so that the addition amounts of the collagen and the sodium hyaluronate are high, and the cost is correspondingly increased.
Disclosure of Invention
The invention aims to solve the technical problem that most of moisturizing products on the market at present are compounded and cooperated with collagen and other humectants to moisturize, and the moisturizing effect is limited.
In order to solve the above problems, the present invention proposes the following technical solutions:
the invention provides a skin moisturizing composition, which comprises mussel mucin and collagen; the collagen is selected from one or two or more of type I collagen, type II collagen, type III collagen, type IV collagen, type V collagen, type VI collagen, type VII collagen, type IX collagen, type X collagen, type XI collagen, type XII collagen, type XIII collagen, type XIV collagen, type XV collagen, type XVI collagen, type XVII collagen and type XVIII collagen.
Furthermore, the composition comprises, by weight, 15.5-80% of mussel mucin and 20.0-85.5% of collagen, and the composition can improve and enhance the barrier function of skin while keeping moisture.
Further, the composition comprises 20-40% of mussel mucin and 60-80% of collagen by weight percentage.
Further, the collagen is selected from the group consisting of type i collagen and type iii collagen.
Furthermore, the mass ratio of the type I collagen to the type III collagen is 1: 1-10.
Further, the collagen is selected from type III collagen.
The invention also provides a cosmetic containing the skin moisturizing composition, and the addition amount of the composition for improving the skin moisturizing activity is 0.035-1.1% by weight of the cosmetic.
Furthermore, the composition for improving the skin moisturizing activity is added in an amount of 0.1-0.60% by weight of the cosmetic.
The skin moisturizing composition disclosed by the invention is very low in addition amount in cosmetics based on the compounding of components, can achieve an excellent moisturizing effect, has the effects of instantly moisturizing and smoothing the skin, can also moisturize and lock water, remarkably improves the barrier function of the skin, and is moist and non-sticky after being absorbed by the skin, so that the skin is continuously moisturized and soft, and is moist and glossy.
Further, the cosmetics include water type cosmetics, emulsion type cosmetics, cream type cosmetics.
Further, the aqueous cosmetic comprises: toning lotion or facial mask liquid; the emulsion type cosmetic comprises: skin lotion, skin care essence, eye essence, sleep mask, moisturizing gel or face cleaning lotion; the cream-type cosmetic includes a skin cream or an eye cream.
The present invention also provides a method for preparing the cosmetic, comprising the steps of: the skin moisturizing composition is pre-dispersed in a solvent and added to a cosmetic system.
Further, the solvent is glycerol.
Because the mussel mucin in the skin moisturizing composition contains 20% of lysine and carries a large amount of positive charges, most raw materials used in the cosmetic are negatively charged, and the mussel mucin with the positive charges is added before the system is formed, so that the stability of the system is easily influenced; the mode of dispersing mussel mucin in the solvent in advance and then adding the system is adopted, so that the charge concentration condition of the mussel mucin can be dispersed, the combination and agglomeration of the mussel mucin and negatively charged components in the system are effectively avoided, the phenomena of layering, aggregation and the like are avoided, and the system stability is improved.
For example, in an emulsion product, the components of the oil phase are weighed according to the formula, the components of oil (such as mineral oil, silicone oil, synthetic petroleum oil and the like) and emulsifier are weighed, the temperature is raised to 75-85 ℃, the mixture is stirred and dissolved, the skin moisturizing composition is dissolved and dispersed in glycerin, and then the oil phase is added; weighing the components of the water phase, the humectant, the thickener, the chelating agent and the like according to the formula, adding into an emulsifying pot, heating to 75-85 ℃, and stirring to dissolve completely; cooling to 45-50 deg.C, adding neutralizer, antiseptic, essence, and effective materials into the emulsifying pan, and mixing;
for example, in an aqueous product, the components such as the humectant, the thickener, the chelating agent and the like are weighed according to the formula and added into a water kettle to be heated and dissolved completely; cooling to 45-50 deg.C, weighing antiseptic, solubilizer, essence, and other materials, and uniformly dispersing; the skin moisturizing composition is weighed and dissolved in glycerin, and is added into the system to be uniformly dispersed.
The above-listed addition mode adds the mussel mucin with positive charge after the system is stabilized, so that the influence of the mussel mucin on the stability of the system is reduced.
Compared with the prior art, the invention can achieve the following technical effects:
according to the skin moisturizing composition obtained by scientifically compounding the collagen and the mussel mucin, the mussel mucin is applied to the field of skin moisturizing for the first time, and the composition with an excellent moisturizing effect is obtained by utilizing the synergistic interaction between the collagen and the mussel mucin, and the moisturizing performance of the composition can be comparable to that of hyaluronic acid with the best moisturizing effect which is acknowledged in the field of skin care products; on the other hand, by scientific compounding of the collagen and the mussel mucin, when the composition is applied to cosmetics, efficient moisture preservation can be realized by adding a small amount of the composition, and the cost is greatly reduced.
In a third aspect, the skin moisturizing composition provided by the invention can be applied to normal skin care products to achieve the effect of efficiently moisturizing by adding a small amount of the composition; in addition, the skin moisturizing composition can be applied to a care product for damaged skin, so that the moisturizing effect is improved, a moist healing microenvironment is provided for tissue repair, and barrier repair is further promoted.
The skin moisturizing composition provided by the invention is compounded with multiple proteins, and has a synergistic moisturizing effect. The invention also provides a cosmetic containing the skin moisturizing composition, which has a high skin feel experience index, effectively improves the problem of water shortage of skin, and deeply moisturizes the skin.

The III type collagen used in the embodiment of the invention is a recombinant humanized III type collagen and a protein mentioned in a pichia pastoris recombinant expression system thereof in Chinese patent CN112552393A, and has the advantages of safe source, high protein purity and good compatibility. The mussel mucin and the type I collagen which are used in the embodiment of the invention are fermentation sources, and the mussel mucin is MFP-151 type mussel mucin which is gene recombination and is recombinant protein consisting of 6 MAP1 type (fp-1) decapeptide repetitive sequences at C end and N end of MAP5 type (fp-5).
The recombinant collagen and the mussel mucin are produced by adopting a biotechnology method, so that the virus infection risk brought by the traditional animal source is solved, the hydrophilicity of the collagen is increased, the adverse factors such as immunity rejection and the like are reduced, and the problem of raw material shortage is solved by mass production of genetic engineering; the collagen obtained by the genetic engineering technology is homologous with human skin cells in the same root, and has gene affinity.
Moisture retention test experiment
The method comprises the following steps: the method comprises the steps of selecting subjects with the age of 20-45 years, numbering every 5 persons, and enabling the subjects not to use other similar cosmetics before experiments, so that skin care behaviors except daily cleaning are avoided. After the experiment, each group of subjects used the designated facial cleanser to clean the face and then pressed with the same facial tissue to suck the moisture on the skin surface;
a test area of 3.0 × 3.0cm and a product amount (0.020 ± 0.001 g) were selected for each test area of the face, and the moisture content of the skin stratum corneum was measured using a Corneometer before the test area was applied to the test area (H0), after the test area was applied for 1 hour (H1), after the test area was applied for 2 hours (H2), after the test area was applied for 4 hours (H4), after the test area was applied for 6 hours (H6), and after the test area was applied for 8 hours (H8), and the moisture content change rate of the skin stratum corneum was calculated (formula: change rate of moisture content of the skin stratum corneum ═ hydration/H0 ÷ hydration/H0, n ═ 1, 2, 3, 4, 5, 6, 7, 8).
Experiment I, screening moisturizing effect of skin moisturizing composition

Comparative example 8 | 37.47 | 43.81 | 40.16 | 41.48 | 40.95 | 34.65 | 36.77 | 31.67 |
As can be seen from the results of tables 2 to 3, in comparative examples 1 to 4, the moisturizing effect is very weak when only one of mussel mucin or collagen is contained, and particularly in comparative examples 3 and 4, even when a large amount of mussel mucin or collagen is added, the moisturizing effect is still poor, which is probably because, although mussel mucin can form an elastic water-permeable and air-permeable membrane on the skin surface, the moisture in the skin is lost through the membrane in a dry environment due to this property, and conversely causes skin water shortage.
In previous researches, the anti-inflammatory and healing promoting effects of mussel mucin are generally proved or utilized, but the mussel mucin is firstly used in the moisturizing field, and the excellent moisturizing effect is realized through scientific proportioning with collagen. The inventor surprisingly found in experiments that when the mussel mucin and the collagen are compounded, the moisturizing performance of the composition is remarkably improved. When the mussel mucin and the collagen are compounded in a proper proportion (preferably 20-40% of the mussel mucin and 60-80% of the collagen), the moisturizing effect is respectively equal to more than 5 times of that of the single mussel mucin or the collagen, and is about 3 times of the sum of the moisturizing effects of various single proteins, and the moisturizing effect is equal to that of sodium hyaluronate accepted in the field; data after 4 hours show that the moisturizing effect of each single protein is obviously reduced, but the moisturizing effect of the skin moisturizing composition of examples 1-8 is not obviously reduced; on the other hand, the data of embodiments 1 to 6 and 7 to 8 also show that the composition containing mussel mucin, type I collagen and type III collagen has better moisturizing effect than the composition containing mussel mucin and type III collagen, and is generally better than the compounding of mussel mucin and other types of collagen, which indicates that the moisturizing effect between several types of proteins has synergistic effect through scientific proportioning of mussel mucin and collagen (especially type I collagen and/or type III collagen), and the moisturizing rate of the composition is improved. This is probably because, among molecules of collagen, a large number of hydrophilic hydroxyl groups and carboxyl groups exist, and thus it can achieve a high moisturizing ability in a humid environment by a high hydration ability; the collagen in the composition is used as an exogenous sebum membrane collagen supplement to repair damaged skin tissues, and the strong positive charge property of the mussel mucin attracts the crawling growth of fibroblasts, epidermal cells and the like, so that the repair of damaged skin barriers is promoted, and the moisturizing effect is further improved. The skin moisturizing composition can be applied to normal skin care products to achieve the effect of efficient moisturizing through adding a small amount of moisturizing composition, has more remarkable effects when being applied to care products for damaged skin, improves moisturizing effects, provides a moist healing microenvironment for tissue repair, and promotes barrier repair, and is also beneficial to barrier repair due to the anti-inflammatory and healing promoting effects of mussel mucin.
Experiment II, application of skin moisturizing composition to moisturizing of cosmetics
The cosmetic of the present invention containing the above-mentioned skin moisturizing composition may further include commonly used excipients such as lipid components, organic solvents, solubilizers, thickeners (gelling agents), surfactants, water, ionic or nonionic emulsifiers, fillers, chelating agents (metal ion chelating agents), preservatives, vitamins, hydrophilic or lipophilic active agents, and the like, which are generally used in cosmetic compositions.
For example, application examples 1 to 2 and comparative examples 1 to 4 of emulsion type cosmetics, and application examples 3 and comparative examples 5 of aqueous type cosmetics, the material composition of each experimental group is shown in table 4.
Table 4 cosmetic base formula containing skin moisturizing composition
The cosmetics of examples 1 to 2 and comparative examples 2 to 4 were prepared by the following method:
the method comprises the following steps: adding the phase D raw material thickener into a pot, dispersing with water, uniformly stirring, adding other raw materials such as a humectant, a chelating agent and the like, heating to 80-85 ℃, and stirring for dissolving for 10-20 min until the raw materials are completely dissolved;
step two: heating the raw materials of the B + C phase to 75-80 deg.C in advance, dissolving completely, adding the raw materials of the A phase, such as oil and emulsifier, etc., stirring and dispersing the raw materials of the A + B + C phase at 70-75 deg.C, adding the raw materials of the A + B + C phase into the D phase, and homogenizing for 5-10 min;
step three: adding the phase E raw material, and uniformly stirring and dispersing;
step four: cooling the above materials to 45 deg.C under stirring, adding F phase material, dispersing, and cooling to 37 deg.C to obtain emulsion type cosmetic.
Comparative example 1: the emulsion type cosmetic containing the skin moisturizing composition of this comparative example was prepared by a method different from that of example 1, specifically, as follows:
the method comprises the following steps: adding the thickener in the B + D phase raw material into a pot, dispersing with water (reserved part of water), stirring uniformly, adding the humectant, chelating agent and other raw materials, heating to 80-85 ℃, stirring for dissolving for 10min, and dissolving completely;
step two: heating the oil and emulsifier of phase A to 75-80 deg.C, stirring, dispersing, adding into phase B + D, and homogenizing for 5-10 min;
step three: adding the phase E raw material, and uniformly stirring and dispersing;
step four: cooling the material body to 45 ℃ while stirring, adding the F-phase raw material, uniformly stirring and dispersing, dissolving and uniformly dispersing the C-phase raw material with the reserved water, adding the mixture into a pot, and uniformly stirring; cooling to 37 deg.C to obtain emulsion type cosmetic.
The preparation methods of the aqueous type cosmetics of example 3 and comparative example 5 were as follows:
the method comprises the following steps: adding the humectant, the chelating agent, the thickening agent and the like of the raw materials of the phase D into a pot, dispersing with water (reserved part of water), heating to 75 ℃, and stirring to dissolve uniformly;
step two: cooling the material body to 45 ℃, adding the F-phase raw material, and stirring and dispersing uniformly;
step three: and (3) uniformly stirring and dispersing the B + C phase raw materials, adding the mixture into a pot, stirring until the material is uniform, and continuously cooling to 37 ℃ to obtain the water-based cosmetic.
The results of the moisturizing effect test on the emulsion type cosmetics (examples 1 to 2, comparative examples 2 to 4) and the aqueous type cosmetics (example 3, comparative example 5) are shown in table 5.

Comparative example 5 | 50.81 | 48.41 | 41.09 | 39.52 | 36.47 | 31.51 | 32.96 | 26.34 |
As can be seen from tables 5 to 6, the moisturizing effect of the skin moisturizing composition provided by the invention in comparative example 2 is obviously smaller than that of example 1, the moisturizing effect of the emulsion type cosmetic in examples 1 to 2 is smaller than that of example 2 although the skin moisturizing composition contains mussel mucin and collagen and has certain moisturizing activity on the skin, and the moisturizing effect of the emulsion type cosmetic in examples 1 to 2 can still reach 60% after the emulsion type cosmetic is used for 8 hours. The above data show that when the skin moisturizing composition is not added or the addition amount of the skin moisturizing composition is too small, the moisturizing effect of the cosmetic is not obvious, and the moisturizing effect of the cosmetic can be remarkably improved by adding the skin moisturizing composition provided by the invention in a proper proportion; the data of example 2 and comparative example 4 also further demonstrate that as the amount of skin moisturizing composition added increases, the rate of change of skin moisture content does not change linearly and the cosmetic moisturizing effect does not change significantly, which in turn increases the cost of the product.
Experiment III, influence of different preparation methods on product stability
Each group of samples in the example 1 and the comparative example 1 are placed in a circulation mode (stored at-18 ℃ for 24 hours, taken out, stored at 25 ℃ for 24 hours, stored at 50 ℃ for 24 hours, and stored at-18 ℃ in a circulation mode sequentially), the appearance change (such as coarse material body, color change, layering and caking) of the samples is observed at 18 ℃, 25 ℃ and 50 ℃ for 1 week, 2 weeks and 4 weeks respectively, the viscosity is detected, if any appearance change occurs, the experimental result is marked as 'x', and if the appearance change does not occur, the experimental result is marked as 'OK'; the viscosity detection conditions are as follows: 25 ℃, 7# rotor, 200rpm, machine: BrookFIELD viscometer, model: DV2 TRV. The results of the experiments were collected to give table 7.
TABLE 7 influence of different preparation methods on the product stability
As shown in table 7, in example 1, the initial viscosity of the cosmetic containing the skin moisturizing composition prepared by the method of dispersing the skin moisturizing composition in advance with the solvent and then adding the composition to the system at last is 6370rpm/min, and under the conditions of circulation, 18 ℃ below zero, 25 ℃ and 50 ℃, the cosmetic has good stability, does not have phenomena of thickening, discoloration, layering, caking, fungal growth and the like, has little change in viscosity when being placed for 1 week, 2 weeks and 4 weeks, and is within an allowable range, and the viscosity of the system is stable; comparative example 1 the skin moisturizing composition was not pre-dispersed, the initial viscosity of the system was 5534rpm/min, a phenomenon of slight flocculation occurred when the composition was left at 50 ℃ for 2 weeks to 4 weeks, and the composition was slightly yellowish; at the 4 th week of circulation, a little flocculation occurred and the viscosity of the system decreased significantly. The results show that the mode of dispersing the skin moisturizing composition in advance and then adding the skin moisturizing composition into the system finally effectively improves the compatibility of the skin moisturizing composition and the system, solves the problems of precipitation and delamination caused by the agglomeration of the skin moisturizing composition and other raw materials in cosmetics, keeps the viscosity of the system stable, and ensures that the obtained cosmetics have good stability.
Experiment four, evaluation of use of different skin moisturizing products
Randomly selecting 40 subjects with oily skin, dry skin and sensitive skin, wherein the total number of the subjects is 120, averagely dividing the subjects into 8 groups, and each group has 15 subjects; wherein each group uses one product. The samples are used in the morning and at night every day, and after the samples are continuously used for 1 month, the scoring condition of the feedback comprehensive evaluation of the testee is recorded.
