Description
Technical Field
The invention belongs to the technical field of skin care products, and particularly relates to a skin care composition with anti-sensitization and repair effects, an application thereof, a cosmetic and a method thereof.
Background
Skin allergy and irritation refer to abnormal immune responses, mainly including physiological dysfunction or tissue cell injury, when the body is stimulated by certain antigens, and the abnormal immune responses are mainly classified into allergic contact dermatitis and irritant contact dermatitis.
At present, the cosmetic has the following ways for preventing and treating skin sensitivity and irritation: 1. inhibiting inflammatory mediator release such as histamine; 2. enhancing body barrier function; 3. scavenging excessive free radical in vivo. Dermatitis is caused by inflammation mediators such as histamine, which is formed by decarboxylation of histidine in vivo, and the histamine in tissues exists in granules of mast cells and basophils in inactive combination, and is contained in a large amount in skin, bronchial mucosa, intestinal mucosa and nervous system. When the organism is subjected to physical and chemical stimulation or anaphylactic reaction, the cells can be caused to be degranulated, so that histamine is released and combined with histamine receptors to generate biological effects; the antihistamine can inhibit the mast cells from releasing allergic substances such as histamine and the like and inhibit the division of eosinophils and mast cells, thereby preventing the aggregation of the cells at an inflammation position, relieving inflammatory reaction and achieving the treatment purpose. Studies have shown that abnormalities in the lipid barrier not only reduce the water storage and moisturization functions of the skin, but also directly affect the regulation of keratinocyte growth and differentiation, affecting the formation of healthy stratum corneum, which is the core of the physical barrier structure of the skin, and repairing or enhancing the barrier function of the skin is an effective way to reduce skin sensitivity. Free radicals in humans destroy cell membranes, inactivate serum antiproteases, and damage genes leading to the appearance and accumulation of cellular variation. The attack of free radicals on the human body begins with the cell membrane. The cell membrane is extremely elastic and flexible, which is determined by its loose chemical structure, and as such, its electrons are easily lost, and thus the cell membrane is extremely vulnerable to free radical attack. Once electrons are taken away by the free radicals, the cell membrane loses elasticity and loses all functions, resulting in cardiovascular diseases. More serious is the attack of free radical on gene, which can destroy the molecular structure of gene and result in mutation of gene, resulting in systematic confusion of whole life. The excellent anti-allergic skin care product can inhibit the release of histamine, can penetrate into cells to fundamentally block allergic reaction, can effectively remove free radicals, and can protect cell membranes from being damaged by the free radicals.
Most of the existing cosmetics for relieving and resisting allergy are used for relieving the symptoms such as burning, stinging, itching and tightness of skin from the aspect of inhibiting histamine, and the products can relieve the symptoms of skin allergy in a short period of time, but are easy to repeatedly attack and cannot effectively improve the skin quality.

2. Test for inhibition of histamine release
Peripheral blood basophils (KU 812) are often used as effector cells for studying allergic reactions, and the present invention uses a KU812 cell histamine release model to examine the effect of samples on KU812 cell pro-inflammatory cytokine expression.
Cell model establishment: cell density was adjusted to 2X 10 by diluting cells with IMDM medium containing 10% fetal bovine serum 6 The resulting cell suspension was then seeded into well plates at an inoculum size of 150 μl per well.
Preparing and treating a tested sample: sample solutions of examples 1-8 and comparative examples 1-6 were formulated in DMSO at a mass concentration of 10% skin care composition, and then diluted to a skin care composition concentration of 0.3% by 10% fetal bovine serum culture medium to give test solutions.
The cell suspension was incubated at 36℃with 5% CO 2 Culturing in incubator for 10min, adding 20 μl of test solution, respectively, providing a control group without test solution, pre-incubating in incubator for 2 hr, adding stimulator (40 nM crotyl alcohol-12-tetradecanoate-13-acetate (PMA), 1 μm calcium ionophore A2318, respectively7) The PMA and a23187 were formulated using DMSO; after 2h, the cells were centrifuged, the cell supernatant was collected, and the histamine content in the supernatant was measured. The experimental results are shown in table 3.
3. Skin moisturization test
The skin care compositions of examples 1 to 5 and comparative examples 1 to 6 were diluted in a mixed solvent of glycerin and water in a volume ratio of 1:5 to a skin care composition mass concentration of 1%, to prepare sample solutions; sample solutions were prepared by diluting the cosmetics of examples 6 to 8 20 times. A supersaturated solution of ammonium sulfate was placed in a sealed container desiccator, the relative humidity of air was maintained at 60%, a medical breathable tape was attached to the slide, the tape was 6 cm. Times.2 cm, and 100. Mu.L of the test substance was applied to the tape. The control group was deionized water and 3 replicates were made per sample. Placing for 4 hours in a 40% humidity environment, weighing and calculating the moisture retention rate; the experimental results are shown in table 3.
4. Test of Water percutaneous loss
The principle of the transdermal moisture loss test is to measure the change in vapor pressure of moisture adjacent to the skin surface based on the diffusion principle. A cylindrical cavity measuring probe with two open ends is specially designed to form a relatively stable test small environment on the skin surface, the water vapor pressure gradient of the near epidermis (within about 1 cm) formed by the water loss of the stratum corneum at different two points is measured through two groups of temperature and humidity sensors, and the water amount evaporated through the epidermis is directly measured, so that the water loss condition of the skin surface is measured, and the moisturizing and skin barrier repairing effects of cosmetics on the skin surface can be evaluated. The calculation formula of the relative value of TEWL is:
TEWL relative value= (skin moisture loss of test sample-skin moisture loss of blank)/(skin moisture loss of positive control-skin moisture loss of blank).
The experimental method comprises the following steps: 36 healthy subjects 20-40 years old were selected and divided into 13 groups, none of the subjects had a history of skin disease, the tested sites were normal, and no experiment-independent medicines or cosmetics were applied during the test. The temperature of the test environment is controlled at 25+/-1 ℃, the relative humidity is 40+/-5%, purified water at 36 ℃ is used for wiping the two arms of the subject before the test, and the subject is allowed to sit still for 30min in the test environment for the test.
Sample solution: the skin care compositions of examples 1 to 5 and comparative examples 1 to 6 were diluted in a mixed solvent of glycerin and water in a volume ratio of 1:5 to a skin care composition mass concentration of 1%, to prepare sample solutions; sample solutions were prepared by diluting the cosmetics of examples 6 to 8 20 times; the positive control was sodium hyaluronate solution at the same concentration, and a blank control was set. Marking a plurality of regions on each subject's arm, the regions being generally 5X 5cm in size 2 . And coating the tested sample, blank control and positive control sample on different areas of the arm of the same subject. The skin moisture loss in the test area was measured 3 hours after application by a skin moisture loss tester, the relative value of TEWL was calculated, and the experimental results are shown in table 3.
