the adjuvant composition consists of a parasitic loranthus extract, a cynanchum paniculatum extract and an almond extract according to a mass ratio of 4: 5: 9;
the messenger medicine composition is composed of wild soybean seed powder and honey extract according to a mass ratio of 4: 3.
Experimental data for the peach blossom extract containing protective composition provided herein and comparative experiments therewith are given below.
First, efficacy test
Normal skin has a strong barrier function and can prevent external harmful substances from entering the skin. When barrier function is compromised, pathways are formed between keratinocytes from which water is lost. Thus, the transdermal loss of water increases when the skin barrier efficacy is diminished and decreases vice versa. The better the skin protection layer, the higher the moisture content and the lower the value of TEWL for skin moisture loss. The test evaluates the strength of the skin barrier function by the trans-dermal water loss (TEWL) and thus the anti-pollution efficacy of the compositions of the present invention.
The test samples were prepared by adding the compositions prepared in examples 1-5 and comparative examples 1-5 to a cream base at 5% each; cream base served as blank control.
The test population is selected from 110 healthy volunteers with dry skin and wrinkles such as dry lines, fine lines, expression lines and the like, the age is 30-45 years, the average age is 38 years, no allergy is caused to cosmetics, no acute inflammation is caused to the face, no hormone drugs or immunosuppressants are used, no other clinical tests are participated, and no external preparation is applied. The test samples were randomly divided into 11 groups of 10 persons each, and used in turn.
Test method three days before the test, the face without any cosmetic product, the TEWL value of the face before the product was used on day 1 was determined using the TEWL probe from CKMPA580 (5 tests, mean values read on computer). Cleaning face of each group of subjects with warm water every day, respectively applying equal amount of cream on facial skin, once in the morning and evening, continuously using for 28 days, stopping using the product in 29 and 30 days, measuring TEWL value with TEWL probe of CKMPA580 on 31 days, and calculating TWEL difference before and after use; the water content in the stratum corneum of the facial skin was measured using a skin moisture meter CorneometerCM825, and the hydration rate (%) was calculated; measuring the oil content of the skin of the forehead of the face by using a SebumetersM815 probe of an oil content tester; the skin elasticity of the face was measured using the skin elasticity tester MPA580 (the negative pressure was constant at 450MPA during the test), and the result was expressed as the ratio of the rebound amount Ur (i.e., the recovery value of the skin after 0.1 second of negative pressure removal) to the maximum tensile amount Uf of the skin, and the closer the value to 1, the better the skin elasticity. The results are shown in tables 1 and 2.

The above test data show that the TEWL value of the skin water loss after the cream added with the composition of the embodiment of the invention is obviously lower than the TEWL value before the use, and the difference value of TWEL before and after the use is obviously higher than that of the cream substrate, which shows that the composition of the invention has the function of enhancing the skin barrier function; and the skin hydration value and the skin elasticity value are obviously higher than those of the cream matrix, which shows that the cream prepared by the composition in the embodiment has good moisturizing effect, can increase the skin elasticity, and achieves the effects of moisturizing and delaying senility.
After the cream added with the composition of the invention in the example 5 is used, the difference of TEWL, the skin hydration rate and the skin elasticity value are obviously higher than those of the cream added with the compositions of the comparative examples 1-2, which indicates that the peach blossom extract plays a main role in the composition of the invention and is an irreplaceable or lacked medicament in the formula; moreover, the difference of TEWL, the skin hydration rate and the skin elasticity value in example 5 are higher than those of the cream added with the compositions of comparative examples 3 to 4, which shows that the efficacy of the wintergreen extract, the asparagus root extract, the rehmannia root extract and the walnut kernel extract in the ministerial drug composition of the invention is obviously better than that of the asparagus root extract, the rehmannia root extract and the walnut kernel extract with the same total mass, and the rigor and scientificity of the formula of the invention is demonstrated; similarly, in order to prove the scientificity and rigor of the formulation of the present application, the applicant further replaces the extract of loranthus parasiticus in the adjuvant composition with the extract of cynanchum paniculatum of the same quality in the research process, namely, comparative example 4, and as a result, the difference of TEWL, the skin hydration rate and the skin elasticity value of the composition in comparative example 4 are also reduced after the composition is added into cream, which is specifically shown in table 1 and table 2.
In order to prove the role of the protease glucan in the technical scheme provided by the application, the applicant removes the protease glucan in the components in the research process, namely, the comparative example 5, and as a result, the difference of TEWL, the skin hydration rate and the skin elasticity value of the composition in the comparative example 5 are also reduced after the composition is added into cream, and particularly, the difference, the skin hydration rate and the skin elasticity value are shown in tables 1 and 2, and the applicant believes that the protease glucan can be matched with wild soybean seed powder and honey extract to improve the permeability of the skin, promote the absorption of nutrient components, and simultaneously form a layer of nutrient film on the surface of the skin to help to enhance the barrier function of the skin.
Second, inhibition of Propionibacterium acnes
Preparation of a sample: the compositions prepared in examples 1 to 5 were mixed uniformly, and then an equal amount of the mixture was used to prepare an aqueous sample solution, which was subjected to the following experiment.
The experimental method comprises the following steps: 1mL of each sample aqueous solution was added to a sterile glass test tube, and 8mL of the previously prepared modified GAM broth was poured. Taking out the purified CO and keeping the temperature of the CO constant at 35 DEG C2Culturing clinical propionibacterium acnes for 48h in an incubator, respectively injecting 1mL of bacterial suspension into a broth culture tube filled with sample aqueous solution, and taking a glass tube without bacterial suspension as a negative control. Placing at 35 deg.C constant temperature CO as soon as possible after inoculation2The incubator is used for 48 hours. After culturing, the liquid in the broth culture medium is extracted for a verification experiment, and the survival condition of the propionibacterium acnes is observed.
The experiment results show that the propionibacterium acnes cannot grow in the culture medium added with the composition of the invention in the examples 1-5, and the composition has obvious inhibition effect on the propionibacterium acnes.
Third, testing the tolerance to stimuli
The skin is stimulated by exogenous pollutants, so that red swelling, pruritus, erythema, wheeze and the like are often caused, the autoimmune function of the skin can be reduced over time to form sensitive skin, the health of the skin is seriously damaged, so that the damaged skin is repaired and the immunity of the skin is enhanced, which is the most fundamental mode for resisting the exogenous pollutants, and the anti-pollution effect of the test composition is evaluated through the skin stimulation effect.
Experimental method: -stimulating the left and right forearm flexor skin with 3% SLS solution and 5% fruit acid as stimuli until the redness is developed, applying a cream containing 8% of the composition of the invention according to example 1 to the left forearm area, applying a cream containing 8% of the composition of the invention according to example 5 to the right forearm area, and observing the degree of regression of the redness of the skin after 2 days.

After 2 days of cream with the composition of example 1, the redness in left forearm area 1 had clearly subsided; after 5% of the fruit acid had irritated the right forearm area 2, the skin was visibly red and swollen in the right forearm area 2 was also visibly resolved after 2 days of application of a cream containing 8% of the composition of example 5 of the invention. The composition of the invention has obvious anti-irritation and anti-inflammation effects.
