背景技术
特应性皮炎或特应性湿疹是一种瘙痒性慢性炎症性皮炎,即一种炎性皮肤疾病,其特征是剧烈和慢性瘙痒以及复发性、偶发性湿疹病变(Mack和Kim,2018年;Weidinger等人,2018年)。这种疾病与皮肤屏障功能异常有关,或者实际上与其他症状,例如过敏性鼻炎、哮喘或对健康受试者通常可以耐受的环境过敏原过敏的可能出现有关。

紧密连接(或闭锁小带)是对上皮粘附(或细胞间粘附)和屏障功能至关重要的细胞间连接。它们是动态的跨膜复合物,可以打开和关闭。这些紧密连接由结构蛋白、与细胞骨架相关的蛋白质以及参与信号传导和细胞极性机制的蛋白质组成(Niessen,2007)。
特别地,紧密连接由跨膜蛋白,如闭合蛋白、闭锁蛋白、连接黏附分子(JAMs)和三细胞紧密连接蛋白(tricellulins),它们是与闭锁蛋白相似的结构蛋白;和胞质蛋白,例如闭锁小带(ZO)蛋白、扣带蛋白(cingulins)、多-PDZ结构域蛋白1(MUPP1)蛋白,或者实际上是具有颠倒方向的膜相关鸟苷酸激酶(MAGI)蛋白构成。
特别是在表皮的粒层(或颗粒层)中发现紧密连接(Brandner等人,2002)。
皮肤中紧密连接的主要功能是建立第二皮肤屏障,其在表皮角质层(或角质层)破裂时立即进行干预。
几项科学研究表明,在患有特应性皮炎的患者的皮肤中,组成紧密连接的几种蛋白质的表达发生了改变;特别是,特应性皮炎患者病灶区中claudin-1的表达显著降低(Batista等人,2015;Gruber等人,2015;)。
前述表明,恢复紧密连接的表达和功能提供了一种加强或重建皮肤屏障功能的新方法,并因此,用于预防和/或治疗特应性皮炎。
例如,文件EP 2 691 090和EP 2 704 704分别描述了化妆品活性成分(植物多酚、类黄酮或顺式尿刊酸)和益生菌在加强紧密连接,目的是治疗特应性皮炎中的用途。
然而,这些文件提出的解决方案对应于植物提取物和细菌裂解物的未定义组合物,这些组合物在施用于人皮肤后可能引起过敏或炎症反应,并且其中,所述组合物在批次之间可能显著不同。
因此,显然需要开发一种包含纯皮肤活性化合物的组合物,这些化合物有助于重建特应性前期和特应性皮肤中紧密连接的功能,还具有高滴度的活性分子,可导致特应性皮炎将被预防和/或治疗,特别是长期的,并且对于局部人类使用是安全的,即与皮肤完全相容,并且理想地是仿生的,即与人类皮肤的成分相同。
发明内容
申请人已经表明,令人惊讶的是,几种完全安全且适合人类化妆品使用的皮肤活性成分可以克服上述缺点并满足上述需求。
一般而言,本发明的目的是响应真正的需要,特别是美容需要,同时滋润和舒缓皮肤,减少不适感,并最终改善皮肤外观。
特别地,本发明旨在治疗特应性皮炎以改善皮肤外观并减少不完美之处。
更准确地说,申请人已经表明,包含在本发明的组合物中的化合物或活性成分以协同方式起作用以诱导克劳丁-1(claudin-1)(紧密连接中的关键结构蛋白)的表达。这些完全仿生的化合物能够用于配制适合预防和/或治疗特应性皮炎的皮肤化妆品产品。
在本发明的上下文中,术语“活性成分”是指具有生物和/或治疗特性的物质或化合物,其具有生理作用。
活性成分不同于存在于本发明的组合物中的至少一种赋形剂。
术语“赋形剂”表示除活性成分之外的物质,其为组合物提供活性成分的特性,特别是黏稠度、盖伦特性和/或矢量化特性。
在本发明的上下文中,术语“仿生化合物”是指天然存在于健康皮肤及其附件上/内的化合物,或者在水解或氧化/还原后转化为也天然存在于健康皮肤及其附件上/内的化合物。根据本发明,认为该化合物处于分离状态、与其天然形式相同的形式,或与其天然形式相比被修饰,但无论其获得或生产的方式如何,都保持与皮肤的细胞相容性。
因此,根据第一方面,本发明涉及一种组合物,其包含:
-至少一种碱性氨基酸,优选选自下组:精氨酸、赖氨酸、组氨酸、鸟氨酸和羟脯氨酸;和
-至少一种糖基神经酰胺。
根据特定的实施方式,本发明的组合物是化妆品或皮肤药物组合物,优选化妆品。
氨基酸是完全仿生的活性成分,因为它们存在于所有生物体中,无论是作为蛋白质成分还是在生物体降解过程后以游离形式存在。
在本发明的上下文中,术语“碱性氨基酸”表示任何形式的氨基酸,包括任何盐。
糖基神经酰胺是由通过β-糖苷键与单个中性糖(neutral ose)(单糖基神经酰胺)或多个中性糖(neutral oses)(多糖基神经酰胺)结合的神经酰胺键构成的糖脂,神经酰胺是鞘氨醇和脂肪酸的酰胺。优选的,中性糖是选自下组:阿洛糖、阿卓糖、葡萄糖、甘露糖、古洛糖、艾杜糖、半乳糖、岩藻糖和果糖的己糖或脱氧己糖。脂肪酸优选具有长链并且通常包含16至18个碳原子,该链任选地被羟基化。优选地,神经酰胺选自下组:神经酰胺-1、神经酰胺-2、神经酰胺-3、神经酰胺-4、神经酰胺-5、神经酰胺-6、神经酰胺-7。
糖基神经酰胺存在于所有生物体中,尤其是人类,它们在神经和脑组织中以髓鞘成分的容量特别丰富。因此,糖基神经酰胺被认为是仿生化合物。
根据特定的实施方式,包含在发明的组合物中的碱性氨基酸是精氨酸或其盐之一。
通常,各种形式的精氨酸是D-精氨酸、D,L-精氨酸和L-精氨酸。可提及的精氨酸的有机或无机盐的实例是盐酸盐、谷氨酸盐、丁酸盐和乙醇酸盐。
优选地,本发明的组合物含有L-精氨酸。
根据一个具体实施方式,本发明的组合物中使用的精氨酸呈纯化形式,优选纯度等于至少60%,优选等于至少70%,有利地等于至少80%,甚至更有利地等于至少90%,或甚至等于至少95%或甚至98%。包含在本发明的组合物中的精氨酸可以来自植物或化学来源,或者实际上通过生物技术获得。因此,日本协和发酵工业株式会社销售的具有INCI名称精氨酸的化妆品原料L-精氨酸可用于本发明的组合物中。
根据特定实施方式,本发明的组合物包含至少一种单糖基神经酰胺和/或多糖基神经酰胺形式的糖基神经酰胺,有利地是单糖基神经酰胺和/或四糖基神经酰胺。
根据优选的实施方式,本发明的组合物包含单糖基神经酰胺和四糖基神经酰胺形式的糖基神经酰胺。
根据一个特定的实施方式,该组合物包含对应于通过β-糖苷键共价结合到神经酰胺的葡糖醛酸(或葡萄糖酸)的单糖基神经酰胺。
根据一个特定的实施方式,本发明的多糖基神经酰胺对应于通过β-糖苷键共价键合到神经酰胺的寡糖。
有利地,本发明的多糖基神经酰胺是四糖基神经酰胺,其寡糖对应于以下糖的串联:葡糖醛酸-氨基葡糖酸-半乳糖-甘露糖(或GlcA-GlcN-Gal-Man)。

根据一个特定的实施方式,本发明的糖基神经酰胺是生物技术来源的,特别是通过从合成它们的细菌物种(例如假单胞菌属)的膜中提取而获得的。有利地,葡糖神经酰胺获自细菌物种动假单胞菌(Pseudomonas paucimobilis)。因此,分别由三乙股份有限公司(THREE B CO.,INC)和龟甲万生物化学公司(KIKKOMAN BIOCHEMIFA COMPANY)销售并具有INCI名称鞘糖脂的化妆品原料鞘糖脂和糖鞘脂(Biosphingo)可用于本发明的组合物中。
根据一个特定的实施方式,至少一种碱性氨基酸,有利地为精氨酸或其盐之一,占组合物总重量的0.001%至2%,有利地为0.01%至0.5%。
根据另一特定实施方式,至少一种糖基神经酰胺,有利地为单糖基神经酰胺和四糖基神经酰胺形式的糖基神经酰胺,占组合物总重量的0.0001%至1%,有利地0.001%至0.1%。
在一个特定的实施方式中,本发明的组合物还包含至少一种三萜皂苷(triterpene saponoside)或其衍生物之一,其选自下组:甘草次酸、甘草甜素、硬脂基甘草亭酸酯和二甘草次酸二钾(dipotassium diglycyrrizinate)。
这些从甘草中获取的化合物具有抗炎特性,通常用于局部治疗特应性皮炎,以对抗与这种皮肤病相关的瘙痒。例如,化妆品起始原料18β-甘草次酸、甘草酸二钾和甘草酸分别对应于INCI名称甘草次酸、甘草酸二钾和甘草酸和硬脂基甘草亭酸酯(均以其INCI名称命名,可从所列的生产商处,例如,在INCI的国际购买者指南中列出的生产商处购得),可用于本发明的组合物中。
有利地,除了碱性氨基酸和至少一种糖基神经酰胺之外,本发明的组合物包含具有抗炎剂能力的甘草次酸和二甘草酸二钾的组合。
根据一个特定的实施方式,至少一种三萜皂苷或其衍生物之一占组合物总重量的0.0001%和10%之间,有利地在0.01%和5%之间,优选地在0.05%和1%之间。
根据一种实施方式,本发明的组合物另外包含至少一种选自蔗糖酯和/或失水山梨醇酯的非离子表面活性剂。
根据一个具体的实施方式,本发明的至少一种非离子表面活性剂占组合物总重量的0.1%至5%,有利地1%至3%。
本发明使用的合适的蔗糖酯和/或失水山梨醇酯详细描述在文件WO2014/023895中,该文件通过引用整体并入。这些药剂单独或联合使用,可用于减少金黄色葡萄球菌在皮肤和/或鼻粘膜上的粘附和/或增殖,随后改善特应性皮炎的一般临床情况。
根据一个优选的实施方式,对应于失水山梨醇酯的本发明的非离子表面活性剂是聚山梨醇酯20(INCI)或聚山梨醇酯80(INCI),有利地是聚山梨醇酯20。
根据另一个优选的实施方式,对应于蔗糖酯的本发明的非离子表面活性剂是具有至少16的亲水亲油平衡值(HLB)的蔗糖硬脂酸酯(sucrostearate),其对应于蔗糖单酯与硬脂酸的相对比例包含在占蔗糖硬脂酸酯(INCI)重量的75%和80%。
根据一个特定的实施方式,本发明的非离子表面活性剂占组合物重量的0.1%至5%,有利地介于1%至3%之间。
根据一个特定的实施方式,本发明的组合物另外包含至少一种能够恢复皮肤屏障的脂质,有利地选自下组:
-至少一种皮肤外源性脂质,优选植物油;
-天然存在于皮肤中的成分混合物,包括神经酰胺-1、神经酰胺-3、神经酰胺-6;胆固醇;游离脂肪酸和植物鞘氨醇;和
-角鲨烷。
在一个特定的实施方式中,皮肤外源性脂质对应于向日葵籽油(INCI:向日葵籽油)、油菜油(INCI:菜籽油)和/或荷荷芭油(INCI:荷荷巴(Simmondsia chinensis(jojoba))种子油)。这三种植物油具有已知的润肤和抗炎特性。
在一个特定的实施方式中,天然存在于皮肤中的成分的混合物呈包含具有以下INCI名称的脂质的组合物的形式:月桂酰基乳酰乳酸钠、神经酰胺NP、神经酰胺AP、植物鞘氨醇、胆固醇、神经酰胺EOP。
在一个特定的实施方式中,能够恢复皮肤屏障的脂质是角鲨烷,一种也存在于皮肤组合物中的脂质。本发明上下文中的角鲨烷有利地是植物来源的。
优选地,能够恢复皮肤屏障的脂质占组合物重量的0.01%和25%之间,有利地在1%和15%之间。
根据一个特定的实施方式,本发明的组合物另外包含至少一种选自鼠李糖、木糖醇和甘露糖醇的额外多羟基化合物。这些多羟基化合物有助于减少致病菌如金黄色葡萄球菌对人体皮肤和粘膜(包括鼻粘膜)的粘附。甘露醇具有自由基清除活性。
在本发明的一个特定的实施方式中,该组合物包含上述三种另外的多羟基化合物的混合物。有利地,鼠李糖占组合物总重量的0.01%至1%,木糖醇占组合物总重量的0.05%至2%,甘露糖醇占组合物总重量的0.005%至1%。
有利地,本发明的组合物另外包含维生素PP,也称为维生素B3、烟酰胺或尼克酰胺,或其衍生物之一;已知这些化合物可刺激角质层脂质的合成,例如神经酰胺、游离脂肪酸和胆固醇。维生素PP的作用是激活丝氨酸棕榈酰转移酶的活性,这是合成鞘氨醇(神经酰胺的前体分子)的关键酶。
根据一个特定的实施方式,烟酰胺和/或其衍生物之一占组合物总重量的0.001%和10%之间,有利地在0.01%和5%之间,优选地在0.1%和2%之间。
根据一个特定的实施方式,本发明的组合物包含益生元物质,特别是低聚糖如低聚果糖或FOS(INCI)、短链糊精(INCI)、阿拉伯半乳聚糖(INCI)、菊粉(INCI)或乳果糖(INCI)。在一个优选的实施方式中,本发明的组合物包含FOS,即聚合度包含在2至10的果糖和蔗糖的低聚物。FOS有助于诱导β-防御素并且还被腐生菌群(saprophytic flora)代谢。FOS具有抗菌特性,可用于减少人工防腐剂的使用,如文件FR 2 831 059中所述。
有利地,FOS占组合物总重量的0.001%和20%之间,有利地在0.01%和10%之间,优选地在0.1%和5%之间。
根据特定实施方式,本发明的组合物另外包含至少一种抗微生物肽诱导剂,即能够诱导人类或微生物群抗微生物肽(特别是β-防御素)的合成和/或分泌。这些特性尤其在植物提取物中得到证实,例如来自波尔多树(boldo)的提取物或来自绣线菊(meadowsweet)的提取物。
有利地,抗微生物肽诱导剂是波尔多树提取物、绣线菊提取物或其混合物。
根据一个特定的实施方式,抗微生物肽诱导剂占组合物总重量的0.001%至5%,优选0.01%至1%。
根据特定实施方式,本发明的组合物呈适于美容和/或皮肤可接受的形式,即与皮肤、粘膜、头发、指甲和/或头皮相容。
根据优选的实施方式,本发明的组合物呈适合局部给药的形式。
在另一个实施方式中,本发明的有利的化妆品组合物另外包含生物活性系统,该系统一方面在水溶液中结合选自ATP(三磷酸腺苷)、Gp4G(四磷酸二鸟苷)和Ap4A(四磷酸二腺苷)的核苷酸的稳定形式;另一方面,至少一种包含至多六个氨基酸的仿生肽,有利地不同于本发明的那些,模仿皮肤多肽或皮肤蛋白质,或所述肽或所述蛋白质的激动或拮抗生物分子。在实践中,这些活性成分的组合意味着由于使用仿生肽,可以在获得皮肤美容或治疗效果的同时催化皮肤细胞的代谢活性。可选择这些后者以获得所需的效果,例如对神经源性刺激的抑制效果、脱色活性、抑制任何不耐受或致敏的效果等。
实际上,在本发明的生物活性系统中,核苷酸占组合物总重量的至多10%,优选在0.001%和5%之间;仿生肽占组合物总重量的0.001%至1%。
根据另一个实施方式,本发明的组合物可以另外包含来自运动节杆菌的提取物,特别是富含类胡萝卜素的提取物(INCI:微球菌溶菌液)。有利地,本发明的组合物包含这种类型的干提取物,其占组合物总重量的0.00001%至0.1%,优选0.0001%至0.001%。
根据另一个实施方式,本发明的组合物还包括其他成分,这些成分可以通过保护DNA、减少由UV辐射诱导的免疫抑制、自由基清除作用或这些作用的组合作用组成的作用而有助于内部保护。
如果本发明的制剂还包含一种或多种抗氧化剂,则可以进一步提高本发明的制剂对氧化应激或对抗自由基作用的保护作用,本领域技术人员将容易地从化妆品领域中使用的起始材料中选择所述抗氧化剂。
在本发明的一个特定的实施方式中,本发明的组合物还包含能够刺激皮肤细胞增殖的活性成分,有利地是成纤维细胞和/或角质形成细胞,优选人或动物成纤维细胞。特别地并且根据该实施方式,本发明的组合物还包括:

本发明的组合物还可另外包含大豆和/或小麦的肽提取物。
在实践中,源自大豆或小麦谷粒的肽提取物是通过肽酶对所述谷粒进行酶水解获得的,可用于回收平均大小为700Da的肽。优选地,大豆肽提取物为CAS号68607-88-5鉴定的提取物和/或小麦肽提取物为CAS号70084-87-6鉴定的提取物。小麦和大豆提取物可分别对应于INCI名称水解小麦蛋白和水解大豆蛋白。
在一个特定的实施方式中,大豆和小麦肽提取物一起使用,例如以分别包含在80/20和20/80之间的重量比,有利地包含在70/30和30/70之间,优选等于60/40。
在一个有利的实施方式中,大豆和/或小麦肽提取物不含合成的GHK三肽(甘氨酰-组氨酰-赖氨酸;INCI:三肽-1)。实际上,大豆和/或小麦的肽提取物占组合物总重量的0.01%和20%之间,有利地在0.1%和10%之间,优选地在0.2%和0.7%之间。
根据替代实施方式并且有利地,本发明的组合物另外包含至少一种紫外线吸收剂。众所周知,过滤紫外线可以改善特应性皮炎患者的皮肤状况。
在本发明的上下文中,术语“紫外线吸收剂”包括能够过滤UV-A、UV-B和/或UV-C的有机或无机化合物。
根据本发明,这些也可以是无机吸收剂而不是化学或有机吸收剂。
本发明的组合物可包含一种或多种广谱紫外线吸收剂,即吸收UV-A、UV-B、UV-C和可能的可见光的化合物或混合物。
可用于本发明上下文中的广谱有机吸收剂的实例是对应于以下INCI名称的吸收剂:三联苯三嗪、双乙基己氧基苯酚甲氧基苯基三嗪、亚甲基双-苯并三唑基四甲基丁基苯酚。适用于本发明的组合物的广谱吸收剂的另一个例子具有INCI名称二乙基己基丁酰氨基三嗪。
因此,在一个特定的实施方式中,本发明的组合物包含至少一种选自以下化合物的吸收剂,这些化合物由它们的INCI名称标识:三联苯三嗪、双乙基己氧基苯酚甲氧基苯基三嗪和亚甲基双-苯并三唑基四甲基丁基苯酚、二乙基己基丁酰胺三氮酮或其混合物。
有利地,该组合物包含过滤器双-乙基己氧基苯酚甲氧基苯基三嗪。
根据另一个实施方式,代替广谱吸收剂或除广谱吸收剂外,该组合物含有至少一种有机和/或无机的UV-A和/或UV-B吸收剂,其可以在水相(亲脂性)和/或油相(脂溶性)中。
因此,例如,本发明的组合物可以包含脂溶性UV-B吸收剂,其能够有助于广谱吸收剂的稳定化或增溶,或者实际上相互稳定,并且因此,增加防晒系数(SPF)。

在一个优选的实施方式中,本发明的组合物包含乙基己基三嗪酮。
在另一个实施方式中,脂溶性UV-B吸收剂是α-(三甲基甲硅烷基)-ω-(三甲基甲硅烷氧基)聚[氧基(二甲基)亚甲硅烷基]-co-[氧基(甲基)(2-{4-[2,2-双(乙氧基羰基)乙烯基]苯氧基}-1-亚甲基乙基)亚甲硅烷基]-co-[氧基(甲基)(2-(4-[2,2-双(乙氧基羰基)乙烯基]苯氧基)丙-1-烯基)亚甲硅烷基],一种能够过滤UV-B的有机硅聚合物(INCI:polysilicone-15)。

在本发明的上下文中有利的UV-A吸收剂是丁基甲氧基二苯甲酰甲烷(INCI)和二乙氨基羟基苯甲酰基苯甲酸己酯(INCI)。
在一个具体实施方式中,UV-A吸收剂是双-(二乙氨基羟基苯甲酰基苯甲酰基)哌嗪(INCI)(CAS号919803-06-8)。
因此,在一个优选的实施方式中,本发明的组合物包含至少一种选自下组的化合物的吸收剂,这些化合物由它们的INCI名称标识:丁基甲氧基二苯甲酰基甲烷、二乙氨基羟基苯甲酰基苯甲酸己酯、双-(二乙氨基羟基苯甲酰基苯甲酰基)哌嗪,或它们的混合物。
在本发明的上下文中有利的其他紫外线吸收剂是水溶性吸收剂,例如:

有利地,无机矿物过滤器或矿物筛(mineral screens)是金属氧化物和/或其他难以溶解或不溶于水的化合物,特别是以下物质的氧化物:钛(TiO2)、锌(ZnO)、铁(Fe2O3)、锆(ZrO2)、硅(SiO2)、锰(例如MnO)、铝(Al2O3)或铈(Ce2O3)。

根据特定的实施方式,本发明的组合物另外包含具有模糊效果的有利着色颗粒或具有模糊效果的几种颗粒的混合物。这些具有模糊效果的粒子也称为“柔焦效果粒子”或“雾霾效果粒子”。
在本发明的上下文中,术语“具有模糊效果的颗粒”表示有利地着色的颗粒,其旨在为肤色提供更高的透明度和模糊效果。特别地,具有模糊效果的颗粒使含有它的组合物能够通过光学效果减弱皮肤的小皱纹,特别是皮肤瑕疵,例如斑点、皱纹和细纹。

优选地,包含在本发明组合物中的模糊效果颗粒占组合物总重量的0.5%至20%,有利地介于1%和10%之间,甚至更有利地介于1%和5%之间。

1.材料和方法
1.1生物模型和细胞系
细胞系:
-细胞类型:正常人表皮角质形成细胞(NHEK;参考:Bioalternatives K34 1),用于第3代-培养条件:37℃,5%CO2
-培养基:角质形成细胞-SFM,辅以浓度为0.25ng/mL的表皮生长因子(或EGF)、浓度为25μg/mL的垂体浸出质(PE)和浓度为25μg/mL的庆大霉素
-用于此示例的测定介质:角质形成细胞-SFM补充浓度为25μg/mL的庆大霉素。
生产正常人角质形成细胞的培养物以测定这些物质;简而言之,方案如下:
-用0.75mM钙诱导角质形成细胞分化24小时;
-在警报素诱导前1小时用化合物处理;
-用以下警报素治疗:组胺、IL-33和IL-1β,分别为以下浓度:50μM、100ng/mL和100ng/mL,以诱导特应性皮炎的表型,其特征是claudin-1的表达下降。

1.2.本发明的组合物的活性化合物
本实施例中单独或组合使用的化合物示于下表4中。
1.3细胞毒性试验
在还包含浓度为0.75mM的CaCl2的测定培养基中对正常人角质形成细胞进行初步细胞毒性测定。这些初步测定将MTT四唑(3-(4,5-二甲基噻唑-2-基)-2,5-二苯基溴化四唑)盐还原方法与显微镜下的形态观察相结合。
在处理结束时(孵育72小时),将细胞在MTT存在下孵育,MTT转化为甲臜蓝色晶体,与琥珀酸脱氢酶(线粒体酶)的活性成正比。在细胞解离并通过添加DMSO溶解甲臜后,用分光光度计(VERSAmax,Molecular Devices)在540nm处测量代表活细胞数量及其代谢活性的光密度(OD)。
MTT存活测定的结果和细胞层的观察导致本发明的组合物选择活性化合物的浓度,以在研究的其余部分中测定感兴趣的化合物(见表2)。
1.4培养和治疗
然后将角质形成细胞接种到96孔板中并在培养基中培养72小时,然后在含有CaCl2分化诱导剂(0.75mM)的测定培养基中再培养24小时。然后用含有CaCl 2诱导剂(0.75mM)以及含有或不含(对照)本发明的活性化合物(单独或组合)或溶剂对照(0.02%测定的DMSO)的测定培养基替换培养基,然后细胞预孵育1小时。然后加入警报素的混合物:组胺、IL-33和IL-1β的混合物(分别为以下浓度:50μM、100ng/mL和100ng/mL)并将细胞孵育72小时。同时,产生非刺激/分化和非抑制对照条件(不含CaCl2且不含警报素的测定培养基)和非抑制对照条件(含CaCl2且不含警报素的测定培养基)。
对于每个条件,除了从60个图像量化(n=60)产生的对照条件之外,从30个图像量化(n=30)进行分析。通过考虑所有量化来对结果进行第一次评估,然后仅对重复培养的结果进行第二次评估,即具有3个重复(n=3),除了对照条件进行6次重复(n=6)。
1.5原位免疫标记
孵育后,除去培养基并冲洗、固定和透化细胞。然后用针对claudin-1的一抗标记细胞。然后通过与荧光染料(GAM-Alexa 488)偶联的二抗显示该抗体。同时,细胞核用Hoechst 33258(双苯酰亚胺)染色。
1.6显微镜观察和图像分析
使用高分辨率成像系统INCell AnalyzerTM 2200(GE Healthcare;20x物镜)获取图像。对于每个孔,获取10个数字图像。
通过测量蛋白质的荧光强度相对于通过双苯酰亚胺染色鉴定的细胞总数(通过Developer Toolbox 1.5软件整合数字数据,GE Health Care)来量化标记。
1.7数据处理
组间比较是在双边未配对学生t测定的帮助下进行的。如果n≥5,则可以解释统计分析。但是,对于n<5,计算数据仅作为指示提供。
本报告中使用的公式:
-均值的标准误差(sem)表示样本均值与真实总体均值之间的差异。sem的计算方法是将标准偏差(Sd)除以样本大小的平方根,按照以下公式计算:sem=标准偏差(Sd)/√n;
-百分比存活率使用以下公式计算:存活率(%)=(OD化合物/OD对照)x100;
-统计显著性阈值如下:ns:>0.0.5(不显著);*=0.01到0.05(显著);**=0.001到0.01(非常显著);***=<0.001(非常显著)。显著性是通过统计比较每个剂量的单独活性成分和组合的结果之间的差异来计算的。
结果如图1所示。
2.结果
在未受钙(CaCl2)刺激/分化的对照条件下,claudin-1的免疫标记在细胞质中的一些细胞中以斑点形式存在,偶尔出现在细胞膜中。
正如预期的那样,用0.75mM浓度的CaCl2进行分化诱导所分析的紧密连接蛋白(claudin-1)的表达增加。这种诱导是强烈且明显显著的,并且导致在膜水平以及细胞质水平上的claudin-1的表达大量增加。
用0.75mM钙(CaCl2)刺激/分化的角质形成细胞用警报素混合物(组胺+IL-1+IL-33测定为50μM+100ng/mL+100ng/mL)处理诱导显著抑制关于claudin-1表达的影响(对照的平均值约为61.5%)。
用本发明的活性化合物单独或组合处理对claudin-1表达的影响示于图1。从这些结果可以看出,单独测定的精氨酸(A)和糖基神经酰胺(GC)(相对于T+的%)显著逆转了警报素混合物对claudin-1表达的抑制作用。
精氨酸和糖基神经酰胺(A+GC)的组合对claudin-1的表达产生诱导作用,这比单独测定这些化合物时更大。在所有测定的浓度组合中,增加是非常显著的(***)或极其显著的(**),并且在每种情况下都不仅仅是互补,表明精氨酸和糖基神经酰胺在诱导claudin-1的表达方面具有意想不到的潜在协同作用,从而增强皮肤屏障功能和预防和/或治疗特应性皮炎方面。

Claims (16)

-与神经酰胺共价结合的寡糖是葡糖醛酸-葡糖胺-半乳糖-甘露糖。
6.如前述权利要求中任一项所述的组合物,其特征在于,所述至少一种糖基神经酰胺通过从至少一种细菌物种的膜中提取而获得。
