本发明涉及一种发酵液型植物生长调节剂及其制备方法和应用,包含至少一种投置于兜唇石斛活性液的发酵菌株,其中,所述发酵菌株包括解淀粉乳杆菌L6,所述兜唇石斛活性液中含有化合物A1和A2中的至少一种。化合物A1和化合物A2能提高植物的抗逆性与生长,加快植物组织细胞分裂和生长,使幼芽快速生长,对植物蛋白生成具有良好的促进效果,该植物生长调节剂可以应用在制备植物的环境胁迫耐性提高剂、制备促进植物种子生根发芽组合物、制备促进与调节植物生长营养组合物,具有环保与绿色功能,来源环保低廉,并且保存简单,易于大量生产。

本发明属于植物生长调节剂领域,具体涉及一种发酵液型植物生长调节剂及其制备方法和应用。
背景技术
农业作物是人类最基本的资源,在生产过程中,农作物的生长与成熟,健康是关注重要的环节。植物的种子萌发、生根、生长、开花、结实、衰老、脱落、休眠等一切生理活动,都离不开植物生长物质的调控。植物生长调节剂是其中重要的生物、化学药剂,常见的有生长素、赤霉酸、脱落酸、乙烯等,但是其实植物生长调节剂远远不止这些,它有许多的种类和作用。常见2,4-D、萘乙酸、吲哚乙酸、吲哚丁酸等生长素类调节剂,具有不同程度促使插条形成不定根的作用;赤霉素类、细胞分裂素类等PGRs处理种子后,可诱导增强水解酶的活性,加速种子萌发;生长素类、赤霉素类、细胞分裂素类、芸苔素内酯等都有促进细胞伸长的作用。在众多的植物生长调节剂中生长调节与抗逆性是最重要的功能,通过添加外源性植物生长调节剂,可以在植物生长过程中,促进与调节植物生长。
随着人口的不断增长,粮食种植行业飞速发展,传统的植物生长调节剂的需求量不断增大,价格也不断增长,相应粮食的价格也出现不小的涨幅。因此,有必要研发出一种新的植物生长调节剂,该植物生长调节剂具有环保与绿色功能,来源环保低廉,并且保存简单,易于大量生产。

本发明通过实验验证,化合物A1和化合物A2能提高植物的抗逆性与生长,加快植物组织细胞分裂和生长,使幼芽快速生长,对植物蛋白生成具有良好的促进效果。
在上述实施例中,所述发酵菌株还可以选择以下至少一种活菌:放线菌、枯草芽孢杆菌、脆壁克鲁维酵母、地衣芽孢杆菌、巨大芽孢杆菌、菌根、棕色固氮菌、胶质芽孢杆菌、泾阳链霉菌。
在本发明的一实施例中,上述发酵液型植物生长调节剂可例如为含有解淀粉乳杆菌L6活菌。
进一步地,所述兜唇石斛活性液中解淀粉乳杆菌L6的浓度为106~107cfu/ml,所述化合物A1的浓度为0~10%,所述化合物A2的浓度为0~10%。
进一步地,一种发酵液型植物生长调节剂制备方法,通过如下步骤制备:
步骤1:将兜唇石斛洗净并剪成5~7cm的小段,置于电热恒温干燥烘箱杀青30min,再将温度调整为55℃~60℃,烘干至恒重,取出磨粉,过60~80目筛,得兜唇石斛粉;
步骤2:将兜唇石斛粉进行超微粉碎处理后,加入到无水乙醇搅拌,搅拌过程中逐步加入与兜唇石斛粉等质量的去离子水,然后置于离心机9000rpm离心,去除上清液,55℃~60℃烘干至恒重,获得纳米石斛粉末;
步骤3:用移液枪向MRS肉汤培养基中滴入50uL已复苏的发酵菌株,塞好瓶塞,并将该发酵菌株培养液置于37℃培养箱中摇床培养36h~48h;
步骤4:将发酵菌株培养液进行离心处理,离心速率为5000r/min,离心处理15~20min后除去上清液,并加入等量的生理盐水,在相同离心条件下重复操作1~2次,制得菌液;
步骤5:取40~100g纳米石斛粉末置于锥形瓶中,加入300~400ml的蒸馏水,在121℃下灭菌30min,冷却至室温后,加入5~10ml菌液,塞好瓶塞后放置于培养箱中30℃~37℃培养48~72h,得发酵液;
步骤6:将发酵液进行高速离心得到获得上清液,离心温度为40~45℃,离心速度为10000rpm,用1μm滤膜将上清液进行抽滤,收集滤液并将滤液置于2~5℃环境下冷冻保存,即可得到兜唇石斛活性液。
进一步地,在所述步骤1中,电热恒温干燥烘箱杀青温度为100℃~130℃。
进一步地,在所述步骤2中,每毫升无水乙醇中添加兜唇石斛粉0.02~0.05g。
进一步地,在所述步骤3中,MRS肉汤培养基的制备:配制好的54g/L MRS肉汤培养基装入2L锥形瓶中,用瓶塞封住瓶口,于121℃灭菌锅杀菌30min,趁热将MRS肉汤培养基等量分装到5个500ml锥形瓶,在121℃灭菌锅杀菌30min。
进一步地,在所述步骤5中,培养箱的培养条件为:厌氧,在150~200rpm下避光振荡培养48~72h。
进一步地,根据本发明的另一目的,提出一种发酵液型植物生长调节剂在制备植物的环境胁迫耐性提高剂中的应用。
进一步地,根据本发明的另一目的,一种发酵液型植物生长调节剂在制备促进植物种子生根发芽组合物中的应用。
进一步地,根据本发明的另一目的,一种发酵液型植物生长调节剂在制备促进与调节植物生长营养组合物中的应用。
本发明中,对于植物生长调节剂的剂型无特殊要求,可以采用本领域公知的各种剂型制备方法制备成相应的植物的环境胁迫耐性提高剂、植物种子生根发芽组合物或植物生长营养组合物,在此不再赘述。本领域技术人员应该理解的是,本发明中在实际使用时可以根据实际需要稀释使用。
根据本发明,可以提供一种发酵液型植物生长调节剂,通过将其施加给植物或植物栽培介质,可以提高植物对环境胁迫的耐性、促进植物的生长,安全且容易制备,进而用于制备植物的环境胁迫耐性提高剂、植物种子生根发芽组合物或植物生长营养组合物的用途。

以下通过具体实施方式的描述对本发明作进一步说明,但这并非是对本发明的限制,本领域技术人员根据本发明的基本思想,可以做出各种修改或改进,但是只要不脱离本发明的基本思想,均在本发明的范围之内。本发明中所涉及的材料、器皿、试剂以及设备,均可通过市售或本领域常规技术手段获得。
本发明中,实施例中公开的制备方法仅例示性说明本发明的原理及其功效,而非用于限制本发明,其中公开的参数数值仅为本实施例的其中一种方式,在本发明公开的参数范围内均落入本申请的保护范围。任何熟悉此技术的人士皆可在不违背本发明的精神及范畴下,对上述实施例进行修饰或改变,在此不再赘述。本领域技术人员应该理解的是,本发明中的制备方法中所使用的各种成分或设备运行参数在实际使用时可以根据实际需要调整使用。
一种发酵液型植物生长调节剂,通过如下步骤制备:
步骤1:将兜唇石斛洗净并剪成5~7cm的小段,置于电热恒温干燥烘箱120℃杀青30min,再将温度调整为55℃~60℃,烘干至恒重,取出磨粉,过60~80目筛,得兜唇石斛粉;
步骤2:将兜唇石斛粉进行超微粉碎处理后,加入到无水乙醇搅拌,每毫升无水乙醇中添加兜唇石斛粉0.02g,搅拌过程中逐步加入与兜唇石斛粉等质量的去离子水,然后置于离心机9000rpm离心,去除上清液,55℃~60℃烘干至恒重,获得纳米石斛粉末;
步骤3:用移液枪向MRS肉汤培养基中滴入50uL已复苏的发酵菌株,塞好瓶塞,并将该发酵菌株培养液置于37℃培养箱中摇床培养48h;
步骤4:将发酵菌株培养液进行离心处理,离心速率为5000r/min,离心处理15min后除去上清液,并加入等量的生理盐水,在相同离心条件下重复操作1次,制得菌液;
步骤5:取50g纳米石斛粉末置于锥形瓶中,加入350ml的蒸馏水,在121°
C下灭菌30min,冷却至室温后,加入10ml菌液,塞好瓶塞后放置于培养箱中37℃培养48h,得发酵液;培养箱的培养条件为:厌氧,在200rpm下避光振荡培养;
步骤6:将发酵液进行高速离心得到获得上清液,离心温度为42℃,离心速度为10000rpm,用1μm滤膜将上清液进行抽滤,收集滤液并将滤液置于4℃环境下冷冻保存,即可得到兜唇石斛活性液。
实施例1:
一种发酵液型植物生长调节剂,包含一种投置于兜唇石斛活性液的发酵菌株,其中,所述发酵菌株包括解淀粉乳杆菌L6,所述兜唇石斛活性液中含有化合物A1,化合物A1的化学结构式如式Ⅰ所示:
所述兜唇石斛活性液中解淀粉乳杆菌L6的浓度为106~107cfu/ml,所述化合物A1的浓度为5%。
实施例2:
一种发酵液型植物生长调节剂,包含一种投置于兜唇石斛活性液的发酵菌株,其中,所述发酵菌株包括解淀粉乳杆菌L6,所述兜唇石斛活性液中含有化合物A2,
A2的化学结构如式Ⅱ所示:
所述兜唇石斛活性液中解淀粉乳杆菌L6的浓度为106~107cfu/ml,所述化合物A2的浓度为5%。
实施例3:
一种发酵液型植物生长调节剂,包含一种投置于兜唇石斛活性液的发酵菌株,其中,所述发酵菌株包括解淀粉乳杆菌L6,所述兜唇石斛活性液中含有化合物A1和化合物A2,化合物A1的化学结构式如式Ⅰ所示:
A2的化学结构如式Ⅱ所示:
所述兜唇石斛活性液中解淀粉乳杆菌L6的浓度为106~107cfu/ml,所述化合物A1的浓度为2%,所述化合物A2的浓度为3%。
实施例4:
一种发酵液型植物生长调节剂,该发酵液型植物生长调节剂与上述实施例3的区别在于:所述化合物A1的浓度为1%,所述化合物A2的浓度为4%。
实施例5:
一种发酵液型植物生长调节剂,该发酵液型植物生长调节剂与上述实施例3的区别在于:所述化合物A1的浓度为4%,所述化合物A2的浓度为1%。
实施例6
一种发酵液型植物生长调节剂,该发酵液型植物生长调节剂与上述实施例3的区别在于:所述化合物A1的浓度为2.5%,所述化合物A2的浓度为2.5%。
试验例
研究发酵液型植物生长调节剂催芽试验:
试验对象:
(1)将蒸馏水作为空白对照例,记为试样1;
(2)以鹤壁全丰生物科技有限公司生产的0.01%24-表芸苔素内酯稀释100倍制得植物生长调节剂作为对照例,记为试样2;
(3)以实施例1~实施例6制得的发酵液型植物生长调节剂分别稀释100倍,分别记为序号试样3~8。
试验方法:
在8个100ml的烧杯中分别放入150粒饱满的绿豆,倒入50ml浓度为3%的次氯酸钠溶液浸泡30min后,用清水将绿豆冲洗至无味,再用去离子水润洗;分别向8个烧杯中倒入100ml试样1~试样8,在恒温生化培养箱中26℃避光培养12h;浸种12h后,倒去溶液用纸巾和橡皮净封住烧杯口,继续在培养箱中培养24h;在8个豆芽发生器中分别加入100ml试样1~试样8,将8个烧杯中已经催好芽的绿豆分别对应放置在8个豆芽发生器内,得处理组1~处理组8,将得处理组1~处理组8移至恒温生化培养箱中26℃避光培养5天后,观察得处理组1~处理组8中豆芽的长势,并测量并记录绿豆芽可食用部分长度、绿豆芽根部长度、绿豆芽鲜重以及绿豆芽蛋白质含量。
(1)绿豆芽可食用部分长度、绿豆芽部总长度测定:将清洗好的豆芽用软尺同时测量其可食用部分、根部长度,单位为cm;
(2)绿豆芽鲜重测定:用分析天平测定每一个豆芽发生器内绿豆芽总重量;
(3)绿豆芽蛋白质含量测定:取各组绿豆芽可食用部分2.0g置于研钵研磨制浆,定容至10ml在冰浴浸提45min,用考马斯亮蓝G250测定其蛋白质含量。
试验结果:
发酵液型植物生长调节剂催芽试验中绿豆芽长势如图1所示:
在图1中,处理组1~处理组4从左到右排列放置在恒温生化培养箱上层,处理组5~处理组8从左到右排列放置在恒温生化培养箱下层,可以看出,采用处理组2~处理组4培植出的绿豆芽长势比处理组1培植出的绿豆芽要茂盛,且处理组3、处理组4培植出的绿豆芽,其生长趋势与处理组2培植出的绿豆芽的生长趋势相当,明显优于采用处理组1培植的绿豆芽,且处理组5~处理组8培植出的绿豆芽的生长趋势、芽长以及均衡性明显优于处理组1~处理组4培植出的绿豆芽。

Claims (1)
1.一种发酵液型植物生长调节剂,其特征在于,包含至少一种投置于兜唇石斛活性液的发酵菌株,其中,所述发酵菌株包括解淀粉乳杆菌L6,所述兜唇石斛活性液中含有化合物A1和化合物A2中的至少一种,所述解淀粉乳杆菌L6在兜唇石斛活性液中的浓度为106~107cfu/ml,所述化合物A1在兜唇石斛活性液中的浓度为2%,所述化合物A2在兜唇石斛活性液中的浓度为3%,
所述兜唇石斛活性液的制备方法为:
步骤1:将兜唇石斛洗净并剪成5~7cm的小段,置于电热恒温干燥烘箱120℃杀青30min,再将温度调整为55℃~60℃,烘干至恒重,取出磨粉,过60~80目筛,得兜唇石斛粉;
步骤2:将兜唇石斛粉进行超微粉碎处理后,加入到无水乙醇搅拌,每毫升无水乙醇中添加兜唇石斛粉0.02g,搅拌过程中逐步加入与兜唇石斛粉等质量的去离子水,然后置于离心机9000rpm离心,去除上清液,55℃~60℃烘干至恒重,获得纳米石斛粉末;
步骤3:用移液枪向MRS肉汤培养基中滴入50uL已复苏的发酵菌株,塞好瓶塞,并将该发酵菌株培养液置于37℃培养箱中摇床培养48h;
步骤4:将发酵菌株培养液进行离心处理,离心速率为5000r/min,离心处理15min后除去上清液,并加入等量的生理盐水,在相同离心条件下重复操作1次,制得菌液;
步骤5:取50g纳米石斛粉末置于锥形瓶中,加入350ml的蒸馏水,在121°C下灭菌30min,冷却至室温后,加入10ml菌液,塞好瓶塞后放置于培养箱中37℃培养48h,得发酵液;培养箱的培养条件为:厌氧,在200rpm下避光振荡培养;
步骤6:将发酵液进行高速离心得到获得上清液,离心温度为42℃,离心速度为10000rpm,用1μm滤膜将上清液进行抽滤,收集滤液并将滤液置于4℃环境下冷冻保存,即可得到兜唇石斛活性液;
