Abstract
The invention relates to a radix pseudostellariae skin lotion for whitening and moisturizing and a preparation method thereof, wherein the radix pseudostellariae skin lotion comprises the following components: radix pseudostellariae extract, glutathione, propylene glycol, an oil phase, a water phase and ginseng essence. The preparation method of the radix pseudostellariae skin lotion comprises the following process steps: (1) preparing a radix pseudostellariae extract; (2) preparing a radix pseudostellariae extract-glutathione solution; (3) preparing an oil phase: uniformly mixing all the raw materials of the oil phase, and heating to obtain the oil phase; (4) preparing an aqueous phase: uniformly mixing raw materials of a water phase, and heating to obtain the water phase; (5) emulsification: and adding the radix pseudostellariae extract-glutathione solution, the oil phase and the water phase into a homogenizer respectively for emulsification to obtain the radix pseudostellariae skin lotion. The radix pseudostellariae skin lotion prepared by the invention has high-efficiency whitening, antioxidant and moisturizing capabilities.
Description
Technical Field
The invention relates to a radix pseudostellariae skin lotion for whitening and moisturizing and a preparation method thereof.
Background
In recent years, the addition of natural substances in the global cosmetics industry is gradually a popular trend, and Chinese herbal medicines contain rich effective components which naturally become common additives. Radix pseudostellariae (pseudostellaria heterophylla) belongs to the plant of the family caryophyllaceae, is also a common traditional Chinese medicine supplement, and is also called as four-leaf ginseng, radix scrophulariae and the like. Some Chinese medicine ancient books record relevant data of radix pseudostellariae, dry root tubers are used for medicine taking, and the radix pseudostellariae traditional Chinese medicine ancient books have certain nourishing effects, such as tonifying qi and strengthening spleen, promoting the production of body fluid and moistening lung, and the like, and can be used for treating corresponding symptoms of weakness and sickliness of human bodies, lassitude and hypodynamia, poor appetite and the like.
According to the relevant verification, ancient China has some works about Chinese herbal medicines, and a plurality of beauty treatment folks and Chinese herbal medicines suitable for skin care are recorded in the works. In the current life, people often adopt some related components in the radix pseudostellariae to lay a corresponding foundation for the development of subsequent products. Wherein the trace elements are effective in controlling the loss of iron from the skin and maintaining a balanced and stable state of some circulatory systems. Radix Pseudostellariae contains many kinds of polysaccharides, glycosides, cyclic peptides, amino acids, volatile components, fatty acids, oils, sterols, etc., and can nourish and protect human skin and hair. In addition, radix pseudostellariae also contains a large amount of sugar saponins, palmitic acid, linoleic acid, and monoglycerides, which are cosmetic additives and are generally used for improving the epidermis. Radix Pseudostellariae contains the above effective components, so that the extract can be used in preparation of skin care products, and can be used as nutritional additive for hair and skin care products. Although the radix pseudostellariae extract has certain moisturizing and skin nourishing effects, the moisturizing effect is to be further improved, and the anti-oxidation and whitening effects are limited and the stability is poor. Therefore, the preparation of the radix pseudostellariae skin lotion with the whitening and high moisturizing effects is needed, the effective components in the radix pseudostellariae extract are stabilized, the nourishing and whitening effects of the skin lotion are improved, and the skin lotion has high whitening, antioxidant and moisturizing capabilities.
Disclosure of Invention
The invention aims to provide a radix pseudostellariae skin lotion for whitening and moisturizing and a preparation method thereof.
The purpose of the invention is realized by the following technical scheme: the radix pseudostellariae skin lotion for whitening and moisturizing comprises the following components in parts by weight: 3-5 parts of radix pseudostellariae extract, 0.4-0.6 part of glutathione, 5-6 parts of propylene glycol, 15-16 parts of oil phase, 71-77 parts of water phase and 0.05-0.15 part of ginseng essence.
Preferably, the oil phase comprises the following components in parts by weight: 5-7 parts of cyclomethicone, 5-6 parts of xanthan gum, 1-3 parts of methyl paraben and 1-3 parts of glycerol;
the water phase comprises the following components in parts by weight: 6-8 parts of panthenol, 5-7 parts of glyceryl stearate and 60-62 parts of water.
Preferably, the oil phase comprises the following components in parts by weight: 1-3 parts of stearic acid, 0.5-1.5 parts of triethanolamine, 7-9 parts of lanolin and 4-6 parts of glycerol;
the water phase comprises the following components in parts by weight: 3-5 parts of cetyl alcohol, 2-4 parts of glyceryl stearate, 0.3-0.4 part of methyl p-hydroxybenzoate and 66-68 parts of water.
The preparation method of the radix pseudostellariae skin lotion for whitening and moisturizing comprises the following process steps:
(1) preparing a radix pseudostellariae extract;
(2) preparation of radix pseudostellariae extract-glutathione solution: dissolving glutathione and radix Pseudostellariae extract in propylene glycol in advance, and performing ultrasonic treatment for 10-20min to obtain stable radix Pseudostellariae extract-glutathione solution;
(3) preparation of oil phase: uniformly mixing the raw materials of the oil phase, and heating to 65-85 ℃ to obtain the oil phase;
(4) preparation of the aqueous phase: uniformly mixing the raw materials of the water phase, and heating to 65-85 ℃ to obtain the water phase;
(5) emulsification: adding radix Pseudostellariae extract-glutathione solution, oil phase and water phase into homogenizer respectively, emulsifying, cooling to 40-50 deg.C, adding Ginseng radix essence, stirring, degassing, and cooling to below 40 deg.C to obtain the final product.
The specific operation method of the step (1) comprises the following steps:
step 1) radix pseudostellariae pretreatment: cleaning radix Pseudostellariae, lyophilizing, pulverizing, and sieving with a third sieve to obtain radix Pseudostellariae powder;
step 2) mixing the radix pseudostellariae powder and water according to the proportion of 1: 3-5 (W/V), then heating to 90-95 ℃, and continuously stirring for 2-3 h to obtain a radix pseudostellariae-hot water mixed solution;
step 3) heating and refluxing the radix pseudostellariae-hot water mixed solution for 4-6 hours, centrifuging, and collecting the radix pseudostellariae extract liquor on the upper layer;
and 4) concentrating the radix pseudostellariae extract liquid under reduced pressure to obtain a paste, thus obtaining the radix pseudostellariae extract.
The radix pseudostellariae lotion prepared by the invention is considered by the inventor to have high whitening, antioxidant and moisturizing capabilities, and the reason may be that: glutathione is a polypeptide compound containing gamma-amido bond and sulfydryl, and consists of glutamic acid, cysteine and glycine, and because the sulfydryl on the cysteine can form hydrogen bonds with active ingredients of the radix pseudostellariae extract, such as hydroxyl in the radix pseudostellariae saponin, the stability of the active ingredients of the radix pseudostellariae extract is improved. In addition, the selenium cysteine can be formed by combining the trace element selenium in the radix pseudostellariae extract and the cysteine in the glutathione, so that the selenium cysteine is an important component of important peroxide decomposition enzyme-glutathione peroxidase widely existing in a body, and the toxic peroxide is promoted to be reduced into a nontoxic hydroxyl compound, so that the structure and the function of a cell membrane are protected from being interfered and damaged by the peroxide, and the nourishing and whitening effects of the skin lotion are improved.
Compared with the prior art, the invention has the advantages that: the radix pseudostellariae skin lotion prepared by the invention has high-efficiency whitening, antioxidant and moisturizing capabilities. The radix pseudostellariae skin lotion obtained in the example 1 has the ROS inhibition rate of 88.3 +/-2.5%, strong free radical scavenging capacity, the DPPH inhibition rate of 93.2 +/-0.5%, the ABTS inhibition rate of 92.0 +/-0.8%, the tyrosinase activity inhibition rate of 86.6 +/-1.2% and the melanin inhibition capacity of 85.4 +/-1.0%; the radix pseudostellariae skin lotion obtained in the embodiment 4 has the ROS inhibition rate of 84.6 +/-3.6%, the DPPH inhibition rate of 92.0 +/-0.8%, the ABTS inhibition rate of 90.1 +/-1.0%, the tyrosinase activity inhibition rate of 84.3 +/-0.8% and the melanin inhibition capacity of 83.3 +/-1.4%; from the data, the radix pseudostellariae skin lotion obtained by compounding the radix pseudostellariae extract and the glutathione has good moisture absorption efficiency and whitening and moisturizing effects.

example 1:

The preparation method of the radix pseudostellariae skin lotion for whitening and moisturizing comprises the following process steps:
(1) preparing a radix pseudostellariae extract;
(2) weighing the raw materials according to a formula;
(3) preparation of radix pseudostellariae extract-glutathione solution: dissolving glutathione and radix Pseudostellariae extract in propylene glycol in advance, and performing ultrasonic treatment for 10-20min to obtain stable radix Pseudostellariae extract-glutathione solution;

the formula of the radix pseudostellariae lotion (named formula 1 as control group 1) is shown in table 2, and the radix pseudostellariae lotion obtained in the embodiment can be used as a control product of the radix pseudostellariae lotion obtained in the embodiment 1. Comparing the formulations of example 1 and example 2, it is clear that glutathione is not added to the formulation of the radix pseudostellariae lotion of example 2.

The preparation method of the radix pseudostellariae skin lotion comprises the following process steps:
(1) preparing a radix pseudostellariae extract;
(2) weighing the raw materials according to a formula;

(5) emulsification: respectively adding the radix pseudostellariae extract, the propylene glycol, the oil phase and the water phase into a homogenizer for emulsification, cooling to 45 ℃, then adding the ginseng essence, uniformly stirring, degassing, and continuously cooling to below 40 ℃ to obtain the radix pseudostellariae skin lotion.

Comparing the formulations of example 1 and example 3, it can be seen that the formula of the skin lotion of example 3 does not contain the radix pseudostellariae extract.

The preparation method of the skin lotion comprises the following process steps:
(1) weighing the raw materials according to a formula;
(2) preparation of oil phase: uniformly mixing the raw materials of the oil phase, and heating to 70 ℃ to obtain the oil phase;
(3) preparation of the aqueous phase: uniformly mixing raw materials of the water phase, and heating to 70 ℃ to obtain the water phase;
(4) emulsification: and adding glutathione, propylene glycol, an oil phase and a water phase into a homogenizer respectively for emulsification, cooling to 45 ℃, adding ginseng essence, uniformly stirring, degassing, and continuously cooling to below 40 ℃ to obtain the skin lotion.
Example 4:

Comparing the formulations of example 4 and example 5, it can be seen that glutathione was not added to the formulation of the radix pseudostellariae skin lotion of example 5.

Comparing the formulations of example 4 and example 6, it can be seen that the skin lotion of example 6 does not contain radix pseudostellariae extract.

The preparation method of the skin lotion comprises the following process steps:
(1) weighing the raw materials according to a formula;
(2) preparation of oil phase: uniformly mixing the raw materials of the oil phase, and heating to 70 ℃ to obtain the oil phase;
(3) preparation of the aqueous phase: uniformly mixing raw materials of the water phase, and heating to 70 ℃ to obtain the water phase;
(4) emulsification: and adding glutathione, propylene glycol, an oil phase and a water phase into a homogenizer respectively for emulsification, cooling to 45 ℃, adding ginseng essence, uniformly stirring, degassing, and continuously cooling to below 40 ℃ to obtain the skin lotion.
Example 7: performance evaluation of radix pseudostellariae skin lotion
7.1 evaluation of antioxidant Capacity
(1) Intracellular Reactive Oxygen Species (ROS) inhibition assay
Culture in 96-well cell culture plates at 1X 105Human skin keratinocytes per mL (HaCaT) in an incubator (temperature: 37 ℃, atmosphere: 5% CO)2) Continuously culturing for 1 day, adding self-made radix Pseudostellariae skin caring lotion, mixing, and reacting. When the reaction time is long enough, the supernatant in the 96-well cell culture plate is completely removed, and then the surface of the 96-well cell culture plate is washed using phosphoric acid as a buffer solution. Followed by addition of fresh culture medium (which contained 0.1 mmol. multidot.L)- 1H2O2Solution), the reaction time was 1 hour. The culture medium was then removed from the inside and fresh culture medium (containing 10. mu. mol. L-12 ', 7' -dichlorofluoroxanthate) was added, requiring 1 hour of further action in the absence of light. Finally, the culture solution is removed from the 96-well cell culture plate, and the absorbance of the radix pseudostellariae-containing skin lotion is measured under the condition that the excitation light is 502nm or the emission light is 524nm by taking phosphoric acid as a buffer solution (adding an appropriate amount).
ROS inhibition Rate results
According to the following results, comparing the ROS inhibition rates of the 6 self-made skin lotions of examples 1 to 6, the self-made radix pseudostellariae skin lotion has the following sequential effects of inhibiting strong intracellular oxidant active oxygen: formula 1, formula 2, control group 2, formula 1, control group 2, formula 2, control group 1, formula 1 and control group 1, wherein the ROS inhibition rates of the formula 1, the formula 2 and the control group 1 are respectively 88.3 +/-2.5% >84.6 +/-3.6% >47.6 +/-3.6% >45.5 +/-4.2% >21.4 +/-0.5% >18.3 +/-0.2%, wherein the ROS inhibition rates of the formula 1 and the formula 2 can reach 88.3 +/-2.5% and 84.6 +/-3.6%, and the fact that after the radix pseudostellariae lotion is added with glutathione is better in antioxidant effect (shown in a table 5-1) and better than the formula with a single component is shown, and the compounded radix pseudostellariae extract and the glutathione synergistically promote the antioxidant capacity of the lotion.

(2) ABTS cationic free radical inhibition assay
Sucking 1mL of self-made radix pseudostellariae skin lotion, placing the self-made radix pseudostellariae skin lotion in a 96-hole cell culture plate, and adding 180 mu L of 750 mu mol.L- 1ABTS+. And finally, performing related determination and analysis by using an enzyme linked immunosorbent assay (ELISA) detector, and detecting the light absorption value of the self-made radix pseudostellariae skin lotion at 734 nm.
(3) Evaluation of inhibition of DPPH free radical
And (4) transferring samples of the radix pseudostellariae skin lotion, wherein the transferred samples have different concentrations. Then, freshly prepared DPPH (10 mmol. multidot.L) was added-1) The control amount was 1 mL. The two were then mixed, with care being taken to uniformly oscillate and allow the reaction to go to completion. And finally, controlling the indoor temperature, standing the reactant for half an hour, and detecting the light absorption value of the radix pseudostellariae skin lotion under the condition of 517nm by using an enzyme linked immunosorbent assay (ELISA) detector.
ABTS and DPPH inhibition Rate results
The inhibition rate results of ABTS free radicals and DPPH free radicals are shown in the following table, and the experimental data show that the anti-oxidation capacity of the radix pseudostellariae lotion is sequentially that the formula 1 is more than the formula 2, the formula 1 is more than the control group 2, the formula 2 is more than the control group 1, and the formula 1 is more than the control group 1. The ABTS inhibition rates are respectively 92.0 +/-0.8% >90.1 +/-1.0% >48.3 +/-0.8% >48.2 +/-0.6% >32.8 +/-1.4% >31.3 +/-0.7%, and the DPPH inhibition rates are respectively 93.2 +/-0.5% >92.0 +/-0.8% >51.3 +/-1.2% >50.6 +/-1.0% >38.7 +/-0.6% >35.2 +/-0.5%. Therefore, the skin lotion developed based on the radix pseudostellariae can achieve the purpose of removing free radicals, wherein the formula 1 has the best effect of removing the free radicals, the ABTS inhibition rate reaches 92.0 +/-0.8%, the DPPH inhibition rate reaches 93.2 +/-0.5% (table 5-2), and the radix pseudostellariae extract and the glutathione can synergistically promote the antioxidant capacity of the skin lotion.

7.2 evaluation of whitening ability
(1) Mushroom tyrosinase Activity assay
Mixing the self-made radix pseudostellariae skin lotion with DMSO (dimethyl sulfoxide), respectively adding 2 muL and 18 muL, taking the mixture of the two as a test group of the experiment, and taking vitamin C as a control group of the experiment for comparison. The samples were then placed in 96-well cell culture plates and 25 μ L of mushroom tyrosinase solution (100U) was added at room temperature for a reaction time of 10 minutes. Detecting background value (set wavelength at 475nm) of tyrosinase in enzyme-linked immunoassay instrument (ELIASA), and adding 155 μ L2.5 mmol. L at room temperature-1L-Dopa solution, mixed evenly (control reaction invisible light). And finally, controlling the wavelength to be 475nm, detecting the light absorption value of the mushroom tyrosinase, and calculating according to the obtained numerical value, thereby analyzing the inhibition effect of the self-made radix pseudostellariae skin lotion with different formulas on the mushroom tyrosinase activity.
(2) Determination of melanin content in B16 cells

a series of related analyses can be carried out on the inhibition rate of the tyrosinase activity and the content of the melanin of the B16 cells, and the conclusion is that the formula 1 and the formula 2 have the highest inhibition capacity on the tyrosinase activity and the B cell 16 melanin, which shows that the whitening effect of the skin lotion is greatly improved after the radix pseudostellariae extract and the glutathione are compounded. 7.3 evaluation of moisturizing ability

Meanwhile, in the environments with several humidity levels, the radix pseudostellariae skin lotion compounded by the radix pseudostellariae extract and the glutathione is superior to hyaluronic acid. Therefore, the radix pseudostellariae skin lotion compounded by the radix pseudostellariae extract and the glutathione has good moisture absorption efficiency (tables 5-4).

Conclusion
According to the invention, 2 skin lotions are developed according to the radix pseudostellariae extract and glutathione, and all show excellent skin care performance, wherein the ROS inhibition rate of the formula 1 can reach 88.3 +/-2.5%, the scavenging capacity for free radicals is strong, the DPPH inhibition rate is 93.2 +/-0.5%, the ABTS inhibition rate is 92.0 +/-0.8%, the tyrosinase activity inhibition rate is 86.6 +/-1.2%, and the melanin inhibition capacity is 85.4 +/-1.0%; the ROS inhibition rate of the formula 2 reaches 84.6 +/-3.6 percent, the DPPH inhibition rate is 92.0 +/-0.8 percent, the ABTS inhibition rate is 90.1 +/-1.0 percent, the tyrosinase activity inhibition rate is 84.3 +/-0.8 percent, and the melanin inhibition capacity is 83.3 +/-1.4 percent; the radix pseudostellariae skin lotion obtained by compounding the radix pseudostellariae extract and the glutathione has good moisture absorption efficiency, and simultaneously shows that the radix pseudostellariae extract and the glutathione can synergistically promote whitening and moisturizing capabilities.
It should be noted that the above mentioned embodiments are only preferred embodiments of the present invention, but the scope of the present invention is not limited thereto, and any person skilled in the art can substitute or change the technical solution of the present invention and its inventive concept within the technical scope of the present invention.
Claims (7)
1. The radix pseudostellariae skin lotion for whitening and moisturizing is characterized in that: the paint comprises the following components in parts by weight: 3-5 parts of radix pseudostellariae extract, 0.4-0.6 part of glutathione, 5-6 parts of propylene glycol, 15-16 parts of oil phase, 71-77 parts of water phase and 0.05-0.15 part of ginseng essence.

The radix pseudostellariae lotion for whitening and moisturizing according to claim 1, which is characterized in that: the water phase comprises the following components in parts by weight: 3-5 parts of cetyl alcohol, 2-4 parts of glyceryl stearate, 0.3-0.4 part of methyl p-hydroxybenzoate and 66-68 parts of water.
6. The preparation method of the radix pseudostellariae lotion for whitening and moisturizing according to any one of claims 1 to 5, characterized by comprising the following steps: the method comprises the following process steps:
(1) preparing a radix pseudostellariae extracting solution;
(2) preparing a radix pseudostellariae extracting solution-glutathione solution: dissolving glutathione and radix Pseudostellariae extract in propylene glycol in advance, and performing ultrasonic treatment for 10-20min to obtain stable radix Pseudostellariae extract-glutathione solution;

step 3) heating and refluxing the radix pseudostellariae-hot water mixed solution for 4-6 hours, centrifuging, and collecting the radix pseudostellariae extract liquor on the upper layer;
and 4) concentrating the radix pseudostellariae extract liquid under reduced pressure to obtain a paste, thus obtaining the radix pseudostellariae extract liquid.
