Abstract
The invention relates to a radix pseudostellariae skin lotion for whitening and moisturizing and a preparation method thereof, and the radix pseudostellariae skin lotion comprises the following components: radix Pseudostellariae extract, glutathione, propylene glycol, oil phase, water phase, and Ginseng radix essence. The preparation method of the radix pseudostellariae skin lotion comprises the following process steps: (1) preparing radix pseudostellariae extract; (2) preparing a radix pseudostellariae extract-glutathione solution; (3) preparing an oil phase: uniformly mixing all raw materials of an oil phase, and heating to obtain the oil phase; (4) preparing an aqueous phase: uniformly mixing the raw materials of the water phase, and heating to obtain the water phase; (5) emulsification: adding radix Pseudostellariae extract-glutathione solution, oil phase and water phase into homogenizer respectively, and emulsifying to obtain radix Pseudostellariae skin caring lotion. The radix pseudostellariae skin-moistening lotion prepared by the invention has high-efficiency whitening, antioxidation and moisturizing capabilities.
Description
Technical Field
The invention relates to a radix pseudostellariae skin lotion for whitening and moisturizing and a preparation method thereof.
Background
In recent years, the addition of natural substances in the global cosmetic industry gradually becomes a popular state, and Chinese herbal medicines contain rich active ingredients, and the ingredients naturally become common additives. Radix pseudostellariae (pseudostellaria alternifolia) belongs to a plant of the family of caryophyllaceae, is also a common traditional Chinese medicine tonic, and is also named as four-leaf ginseng, rice ginseng and the like. Some related data of radix pseudostellariae are recorded in ancient books of traditional Chinese medicines, and the radix pseudostellariae is used as a medicine by using dry root tuber, has certain nourishing effects, such as replenishing qi to invigorate the spleen, promoting the production of body fluid to moisten the lung, and the like, and can be used for treating symptoms such as weakness, fatigue, weakness, poor appetite, and the like of a human body.
According to the related verification, the ancient times of China have some works related to Chinese herbal medicines, wherein a plurality of beauty bias and Chinese herbal medicines suitable for skin care are recorded. In the present life, people often adopt some relevant components in the radix pseudostellariae to lay a corresponding foundation for the development of subsequent products. Wherein, microelements are beneficial to controlling the loss of iron element in skin, and maintain certain circulatory system in balanced and stable state. The radix pseudostellariae contains various polysaccharides, glycosides, cyclic peptides, amino acids, volatile components, fatty acids, oils, sterols and other components, and can nourish and protect the skin and hair of people. In addition, radix Pseudostellariae contains more sugar saponins, hexadecanoic acid, linoleic acid and monoglyceride, which are cosmetic additives commonly used for improving epidermis. The radix pseudostellariae contains the active ingredients, so that the extract can be applied to the preparation of skin care products and used as a nutrition additive of hair and skin care products. The invention aims to provide a radix pseudostellariae skin lotion for whitening and moisturizing and a preparation method thereof.
The aim of the invention is realized by the following technical scheme: the radix pseudostellariae skin lotion for whitening and moisturizing comprises the following components in parts by weight: 3-5 parts of radix pseudostellariae extract, 0.4-0.6 part of glutathione, 5-6 parts of propylene glycol, 15-16 parts of oil phase, 71-77 parts of water phase and 0.05-0.15 part of ginseng essence.
Preferably, the oil phase comprises the following components in parts by weight: 5-7 parts of cyclomethicone, 5-6 parts of xanthan gum, 1-3 parts of methyl benzoate and 1-3 parts of glycerol;
the water phase comprises the following components in parts by weight: 6-8 parts of panthenol, 5-7 parts of glyceryl stearate and 60-62 parts of water.
Preferably, the oil phase comprises the following components in parts by weight: 1-3 parts of stearic acid, 0.5-1.5 parts of triethanolamine, 7-9 parts of lanolin and 4-6 parts of glycerol;
the water phase comprises the following components in parts by weight: 3-5 parts of cetyl alcohol, 2-4 parts of glyceryl stearate, 0.3-0.4 part of methyl parahydroxybenzoate and 66-68 parts of water.
The preparation method of the radix pseudostellariae skin-moistening lotion for whitening and moisturizing comprises the following process steps:
(1) Preparing radix pseudostellariae extracts;
(2) Preparing a radix pseudostellariae extract-glutathione solution: dissolving glutathione and radix Pseudostellariae extract in propylene glycol in advance, and performing ultrasonic treatment for 10-20min to form stable radix Pseudostellariae extract-glutathione solution;
(3) Preparation of an oil phase: uniformly mixing all the raw materials of the oil phase, and heating to 65-85 ℃ to obtain the oil phase;
(4) Preparation of an aqueous phase: uniformly mixing the raw materials of the water phase, and heating to 65-85 ℃ to obtain the water phase;
(5) Emulsification: adding radix Pseudostellariae extract, glutathione solution, oil phase and water phase into homogenizer respectively for emulsification, cooling to 40-50deg.C, adding Ginseng radix essence, stirring, degassing, and cooling to below 40deg.C to obtain radix Pseudostellariae skin caring lotion.
The specific operation method of the step (1) comprises the following steps:
step 1) preprocessing the radix pseudostellariae: washing radix Pseudostellariae, lyophilizing, pulverizing, and sieving with No. three sieve to obtain radix Pseudostellariae powder;
step 2) mixing the radix pseudostellariae powder and water according to the ratio of 1:3-5 (W/V), then heating to 90-95 ℃, and continuously stirring for 2-3 hours to obtain a radix pseudostellariae-hot water mixed solution;
step 3) heating and refluxing the radix pseudostellariae-hot water mixed solution for 4-6 hours, centrifuging, and collecting the radix pseudostellariae extract at the upper layer;
and 4) concentrating the radix pseudostellariae extract liquid under reduced pressure to obtain a paste, thereby obtaining the radix pseudostellariae extract.
The inventor considers that the radix pseudostellariae skin lotion prepared by the invention has high-efficiency whitening, antioxidation and moisturizing capabilities, and the reason is probably that: glutathione is a polypeptide compound containing gamma-amide bond and sulfhydryl, and consists of glutamic acid, cysteine and glycine, and because the sulfhydryl on the cysteine can form hydrogen bond with hydroxyl in the active ingredient of radix pseudostellariae extract such as radix pseudostellariae saponin, the stability of the active ingredient of radix pseudostellariae extract is improved. In addition, the microelement selenium in the radix pseudostellariae extract is combined with cysteine in glutathione to form selenium cysteine, so that the selenium cysteine is an important component of peroxide decomposing enzyme-glutathione peroxidase widely existing in the body, toxic peroxide is promoted to be reduced into a nontoxic hydroxyl compound, the structure and the function of a cell membrane are protected from being interfered and damaged by the peroxide, and the nourishing and whitening effects of the skin lotion are improved.
Compared with the prior art, the invention has the advantages that: the radix pseudostellariae skin-moistening lotion prepared by the invention has high-efficiency whitening, antioxidation and moisturizing capabilities. The radix pseudostellariae skin lotion obtained in the embodiment 1 has the ROS inhibition rate of 88.3+/-2.5%, strong free radical scavenging capacity, DPPH inhibition rate of 93.2+/-0.5%, ABTS inhibition rate of 92.0+/-0.8%, tyrosinase activity inhibition rate of 86.6+/-1.2% and melanin inhibition capacity of 85.4+/-1.0%; the radix pseudostellariae skin lotion obtained in example 4 has an ROS inhibition rate of 84.6+/-3.6%, a DPPH inhibition rate of 92.0+/-0.8%, an ABTS inhibition rate of 90.1+/-1.0%, a tyrosinase activity inhibition rate of 84.3+/-0.8% and a melanin inhibition capability of 83.3+/-1.4%; from the data, the radix pseudostellariae skin-moistening lotion obtained by compounding the radix pseudostellariae extract and the glutathione has good moisture absorption efficiency and whitening and moisturizing effects.

As can be seen from the formulas of comparative example 1 and example 2, the formulation of the pseudostellaria root skin lotion of example 2 is free of added glutathione.

As can be seen from the formulas of comparative example 4 and example 6, the skin lotion of example 6 was formulated without adding radix Pseudostellariae extract.

The preparation method of the skin lotion comprises the following process steps:
(1) Weighing the raw materials according to a formula;
(2) Preparation of an oil phase: uniformly mixing all the raw materials of the oil phase, and heating to 70 ℃ to obtain the oil phase;
(3) Preparation of an aqueous phase: uniformly mixing the raw materials of the water phase, and heating to 70 ℃ to obtain the water phase;
(4) Emulsification: adding glutathione, propylene glycol, oil phase and water phase into a homogenizer respectively for emulsification, cooling to 45 ℃, adding ginseng essence, stirring uniformly, degassing, and continuously cooling to below 40 ℃ to obtain the skin lotion.
Example 7: performance evaluation of radix pseudostellariae skin lotion
7.1 evaluation of antioxidant Capacity
(1) Intracellular Reactive Oxygen Species (ROS) inhibition assay
Culturing 1X 10 cells in 96 well cell culture plates 5 Human skin keratinocytes (HaCaT) at a volume of one mL were cultured in an incubator (temperature: 37 ℃ C., atmosphere: 5% CO) 2 ) Continuously culturing for 1 day, and adding self-made radix Pseudostellariae skin lotion after culturing, and mixing thoroughly for reaction. When the reaction time is enough, the supernatant in the 96-well cell culture plate is completely removed, and then phosphoric acid is used as a buffer solution to wash the surface of the 96-well cell culture plate. Fresh broth (which contained 0.1 mmol.L) was then added - 1 H 2 O 2 Solution) for a period of 1 hour. Next, the culture broth was removed from the inside, and fresh culture broth (containing 10. Mu. Mol. L-12',7' was added 'Dichloro-fluoroxanthodiacetate) for a further 1 hour under conditions of no light. Finally, the culture solution is removed from the 96-well cell culture plate, phosphoric acid is taken as buffer solution (proper amount is added), and the absorbance value of the radix pseudostellariae-containing skin lotion is measured under the condition that the excitation light is 502nm or the emission light is 524 nm.
ROS inhibition rate results
According to the following results, the ROS inhibition rates of the 6 self-made skin lotions of examples 1 to 6 were compared, and the self-made radix pseudostellariae skin lotions have the following active oxygen inhibition abilities to strong oxidants in cells: formula 1> formula 2> control group 2> formula 1> control group 2> formula 2> control group 1> formula 1 control group 1, their ROS inhibition ratios are 88.3+/-2.5% > 84.6+/-3.6% > 47.6+/-3.6% > 45.5+/-4.2% > 21.4+/-0.5% > 18.3+/-0.2%, respectively, wherein the ROS inhibition ratios of formula 1 and formula 2 can reach 88.3+/-2.5% and 84.6+/-3.6%, which indicates that the antioxidation effect is better after the pseudostellaria root skin lotion is added with glutathione (Table 5-1), which is superior to the formula of single component, and the pseudostellaria root extract and the glutathione cooperate to promote the antioxidation capability of the skin lotion after compounding.

(2) ABTS cationic radical inhibition assay
1mL of self-made radix pseudostellariae skin lotion is sucked and placed in a 96-well cell culture plate, and 180 mu L of 750 mu mol.L is added - 1 ABTS + . And finally, performing relevant measurement and analysis by using an enzyme-linked immunosorbent assay (ELISA) and detecting the absorbance of the self-made radix pseudostellariae skin lotion at 734 nm.
(3) Inhibition of DPPH radical assessment

ABTS and DPPH inhibition results
The inhibition rate results of the ABTS free radical and the DPPH free radical are shown in the following table, and experimental data show that the antioxidant capacity of the radix pseudostellariae skin lotion is sequentially shown as formula 1> formula 2> formula 1 control group 2> formula 2 control group 1> formula 1 control group 1 under the same volume. ABTS inhibition rates are 92.0±0.8% >90.1±1.0% >48.3±0.8% >48.2±0.6% >32.8±1.4% >31.3±0.7%, DPPH inhibition rates are 93.2±0.5% >92.0±0.8% >51.3±1.2% >50.6±1.0% >38.7±0.6% >35.2±0.5%, respectively. Therefore, the skin lotion developed based on the radix pseudostellariae can achieve the purpose of scavenging free radicals, wherein the formula 1 has the best effect of scavenging free radicals, has the inhibition rate of 92.0+/-0.8% on ABTS and 93.2+/-0.5% on DPPH (Table 5-2), and shows that the radix pseudostellariae extract and the glutathione can cooperate to promote the antioxidation capability of the skin lotion.

7.2 evaluation of whitening ability
(1) Determination of mushroom tyrosinase activity
The self-made radix pseudostellariae skin lotion and DMSO are mixed, the addition amount of the self-made radix pseudostellariae skin lotion and DMSO is 2 mu L and 18 mu L respectively, the mixture of the self-made radix pseudostellariae skin lotion and DMSO is used as an experimental group of the experiment, and vitamin C is used as a control group of the experiment for comparison. The samples were then placed in 96-well cell culture plates and 25. Mu.L of mushroom tyrosinase solution (100U) was added at room temperature for a reaction period of 10 minutes. (2) B16 cell melanin assay

Whitening efficacy:
a series of related analysis can be carried out on the inhibition rate of tyrosinase activity and the content of B16 cell melanin, so that the conclusion is drawn that the ability of the formula 1 and the formula 2 to inhibit tyrosinase activity and B cell 16 melanin is the highest, which indicates that the whitening effect of the skin lotion is greatly improved after the radix pseudostellariae extract and the glutathione are compounded. 7.3 evaluation of moisturizing ability
Moisture absorption rate measurement: weigh the sample, which needs to be dried to constant mass, apply it with M 0 And (3) representing. Under the environment conditions of 44%, 65% and 80% of relative humidity, the capability of the 6 self-made radix pseudostellariae skin lotions and the hyaluronic acid for absorbing water is gradually enhanced along with the increase of time, and the hygroscopic efficacy data show that the hygroscopic effects of the 6 self-made radix pseudostellariae skin lotions are also higher and higher along with the gradual rise of the humidity in the environment. Meanwhile, in the environments with the humidity, the radix pseudostellariae skin lotion compounded by the radix pseudostellariae extract and the glutathione is superior to hyaluronic acid. Thus, the radix pseudostellariae skin lotion compounded by the radix pseudostellariae extract and the glutathione has good moisture absorption efficacy (tables 5-4).

Conclusion(s)
According to the invention, 2 skin lotions are developed according to the radix pseudostellariae extract and the glutathione, and the skin lotions all show excellent skin care performance, wherein the ROS inhibition rate of the formula 1 can reach 88.3+/-2.5%, the scavenging capability to free radicals is stronger, the DPPH inhibition rate is 93.2+/-0.5%, the ABTS inhibition rate is 92.0+/-0.8%, the tyrosinase activity inhibition rate is 86.6+/-1.2%, and the melanin inhibition capability is 85.4+/-1.0%; the ROS inhibition rate of formula 2 reaches 84.6+/-3.6%, the DPPH inhibition rate is 92.0+/-0.8%, the ABTS inhibition rate is 90.1+/-1.0%, the tyrosinase activity inhibition rate is 84.3+/-0.8%, and the melanin inhibition capacity is 83.3+/-1.4%; the radix pseudostellariae skin-moistening lotion obtained by compounding the radix pseudostellariae extract and the glutathione has good moisture absorption efficiency, and meanwhile, the radix pseudostellariae extract and the glutathione are proved to be capable of synergistically promoting the whitening and moisturizing abilities.
It should be noted that the foregoing is only a preferred embodiment of the present invention, but the scope of the present invention is not limited thereto, and any person skilled in the art should be able to apply equivalent substitutions or alterations to the technical solution and the inventive concept thereof within the scope of the present invention.
Claims (7)
1.A radix pseudostellariae skin lotion for whitening and moisturizing is characterized in that: the composite material comprises the following components in parts by weight: 3-5 parts of radix pseudostellariae extract, 0.4-0.6 part of glutathione, 5-6 parts of propylene glycol, 15-16 parts of oil phase, 71-77 parts of water phase and 0.05-0.15 part of ginseng essence;
the preparation method of the radix pseudostellariae extracting solution comprises the following steps:
step 1) preprocessing the radix pseudostellariae: washing radix Pseudostellariae, lyophilizing, pulverizing, and sieving with No. three sieve to obtain radix Pseudostellariae powder;
step 2) mixing the radix pseudostellariae powder and water according to the ratio of W/V of 1:3-5, then heating to 90-95 ℃, and continuously stirring for 2-3 hours to obtain a radix pseudostellariae-hot water mixed solution;
step 3) heating and refluxing the radix pseudostellariae-hot water mixed solution for 4-6 hours, centrifuging, and collecting the radix pseudostellariae extract at the upper layer;
step 4) concentrating the radix pseudostellariae extract under reduced pressure to obtain a paste, thereby obtaining the radix pseudostellariae extract;
the preparation method of the radix pseudostellariae skin-moistening lotion for whitening and moisturizing comprises the following process steps:
(1) Preparing radix pseudostellariae extract;
(2) Preparing a radix pseudostellariae extracting solution-glutathione solution: dissolving glutathione and radix pseudostellariae extract in propylene glycol in advance, and performing ultrasonic treatment for 10-20min to form a stable radix pseudostellariae extract-glutathione solution;
(3) Preparation of an oil phase: uniformly mixing all the raw materials of the oil phase, and heating to 65-85 ℃ to obtain the oil phase;
(4) Preparation of an aqueous phase: uniformly mixing the raw materials of the water phase, and heating to 65-85 ℃ to obtain the water phase;
(5) Emulsification: adding radix Pseudostellariae extract, glutathione solution, oil phase and water phase into homogenizer respectively for emulsification, cooling to 40-50deg.C, adding Ginseng radix essence, stirring, degassing, and cooling to below 40deg.C to obtain radix Pseudostellariae skin caring lotion.
2. The skin lotion for whitening and moisturizing according to claim 1, wherein: the oil phase comprises the following components in parts by weight: 5-7 parts of cyclomethicone, 5-6 parts of xanthan gum, 1-3 parts of methyl benzoate and 1-3 parts of glycerin.
3. The skin lotion for whitening and moisturizing according to claim 1, wherein: the water phase comprises the following components in parts by weight: 6-8 parts of panthenol, 5-7 parts of glyceryl stearate and 60-62 parts of water.
4. The skin lotion for whitening and moisturizing according to claim 1, wherein: the oil phase comprises the following components in parts by weight: 1-3 parts of stearic acid, 0.5-1.5 parts of triethanolamine, 7-9 parts of lanolin and 4-6 parts of glycerol.
5. The skin lotion for whitening and moisturizing according to claim 1, wherein: the water phase comprises the following components in parts by weight: 3-5 parts of cetyl alcohol, 2-4 parts of glyceryl stearate, 0.3-0.4 part of methyl parahydroxybenzoate and 66-68 parts of water.
6. The method for preparing the skin lotion for whitening and moisturizing as claimed in any one of claims 1 to 5, wherein: the method comprises the following process steps:
(1) Preparing radix pseudostellariae extract;
(2) Preparing a radix pseudostellariae extracting solution-glutathione solution: dissolving glutathione and radix pseudostellariae extract in propylene glycol in advance, and performing ultrasonic treatment for 10-20min to form a stable radix pseudostellariae extract-glutathione solution;
(3) Preparation of an oil phase: uniformly mixing all the raw materials of the oil phase, and heating to 65-85 ℃ to obtain the oil phase;
(4) Preparation of an aqueous phase: uniformly mixing the raw materials of the water phase, and heating to 65-85 ℃ to obtain the water phase;
(5) Emulsification: adding radix Pseudostellariae extract, glutathione solution, oil phase and water phase into homogenizer respectively for emulsification, cooling to 40-50deg.C, adding Ginseng radix essence, stirring, degassing, and cooling to below 40deg.C to obtain radix Pseudostellariae skin caring lotion.
