Description

Fermented traditional Chinese medicine microecological preparation for preventing and treating piglet diarrhea and preparation method thereof
Technical Field
The invention relates to the field of fermented traditional Chinese medicine preparations, in particular to a fermented traditional Chinese medicine microecological preparation for preventing and treating piglet diarrhea and a preparation method thereof.
Background
In the traditional breeding industry, the genetic breeding improvement of the pigs focuses more on the improvement of growth and reproductive performance, but the disease resistance is not obviously improved. The diarrhea of the piglets is one of the most common diseases in the breeding industry, is one of the reasons influencing the development of the pig industry, can commonly occur the diseases, has wide prevalence, has slow growth speed after the disease occurrence, is easy to generate other secondary infectious diseases because the diarrhea causes the physique decline and the immunologic function decline, greatly weakens the resistance to the epidemic diseases, seriously causes the higher mortality of the piglets, and causes great loss to the pig industry.
Antibiotics or synthetic antibacterial drugs are mainly adopted in the aspect of piglet diarrhea drug treatment at the present stage, but drug-resistant bacteria can be caused by long-term use of the drugs, so that the piglet diarrhea treatment difficulty is increased. The biological feed as a live bacterial preparation plays an active role in improving the growth performance and the production performance of pigs and preventing and treating diseases, and can improve the digestibility of the feed while ensuring the balance of the flora in intestinal tracts of piglets and improving the immunity of the intestinal tracts. The Chinese herbal medicine contains natural active substances, has the advantages of no drug resistance, small toxic and side effects, no drug residue and the like compared with antibiotics, and can enhance the resistance of livestock and poultry and improve the immunity of the livestock and poultry.
The modern Chinese medicine fermentation technology is a novel Chinese medicine fermentation process formed by combining modern microecology, fermentation engineering technology, bioengineering and the like on the basis of the traditional Chinese medicine fermentation. At present, a large number of test results show that the modern traditional Chinese medicine fermentation technology can improve the curative effect to a greater extent, change the performance of the medicine and expand the indications of the medicine. Meanwhile, enzymes with various effects generated by microbial growth and metabolism may decompose and convert certain components of traditional Chinese medicines into new effective components or decompose toxic components to reduce toxic and side effects of the medicines. The fermented traditional Chinese medicine microecologics have a lot of research on the influence of the fermented traditional Chinese medicine microecologics on the production performance of live pigs, and a large number of literature reports show that the fermented traditional Chinese medicine microecologics can obviously improve the production performance of the live pigs, but the research on the influence of the fermented microecologics on the diarrhea of piglets is not many.

1 materials and methods
1.1 test drugs, test animals
Test drugs: the fermented traditional Chinese medicine micro-ecological preparation prepared in example 5, florfenicol soluble powder.
Test animals: 60 weaned piglets suffering from diarrhea and about 20 days old are selected from the pig group.
1.2 test methods
Randomly dividing 60 weaned piglets suffering from diarrhea into 3 groups, wherein each group comprises 20 piglets, namely a blank group, a positive control group and a test group; healthy and disease-free 20 piglets are taken as a prevention group; the blank group is administrated with 2mL of physiological saline per kilogram of piglet weight; the positive control group is administrated with florfenicol soluble powder 15mg per kilogram of piglet weight; the experimental group is administrated with 15mg of the fermented traditional Chinese medicine microecological preparation per kilogram of piglet weight; during the test period, all the test piglets are raised under the same condition, are fed freely, drink freely and are fed with less feeding; the mental, fecal and appetite status and death status of the piglets are observed every day, the treatment process is carried out until the piglets are cured or die, and the test is also terminated if the piglets are not cured or die within one week.
1.3 therapeutic efficacy determination
The method has the following advantages: the shape of the excrement is improved, the diarrhea frequency is reduced, and the food intake is increased;
and (3) curing: the food intake is normal, the mental state is good, and the diarrhea symptom disappears;
and (4) invalidation: food intake and diarrhea symptoms did not improve, and diarrhea symptoms even worsened, or died.

Table 2 shows the treatment results of the piglets of each group during the trial. As can be seen from the results in Table 2, the mortality rate of piglets suffering from diarrhea is as high as 65% without drug treatment, which indicates that the diarrhea of piglets has serious harm to the growth of piglets; compared with the blank group, after the piglets are treated, the florfenicol soluble powder and the fermented traditional Chinese medicine microecological preparation have obvious treatment effect on the diarrhea of the piglets, and the effective rate can reach 85 percent; compared with a positive control group, the effective rate of the test group on the treatment of the piglet diarrhea can reach 100 percent, is obviously higher than that of the positive control group, the cure rate can reach 90 percent, is also obviously higher than that of the positive control group, and meanwhile, the test group has no dead piglet cases. The test results show that the treatment effect of the fermented traditional Chinese medicine microecological preparation for treating the diarrhea of the piglets is better than the effective rate and the cure rate of the florfenicol soluble powder for treating the diarrhea of the piglets, the death of the piglets caused by the diarrhea can be effectively avoided, and the fermented traditional Chinese medicine microecological preparation can replace antibiotics to treat the diarrhea of the piglets.

4. the preparation method of the fermented traditional Chinese medicine microecological preparation for preventing and treating piglet diarrhea according to any one of claims 1 to 3, wherein the preparation method comprises the following steps:
(1) selecting genuine medicinal materials such as astragalus, liquorice, madder, purslane, fructus alpiniae oxyphyllae and poplar flower, respectively carrying out immersion extraction for 2 times, then carrying out spray drying on extract liquor, and sieving with a 200-mesh sieve for later use after crushing;
(2) respectively inoculating the bacterial liquid of clostridium butyricum, bacillus subtilis, acidophilic lactobacillus, geobacillus and enterococcus faecalis into respective liquid culture medium by the inoculation amount of 2%, fermenting for 36-72 hours at 30-37 ℃ to obtain each strain culture liquid, wherein the viable bacteria number of each strain culture is 0.5 multiplied by 1010CFU/mL~1.0×1012CFU/mL;
(3) Accurately weighing the traditional Chinese medicine extracts in the step (1) according to the following parts by weight: 10-20 parts of astragalus membranaceus crude extract, 5-10 parts of licorice root crude extract, 5-15 parts of madder root crude extract, 10-15 parts of purslane crude extract, 10-20 parts of fructus alpiniae oxyphyllae crude extract and 5-10 parts of poplar flower crude extract, and uniformly mixing to obtain a traditional Chinese medicine composition for later use;
(4) mixing the traditional Chinese medicine composition with each strain culture solution obtained in the step (2) according to the proportion of adding 30mL of strain culture solution into every 150g of the traditional Chinese medicine composition, and performing closed anaerobic fermentation at the fermentation temperature of 30-37 ℃ for 48-72 hours to obtain fermentation products of each strain for later use;
(5) and (3) fermenting products of clostridium butyricum, bacillus subtilis, acidophilic lactobacillus, geobacillus and enterococcus faecalis obtained in the step (3) according to the viable bacteria number ratio of 0.5-1: 1-2: 0.5-2: 1-2: 0.5-2, uniformly stirring, and freeze-drying to finally obtain the fermented traditional Chinese medicine microecological preparation.
