本申请涉及一种羟丙基淀粉空心胶囊及其制备方法,属于胶囊制剂技术领域。
背景技术
空心胶囊是一种常用要用辅料,除以羟丙基甲基纤维素为原料外,并未见有其他材质的空心胶囊。尽管羟丙甲基纤维素价格并不昂贵,但生产过程中胶液水料较多,除水困难,且胶液糊丝较长,成型难度大,崩解时限也较长。特别是淀粉分子间易形成很强的氢键,淀粉膜易脆易碎,难以制得强度高、耐填装的空心胶囊。
发明内容
有鉴于此,本申请提供一种羟丙基淀粉空心胶囊,针对淀粉膜存在的问题,在淀粉膜中添加常见的碳酸盐及柠檬酸等,调整淀粉胶囊制造工艺,改变淀粉胶囊膜的物理结构,在淀粉膜中生成缓冲结构,从而增加淀粉胶囊的韧性,不仅具有很好的的韧性和强度,脆碎度指标也可以大幅提高。
具体地,本申请是通过以下方案实现的:
一种羟丙基淀粉空心胶囊,由下述质量份的各组分构成:
本申请在溶胶羟丙基淀粉中加入碳酸氢盐、碳酸盐、柠檬酸等辅料,利用碳酸盐受热易分解的原理,控制烘干过程,使胶囊壁中生成气泡微囊,这种气泡微囊会增加胶囊的韧性,使胶囊强度更大,脆碎度指标也有大幅的提高,同时,卡拉胶作为凝胶剂,具有凝胶及促悬浮功能,氯化钾作为卡拉胶的助凝剂,钾离子的存在可以促进卡拉胶的凝胶,其他可溶性钾盐也可以提供钾离子,如本申请中用到碳酸钾或碳酸氢钾,则氯化钾的用量可以减少,还可以减轻胶囊重量,降低原料成本。
进一步的,作为优选:
所述柠檬酸为一种弱酸,可以调节胶液pH值,促进碳酸盐的分解,在本申请中起到有利于胶囊中气泡的生成,降低凝胶温度等作用。也可以考虑用乙酸替代。
所述碳酸氢盐为碳酸氢钠、碳酸氢钾中的一种或两者的混合物,碳酸盐为碳酸钠、碳酸钾中的一种或两者的混合物。碳酸盐及碳酸氢盐受热到45-60℃,可以分解,分解所生成气体会在胶囊膜中生成气泡微囊结构,气泡微囊直径在1-10微米之间。更优选的,所述混合物中,碳酸氢钠与碳酸氢钾为1:1,碳酸钠与碳酸盐的添加比为1:0.6-1。
现有胶囊溶胶工艺中,所有原辅料一次性加入溶胶罐,升温溶胶,作为一种优选,本申请提供一种上述羟丙基淀粉空心胶囊的制备方法,在该制备方法中,首先采用两次添加的方法溶胶:
第一次将羟丙基淀粉、卡拉胶、氯化钾、柠檬酸和水混合,边搅拌边加热至85-95℃,保温1-3小时,降温至35-50℃,再次向胶液中加入碳酸盐、碳酸氢盐,待充分溶解混合后,于40-50℃蘸胶。
其次,现有胶囊制造工艺,烘干过程固定烘干温度为25-30℃,烘干2-3小时,本申请则采用分段烘干工艺:第一阶段25-35℃烘20-40min,第二阶段30-45℃烘20-40min,第三阶段45-60℃烘30-90min,第二阶段15-30℃烘40-90min,并保持第一阶段到第三阶段温度递增,第三阶段到第四阶段递减。第一阶段烘干使胶囊定型,第二阶段使胶囊水分含量在30-60%,第三阶段使胶囊膜中碳酸盐分解形成气泡微囊结构,第四阶段使胶囊烘干至含水量不超过15%。
具体实施方式
实施例1
取重量比例为95.1份的羟丙基淀粉、2.5份的卡拉胶、1.5份的氯化钾、0.5份的碳酸氢钠、0.3份的碳酸钠和0.1份的柠檬酸。将羟丙基淀粉、卡拉胶、氯化钾和柠檬酸与水混合,含固量为25%,边搅拌边加热到90℃,保温2小时,降温到45℃,加入碳酸钾、碳酸钠、碳酸氢钠,使其充分溶解,混合均匀,再进行蘸胶、烘干、剪切、套胶。蘸胶温度45℃。烘干过程分四个阶段,第一阶段烘干温度为30℃,烘干时间为0.5小时;第二阶段烘干温度为40℃,烘干时间为0.5小时;第三阶段烘干温度为50℃,烘干时间为1小时;第四阶段烘干温度为25℃,烘干时间为1小时。
实施例2
取重量比例为95.8份的羟丙基淀粉、2.5份的卡拉胶、0.8份的氯化钾、0.5份的碳酸氢钾、0.3份的碳酸钾和0.1份的柠檬酸。将羟丙基淀粉、卡拉胶、氯化钾,和柠檬酸与水混合,含固量为25%,边搅拌边加热到90℃,保温2小时,降温到45℃,加入碳酸氢钾、碳酸钾,使其充分溶解,混合均匀,再进行蘸胶、烘干、剪切、套胶。蘸胶温度45℃。烘干过程,第一阶段烘干温度为30℃,烘干时间为0.5小时;第二阶段烘干温度为40℃,烘干时间为0.5小时;第三阶段烘干温度为55℃,烘干时间为1小时;第四阶段烘干温度为25℃,烘干时间为1小时。
实施例3
取重量比例为95.6份的羟丙基淀粉、2.0份的卡拉胶、0.8份的氯化钾、0.5份的碳酸氢钠,0.5份的碳酸氢钾、0.3份的碳酸钾和0.3份的柠檬酸。将羟丙基淀粉、卡拉胶、氯化钾,和柠檬酸与水混合,含固量为25%,边搅拌边加热到90℃,保温2小时,降温到45℃,加入碳酸钾氢、碳酸氢钾、碳酸钾,使其充分溶解,混合均匀,再进行蘸胶、烘干、剪切、套胶。蘸胶温度45℃。烘干过程,第一阶段烘干温度为30℃,烘干时间为0.5小时;第二阶段烘干温度为40℃,烘干时间为0.5小时;第三阶段烘干温度为52℃,烘干时间为1小时;第四阶段烘干温度为25℃,烘干时间为1小时。

其中,对比样羟丙基淀粉胶囊的配方为:羟丙基淀粉97份,卡拉胶2份,氯化钾1份,配置成20%含量水悬浮液,90℃溶胶,40℃蘸胶,25-30℃烘干3小时。
由表1可见,对比样与实施例胶囊含水量都符合要求;胶囊重量,实施例比对比样减少了1-2%,显著减少了原材料的用量;对比样羟丙基淀粉胶囊的脆碎度指标不符合药典要求,而实施例通过在胶囊制造过程中产生微囊结构,起到了增强韧性的效果,脆碎度指标符合中国药典要求。故本申请对于提高胶囊的脆碎度指标具有良好的应用效果。
Claims (5)
1.一种羟丙基淀粉空心胶囊,其特征在于,由下述质量份的各组分构成:
羟丙基淀粉: 75-96,
卡拉胶: 1-5,
氯化钾: 0.5-3,
柠檬酸: 0.1-3,
碳酸盐: 0.1-3,
碳酸氢盐: 0.1-5,
所述碳酸盐为碳酸钠、碳酸钾中的一种或两者的混合物,
将羟丙基淀粉、卡拉胶、氯化钾、柠檬酸和水混合,边搅拌边加热至85-95℃,保温1-3小时,降温至35-50℃,加入碳酸盐、碳酸氢盐,待充分溶解后,于40-50℃蘸胶处理后,分段烘干,
所述分段烘干具体工艺为:第一阶段25-35℃烘20-40min,第二阶段30-45℃烘20-40min,第三阶段45-60℃烘30-90min,第二阶段15-30℃烘40-90min,并保持第一阶段到第三阶段温度递增,第三阶段到第四阶段递减。
2.根据权利要求1所述的一种羟丙基淀粉空心胶囊,其特征在于:所述柠檬酸采用乙酸替代。
3.根据权利要求1所述的一种羟丙基淀粉空心胶囊,其特征在于:所述碳酸氢盐为碳酸氢钠、碳酸氢钾中的一种或两者的混合物。
4.根据权利要求3所述的一种羟丙基淀粉空心胶囊,其特征在于:碳酸氢钠与碳酸氢钾为1:1。
5.根据权利要求1所述的一种羟丙基淀粉空心胶囊,其特征在于:所述碳酸钠与碳酸盐的添加比为1:0.6-1。
