技术领域
本发明属于功能食品及保健食品领域,具体涉及一种抗衰老组合物及其制备方法。
背景技术
人体衰老是身体各个系统及各种器官和组织在生长发育成熟后,随增龄而逐步出现的各种生理的、代谢的和功能的改变。衰老作为一种复杂的生理机制,目前存在损伤学说、遗传学说、自由基学说、免疫学说、端粒学说等。衰老的自由基学说是Denham Harman在1956年提出的,认为衰老过程中的退行性变化是由于细胞正常代谢过程中产生自由基的有害作用造成的。这里所说的自由基主要就是氧自由基,例如超氧阴离子自由基、羟自由基、脂氧自由基、二氧化氮和一氧化氮自由基。而维持体内适当水平的抗氧化剂和自由基消除剂水平可以延长寿命和推迟衰老。
人衰老是个缓慢的过程,是因为人机体内存在自由基清除系统,其可以最大限度地防御自由基的损伤。自由基清除系统包括酶促反应和非酶促反应两部分。酶促反应所需酶有谷胱甘肽过氧化物酶(GSH-PX)、超氧化物歧化酶(superoxide dismutase,SOD)、过氧化物酶(POP)及过氧化氢酶(catalase,CAT)。非酶促反应的作用物质主要为一些低分子的化合物,是一些抗氧化作用的物质,统称抗氧化剂(antioxidant),主要有谷胱甘肽(GSH)、维生素C、β~胡萝卜素、维生素E、半胱氨酸、硒化物、巯基乙醇等。
正常的我们人体是处于产生和消除自由基的正常平衡的,但是由于生活中的污染严重,生活习惯的改变影响等,自由基会产生过多或者是清除速度减慢,从而破坏了正常的细胞,加快人体的衰老。为了延缓衰老,需要从食物中摄取合适的抗氧化物质,消除过多的氧自由基,及时准确修复自由基带来的损伤。
例如,中国专利CN110663955公开了一种抗糖化防衰老的组合物及其制备方法,其包括金银花10-20份、番石榴叶2-9份、玉竹8-15份、桑叶3.5-14份、针叶樱桃果7-20份、抗氧化物质12-47份、抗糖化物质2-9份和营养增补剂7.76-32.5份。其中抗氧化物质包括黑果腺肋花楸果2-8等,营养增补包括胶原蛋白肽3-10份等。其通过金银花,番石榴叶,玉竹,桑叶和针叶樱桃果为主材料,配以抗氧化物质和抗糖化物质,共同起到抗糖化防衰老的功效,但该专利主要功效为抗糖化,防衰老功效有效进一步提高。
发明内容
本发明的目的在于提供一种抗衰老组合物及其制备方法,本发明提供的组合物消除自由基能力强,具有很好的抗衰老作用。
本发明的技术方案为:
一种抗衰老组合物,按重量份计,包括鱼胶原蛋白肽80~100份、黑果腺肋花楸果浓缩汁8~20份、维生素C 4~10份、百香果浓缩粉0.03~3份、米胚芽粉0.03~3份、γ-氨基丁酸1-8份和烟酰胺0.008~0.07份。
优选的,还包括柠檬浓缩汁4~10份。
进一步优选的,还包括酸度调节剂2~8份、增稠剂10~20份、甜味剂30~40份、香精1~5份和抗性糊精10~20份。
进一步优选的,所述酸度调节剂为无水柠檬酸、苹果酸、酒石酸或富马酸中的一种或多种;所述增稠剂为果胶、魔芋粉、圆苞车前子壳、黄原胶、海藻酸钠中的一种或多种;所述甜味剂由甜菊糖苷和赤藓糖醇组成。

一种制备上述的抗衰老的组合物的方法,包括如下步骤:(1)将增稠剂和甜味剂加入到65~75℃水中搅拌,得液体a;(2)将鱼胶原蛋白肽、百香果浓缩粉、米胚芽粉、烟酰胺加入到液体a中,制得液体b;(3)将酸味剂、维生素C、柠檬浓缩汁、黑果腺肋花楸果浓缩汁加入到水中,制得酸液;(4)将所述酸液加入到所述液体b中,即得。
一种上述抗衰老的组合物,制备方法为,将所述原料添加适宜的辅料,用常规的方法制备为片剂,胶囊剂,粉剂或颗粒剂。

黑果腺肋花楸果又称野樱莓、不老莓,是集食用、园林和生态价值于一身的珍贵树种。其果实中花青素、黄酮、多酚是已知植物中含量最高的。多酚具有强抗氧化能力和清除自由基能力。花青素具有抗皱作用是基于它能维护胶原的合成,抑制破坏弹性蛋白的酶,协助机体保护胶原蛋白和改善皮肤的弹性,改善皮肤的健康循环,从而减缓皱纹和过早老化。
维生素C可使脯氨酸与赖氨酸羟化为羟脯氨酸与羟赖氨酸,羟脯氨酸与羟赖氨酸是胶原蛋白的重要成分。是高效的抗氧化剂。它能逐级供给电子而变成三脱氢抗坏血酸和脱氢抗坏血酸,以清除氧自由基和羟基自由基。维生素C能抑制酪氨酸酶的活性,阻碍黑色素的生成,保护皮肤不受紫外线的伤害,并改善皮肤暗沉。

本发明中鱼胶原蛋白肽的制备方法为以鱼鳞或鱼皮为主要原料经过酶处理、透析、干燥等工艺得到,分子量1000Da。黑果腺肋花楸果浓缩汁的制备方法为果实经清洗、拣选、破碎、酶处理、压汁、巴氏灭菌、脱胶、超滤、蒸发浓缩得到,浓缩比例为6:1。柠檬浓缩汁为为果实经清洗、拣选、破碎、酶处理、压汁、巴氏灭菌、脱胶、超滤、蒸发浓缩得到,浓缩比例为5:1。百香果浓缩粉为经提取、浓缩制成的粉末,选用为日本森永制药株式会社。米胚芽粉为米胚芽粉为大米的胚芽制成的粉末,选用为日本欧力嘉生化有限责任公司。
实施例1
本实施例提供了一种抗衰老组合物,包括鱼胶原蛋白肽89g、黑果腺肋花楸果浓缩汁12g,维生素C 7g,柠檬浓缩汁5g,百香果浓缩粉0.2g,米胚芽粉0.3g,烟酰胺0.03g,γ-氨基丁酸3g,无水柠檬酸2.6g,DL-酒石酸2g,果胶15g,抗性糊精17g,甜菊糖苷0.2g,赤藓糖醇35g,香精3g。
制备方法为:
(1)将果胶15g,赤藓糖醇35g,甜菊糖苷0.2g,加入到温度为70℃的60mL水中搅拌均匀,至温度冷却至室温后得液体a;

本实施例提供了一种抗衰老组合物,包括鱼胶原蛋白肽85g、黑果腺肋花楸果浓缩汁10g,维生素C 6g,柠檬浓缩汁6g,百香果浓缩粉0.1g,米胚芽粉0.5g,烟酰胺0.01g,γ-氨基丁酸4g,无水柠檬酸2.4g,DL-酒石酸2g,果胶14g,抗性糊精15g,甜菊糖苷0.4g,赤藓糖醇30g,香精3g。
制备方法为:
(1)将果胶14g,赤藓糖醇30g,甜菊糖苷0.4g,加入到温度为70℃的60mL水中搅拌均匀,至温度冷却至室温后得液体a;
(2)将鱼胶原蛋白肽85g,百香果浓缩粉0.1g,米胚芽粉0.5g,抗性糊精15g,烟酰胺0.01g加入液体a中,混合均匀得液体b;
(3)将无水柠檬酸2.4g,DL-酒石酸2g,维生素C 6g,柠檬浓缩汁6g,黑果腺肋花楸果浓缩汁10g,加入到20mL水中,制得酸液;
(4)将酸液加入到液体b中,加入香精3g,混合均匀,定容至100mL得产品。
实施例3
本实施例提供了一种抗衰老组合物,包括鱼胶原蛋白肽80g、黑果腺肋花楸果浓缩汁20g,维生素C 4g,柠檬浓缩汁10g,百香果浓缩粉0.03g,米胚芽粉3g,烟酰胺0.008g,γ-氨基丁酸1g,无水柠檬酸1g,DL-酒石酸4g,果胶10g,抗性糊精20g,甜菊糖苷0.1g,赤藓糖醇32g,香精3g。
制备方法为:
(1)将果胶10g,赤藓糖醇32g,甜菊糖苷0.1g,加入到温度为70℃的60mL水中搅拌均匀,至温度冷却至室温后得液体a;
(2)将鱼胶原蛋白肽80g,百香果浓缩粉0.03g,米胚芽粉3g,抗性糊精20g,烟酰胺0.008g加入液体a中,混合均匀得液体b;
(3)将无水柠檬酸1g,DL-酒石酸4g,维生素C 4g,柠檬浓缩汁10g,黑果腺肋花楸果浓缩汁20g,加入到20mL水中,制得酸液;
(4)将酸液加入到液体b中,加入香精3g,混合均匀,定容至100mL得产品。
实施例4
本实施例提供了一种抗衰老组合物,包括鱼胶原蛋白肽100g、黑果腺肋花楸果浓缩汁8g,维生素C10g,柠檬浓缩汁4g,百香果浓缩粉3g,米胚芽粉0.03g,烟酰胺0.07g,γ-氨基丁酸8g,无水柠檬酸4g,DL-酒石酸1g,果胶20g,抗性糊精10g,甜菊糖苷3g,赤藓糖醇35g,香精3g。
制备方法为:
(1)将果胶20g,赤藓糖醇35g,甜菊糖苷3g,加入到温度为70℃的60mL水中搅拌均匀,至温度冷却至室温后得液体a;
(2)将鱼胶原蛋白肽100g,百香果浓缩粉3g,米胚芽粉0.03g,抗性糊精10g,烟酰胺0.07g加入液体a中,混合均匀得液体b;
(3)将无水柠檬酸4g,DL-酒石酸1g,维生素C10g,柠檬浓缩汁4g,黑果腺肋花楸果浓缩汁8g,加入到20mL水中,制得酸液;
(4)将酸液加入到液体b中,加入香精3g,混合均匀,定容至100mL得产品。
实施例5
本实施例提供了一种抗衰老组合物,包括鱼胶原蛋白肽80g、黑果腺肋花楸果浓缩汁12g,维生素C 4g,柠檬浓缩汁7g,百香果浓缩粉0.03g,米胚芽粉0.8g,烟酰胺0.02g,γ-氨基丁酸5g,无水柠檬酸3g,DL-酒石酸1g,果胶15g,抗性糊精12g,甜菊糖苷0.5g,赤藓糖醇34g,香精3g。
制备方法为:
(1)将果胶15g,赤藓糖醇34g,甜菊糖苷0.5g,加入到温度为70℃的60mL水中搅拌均匀,至温度冷却至室温后得液体a;
(2)将鱼胶原蛋白肽80g,百香果浓缩粉0.03g,米胚芽粉0.8g,抗性糊精12g,烟酰胺0.02g加入液体a中,混合均匀得液体b;
(3)将无水柠檬酸3g,DL-酒石酸1g,维生素C 4g,柠檬浓缩汁3g,黑果腺肋花楸果浓缩汁12g,加入到20mL水中,制得酸液;
(4)将酸液加入到液体b中,加入香精3g,混合均匀,定容至100mL得产品。
实施例6
本实施例提供了一种抗衰老组合物,包括鱼胶原蛋白肽90g、黑果腺肋花楸果浓缩汁8g,维生素C 8g,柠檬浓缩汁4g,百香果浓缩粉1.5g,米胚芽粉0.03g,烟酰胺0.03g,γ-氨基丁酸2g,无水柠檬酸2g,DL-酒石酸3g,果胶13g,抗性糊精18g,甜菊糖苷0.3g,赤藓糖醇33g,香精3g。
制备方法为:
(1)将果胶13g,赤藓糖醇33g,甜菊糖苷0.3g,加入到温度为70℃的60mL水中搅拌均匀,至温度冷却至室温后得液体a;
(2)将鱼胶原蛋白肽90g,百香果浓缩粉1.5g,米胚芽粉0.03g,抗性糊精18g,烟酰胺0.03g加入液体a中,混合均匀得液体b;
(3)将无水柠檬酸2g,DL-酒石酸3g,维生素C 8g,柠檬浓缩汁4g,黑果腺肋花楸果浓缩汁8g,加入到20mL水中,制得酸液;
(4)将酸液加入到液体b中,加入香精3g,混合均匀,定容至100mL得产品。
实施例7
本实施例提供了一种抗衰老组合物,包括鱼胶原蛋白肽89g、黑果腺肋花楸果浓缩汁12g,维生素C 7g,百香果浓缩粉0.2g,米胚芽粉0.3g,烟酰胺0.03g,γ-氨基丁酸3g,无水柠檬酸2.6g,DL-酒石酸2g,果胶15g,抗性糊精17g,甜菊糖苷0.2g,赤藓糖醇35g,香精3g。
制备方法为:
(1)将果胶15g,赤藓糖醇35g,甜菊糖苷0.2g,加入到温度为70℃的60mL水中搅拌均匀,至温度冷却至室温后得液体a;
(2)将鱼胶原蛋白肽89g,百香果浓缩粉0.2g,米胚芽粉0.3g,抗性糊精17g,烟酰胺0.03g加入液体a中,混合均匀得液体b;
(3)将无水柠檬酸2.6g,DL-酒石酸2g,维生素C 7g,黑果腺肋花楸果浓缩汁12g,加入到20mL水中,制得酸液;
(4)将酸液加入到液体b中,加入香精3g,混合均匀,定容至100mL得产品。
对比例1
本对比例提供了一种抗衰老组合物,包括黑果腺肋花楸果浓缩汁36g,无水柠檬酸2.6g,DL-酒石酸2g,果胶15g,抗性糊精17g,甜菊糖苷0.2g,赤藓糖醇35g,香精3g。
制备方法为:
(1)将果胶15g,赤藓糖醇35g,甜菊糖苷0.2g,加入到温度为70℃的60mL水中搅拌均匀,至温度冷却至室温后得液体a;
(2)将抗性糊精17g加入液体a中,混合均匀得液体b;
(3)将无水柠檬酸2.6g,DL-酒石酸2g,黑果腺肋花楸果浓缩汁36g,加入到20mL水中,制得酸液;
(4)将酸液加入到液体b中,加入香精3g,混合均匀,定容至100mL得产品。
本对比例1同实施例1的区别在于,功效成分仅为黑果腺肋花楸果浓缩汁,且含量为实施例1中的3倍。
对比例2
该对比例同实施例1的区别在于,无米胚芽粉和γ-氨基丁酸,其余组分含量及制备方法与实施例1相同。
实验例1羟自由基清除率实验
方法:取1.0mL浓度为1.865mmol/L的邻二氮菲的无水乙醇溶于带塞试管里,分别加入浓度为0.2mol/L的pH 7.4的磷酸盐缓冲液2mL和1mL实施例1-7和对比例1-2的抗衰老组合物,充分混匀后加入1.0mL浓度为1.865mmol/L的FeSO4.7H2O溶液,再次混匀后加入1.0mL 0.03%(v/v)的H2O2,于37℃恒温水浴,60min后,在536nm下分别测量各组混合溶液的吸光度值,A1,以蒸馏水代替样实施例和对比例组合物为空白组测吸光度值,得A2,以蒸馏水替代H2O2作为损伤组,测其吸光度值为A3,样品的羟基自由基的清除率按以下公式计算羟基自由基清除率(%)=[(A1-A3)/(A2-A3)]*100%,取三次测量的平均值,检测结果如表1所示。
实验例2 DPPH自由基清除率实验
方法:取3mL浓度为0.0178mmol/L的DPPH-无水甲醇溶液于比色皿中,再加入3mL实施例1-7和对比例1-2的抗衰老组合物,混匀立即在517nm波长处测吸光值测,吸光值记为A1,再在37℃下避光保存30min后测吸光值,记为A2,空白对照为只加DPPH-无水甲醇溶液,其吸光值记为A3。按下式计算DPPH自由基清除率:DPPH自由基清除率(%)=[1-(A1-A2)/A3]*100%,检测结果如表1所示。

由表1可以看出,实施例1-7均具有较好的羟自由基消除率和DPPH自由基消除率,其中实施例1和2具有较优的羟基自由基和DPPH自由基消除能力。实施例1中羟自由基消除率和DPPH自由消除能力是对比例1的1.597倍和1.6倍,表明黑果腺肋花楸果和鱼胶原蛋白肽、维生素C、柠檬浓缩汁、百香瓜浓缩粉、米胚芽粉、烟酰胺和γ-氨基丁酸协同配合作用要优于黑果腺肋花楸果单独效果。实施例1中羟自由基消除率和DPPH自由消除能力是对比例2的1.47倍和1.46倍,表明米胚芽粉和γ-氨基丁酸的加入提高了自由基消除能力,更利于抗氧化。
实验例3人体试用评价试验
试验总人数300人,分为5组,每组60人。分别服用实施例1,实施例2,实施例7和对比例1-2的抗衰老组合物,每人每天服用30mL/2次,为期28天。其中男性144人,女性156人。男女比例:男性144/300人=48%,女性156/300人=52%。年龄范围:25-65岁。年龄比例:25-35岁:83人/300人=27.67%。36-45岁:77人/300人=25.67%。46-55岁:84人/300人=28%。56-65岁:56人/300人=18.67%。试服结束后,以回访的方式进行记录。回访内容如表2所示

试验结果
试验过程中有女性8人,男性3人中途退出。总计试用人数289人。年龄比例:25-35岁:81人/289人=28.03%;36-45岁:73人/289人=25.26%;46-55岁:83人/289人=28.72%;56-65岁:52人/289人=17.99%。上述症状有缓解的人数及比例汇总结果如表3所示

结论:体感反馈(服用28天):本发明组合物在皮肤,睡眠、体力方面有明显的提高,关节和头发方面有一些改善,具有抗衰效果。整体效果较好。通过分组对比试验,本发明产品(实施例1)在改善睡眠、皮肤状态、改善精力、改善关节、整体年轻感方面,具有最佳抗衰效果。
显然,上述实施例仅仅是为清楚地说明所作的举例,而并非对实施方式的限定。对于所属领域的普通技术人员来说,在上述说明的基础上还可以做出其它不同形式的变化或变动。这里无需也无法对所有的实施方式予以穷举。而由此所引伸出的显而易见的变化或变动仍处于本发明创造的保护范围之中。
Claims (10)
1.一种抗衰老组合物,其特征在于,按重量份计,包括鱼胶原蛋白肽80~100份、黑果腺肋花楸果浓缩汁8~20份、维生素C 4~10份、百香果浓缩粉0.03~3份、米胚芽粉0.03~3份、γ-氨基丁酸1-8份和烟酰胺0.008~0.07份。
2.根据权利要求1所述的抗衰老组合物,其特征在于,还包括柠檬浓缩汁4~10份。
3.根据权利要求1或2所述的抗衰老组合物,其特征在于,还包括酸度调节剂2~8份、增稠剂10~20份、甜味剂30~40份、香精1~5份和抗性糊精10~20份。
4.根据权利要求3所述的抗衰老组合物,其特征在于,所述酸度调节剂为无水柠檬酸、苹果酸、酒石酸或富马酸中的一种或多种;所述增稠剂为果胶、魔芋粉、圆苞车前子壳、黄原胶、海藻酸钠中的一种或多种;所述甜味剂由甜菊糖苷和赤藓糖醇组成。
5.根据权利要求4所述的抗衰老的组合物,其特征在于,按重量份计,包括鱼胶原蛋白肽80~100份、黑果腺肋花楸果浓缩汁8~20份、维生素C 4~10份、柠檬浓缩汁4~10份、百香果浓缩粉0.03~3份、米胚芽粉0.03~3份、γ-氨基丁酸1-8份、烟酰胺0.008~0.07份、无水柠檬酸1~4份、酒石酸1~4份、果胶10~20份、抗性糊精10~20份、甜菊糖苷0.1~3份、赤藓糖醇30~35份和香精1~5份。
6.根据权利要求4所述的抗衰老的组合物,其特征在于,按重量份计,包括鱼胶原蛋白肽80~90份、黑果腺肋花楸果浓缩汁8~12份、维生素C 4~8份、柠檬浓缩汁4~7份、百香果浓缩粉0.03~1.5份、米胚芽粉0.03~0.8份、γ-氨基丁酸2-5份、烟酰胺0.008~0.03份、无水柠檬酸2~4份、酒石酸1~3份、果胶10~15份、抗性糊精12~18份、甜菊糖苷0.1~0.5份、赤藓糖醇30~34份和香精2~4份。
7.根据权利要求4所述的抗衰老的组合物,其特征在于,按重量份计,包括鱼胶原蛋白肽89份、黑果腺肋花楸果浓缩汁12份、维生素C 7份、柠檬浓缩汁5份、百香果浓缩粉0.2份、米胚芽粉0.3份、烟酰胺0.03份、γ-氨基丁酸3份,无水柠檬酸2.6份、酒石酸2份、果胶15份、抗性糊精17份、甜菊糖苷0.2份、赤藓糖醇35份和香精3份。
8.根据权利要求4所述的抗衰老的组合物,其特征在于,按重量份计,包括鱼胶原蛋白肽85份、黑果腺肋花楸果浓缩汁10份、维生素C 6份、柠檬浓缩汁6份、百香果浓缩粉0.1份、米胚芽粉0.5份、烟酰胺0.01份、γ-氨基丁酸4份、无水柠檬酸3份、酒石酸2份、果胶14份、抗性糊精15份、甜菊糖苷0.4份、赤藓糖醇30份和香精3份。
9.一种制备权利要求1-8任一所述的抗衰老的组合物的方法,其特征在于,包括如下步骤:
(1)将增稠剂和甜味剂加入到65~75℃水中搅拌,得液体a;
(2)将鱼胶原蛋白肽、百香果浓缩粉、米胚芽粉、烟酰胺加入到液体a中,制得液体b;
(3)将酸味剂、维生素C、柠檬浓缩汁、黑果腺肋花楸果浓缩汁加入到水中,制得酸液;
(4)将所述酸液加入到所述液体b中,即得。
10.根据权利要求1所述的抗衰老的组合物,其特征在于,制备方法为,将所述原料添加适宜的辅料,用常规的方法制备为片剂,胶囊剂,粉剂或颗粒剂。
