Abstract
本发明提供一种具有清除自由基功能的抗衰老组合物及其制备方法,主要由鱼胶原蛋白肽粉、西柚粉、蔓越莓粉、L‑阿拉伯糖、针叶樱桃果粉、雨生红球藻、燕窝肽、罗汉果甜苷等原料制备而成,采用特定的工艺对鱼皮进行处理制备得到独特的鱼胶原蛋白肽粉,以及特定的工艺对西柚、蔓越莓和针叶樱桃果进行处理,复配以其他成分,制备得到一种具有高清除自由基活性,和高抗衰老性能的,便捷食用,口感愉悦的组合物,并意外的发现所述组合物无需低温条件下进行保存,在常温下保存也能长期的保持高的清除自由基活性。

技术领域
本发明涉及医药保健品领域,尤其涉及一种具有清除自由基功能的抗衰老组合物及其制备方法。
背景技术
自由基,化学上称为“游离基”,它是含有一个不成对电子的原子团,是人体细胞在代谢过程中产生一类非常活泼、有很强氧化作用的化学物质,也叫活性氧,如羟自由基、超氧阴离子自由基、过氧化氢等。体内活性氧自由基具有一定的功能,如免疫和信号传导过程,但过多的活性氧自由基就会有破坏行为,攻击细胞膜,氧化血清抗蛋白酶,甚至抢夺DNA的电子,损伤人体健康细胞,诱发心血管疾病、糖尿病、老年性黄斑病变、癌症等疾病。此外,外界环境中的阳光辐射、空气污染、不良生活嗜好(吸烟、酗酒)、各种食材的农药残留等都会使人体产生更多活性氧自由基,使核酸突变,这是人类衰老和患病的根源。
近年来,国内外对自由基及自由基清除剂的研究十分活跃,在各类食品科学、生命科学及医学书籍上都有许多关于自由基及其清除剂的研究报道,自由基清除剂作为功能性食品的重要原料成分之一,通过人们日常消费的食品来调节人体内自由基的平衡,已经受到食品营养学的广泛重视。人体内自由基的清除,一是靠生物合成的抗氧化酶与内源性抗氧化剂,另一是靠外源性的抗氧化剂。抗氧化酶主要昧超氧化物歧化酶、过氧化氢酶、谷胱甘肽过氧化物酶与谷胱甘肽转硫酶;外源性抗氧化物来自食物与饮料。在营养素中显示清除活性氧自由基的有维生素C、维生素E以及β-胡萝卜素。但在食物中尚有一些可在体内发挥重要抗氧化功能的化合物,统称为植物化合物,包括类胡萝卜素、生物黄酮、植物固醇等。
在日常的生理情况下,人体内氧自由基虽不断产生,但也不断被清除。处于平衡状态自由基浓度是较低的,它们不仅不会损伤机体,而且会显示独特的生理作用,对皮肤中生物大分子的损伤及修复启动一系列程序。但人体处于衰老、应激、压力过大的特殊情况下,人体内氧自由基的产量会有所增加或清除的能力有所减弱。而为了维持或提高皮肤的质量,延缓衰老,就不仅要适当提高胶原蛋白的营养供给量,还要使营养有助于防止或纠正氧自由基产生与清除的失衡,从食物中摄取合适的抗氧化物质,利于清除过多的氧自由基,及时准确修复自由基带来的损伤。
由于具有清除自由基功能的活性较高,存储条件苛刻,大多需要低温、干燥或避光条件下进行保存,导致生成运输成本升高,且消费者购买后由于无法严格按照保存条件进行保存,会导致产品的活性降低,无法起到预期的效果。因此,研制出一款产品配方合理,存储条件简单、便捷食用,口感愉悦,适用于想维持青春容貌、清除自由基、抗氧化、抗衰老的广大人群食用的产品,特别符合女性市场的需求。
发明内容
为了克服现有技术的不足,本发明提供一种具有清除自由基功能的抗衰老组合物,主要由鱼胶原蛋白肽粉、西柚粉、蔓越莓粉、L-阿拉伯糖、针叶樱桃果粉、雨生红球藻、燕窝肽、罗汉果甜苷等原料制备而成,采用特定的工艺对鱼皮进行处理制备得到独特的鱼胶原蛋白肽粉,以及特定的工艺对西柚、蔓越莓和针叶樱桃果进行处理,复配以其他成分,制备得到一种具有高清除自由基活性,和高抗衰老性能的,便捷食用,口感愉悦的组合物,并意外的发现所述组合物无需低温条件下进行保存,在常温下保存也能长期的保持高的清除自由基活性。
为了达到上述技术效果,本发明提供一种具有清除自由基功能的抗衰老组合物,包括以下重量份的原料:
鱼胶原蛋白肽粉50-100份、西柚粉10-40份、蔓越莓粉5-20份、L-阿拉伯糖2-15份、针叶樱桃果粉2-15份、雨生红球藻提取物0.2-4份、燕窝肽0.2-4份、罗汉果甜苷0.02-1份。
作为优选,所述具有清除自由基功能的抗衰老组合物,包括以下重量份的原料:鱼胶原蛋白肽粉50-80份、西柚粉10-25份、蔓越莓粉5-10份、L-阿拉伯糖2-8份、针叶樱桃果粉2-10份、雨生红球藻提取物0.2-2份、燕窝肽0.2-2份、罗汉果甜苷0.02-0.5份。
进一步,所述鱼胶原蛋白肽粉的制备方法,包括以下步骤:
(1)将鱼皮放入质量分数为1-2%的氯化钠溶液中,在15-25℃的低温下浸泡5-8小时,然后水洗至中性,沥干水分,粉碎,得到鱼胶原蛋白粗提物;
(2)向鱼胶原蛋白粗提物中加入去离子水,调节pH,加入碱性蛋白酶,在50-65℃下酶解处理3-5小时;然后加入木瓜和无花果酶原,在45-60℃条件下继续酶解处理2-4小时;灭酶,离心过滤得到胶原蛋白肽液
(3)向胶原蛋白肽液中加入pH值为5.5-6.5的磷酸盐缓冲液,以及脱乙酰度大于85%的壳聚糖,其中,三者的重量份数如下:胶原蛋白肽液10-20份,磷酸盐缓冲液20-40份,壳聚糖5-10份,经超声波分散30-60min,离心过滤后,透析48h后,喷雾干燥得到鱼胶原蛋白肽粉。
进一步,所述西柚粉和针叶樱桃果粉的制备方法,包括以下步骤:
(1)将西柚、或针叶樱桃果制成浆液,再加入脱涩液搅拌,经过胶磨、均质、过滤后得到第一过滤液,所述脱涩液为质量分数0.5-1%的柠檬酸和质量分数0.2-0.4%份氯化钠按照质量比3∶1的混合物;
(2)向步骤(1)制备得到的滤渣中加入占滤渣质量0.2-0.4%的果胶酶和0.1-0.2%的纤维素酶,加热至40-55℃酶解6-10h;再次胶磨均质、过滤后得到第二过滤液;
(3)将第一滤液和第二滤液混合,加入占滤液质量的1-3%的海藻糖,经真空浓缩,冷冻干燥后得到西柚粉、或针叶樱桃果粉。
进一步,所述蔓越莓粉的制备方法,包括以下步骤:
(1)将蔓越莓洗净后置于汽爆罐内,通入氮气至汽爆罐内压力为1.7-2.3MPa,蒸汽爆破处理1-3min后,冷却至室温后,加入由纤维素酶∶果胶酶∶碱性蛋白酶按质量比2∶2∶3混合所得的复合酶,复合酶用量为原料重量的0.7-2.3%,保持在30-45℃下酶解0.5-2h,过滤,得第一滤液
(2)将步骤(1)制备得到的滤渣加入萃取釜中进行超临界CO2萃取,乙醇为夹带剂,萃取压力40-45Mpa,萃取温度为40-45℃,CO2流量15-20L/h,萃取时间40-60min,得到萃取物;
(3)将步骤(1)得到的第一滤液和步骤(2)得到的萃取物混合均匀,经真空浓缩,冷冻干燥后得到粉末,加入占粉末质量5-10%的麦芽糊精,混合均匀制备得到蔓越莓粉。
进一步,所述雨生红球藻提取物的制备方法,包括以下步骤:
(1)采用人工LED光源对绿色藻液进行光照积累虾青素,直到藻液呈鲜红色时进行离心或沉降采收;
(2)将步骤(2)采收的雨生红球藻在80-90℃下干燥至含水量低于10%,加入汽爆罐内,通入氮气至汽爆罐内压力为1.7-23MPa,蒸汽爆破处理1-3min后,冷却至室温后;
(3)将步骤(2)制备得到混合物加入萃取釜中进行超临界CO2萃取,乙醇为夹带剂,萃取压力40-45Mpa,萃取温度为45-55℃,CO2流量20-25L/h,萃取时间60-80min,得到萃取物;
(4)向步骤(3)得到的萃取物中加入占萃取物质量的1-3%的海藻糖,经冷冻干燥制备得到雨生红球藻提取物。
进一步,所述燕窝肽的制备方法,包括以下步骤:
(1)将燕窝用去离子水清洗干净后,在35-40℃下风干,粉碎成60-120目,再采用气流粉碎法粉碎至颗粒直径在4-12μm,得细燕窝粉,然后按照料液比1∶5-10加入去离子水,在搅拌速度为120-150r/min的条件下搅拌20-30min,得到燕窝混合浆;
(2)将步骤(1)中的燕窝混合浆放置纳米湿法粉碎机中粉碎、乳化30-60min,粉碎至颗粒直径在30-120nm之间,然后在40-50℃的条件下干燥,得到饼状燕窝;
(3)将步骤(2)中的饼状燕窝放置于研磨机中研磨1-2h后移至超细粉碎机深度粉碎至颗粒的直径范围在30-180nm,得超细燕窝粉,然后按照料液比1∶10-15加入去离子水搅拌均匀,再置于水浴罐中进行超声水浴震荡,水浴时间为3-5h,温度为70-90℃,得到超细燕窝混合液;
(4)将步骤(3)中的超细燕窝混合液用7000-8000道尔顿的超滤膜进行离心超滤,再用葡聚糖凝胶柱层析,收集300nm处吸收峰组分,然后冷冻干燥,得到燕窝肽粉。
本发明的另一个目的在于提供一种具有清除自由基功能的抗衰老组合物的制备方法,将所有原料混合均匀,然后使用40-60目筛网过筛,即可。
本发明所述的一种具有清除自由基功能的抗衰老组合物可制备成固体或液体制剂,所述固体制剂为胶囊剂、片剂、粉剂、泡腾剂或颗粒剂。
本发明所述的一种具有清除自由基功能的抗衰老组合物在制备制剂时可加入辅料填充剂、崩解剂、润滑剂、泡腾崩解剂等,填充剂选自淀粉、可压性淀粉、微晶纤维素、糊精、蔗糖、乳糖、甘露醇、无机盐类等一种或多种,崩解剂选自干淀粉、羧甲淀粉钠、低取代羟丙基纤维素、交联羧甲基纤维素钠等的一种或多种,泡腾崩解剂选自碳酸氢钠、柠檬酸等,润滑剂选择硬脂酸镁、微粉硅胶、滑石粉、聚乙二醇400、聚乙二醇6000、氢化植物油等的一种或多种。
本发明所述的一种具有清除自由基功能的抗衰老组合物具有抗氧化功效,可作为食品、药品、保健品、化妆品的原料。
本发明与现有技术相比,具有如下的优点和有益效果:
(1)本发明选用鱼胶原蛋白肽粉、西柚粉、蔓越莓粉、L-阿拉伯糖、针叶樱桃果粉、雨生红球藻、燕窝肽、罗汉果甜苷等原料,结合特定的制备工艺得到一种具有清除自由基功能的抗衰老组合物,所述组合物具有高清除自由基活性,和高抗衰老性能的,便捷食用,口感愉悦的组合物,并意外的发现所述组合物无需低温条件下进行保存,在常温下保存也能长期的保持高的清除自由基活性。
其中,鱼胶原蛋白肽粉含有天然的保湿因子,可以有效锁住水分,令皮肤光鲜亮丽,并且可以改善皮肤的新陈代谢,滋润皮肤。当胶原蛋白被皮肤吸收后,填充在皮肤与真皮之间,不仅能够增加皮肤的紧密度,还可以淡化细纹,让皮肤紧绷而富有弹性。胶原蛋白富含酪氨酸残基,能抑制黑色素的生成,达到美白皮肤的目的。当鱼胶原蛋白肽通过表皮进入真皮层组织后,能迅速填补局部塌陷,改善松弛的皮肤,收紧皮肤,减少深皱纹、平复细纹,缩小毛孔。胶原蛋白使细胞变得丰满,从而使肌肤充盈,保持肌肤弹性与润泽,维持肌肤细腻光滑,皱纹舒展,呈现质感和透明感,有效防止老化。
燕窝肽,是由燕窝利用酶解技术而得到的小分子肽的粉末,较大程度的保留了燕窝中所含的营养成分,分子量小更容易吸收。富含珍贵的燕窝酸成分,为神经氧酸的衍生物,对人体的生理和生化功能具有重要的调节作用,能够增强人体免疫力,具有抗氧化、抗衰老的作用。
雨生红球藻是自然界中虾青素含量最丰富的生物,其虾青素的含量可达干重的2.0%,甚至更高,被誉为天然虾青素的“浓缩品”。虾青素属于类胡萝卜素,它与β-胡萝卜素具有相同的生物合成途径,他们都拥有能淬灭自由基的多烯链。并且虾青素是叶黄素支群的一部分,它额外的酮基和羟基附在了异戊二烯环上,这使得虾青素能定位亲水和疏水位置的能力,为细胞膜磷脂双脂质层提供优质的抗过氧化保护。有研究表明,雨生红球藻中的虾青素的抗氧化性是维生素C的6000倍,是维生素E的1000倍,被称为“超级抗氧化剂”。但雨生红球藻用于固体饮料制剂中,冲调溶解后存在一定的腥昧,使人产生不愉悦的口感体验,这时配伍西柚粉,利用西柚清新、昧酸的口感就能够明显改善口味。另外西柚也含有丰富的维生素C和纤维素,含糖量和热量较低,与其它具有抗衰老美白的原料起到协同增效的作用。
针叶樱桃原产于热带美洲西印度群岛加勒比海地区,又称西印度樱桃;针叶樱桃富含强抗氧化维生素C,是目前世界上已知含维生素C最丰富的水果,是名副其实的“维生素C之王”。源自天然的维生素C是一种很好的抗氧化剂,比合成的维生素C更能促进血浆中对其的吸收及限制尿液中的排泄,可以有效清除人体活动中产生的自由基,起到抗氧化的作用。
蔓越莓为一种营养价值很高的水果,富含维生素C、花青素等抗氧化物质及丰富果胶,能养颜美容。生物黄酮是有一种非常强力的抵抗自由基的物质,蔓越莓中生物黄酮的含量高居于一般常见的20种蔬果之冠。蔓越莓除了含有高量的单元不饱和脂肪酸和生育三烯醇外,还有另一种抗氧化尖兵-浓缩单宁酸,能起到保护心血管健康的作用。
L-阿拉伯糖最具代表性的生理作用是有选择性地影响小肠中的蔗糖酶,从而抑制蔗糖的吸收,减少过高的游离血糖的生成以降低晚期糖基化终产物,进而达到缓解衰老、抑制皮肤老化等目的。
(2)本发明采用特定的工艺制备鱼胶原蛋白肽粉,通过鱼胶原蛋白肽与高脱乙酰度的壳聚糖进行美拉德反应,实现了鱼胶原蛋白肽的耐热性和稳定性,且不破坏鱼胶原蛋白肽的清除自由基的能力。
(3)本发明选用的原料均为绿色原料,具有安全性好和利于吸收等优势,便捷食用,口感愉悦,能够满足不同人群的抗衰老需求。
具体实施方式
为了进一步理解本发明,下面结合实施例对本发明优选实施方案进行描述,但是应当理解,这些描述只是为了进一步说明本发明的特征和优点,而不是对本发明权利要求的限制。下面实施例中涉及的物料均可从商业渠道获得。
本发明以下实施例提供一种具有清除自由基功能的抗衰老组合物及其制备方法,首先制备相应的原料。
其中,所述鱼胶原蛋白肽粉的制备方法,包括以下步骤:
(1)将鱼皮放入质量分数为1.5%的氯化钠溶液中,在20℃的低温下浸泡8小时,然后水洗至中性,沥干水分,粉碎,得到鱼胶原蛋白粗提物;
(2)向鱼胶原蛋白粗提物中加入去离子水,调节pH,加入碱性蛋白酶,在65℃下酶解处理5小时;然后加入木瓜和无花果酶原,在60℃条件下继续酶解处理3小时;灭酶,离心过滤得到胶原蛋白肽液
(3)向胶原蛋白肽液中加入pH值为5.5的磷酸盐缓冲液,以及脱乙酰度大于85%的壳聚糖,其中,三者的重量份数如下:胶原蛋白肽液20份,磷酸盐缓冲液40份,壳聚糖5份,经超声波分散60min,离心过滤后,透析48h后,喷雾干燥得到鱼胶原蛋白肽粉。
所述西柚粉和针叶樱桃果粉的制备方法,包括以下步骤:
(1)将西柚、或针叶樱桃果制成浆液,再加入脱涩液搅拌,经过胶磨、均质、过滤后得到第一过滤液,所述脱涩液为质量分数1%的柠檬酸和质量分数0.4%份氯化钠按照质量比3∶1的混合物;
(2)向步骤(1)制备得到的滤渣中加入占滤渣质量0.4%的果胶酶和0.2%的纤维素酶,加热至55℃酶解10h;再次胶磨均质、过滤后得到第二过滤液;
(3)将第一滤液和第二滤液混合,加入占滤液质量的2%的海藻糖,经真空浓缩,冷冻干燥后得到西柚粉、或针叶樱桃果粉。
所述蔓越莓粉的制备方法,包括以下步骤:
(1)将蔓越莓洗净后置于汽爆罐内,通入氮气至汽爆罐内压力为2MPa,蒸汽爆破处理3min后,冷却至室温后,加入由纤维素酶∶果胶酶∶碱性蛋白酶按质量比2∶2∶3混合所得的复合酶,复合酶用量为原料重量的2.3%,保持在45℃下酶解2h,过滤,得第一滤液
(2)将步骤(1)制备得到的滤渣加入萃取釜中进行超临界CO2萃取,乙醇为夹带剂,萃取压力45Mpa,萃取温度为45℃,CO2流量20L/h,萃取时间60min,得到萃取物;
(3)将步骤(1)得到的第一滤液和步骤(2)得到的萃取物混合均匀,经真空浓缩,冷冻干燥后得到粉末,加入占粉末质量10%的麦芽糊精,混合均匀制备得到蔓越莓粉。
所述雨生红球藻提取物的制备方法,包括以下步骤:
(1)采用人工LED光源对绿色藻液进行光照积累虾青素,直到藻液呈鲜红色时进行离心或沉降采收;
(2)将步骤(2)采收的雨生红球藻在90℃下干燥至含水量低于10%,加入汽爆罐内,通入氮气至汽爆罐内压力为2.3MPa,蒸汽爆破处理3min后,冷却至室温后;
(3)将步骤(2)制备得到混合物加入萃取釜中进行超临界CO2萃取,乙醇为夹带剂,萃取压力45Mpa,萃取温度为55℃,CO2流量25L/h,萃取时间80min,得到萃取物;
(4)向步骤(3)得到的萃取物中加入占萃取物质量的3%的海藻糖,经冷冻干燥制备得到雨生红球藻提取物。
所述燕窝肽的制备方法,包括以下步骤:
(1)将燕窝用去离子水清洗干净后,在40℃下风干,粉碎成100目,再采用气流粉碎法粉碎至颗粒直径在4-12μm,得细燕窝粉,然后按照料液比1∶5加入去离子水,在搅拌速度为150r/min的条件下搅拌30min,得到燕窝混合浆;
(2)将步骤(1)中的燕窝混合浆放置纳米湿法粉碎机中粉碎、乳化60min,粉碎至颗粒直径在30-120nm之间,然后在50℃的条件下干燥,得到饼状燕窝;
(3)将步骤(2)中的饼状燕窝放置于研磨机中研磨2h后移至超细粉碎机深度粉碎至颗粒的直径范围在30-180nm,得超细燕窝粉,然后按照料液比1∶10加入去离子水搅拌均匀,再置于水浴罐中进行超声水浴震荡,水浴时间为5h,温度为90℃,得到超细燕窝混合液;
(4)将步骤(3)中的超细燕窝混合液用7000-8000道尔顿的超滤膜进行离心超滤,再用葡聚糖凝胶柱层析,收集300nm处吸收峰组分,然后冷冻干燥,得到燕窝肽粉。
所述一种具有清除自由基功能的抗衰老组合物的制备方法,将所有原料按照表1的配方量混合均匀,然后使用40-60目筛网过筛,即可。

准确量取0.2mmol/L的DPPH·乙醇溶液2.5mL置于五支试管中,然后依次加入1mL质量浓度分别为0.01mg/mL的待测液体,静置30min,使其充分反应后在517nm处测其吸光值,数值记为Da;同时准确量取0.2mmol/L的DPPH·乙醇溶液2.5mL,随后加入1mL的乙醇溶液,测其吸光值记为Dc;同时准确量取2.5mL的无水乙醇,然后加入1mL质量浓度分别为0.01mg/mL的待测液体,测其吸光值记为Db。其中,清除率(%)=[1-(Da-Db)/Dc]×100%。
(2)羟自由基(·OH)活性测试
准确量取1mL质量浓度分别为0.01mg/mL的待测液体于不同编号的试管中,依次加入2mL的9mmol/L的FeSO4溶液和水杨酸-乙醇溶液、8.8mmol/L的H2O2溶液,用超纯水定容至10mL,静置30min,于510nm处测其吸光值(A1);测量超纯水(空白对照液)于510nm处的吸光值(A0)。同时准确量取1mL质量浓度分别为0.01mg/mL的待测液体于不同编号的试管中,先加入9mmol/L的FeSO4溶液2mL,随后加入2nL9mmol/L的水杨酸-乙醇溶液,超纯水定容至10mL,测其于510nm处的吸光值(A2)。其中,清除率(%)=[1-(A1-A2)/A0]×100%。
(3)超氧阴离子自由基(O2·)活性测试
于试管中依次准确加入50mmol/L Tris-HC1溶液(pH=8.2)4.5mL、蒸馏水4.2mL,静置25min,充分反应后加入3mmol/L邻苯三酚0.3mL,在325nm处每30s测一次吸光值,5min时停止,10mmol/L的HC1作为邻苯三酚的空白对照,计算空白溶液吸光值随时间的变化率V0。同时准确量取1mL质量浓度分别为0.01mg/mL的待测液体于试管中,加入pH为8.2的50mmol/L Tris-HCl溶液4.5mL,3.2mL蒸馏水,反应25min后加入3mmol/L的邻苯三酚0.3mL。于325nm处每隔30s测一组吸光值,5min时测量停止。其吸光值随时间的变化率记为V1。其中,清除率(100%)=(V0-V1)/V0×100%。
将实施例1-10制备得到的具有清除自由基功能的抗衰老组合物制备成质量浓度为0.01mg/mL,作为待测液体,相关测试结果见表2。

从表2的数据可知,本发明制备得到的抗衰老组合物具有良好的体外抗氧化效果。
本发明还提供以下对比实施例,测试不同原料的组合对自由基清除能力的影响,以实施例3为基础,改变不同原料的组合制备对比实施例,相应的配方量见表3,制备方法与实施例2完全一致。

基于上述测试方法,测试实施例3和对比例1-5制备得到的抗衰老组合物的抗氧化效果,测试数据见表4。

从表4的实验数据来看,本发明通过各种原料的特定配合实现了高清除自由基性能,所述各种原料的配合起到了相互协同的效果。
进一步,本发明还测试了保存环境对抗衰老组合物的性能稳定性的影响,测试方法如下:将实施例3制备得到的具有清除自由基功能的抗衰老组合物制备成质量浓度为0.01mg/mL,作为待测液体,按照不同温度进行保存:(1)0℃,避光保存;(2)20℃,避光保存;(3)40℃,避光保存,分别测试0周、2周、4周、8周、10周后的抗氧化性能。测试结果见表5。

从表5的实验数据来看,本发明制备的具有清除自由基功能的抗衰老组合物,在室温(20℃)时具有较好的稳定性,与低温(0℃)保存时的效果相差较小,较高温(40℃)保存时具有明显的改进。
本领域的技术人员容易理解,以上所述仅为本发明的较佳实施例而已,并不用以限制本发明,凡在本发明的精神和原则之内所作的任何修改、等同替换和改进等,均应包含在本发明的保护范围之内。
Claims (10)
1.一种具有清除自由基功能的抗衰老组合物,其特征在于:包括以下重量份的原料:
鱼胶原蛋白肽粉50-100份、西柚粉10-40份、蔓越莓粉5-20份、L-阿拉伯糖2-15份、针叶樱桃果粉2-15份、雨生红球藻提取物0.2-4份、燕窝肽0.2-4份、罗汉果甜苷0.02-1份。
2.根据权利要求1所述的一种具有清除自由基功能的抗衰老组合物,其特征在于:包括以下重量份的原料:鱼胶原蛋白肽粉50-80份、西柚粉10-25份、蔓越莓粉5-10份、L-阿拉伯糖2-8份、针叶樱桃果粉2-10份、雨生红球藻提取物0.2-2份、燕窝肽0.2-2份、罗汉果甜苷0.02-0.5份。
3.根据权利要求1-2任一项权利要求所述的一种具有清除自由基功能的抗衰老组合物,其特征在于:所述鱼胶原蛋白肽粉的制备方法,包括以下步骤:
(1)将鱼皮放入质量分数为1-2%的氯化钠溶液中,在15-25℃的低温下浸泡5-8小时,然后水洗至中性,沥干水分,粉碎,得到鱼胶原蛋白粗提物;
(2)向鱼胶原蛋白粗提物中加入去离子水,调节pH,加入碱性蛋白酶,在50-65℃下酶解处理3-5小时;然后加入木瓜和无花果酶原,在45-60℃条件下继续酶解处理2-4小时;灭酶,离心过滤得到胶原蛋白肽液;
(3)向胶原蛋白肽液中加入pH值为5.5-6.5的磷酸盐缓冲液,以及脱乙酰度大于85%的壳聚糖,其中,三者的重量份数如下:胶原蛋白肽液10-20份,磷酸盐缓冲液20-40份,壳聚糖5-10份,经超声波分散30-60min,离心过滤后,透析48h后,喷雾干燥得到鱼胶原蛋白肽粉。
4.根据权利要求1-2任一项权利要求所述的一种具有清除自由基功能的抗衰老组合物,其特征在于:所述西柚粉和针叶樱桃果粉的制备方法,包括以下步骤:
(1)将西柚、或针叶樱桃果制成浆液,再加入脱涩液搅拌,经过胶磨、均质、过滤后得到第一过滤液,所述脱涩液为质量分数0.5-1%的柠檬酸和质量分数0.2-0.4%份氯化钠按照质量比3∶1的混合物;
(2)向步骤(1)制备得到的滤渣中加入占滤渣质量0.2-0.4%的果胶酶和0.1-0.2%的纤维素酶,加热至40-55℃酶解6-10h;再次胶磨均质、过滤后得到第二过滤液;
(3)将第一滤液和第二滤液混合,加入占滤液质量的1-3%的海藻糖,经真空浓缩,冷冻干燥后得到西柚粉、或针叶樱桃果粉。
5.根据权利要求1-2任一项权利要求所述的一种具有清除自由基功能的抗衰老组合物,其特征在于:所述蔓越莓粉的制备方法,包括以下步骤:
(1)将蔓越莓洗净后置于汽爆罐内,通入氮气至汽爆罐内压力为1.7-2.3MPa,蒸汽爆破处理1-3min后,冷却至室温后,加入由纤维素酶:果胶酶:碱性蛋白酶按质量比2∶2∶3混合所得的复合酶,复合酶用量为原料重量的0.7-2.3%,保持在30-45℃下酶解0.5-2h,过滤,得第一滤液;
(2)将步骤(1)制备得到的滤渣加入萃取釜中进行超临界CO2萃取,乙醇为夹带剂,萃取压力40-45Mpa,萃取温度为40-45℃,CO2流量15-20Lh,萃取时间40-60min,得到萃取物;
(3)将步骤(1)得到的第一滤液和步骤(2)得到的萃取物混合均匀,经真空浓缩,冷冻干燥后得到粉末,加入占粉末质量5-10%的麦芽糊精,混合均匀制备得到蔓越莓粉。
6.根据权利要求1-2任一项权利要求所述的一种具有清除自由基功能的抗衰老组合物,其特征在于:所述雨生红球藻提取物的制备方法,包括以下步骤:
(1)采用人工LED光源对绿色藻浓进行光照积累虾青素,直到藻液呈鲜红色时进行离心或沉降采收;
(2)将步骤(2)采收的雨生红球藻在80-90℃下干燥至含水量低于10%,加入汽爆罐内,通入氮气至汽爆罐内压力为1.7-23MPa,蒸汽爆破处理1-3min后,冷却至室温后;
(3)将步骤(2)制备得到混合物加入萃取釜中进行超临界CO2萃取,乙醇为夹带剂,萃取压力40-45Mpa,萃取温度为45-55℃,CO2流量20-25L/h,萃取时间60-80min,得到萃取物;
(4)向步骤(3)得到的萃取物中加入占萃取物质量的1-3%的海藻糖,经冷冻干燥制备得到雨生红球藻提取物。
7.根据权利要求1-2任一项权利要求所述的一种具有清除自由基功能的抗衰老组合物,其特征在于:所述燕窝肽的制备方法,包括以下步骤:
(1)将燕窝用去离子水清洗干净后,在35-40℃下风干,粉碎成60-120目,再采用气流粉碎法粉碎至颗粒直径在4-12μm,得细燕窝粉,然后按照料液比1∶5.10加入去离子水,在搅拌速度为120-150r/min的条件下搅拌20-30min,得到燕窝混合浆;
(2)将步骤(1)中的燕窝混合浆放置纳米湿法粉碎机中粉碎、乳化30-60min,粉碎至颗粒直径在30-120nm之间,然后在40-50℃的条件下干燥,得到饼状燕窝;
(3)将步骤(2)中的饼状燕窝放置于研磨机中研磨1-2h后移至超细粉碎机深度粉碎至颗粒的直径范围在30-180nm,得超细燕窝粉,然后按照料液比1∶10-15加入去离子水搅拌均匀,再置于水浴罐中进行超声水浴震荡,水浴时间为3-5h,温度为70-90℃,得到超细燕窝混合液;
(4)将步骤(3)中的超细燕窝混合液用7000-8000道尔顿的超滤膜进行离心超滤,再用葡聚糖凝胶柱层析,收集300nm处吸收峰组分,然后冷冻干燥,得到燕窝肽粉。
8.根据权利要求1-7任一项权利要求所述的一种具有清除自由基功能的抗衰老组合物,其特征在于:该组合物制备成固体或液体制剂。
9.根据权利要求8所述的一种具有清除自由基功能的抗衰老组合物,其特征在于:所述固体制剂为胶囊剂、片剂、粉剂、泡腾剂或颗粒剂。
10.一种权利要求1-7任一项权利要求所述的具有清除自由基功能的抗衰老组合物的制备方法,其特征在于:将所有原料混合均匀,然后使用40-60目筛网过筛,即可。
