according to the invention, researches show that the water replenishing and moisturizing performances of the coarse bletilla polysaccharide extract can be obviously improved by compounding the coarse gymnadenia conopsea polysaccharide extract, the coarse astragalus polysaccharide extract and the coarse bletilla polysaccharide extract, and one of the three is not necessary, so that the water replenishing and moisturizing performances superior to those of two compounded extracts are obtained through mutual cooperation. Meanwhile, the skin care composition has a simple formula, all the components are from plant components, so that the stimulation of chemical synthesis components is avoided, the skin comfort can be improved, the skin burden is light, and the use is safe.

The term "crude gymnadenia conopsea polysaccharide extract" as used herein refers to an extract containing gymnadenia conopsea polysaccharide compounds, and it is understood that the extract may contain other active ingredients besides gymnadenia conopsea polysaccharide compounds, and is not particularly limited. The content of the crude polysaccharide extract of gymnadenia conopsea prepared in the examples can be determined by conventional methods such as ultraviolet spectrophotometer. The same applies to the bletilla striata crude polysaccharide extract and the astragalus membranaceus crude polysaccharide extract.
Specifically, in the examples of the present invention and the comparative examples:
the content of crude polysaccharide in the gymnadenia conopsea crude polysaccharide extract is determined to be in the range of 85.154-92.729% by adopting a phenol-sulfuric acid method, and the content of protein in the gymnadenia conopsea crude polysaccharide extract is determined to be in the range of 1.165-1.862% by adopting a Kjeldahl method;
the content of crude polysaccharide in the bletilla crude polysaccharide extract is determined to be within the range of 65.215-74.697% by adopting a phenol-sulfuric acid method, and the content of protein in the bletilla crude polysaccharide extract is determined to be within the range of 3.497-4.185% by adopting a Kjeldahl method;
the content of crude polysaccharide in the astragalus crude polysaccharide extract is determined to be within the range of 68.317-75.532% by adopting a phenol-sulfuric acid method, and the content of protein in the astragalus crude polysaccharide extract is determined to be within the range of 14.682-19.532% by adopting a Kjeldahl method.
The boiling-maintaining method is characterized in that after water is boiled, the water is kept in a slightly boiling state, and the temperature is controlled to be 95-100 ℃.
The invention provides a skin care composition with a moisturizing effect, which comprises the following components in parts by weight: 1 to 5 parts of gymnadenia conopsea crude polysaccharide extract, 1 to 5 parts of bletilla striata crude polysaccharide extract and 2 to 8 parts of astragalus membranaceus crude polysaccharide extract.
Generally, plant polysaccharides have the effects of regulating immunity, resisting aging, resisting tumors and the like. Astragalus membranaceus, also known as astragalus membranaceus. The astragalus root has been used for more than 2000 years, and has the functions of enhancing the immunologic function of organisms, protecting the liver, promoting urination, resisting aging, resisting stress, reducing blood pressure and having wider antibacterial action. In the traditional method, astragalus polysaccharide can be prepared by extracting, concentrating and purifying dried roots of astragalus, and research proves that the astragalus polysaccharide can be used as an immune promoter or regulator and has the effects of resisting virus, tumors, aging, radiation, stress, oxidation and the like. Conic gymnadenia rhizome, also called conic gymnadenia rhizome, is a perennial herb of gymnadenia genus of orchidaceae family, is a commonly used traditional Chinese medicine because the tuber of the conic gymnadenia rhizome is shaped like a palm, and can be used for treating diseases such as weakness after illness, cough, asthma, chronic diarrhea, blood loss, hepatitis, brucellosis and the like by being used alone or in combination with other traditional Chinese medicines. In the traditional method, the gymnadenia conopsea polysaccharide can be prepared by the procedures of crushing, degreasing by petroleum ether, removing impurities by ethanol, extracting with water and precipitating with ethanol, removing protein, precipitating with ethanol, washing and the like. The research on the action of the gymnadenia conopsea polysaccharide mainly focuses on the aspects of delaying senility, regulating immunity, relieving fatigue, resisting anoxia and the like.
In the process of carrying out systematic research on plant polysaccharide, the invention discovers that compounding the gymnadenia conopsea crude polysaccharide extract, the astragalus root crude polysaccharide extract and the bletilla striata crude polysaccharide extract can obviously improve the water replenishing and moisturizing performances of the bletilla striata crude polysaccharide extract. Meanwhile, tests prove that the gymnadenia conopsea crude polysaccharide extract, the astragalus root crude polysaccharide extract and the bletilla striata crude polysaccharide extract have a synergistic interaction effect, but the water replenishing and moisturizing performances which can be achieved by the synergistic interaction of the gymnadenia conopsea crude polysaccharide extract, the astragalus root crude polysaccharide extract and the bletilla striata crude polysaccharide extract are obviously higher than those of any two of the complex combinations. Meanwhile, the skin care composition has the advantages of simple formula, easiness in preparation and low cost, all the components are from plant components, the stimulation of chemical synthesis components is avoided, the skin comfort can be improved, the skin burden is light, and the use is safe.
In one specific example, the skin care composition with moisturizing effect comprises the following components in parts by weight: 2 to 3 parts of gymnadenia conopsea crude polysaccharide extract, 2 to 3 parts of bletilla striata crude polysaccharide extract and 5 to 6 parts of astragalus membranaceus crude polysaccharide extract.
In one specific example, the skin care composition with moisturizing effect comprises the following components in parts by weight: 2.3 to 2.7 parts of gymnadenia conopsea crude polysaccharide extract, 2.5 to 3 parts of bletilla striata crude polysaccharide extract and 5 to 5.5 parts of astragalus membranaceus crude polysaccharide extract.
Furthermore, the extraction processes of the gymnadenia conopsea crude polysaccharide extract, the bletilla striata crude polysaccharide extract and the astragalus membranaceus crude polysaccharide extract, particularly the volume concentration of ethanol and the standing temperature in a system are reasonably controlled, so that the moisture retention property of the prepared composition can be optimized.
In one specific example, the preparation method of the gymnadenia conopsea crude polysaccharide extract comprises the following steps:
extracting rhizoma Gymnadeniae decoction pieces with water, concentrating the extractive solution, and preparing into concentrated solution;
adding ethanol into the concentrated solution to ensure that the volume concentration of the ethanol in the solution system is 50-80%, standing the obtained solution system at the temperature of 1-10 ℃, and collecting the solid.

Example 1
The embodiment is a skin care product with moisturizing effect and capable of improving skin comfort, and the skin care product comprises the following raw materials in parts by weight:
2.5 parts of gymnadenia conopsea crude polysaccharide extract, 3 parts of bletilla striata crude polysaccharide extract, 5 parts of astragalus membranaceus crude polysaccharide extract, 5 parts of glycerol, 0.2 part of trimethylglycine, 0.08 part of baicalin, 0.1 part of glycyrrhizin, 0.2 part of asiaticoside and 60 parts of deionized water.
The preparation method comprises the following steps:
(a) preparing a gymnadenia conopsea crude polysaccharide extract:
taking gymnadenia conopsea decoction pieces, crushing into 80-mesh fine powder, adding water for boiling-maintaining extraction for 2 times, adding 40 times of water for extraction for 2.5 hours for the first time, and adding 20 times of water for extraction for 1 hour for the second time; respectively sieving the two extracting solutions with a 350-mesh sieve, mixing the extracting solutions, and concentrating until the solid content is 4%; adding 95 vol% ethanol into the concentrated solution until the volume concentration of ethanol in the solution system is 60%, standing in a refrigerator at 4 deg.C for 16 hr, centrifuging at 4000r/min for 10 min, removing supernatant, oven drying the precipitate at 50 deg.C until the water content is less than 10%, and pulverizing to 80 mesh to obtain rhizoma Gymnadeniae crude polysaccharide extract.
(b) Preparing a bletilla striata crude polysaccharide extract:

2.5 parts of gymnadenia conopsea crude polysaccharide extract, 3 parts of bletilla striata crude polysaccharide extract, 5 parts of astragalus membranaceus crude polysaccharide extract, 5 parts of glycerol, 0.2 part of trimethylglycine, 0.08 part of baicalin, 0.1 part of glycyrrhizin, 0.2 part of asiaticoside and 60 parts of deionized water.
The preparation method comprises the following steps:
(a) preparing the gymnadenia conopsea polysaccharide extract:
taking gymnadenia conopsea decoction pieces, crushing into 80-mesh fine powder, adding water for boiling-maintaining extraction for 2 times, adding 40 times of water for extraction for 2.5 hours for the first time, and adding 20 times of water for extraction for 1 hour for the second time; respectively sieving the two extracting solutions with a 350-mesh sieve, mixing the extracting solutions, and concentrating until the solid content is 4%; concentrating 95% ethanol solution to volume concentration of 70%, standing in refrigerator at 4 deg.C for 16 hr, centrifuging at 4000r/min for 10 min, removing supernatant, oven drying the precipitate at 50 deg.C until water content is less than 10%, and pulverizing to 80 mesh to obtain rhizoma Gymnadeniae crude polysaccharide extract.
(b) Preparing a bletilla striata crude polysaccharide extract:
pulverizing rhizoma bletilla decoction pieces into 40 mesh fine powder, adding water, boiling and extracting for 2 times, adding 30 times of water for the first time, extracting for 3 hr, adding 20 times of water for the second time, and extracting for 1.5 hr; respectively sieving the two extracting solutions with a 350-mesh sieve, mixing the extracting solutions, and concentrating until the solid content is 3%; adding 95 vol% ethanol into the concentrated solution until the volume concentration of ethanol in the solution system is 70%, standing in a refrigerator at 4 deg.C for 16 hr, centrifuging at 4000r/min for 10 min, removing supernatant, oven drying the precipitate at 50 deg.C until the water content is less than 10%, and pulverizing to 80 mesh to obtain rhizoma bletilla crude polysaccharide extract.
(c) Preparing the astragalus crude polysaccharide extract:
cutting radix astragali decoction pieces into 5mm pieces, adding water, boiling for 2 times, extracting for 1.5 hr with 20 times of water for the first time, and extracting for 0.5 hr with 12 times of water for the second time; respectively sieving the two extracting solutions with 350 mesh sieve, mixing the extracting solutions, and concentrating until the solid content is 40%; adding 95 vol% ethanol into the concentrated solution until the volume concentration of ethanol in the solution system is 70%, standing in a refrigerator at 4 deg.C for 16 hr, centrifuging at 4000r/min for 10 min, removing supernatant, oven drying the precipitate at 50 deg.C until the water content is less than 10%, and pulverizing to 80 mesh to obtain crude polysaccharide extract of radix astragali.
(d) Taking glycerol according to the weight parts, adding the gymnadenia conopsea polysaccharide crude extract, the bletilla striata crude polysaccharide extract and the astragalus membranaceus crude polysaccharide extract, and uniformly stirring;
(e) adding trimethylglycine, baicalin, glycyrrhizin, asiaticoside and deionized water into the mixture obtained in the step (d) according to the weight parts;
(f) passing the mixture of step (e) through a 200 mesh screen;
(g) UV sterilization for 50 minutes.
Moisturizing efficacy tests of examples 1-5 and comparative examples 1-5.
Firstly, testing the moisture retention performance

From the test results of the embodiment 1 and the comparative examples 2-4, the compound of the gymnadenia conopsea crude polysaccharide extract, the bletilla striata crude polysaccharide extract and the astragalus membranaceus crude polysaccharide extract has obviously better moisturizing effect than that of compound of two of the gymnadenia conopsea crude polysaccharide extract and the bletilla striata crude polysaccharide extract; example 1 compared with the test results of comparative example 5, shows the decisive effect of the polysaccharide preparation technology means of the patent on the moisture retention. Therefore, the composition provided by the invention can be used as a water replenishing and water retaining agent to effectively improve the moisture content of skin, and the compounding of the three polysaccharide extracts has the moisture-retaining synergistic effect, so that the composition is a natural safe skin care product which can effectively help the skin to replenish water and retain moisture and improve the comfort level of the skin.

The skin care composition with moisturizing effect according to claim 1, which is characterized by comprising the following components in parts by weight: 2 to 3 parts of gymnadenia conopsea crude polysaccharide extract, 2 to 3 parts of bletilla striata polysaccharide crude extract and 5 to 6 parts of astragalus membranaceus crude polysaccharide extract.

The skin care composition with moisturizing effect according to claim 2, which is characterized by comprising the following components in parts by weight: 2.3 to 2.7 parts of gymnadenia conopsea crude polysaccharide extract, 2.5 to 3 parts of bletilla striata crude polysaccharide extract and 5 to 5.5 parts of astragalus membranaceus crude polysaccharide extract.
4. The skin care composition with moisturizing effect as claimed in any one of claims 1 to 3, wherein the preparation method of the gymnadenia conopsea crude polysaccharide extract comprises the following steps:
extracting rhizoma Gymnadeniae decoction pieces with water, concentrating the extractive solution, and preparing into concentrated solution;
adding ethanol into the concentrated solution to ensure that the volume concentration of the ethanol in the solution system is 50-80%, standing the obtained solution system at the temperature of 1-10 ℃, and collecting solids.

The skin care composition with moisturizing effect as claimed in claim 4, wherein the water extraction is performed twice by adding water to preserve boiling, the water added for the first time is 25-40 times of the weight of the gymnadenia conopsea decoction pieces and extracted for 1.5-3 hours, and the water added for the second time is 15-30 times of the weight of the gymnadenia conopsea decoction pieces and extracted for 1-1.5 hours.

extracting rhizoma bletilla decoction pieces with water, concentrating the extractive solution, and making into concentrated solution;
adding ethanol into the concentrated solution to ensure that the volume concentration of the ethanol in the solution system is 50-80%, standing the obtained solution system at the temperature of 1-10 ℃, and collecting solids.

The skin care composition with moisturizing effect as claimed in claim 6, wherein the water extraction is performed twice by adding water to preserve boiling, the water added in the amount 20-35 times of the weight of the bletilla striata decoction pieces is added for the first time for 1.5-3 hours, and the water added in the amount 15-30 times of the weight of the bletilla striata decoction pieces is added for the second time for 1-1.5 hours.

extracting radix astragali decoction pieces with water, concentrating the obtained extractive solution, and preparing into concentrated solution;
adding ethanol into the concentrated solution to ensure that the volume concentration of the ethanol in the solution system is 50-80%, standing the obtained solution system at the temperature of 1-10 ℃, and collecting solids.

The skin care composition with moisturizing effect as claimed in claim 8, wherein the water extraction is performed twice by adding water to preserve boiling, the water added in an amount 10-20 times the weight of the astragalus decoction pieces is added for the first time for 1-2 hours, and the water added in an amount 10-15 times the weight of the bletilla striata decoction pieces is added for the second time for 0.5-1 hour.

12. The skin care product of claim 11, comprising the following components in parts by weight:
1 to 5 parts of gymnadenia conopsea crude polysaccharide extract, 1 to 5 parts of bletilla striata crude polysaccharide extract, 2 to 8 parts of astragalus membranaceus crude polysaccharide extract, 0.1 to 10 parts of glycerol, 0.1 to 5 parts of trimethylglycine, 0.01 to 3 parts of baicalin, 0.01 to 5 parts of glycyrrhizin, 0.01 to 5 parts of asiaticoside and 0 to 70 parts of water.
