Abstract
The invention discloses an application of bletilla striata polysaccharide in preparing a product with a moisturizing effect, and relates to the technical field of plant extraction, wherein the preparation of the bletilla striata polysaccharide comprises the steps of heating and extracting a mixed solution of bletilla striata powder and water, and concentrating a supernatant of the extracted product to obtain colloidal bletilla striata polysaccharide; alcohol precipitation is carried out on the colloidal bletilla striata polysaccharide to obtain the bletilla striata polysaccharide; wherein the temperature of the heating extraction is 60-80 ℃ and the time is 1-8 h. Example 1
An extraction method of bletilla striata polysaccharide comprises the following steps:
(1) Pulverizing rhizoma Bletillae tuber to obtain rhizoma Bletillae tuber powder, sieving with 200 mesh sieve, adding 20 times of distilled water, swelling, homogenizing, extracting at 80deg.C for 4 hr, filtering, adding 20 times of distilled water into the residue, repeating the extraction steps, extracting twice, mixing the supernatants, and concentrating to obtain rhizoma Bletillae polysaccharide gum.
(2) Slowly adding 1 time of 95% ethanol precooled to 4 ℃ into the bletilla striata polysaccharide gum in the step (1), placing the mixture in a refrigerator at 4 ℃ overnight, collecting precipitate, and freeze-drying the precipitate in a freeze dryer to obtain the bletilla striata crude polysaccharide.
(3) Preparing the crude polysaccharide of the bletilla striata in the step (2) into a solution with the concentration of 50mg/mL in advance, centrifuging, absorbing supernatant, separating and purifying by using DE-252 anion column chromatography, eluting sodium chloride with the concentration of 0mol/mL, 0.5mol/mL, 1mol/mL and 2mol/mL, eluting the solution with the volume of about 1-2 times of the column volume, collecting polysaccharide eluents under sodium chloride solutions with different concentrations, concentrating, precipitating with 95% ethanol precooled (4 ℃), refrigerating overnight at 4 ℃, collecting precipitate, and freeze-drying.
Example 2
An extraction method of bletilla striata polysaccharide comprises the following steps:
(1) Pulverizing rhizoma Bletillae tuber to obtain rhizoma Bletillae tuber powder, adding 20 times of distilled water, homogenizing, extracting at 80deg.C for 2 hr, filtering, adding 20 times of distilled water into the residue, repeating the extraction steps, extracting twice, mixing the supernatants, and concentrating to obtain rhizoma Bletillae polysaccharide gum.
(2) Slowly adding 1 time volume of pre-cooled (4 ℃) 95% ethanol into the bletilla striata polysaccharide gum in the step (1), placing the mixture in a refrigerator at 4 ℃ overnight, collecting precipitate, and freeze-drying the precipitate in a freeze dryer to obtain the bletilla striata crude polysaccharide.
(3) Preparing the crude polysaccharide of the bletilla striata in the step (2) into a solution with the concentration of 50mg/mL in advance, centrifuging, absorbing supernatant, separating and purifying by using DE-252 anion column chromatography, eluting sodium chloride with the concentration of 0mol/mL, 0.5mol/mL, 1mol/mL and 2mol/mL, eluting the solution with the volume of about 1-2 times of the column volume, collecting polysaccharide eluents under sodium chloride solutions with different concentrations, concentrating, precipitating with 95% ethanol precooled (4 ℃), refrigerating overnight at 4 ℃, collecting precipitate, and freeze-drying.
Example 3
An extraction method of bletilla striata polysaccharide comprises the following steps:
(1) Pulverizing rhizoma Bletillae tuber to obtain rhizoma Bletillae tuber powder, adding 20 times of distilled water, homogenizing, extracting at 60deg.C for 4 hr, filtering, adding 20 times of distilled water, repeating the extraction steps, extracting twice, mixing the supernatants, and concentrating to obtain rhizoma Bletillae polysaccharide gum.
(2) Slowly adding 1 time volume of pre-cooled (4 ℃) 95% ethanol into the bletilla striata polysaccharide gum in the step (1), placing the mixture in a refrigerator at 4 ℃ overnight, collecting precipitate, and freeze-drying the precipitate in a freeze dryer to obtain the bletilla striata crude polysaccharide.
(3) Preparing the crude polysaccharide of the bletilla striata in the step (2) into a solution with the concentration of 50mg/mL in advance, centrifuging, absorbing supernatant, separating and purifying by using DE-252 anion column chromatography, eluting sodium chloride with the concentration of 0mol/mL, 0.5mol/mL, 1mol/mL and 2mol/mL, eluting the solution with the volume of about 1-2 times of the column volume, collecting polysaccharide eluents under sodium chloride solutions with different concentrations, concentrating, precipitating with 95% ethanol precooled (4 ℃), refrigerating overnight at 4 ℃, collecting precipitate, and freeze-drying.
Example 4
An extraction method of bletilla striata polysaccharide comprises the following steps:
(1) Pulverizing rhizoma Bletillae tuber to obtain rhizoma Bletillae tuber powder, adding 20 times of distilled water, swelling, homogenizing, extracting at 60deg.C for 2 hr, filtering, adding 20 times of distilled water, repeating the extraction steps, extracting twice, mixing the supernatants, and concentrating to obtain rhizoma Bletillae polysaccharide gum.
(2) Slowly adding 1 time volume of pre-cooled (4 ℃) 95% ethanol into the bletilla striata polysaccharide gum in the step (1), placing the mixture in a refrigerator at 4 ℃ overnight, collecting precipitate, and freeze-drying the precipitate in a freeze dryer to obtain the bletilla striata crude polysaccharide.
(3) Preparing the crude polysaccharide of the bletilla striata in the step (2) into a solution with the concentration of 50mg/mL in advance, centrifuging, absorbing supernatant, separating and purifying by using DE-252 anion column chromatography, eluting sodium chloride with the concentration of 0mol/mL, 0.5mol/mL, 1mol/mL and 2mol/mL, eluting the solution with the volume of about 1-2 times of the column volume, collecting polysaccharide eluents under sodium chloride solutions with different concentrations, concentrating, precipitating with 95% ethanol precooled (4 ℃), refrigerating overnight at 4 ℃, collecting precipitate, and freeze-drying.
Example 5
An extraction method of bletilla striata polysaccharide comprises the following steps:
(1) Pulverizing rhizoma Bletillae tuber to obtain rhizoma Bletillae tuber powder, adding 10 times of distilled water, homogenizing, extracting at 80deg.C for 2 hr, filtering, adding 10 times of distilled water into the residue, repeating the extraction steps, extracting twice, mixing the supernatants, and concentrating to obtain rhizoma Bletillae polysaccharide gum.
(2) Slowly adding 1 time volume of pre-cooled (4 ℃) 95% ethanol into the bletilla striata polysaccharide gum in the step (1), placing the mixture in a refrigerator at 4 ℃ overnight, collecting precipitate, and freeze-drying the precipitate in a freeze dryer to obtain the bletilla striata crude polysaccharide.
(3) Preparing the crude polysaccharide of the bletilla striata in the step (2) into a solution with the concentration of 50mg/mL in advance, centrifuging, absorbing supernatant, separating and purifying by using DE-252 anion column chromatography, eluting sodium chloride with the concentration of 0mol/mL, 0.5mol/mL, 1mol/mL and 2mol/mL, eluting the solution with the volume of about 1-2 times of the column volume, collecting polysaccharide eluents under sodium chloride solutions with different concentrations, concentrating, precipitating with 95% ethanol precooled (4 ℃), refrigerating overnight at 4 ℃, collecting precipitate, and freeze-drying.
Example 6
An extraction method of bletilla striata polysaccharide comprises the following steps:
(1) Pulverizing rhizoma Bletillae tuber to obtain rhizoma Bletillae tuber powder, adding 10 times of distilled water, homogenizing, extracting at 80deg.C for 4 hr, filtering, adding 10 times of distilled water into the residue, repeating the extraction steps, extracting twice, mixing the supernatants, and concentrating to obtain rhizoma Bletillae polysaccharide gum.
(2) Slowly adding 1 time volume of pre-cooled (4 ℃) 95% ethanol into the bletilla striata polysaccharide gum in the step (1), placing the mixture in a refrigerator at 4 ℃ overnight, collecting precipitate, and freeze-drying the precipitate in a freeze dryer to obtain the bletilla striata crude polysaccharide.
(3) Preparing the crude polysaccharide of the bletilla striata in the step (2) into a solution with the concentration of 50mg/mL in advance, centrifuging, absorbing supernatant, separating and purifying by using DE-252 anion column chromatography, eluting sodium chloride with the concentration of 0mol/mL, 0.5mol/mL, 1mol/mL and 2mol/mL, eluting the solution with the volume of about 1-2 times of the column volume, collecting polysaccharide eluents under sodium chloride solutions with different concentrations, concentrating, precipitating with 95% ethanol precooled (4 ℃), refrigerating overnight at 4 ℃, collecting precipitate, and freeze-drying.
Example 7
An extraction method of bletilla striata polysaccharide comprises the following steps:
(1) Pulverizing rhizoma Bletillae tuber to obtain rhizoma Bletillae tuber powder, adding 10 times of distilled water, homogenizing, extracting at 60deg.C for 4 hr, filtering, adding 10 times of distilled water into the residue, repeating the extraction steps, extracting twice, mixing the supernatants, and concentrating to obtain rhizoma Bletillae polysaccharide gum.
(2) Slowly adding 1 time volume of pre-cooled (4 ℃) 95% ethanol into the bletilla striata polysaccharide gum in the step (1), placing the mixture in a refrigerator at 4 ℃ overnight, collecting precipitate, and freeze-drying the precipitate in a freeze dryer to obtain the bletilla striata crude polysaccharide.
(3) Preparing the crude polysaccharide of the bletilla striata in the step (2) into a solution with the concentration of 50mg/mL in advance, centrifuging, absorbing supernatant, separating and purifying by using DE-252 anion column chromatography, eluting sodium chloride with the concentration of 0mol/mL, 0.5mol/mL, 1mol/mL and 2mol/mL, eluting the solution with the volume of about 1-2 times of the column volume, collecting polysaccharide eluents under sodium chloride solutions with different concentrations, concentrating, precipitating with 95% ethanol precooled (4 ℃), refrigerating overnight at 4 ℃, collecting precipitate, and freeze-drying.
Example 8
An extraction method of bletilla striata polysaccharide comprises the following steps:
(1) Pulverizing rhizoma Bletillae tuber to obtain rhizoma Bletillae tuber powder, adding 10 times of distilled water, swelling, homogenizing, extracting at 60deg.C for 2 hr, filtering, adding 10 times of distilled water, repeating the extraction steps, extracting twice, mixing the supernatants, and concentrating to obtain rhizoma Bletillae polysaccharide gum.
(2) Slowly adding 4 times of pre-cooled (4 ℃) 95% ethanol into the bletilla striata polysaccharide gum in the step (1), placing the mixture in a refrigerator at 4 ℃ overnight, collecting precipitate, and freeze-drying the precipitate in a freeze dryer to obtain the bletilla striata crude polysaccharide.
(3) Preparing the crude polysaccharide of the bletilla striata in the step (2) into a solution with the concentration of 50mg/mL in advance, centrifuging, absorbing supernatant, separating and purifying by using DE-252 anion column chromatography, eluting sodium chloride with the concentration of 0mol/mL, 0.5mol/mL, 1mol/mL and 2mol/mL, eluting the solution with the volume of about 1-2 times of the column volume, collecting polysaccharide eluents under sodium chloride solutions with different concentrations, concentrating, precipitating with 95% ethanol precooled (4 ℃), refrigerating overnight at 4 ℃, collecting precipitate, and freeze-drying.
Example 9
A method for extracting bletilla striata polysaccharide is similar to that of example 1, except that the heating extraction temperature of step (1) is 70 ℃.
Example 10
A method for extracting bletilla striata polysaccharide is similar to that of example 1, except that the heating extraction temperature of step (1) is 70 ℃ and the time is 2 hours.
Example 11
A method for extracting bletilla striata polysaccharide is similar to that of example 1, except that the heating extraction temperature of step (1) is 70 ℃ and the time is 6 hours.
Example 12
A method for extracting bletilla striata polysaccharide is similar to that of example 1, except that the heating extraction temperature of step (1) is 75 ℃ and the time is 2 hours.
Example 13
A method for extracting bletilla striata polysaccharide is similar to that of example 1, except that the heating extraction temperature of step (1) is 75 ℃ and the time is 4 hours.
Example 14
A method for extracting bletilla striata polysaccharide is similar to that of example 1, except that the heating extraction temperature of step (1) is 75 ℃ and the time is 6 hours.
Example 15
A method for extracting bletilla striata polysaccharide is substantially the same as in example 1, except that the heating extraction in step (1) is performed in a vacuum distillation tank.

An extraction method of bletilla striata polysaccharide is similar to that of example 1, except that the pre-cooled 95% ethanol is replaced by the normal-temperature 95% ethanol in the step (2).

A method for extracting bletilla striata polysaccharide is similar to that of example 1, except that the heating extraction temperature of step (1) is 50 ℃ and the time is 4 hours.
Comparative example 3
A method for extracting bletilla striata polysaccharide is similar to that of example 1, except that the heating extraction temperature of step (1) is 50 ℃ and the time is 6 hours.
Test example 1
1. The polysaccharide extraction yield and purity of examples 1 to 8 were verified.
Experimental procedure
Crude polysaccharide extraction (%) = extract dry weight (g)/bletilla tuber powder (g); purity (%) refers to the milligrams of sugar contained in 1g of extract, and the specific implementation steps include: weighing 50mg of glucose which is dried to constant weight at 105 ℃, adding 500 mL of distilled water to prepare 0.1mg/mL of standard solution, diluting the distilled water to 0,0.02,0.04,0.06,0.08 and 0.1mg/mL, then adding 300 mu L of 6% phenol and 1.5mL of sulfuric acid respectively, shaking uniformly, standing for 10 minutes, detecting absorbance at 490nm, and drawing a standard curve by taking glucose content as an abscissa and OD value as an ordinate. The sample was prepared at 1mg/mL, the above procedure was repeated, absorbance was measured at 490nm, and the total sugar content of the sample was calculated by taking the standard curve.

2. Measurement of moisturizing Property of moisturizer.
Measurement of moisture absorption Rate: the bletilla striata polysaccharide (examples 1 to 19 and comparative examples 1 to 3), glycerin and chitosan were weighed 500mg each, and then placed in a closed environment with a Relative Humidity (RH) of 43% and 81% for moisture absorption effect comparison, and after 48 hours, the weight of the three types of moisture absorbents after moisture absorption was weighed and recorded. Moisture absorption rate (X) = (W1-W0) ×100/W0, W0 and W1 represent the weight (g) of the humectant before and after moisture absorption, respectively.
Measurement of moisture retention: bletilla striata polysaccharide (examples 1-19 and comparative examples 1-3), glycerol, and chitosan were prepared into solutions of 1.0%, 5.0%, and 1.0%, respectively, and then subjected to Relative Humidity (RH)
The moisture absorption effect was compared in a closed environment of 43% and 81%, and the weight of the three types of moisture absorbents after moisture absorption was weighed and recorded after 48 hours. Moisture retention (%) =h1×100/H0, H1 and H0 represent the weight (g) of the aqueous solution before and after 48H of standing, respectively.

The data show that the extraction rate of the bletilla striata polysaccharide humectant separated from the bletilla striata tuber water extract is 42.39%, and the moisture retention performance of the bletilla striata polysaccharide humectant is better than that of glycerol and chitosan.

Claims (10)
1. The application of the bletilla striata polysaccharide in preparing a product with moisturizing effect is characterized in that the extraction method of the bletilla striata polysaccharide comprises the following steps:
heating and extracting the mixed solution of the bletilla striata powder and water, and concentrating the supernatant of the extracted product to obtain colloidal bletilla striata polysaccharide;
alcohol precipitation is carried out on the colloidal bletilla striata polysaccharide to obtain the bletilla striata polysaccharide;
wherein the temperature of the heating extraction is 60-80 ℃ and the time is 1-8 h.
2. The use according to claim 1, wherein the temperature of the heat extraction is: 65-80 ℃;
preferably, the temperature of the heat extraction is: 65-78 ℃;
preferably, the time of the heat extraction is: 1-4 h;
preferably, the time of the heat extraction is: 2-4 h;
preferably, the pressure of the heating extraction is normal pressure.
3. The use according to claim 1, wherein the number of heat extractions is at least 1 or more than 1;
preferably, the number of times of the heat extraction is 2 to 4.
4. The use according to claim 3, wherein the ratio of feed liquid extracted by heating is 1: (10-30); wherein the feed liquid ratio is a volume ratio;
preferably, the feed liquid ratio is 1: (10-20).
5. The use according to any one of claims 1 to 4, wherein the step of alcohol precipitation comprises: adding the colloidal bletilla striata polysaccharide into an ethanol solution, and collecting precipitate.
6. The use according to claim 5, wherein the concentration of the ethanol solution is greater than or equal to 95%;
preferably, when alcohol precipitation is performed, the volume ratio of the colloidal bletilla striata polysaccharide to the ethanol solution is 1: (1-4);
preferably, the volume ratio of the colloidal bletilla striata polysaccharide to the ethanol solution is 1:3, a step of;
preferably, the time of the alcohol precipitation is: 4-12 h;
preferably, the ethanol solution is a precooled ethanol solution;
preferably, the pre-cooling refers to pre-cooling to 4-8 ℃;
preferably, the alcohol precipitation further comprises drying the collected precipitate to obtain the bletilla striata polysaccharide.
7. The use according to any one of claims 1 to 4, wherein the extraction method further comprises: separating and purifying the bletilla striata polysaccharide;
preferably, the separation and purification comprises: preparing bletilla striata polysaccharide solution from the bletilla striata polysaccharide obtained after alcohol precipitation, and obtaining purified bletilla striata polysaccharide after anion column chromatography;
preferably, before the anion column chromatography, the separation and purification further comprises centrifuging the bletilla striata polysaccharide solution, and taking supernatant after centrifugation for anion column chromatography;
preferably, the anion column comprises a DE-252 anion column;
preferably, the concentration of the bletilla striata polysaccharide solution is 40-60 mg/mL, and the solvent is water;
preferably, the eluent used for anion column chromatography is: naCl solutions with different gradients;
preferably, the concentration of the NaCl solution of different gradients comprises: 0mol/mL, 0.5mol/mL, 1mol/mL and 2mol/mL;
preferably, the separation and purification further comprises: concentrating the eluted component obtained after anion column chromatography, carrying out alcohol precipitation after concentrating, and collecting precipitate to obtain the final bletilla striata polysaccharide.
8. The use according to any one of claims 1 to 4, wherein the product comprises: skin care and/or maintenance products;
preferably, the skin care product comprises: any one of face cream, hand cream, lotion and hand leave-on gel.
9. A bletilla striata polysaccharide, characterized in that it is extracted by the extraction method according to any one of claims 1 to 8.
10. A moisturizing product, characterized in that it comprises the bletilla striata polysaccharide of claim 9 or obtained by extraction according to any one of the extraction methods of claims 1 to 8.
