Abstract
The invention discloses a preparation method of a bletilla striata extract and application of the bletilla striata extract in preparation of antibacterial products, and relates to the technical field of plant extraction. The preparation method comprises extracting the mixture of bletilla tuber powder and water at 60-80deg.C for 1-4 hr, concentrating the supernatant of the extracted product to obtain bletilla polysaccharide gum; and (3) carrying out alcohol precipitation on the bletilla striata polysaccharide gum to obtain a bletilla striata extract. Aiming at antibacterial performance, the invention optimizes the preparation process of the bletilla striata, improves the antibacterial efficacy of the bletilla striata polysaccharide under the condition of keeping the extraction rate and purity of the bletilla striata polysaccharide, and provides a way for preparing natural antibacterial products.

Background
The bletilla striata is a traditional Chinese medicine with pharmacological action, and in the tuber water extract, the bletilla striata polysaccharide is used as one of main active substances, so that the bletilla striata has remarkable medicinal value. The antibacterial efficacy, extraction efficiency and purity of the prepared bletilla striata extract are required to be improved.
In view of this, the present invention has been made.

The invention is realized in the following way:
in a first aspect, an embodiment of the present invention provides an application of a bletilla striata extract in preparing a product with an antibacterial effect, where the preparation method of the bletilla striata extract includes: extracting the mixed solution of the tuber powder of bletilla striata and water, and concentrating the supernatant of the extracted product to obtain bletilla striata polysaccharide gum; wherein the extraction temperature is 60-80 ℃ and the extraction time is 1-4 h; and (3) carrying out alcohol precipitation on the bletilla striata polysaccharide gum to obtain a bletilla striata extract.
In a second aspect, an embodiment of the present invention provides a method for preparing a bletilla striata extract, where the method is as described in the previous embodiment.
In a third aspect, the embodiment of the invention provides a product with antibacterial effect, which comprises the bletilla striata extract prepared by the preparation method in the previous embodiment.

Optionally, the extracting time is: 2-4 h, and suitable extraction time is favorable for fully dissolving the bletilla polysaccharide gum.
Optionally, the pressure of the extraction is normal pressure. The normal pressure is one atmosphere, which is the pressure of the gas generated by the atmosphere in normal life, and can be approximately regarded as one standard atmosphere, namely 100KPa or 0.1MPa. The process can ensure the dissolution of most of polysaccharide in the bletilla striata powder particles under normal pressure, is safer than pressurizing, has low requirements on equipment, and is easier to operate.

Optionally, the pre-cooling refers to pre-cooling to 4-8 ℃. The ethanol cooled to 4 ℃ in advance can rapidly precipitate the effective active ingredients in the bletilla striata powder, and finally the effective active ingredients are separated from other substances in a precipitation form.

step (2) adding distilled water with the volume of 20 times to the bletilla striata powder, and extracting for 2 hours at the temperature of 60 ℃;
after the heating in the step (3), cooling to room temperature, filtering, adding distilled water with 20 times of volume into filter residues, continuously extracting for 1 time according to the reaction temperature and time in the step (2), and merging and concentrating supernatant;
adding cold 95% ethanol which is 4 times of the volume of the concentrated solution into the concentrated solution, stirring, standing overnight at 4 ℃, centrifuging, collecting precipitate, and recovering ethanol supernatant;
and (5) drying for 48 hours by a freeze dryer, and crushing to obtain the tuber extract of bletilla.

Verification example 1
The crude polysaccharides of examples 1 to 8 were examined for extraction efficiency and purity.
Experimental procedure
Crude polysaccharide extraction (%) = extract dry weight (g)/bletilla tuber powder (g); purity (%) refers to the milligrams of sugar contained in 1g of extract, and the specific implementation steps include: weighing 50mg of glucose which is dried to constant weight at 105 ℃, adding 500 mL of distilled water to prepare 0.1mg/mL of standard solution, diluting the distilled water to 0,0.02,0.04,0.06,0.08 and 0.1mg/mL, then adding 300 mu L of 6% phenol and 1.5mL of sulfuric acid respectively, shaking uniformly, standing for 10 minutes, detecting absorbance at 490nm, and drawing a standard curve by taking glucose content as an abscissa and OD value as an ordinate. The sample was prepared at 1mg/mL, the above procedure was repeated, absorbance was measured at 490nm, and the total sugar content of the sample was calculated by taking the standard curve.

The inhibition effect of the tuber extract of bletilla tuber prepared in example 1 on escherichia coli was verified.
Two common pathogenic bacteria of escherichia coli (ATCC 8739) and staphylococcus aureus (CICC 10384) are selected as test bacteria, and an agar diffusion method is adopted to diffuse the bletilla tuber extract in an agar plate, so that the bacterial growth around the bletilla tuber extract is inhibited to form a transparent circle, namely a bacteriostasis circle, and the bacteriostasis titer of the bletilla tuber extract to be tested is judged according to the size of the bacteriostasis circle. As can be seen from Table 2, the tuber extracts of bletilla striata obtained in examples 1 to 8 have inhibitory effects on 2 test bacteria.

Claims (10)
1. The application of the bletilla striata extract in preparing a product with an antibacterial effect is characterized in that the preparation method of the bletilla striata extract comprises the following steps:
extracting the mixed solution of the tuber powder of bletilla striata and water, and concentrating the supernatant of the extracted product to obtain bletilla striata polysaccharide gum; wherein the extraction temperature is 60-80 ℃ and the extraction time is 1-4 h;
and (3) carrying out alcohol precipitation on the bletilla striata polysaccharide gum to obtain a bletilla striata extract.
2. The use according to claim 1, wherein the extraction temperature is: 65-80 ℃;
preferably, the extraction temperature is: 62-78 ℃;
preferably, the extraction time is: 2-4 h;
preferably, the pressure of the extraction is normal pressure.
3. The use according to claim 1, wherein the ratio of extracted feed to liquid is (0.5-1.5): (10-30), wherein the feed liquid ratio is the volume ratio;
preferably, the feed liquid ratio is 1: (10-25).
4. The use according to claim 1, wherein the alcohol precipitation comprises: adding the bletilla striata polysaccharide gum into ethanol solution, and collecting precipitate.
5. The use according to claim 4, wherein the concentration of the ethanol solution is 95-100%;
preferably, when alcohol precipitation is performed, the volume ratio of the bletilla striata polysaccharide gum to the ethanol solution is 1: (1-4);
preferably, the time of the alcohol precipitation is: 4-12 h;
preferably, the ethanol solution is a precooled ethanol solution;
preferably, the pre-cooling refers to pre-cooling to 4-8 ℃.
6. The use according to claim 5, wherein the preparation method further comprises: drying the precipitate after alcohol precipitation to obtain the bletilla striata extract.
7. The use according to any one of claims 1 to 6, wherein the antibacterial means: has inhibitory effect on at least one of Escherichia coli and Staphylococcus aureus.
8. Use according to any one of claims 1 to 6, wherein the product is selected from: skin care and/or maintenance products;
preferably, the skin care product comprises: any one of face cream, hand cream, lotion and hand leave-on gel.
9. A method for preparing a bletilla striata extract, characterized in that the method is a method for preparing a bletilla striata extract according to any one of claims 1-8;
preferably, the bletilla striata extract is a bletilla striata extract with antibacterial effect.
