Detailed Description
In order to make the objects, technical solutions and advantages of the embodiments of the present invention more clear, the technical solutions of the embodiments of the present invention will be clearly and completely described below. The specific conditions are not noted in the examples and are carried out according to conventional conditions or conditions recommended by the manufacturer. The reagents or apparatus used were conventional products commercially available without the manufacturer's attention.
Firstly, the embodiment of the invention provides an application of a bletilla striata extract in preparing a product with an antibacterial effect, and the preparation method of the bletilla striata extract comprises the following steps: extracting the mixed solution of the tuber powder of bletilla striata and water, and concentrating the supernatant of the extracted product to obtain bletilla striata polysaccharide gum; wherein the extraction temperature is 60-80 ℃ and the extraction time is 1-4 h; and (3) carrying out alcohol precipitation on the bletilla striata polysaccharide gum to obtain a bletilla striata extract.
Compared with the bletilla striata polysaccharide extracted by other processes, the preparation method can prepare the bletilla striata extract with excellent antibacterial effect under the condition of keeping good polysaccharide extraction rate and purity, and has the advantages of high preparation speed, low cost, easiness in implementation, good antibacterial effect and the like.
Optionally, the mesh number of the tuber powder of the bletilla striata is 100-400 mesh. Specifically, the range between any one or any two of 100, 150, 200, 250, 300, 350 and 400 mesh is possible.
Alternatively, the extracted temperature may be in a range between any one or any two of 60, 62, 64, 66, 68, 70, 72, 74, 76, 78, and 80.
Optionally, the extracting temperature is: 65-80 ℃.
Optionally, the extracting temperature is: 62-78 ℃. The temperature range can increase the solubility of active substances in rhizoma bletilla powder, so that the prepared rhizoma bletilla extract has high extraction rate and purity, and has better antibacterial effect.
Alternatively, the time of the extraction may be in a range between any one or any two of 1h, 2h, 3h, and 4 h.
Optionally, the extracting time is: 2-4 h, and suitable extraction time is favorable for fully dissolving the bletilla polysaccharide gum.
Optionally, the pressure of the extraction is normal pressure. The normal pressure is one atmosphere, which is the pressure of the gas generated by the atmosphere in normal life, and can be approximately regarded as one standard atmosphere, namely 100KPa or 0.1MPa. The process can ensure the dissolution of most of polysaccharide in the bletilla striata powder particles under normal pressure, is safer than pressurizing, has low requirements on equipment, and is easier to operate.
Alternatively, the number of extractions is at least 1 or more, such as 2-4. The number of times may specifically be any one or any range between two of 2 times, 3 times and 4 times.
Optionally, the ratio of the extracted feed liquid is (0.5-1.5): (10-30), wherein the feed liquid ratio is the volume ratio. The low feed-liquid ratio can cause the bletilla powder to form lump and influence the dissolution of polysaccharide gum; the high feed-to-liquid ratio can lead to the dissolution of other water-soluble substances, and more non-polysaccharide impurities are introduced.
The extracted feed ratio refers to a volume ratio between the extracted material and the solution (water), and specifically may be in a range between any one or any two of 0.5:10, 0.5:12, 0.5:14, 0.5:16, 0.5:18, 0.5:20, 0.5:22, 0.5:24, 0.5:26, 0.5:28, 0.5:30, 1:10, 1:12, 1:14, 1:16, 1:18, 1:20, 1:22, 1:24, 1:26, 1:28, 1:30, 1.5:10, 1.5:12, 1.5:14, 1.5:16, 1.5:18, 1.5:20, 1.5:22, 1.5:24, 1.5:26, 1.5:28, and 1.5:30.
Optionally, the feed liquid ratio is 1: (10-25).
Optionally, the alcohol precipitation includes: adding the bletilla striata polysaccharide gum into ethanol solution, and collecting precipitate.
Optionally, the concentration of the ethanol solution is 95% -100%. It will be appreciated that the concentration of the ethanol solution is the volume fraction of ethanol in the solution and may specifically be in the range between any one or any two of 95%, 96%, 97%, 98%, 99% and 100%.
Optionally, during alcohol precipitation, the volume ratio of the bletilla striata polysaccharide gum to the ethanol solution is 1: (1-4). The volume ratio may specifically be in a range between any one or any two of 1:1, 1:2, 1:3, and 1:4.
Optionally, the time of the alcohol precipitation is: 6-12h;
optionally, the ethanol solution is a precooled ethanol solution.
Optionally, the pre-cooling refers to pre-cooling to 4-8 ℃. The ethanol cooled to 4 ℃ in advance can rapidly precipitate the effective active ingredients in the bletilla striata powder, and finally the effective active ingredients are separated from other substances in a precipitation form.
Optionally, the preparation method further comprises: drying the precipitate after alcohol precipitation to obtain the bletilla striata extract.
Optionally, the antibacterial means: the bletilla striata extract has an inhibition effect on the growth of escherichia coli and staphylococcus aureus, and is shown that the growth of the escherichia coli and the staphylococcus aureus is inhibited within the range of coating the bletilla striata extract on an agar plate to form a inhibition zone, wherein the inhibition on the escherichia coli is stronger.
Optionally, the product is selected from: skin care and/or maintenance products. The skin care product comprises: any one of face cream, hand cream, lotion and hand leave-on gel.
The embodiment of the invention also provides a preparation method of the bletilla striata extract, which is the preparation method of the bletilla striata extract in any embodiment.
Optionally, the bletilla striata extract is a bletilla striata extract with antibacterial effect.

Two common pathogenic bacteria of escherichia coli (ATCC 8739) and staphylococcus aureus (CICC 10384) are selected as test bacteria, and an agar diffusion method is adopted to diffuse the bletilla tuber extract in an agar plate, so that the bacterial growth around the bletilla tuber extract is inhibited to form a transparent circle, namely a bacteriostasis circle, and the bacteriostasis titer of the bletilla tuber extract to be tested is judged according to the size of the bacteriostasis circle. As can be seen from Table 2, the tuber extracts of bletilla striata obtained in examples 1 to 8 have inhibitory effects on 2 test bacteria.
